Variation of Microbial Community Along Depth in Paddy and Upland Field (논과 밭 토양에서 토층간 미생물 군집의 차이)

  • We examined the vertical distribution of specific microbial groups and the patterns of microbial community structure within the soil profile using phospholipid fatty acid (PLFA). Samples were collected from the soil surface down to 15 cm in depth from paddy and upland fields located in Daegu, Korea. The two fields have been fertilized with only chemical fertilizers N, P, K for 33 years. Principal component analysis of the PLFA signatures indicated that the composition of the soil microbial communities changed significantly with the cultivation practices and soil depth, suggesting that cultivation practices of paddy and upland fields had more significant influence on soil microbial community than the soil depth did. The soil microbial communities changed more drastically with soil depth in upland field than in paddy field, with making thicker soil surface in paddy field in terms of soil microbial community. The ratios of cyclopropyl/monoenoic precursors and total saturated/total monounsaturated fatty acids increased with soil depth, suggesting that the deeper soil horizons are more carbon-limited and anaerobic than surface soil. The community analysis using PLFAs as biomarkers revealed that Gram-positive bacteria and actinomycetes tended to increase in proportional abundance with increasing soil depth, while the abundance of Gram-negative bacteria and fungi were highest at the soil surface and substantially lower in the subsurface.

Effect of Nitrogen component and Ge Composition on Growth in the Cultivation of ICT-based Ginseng Process (ICT 기반의 인삼 공정 육묘 시 질소 형태와 게르마늄(Ge) 유무가 생장에 미치는 영향)

  • Ginseng hydroponic cultivation was cultivated as a nutrient solution and the growth was investigated 60 days later. The length(cm) increased from EC1.0 and EC2.0 concentrations to 5.47cm before, followed by EC0.5, 2.5, 1.5, and 0.0. The weight(g) increased from EC1.0 concentration to 2.39g before, followed by EC1.5, 2.5, 0.5, 0.0 and 2.0. The width(mm) increased from EC1.0 concentration to 1.9mm than before, followed by EC2.5, 0.5, 1.5, 2.0, and 0.0. In the ginseng growth experiment according to nutritional components, the average length of NO3-N mixed experiment increased to 0.33cm, the average weight of 0.04g, and the average width of 0.35mm. In the experiment in which NH4-N was mixed without NO3-N, the growth was reduced to 0.37cm in average length, 0.03g in average weight, and 0.22mm in average width. In an experiment in which germanium(Ge) is mixed with a nutrient component, the average length is increased to 0.33cm, the average weight is increased to 0.04g, and the average width is increased to 0.35mm, but in an experiment excluding germanium(Ge), the growth is reduced to 0.11cm, the average weight is 0.04g, and the average width is 0.03mm.

  • This investigation was carried out to define the cultivation state of Ginseng seedling and to study the growth of important characters by location and correlations between characters, the results obtained are summarized as follows; 1. Most of farmers culitvating ginseng seedling had not any special consideration for the improvement of soil, and they applied green manure only as basic fertilizer but they did not apply chemical fertilizer. 2. Any efforts on the improvement of ginseng seed were not found. Seed was harvested from 3 years old ginseng plant. Seedling density was 1.8 cm by 2 cm. 3. Growth of all characters showed significant differences among farms. Coefficients of variation among individual seedling plants was largest at root diameter and smallest at root length. 4. Gromth of all characters without root diameter by planted location under the shadow was poor at front row, however root diameter was shortest at back row. 5. Significant high correlatons were rocognized between root diameter and stem diameter, between leaf blade and stem diameter and between leaf blade and leaf length, but stem length were not correlated with any characters. It is noticable that negative correlations were found between stem length and root diameter, between stem length and root length and between stem length and stem diameter.

  • The soil management practices before transplanting the ginseng plant were studied with two organic matter sources such as a traditional organic matter (wild grass) and commercial organic fertilizer (byproducts of amino acid fermantation) during the late spring to late autumn. During the soil management practices, the soil received 40kg N/10a from five different combination treatments with above two organic matter sources, a wild grass and a commercial organic fertilizer. After the application of the treatments, the soil were ploughed regularly at the interval of 20 days and the changes of physicochemical properties during the soil management practices were investigated. The next year after soil management practices, ginseng plants were transplanted to each treatment, growth and the content of some organic components of ginseng plant were measured for comparision of the different treatments. 1. The decrease in bulk density observed during the first 40 days of management was considered to be the effect of the improved physical conditions caused by ploughing, The decrease in bulk density observed after 40 days of management was considered to be the effect of organic matter. Similar results were observed in particle density, however porosity increased with time. 2. Soil pH tended to decrease during the first 40 days of management, after which period the pH increased and was stabilized. However, CEC increased with organic matter treatment and the exchangeable $NH^+_4-N$ and $NO^-_3-N$ increased in 20 and 40 days after the management practices, respectively, and after that period it became steady. 3. The decomposition rate of treated organic matter was measured by the incubation test in laboratory conditions. The rate of decomposition was rapid during the first 20 days of management, after which period it showed slight changes. 4. The weight of ginseng root significantly increased in the treatment of 10kg N/10a organic fertilizer and 30kg N/10a wild grass. 5. The saponin content of ginseng root was highest in the 40kg N/10a wild grass treatment. The addition of organic fertilizer at the rate of more than 20kg N/10a caused the decrease in the saponin content.

Changes of Soil Chemical Properties and Root Injury Ratio by Progress years of Post-Harvest in Continuous Cropping Soils of Ginseng (인삼 연작지 수확 경과년수에 따른 토양화학성 및 뿌리썩음병 발생율의 변화)

  • To study the cause of the injury by continuous cropping and the countermeasure for reduction on it's injury in ginseng cultivation by the period passed since post-harvest, and previous soil management far planting, the injury by continuous cropping was investigated from 2004 to 2005 at the continuous cropping field of National Institute of crop Science in Jungpyong county, Korea. There was negative correlation between the ratio of survived root and the Period Passed since post-harvest. The ratio infected by Cylindrocapon destructans and Fusarium spp. in root was about 81${\sim}$95% at continuous cropping field that passed by 3${\sim}$4 years since post-harvest. Though the previous soil management for planting was somewhat effective to increase the ratio of survival root, but which was not effective to protect the infection of Cylindrocapon destructans and Fusarium spp. in root. There was no significant difference in reducing rotted root between management and nonmanagement. When previous soil management for planting was programed, it should be considered that Cylindrocapon destructans was detected at continuous cropping field that passed by above 10 years after harvesting.

Changes in Major Taste Components of Soybean Sprout Germinated with Extract of Korean Panax ginseng (인삼 추출물로 발아시킨 콩나물의 식품성분 변화)

  • Changes in weight, length, amino acids, organic acids and free sugars of soybean sprouts germinated with extract of Panax ginseng(PGE, 100∼400 ppm) were investigated. PGE increased the weight and length of soybean sprouts. Content of total amino acid in soybean sprout germinated at 100 ppm of PGE after cultivation for 3 days was the most abundant and then decreased by increasing the concentration of PGE. Content of aspartic acid was increased with culture time, but that of glutamic acid was shown to be an opposite trend. Content of total free sugar was increased by increasing culture time and not affected by concentration of PGE. Content of sucrose in control group during growth of soybean sprout was decreased, but sucrose contents in PGE groups were increased to 3 days and decreased thereafter. However, the other sugars were continuously increased for 4 days. Content of total organic acids was the most abundant in soybean sprouts germinated with 200 ppm of PGE and cultured for 3 days. Phytic acid was a major organic acid, showing the range of 45 to 60% for total organic acids. In conclusion, PGE as sprouting water of soybean was effective to increase of growth, contents of amino acids and organic acids in soybean sprouts, indicating that PGE accelerated the quality of soybean sprouts.

Isolation and Characterization of 𝛽-Glucosidase-Producing Yeast, Rhodotorula sp. GYP-1 (𝛽-Glucosidase 생성 효모 Rhodotorula sp. GYP-1의 분리 및 특성)

  • Nine microbial strains were isolated from the byproduct of ginseng processing and field of ginseng cultivation. Two strains among them were confirmed. Phylogenetic analysis of these 𝛽-Glucosidase strains confirmed that strain GYP-1 belongs to the Rhodotorula and strain GYP-3-3 belong to genus Brachybacterium. Rhodotorula sp. GYP-1 was finally selected due to its high biomass production. The 𝛽-Glucosidase activity of Rhodotorula sp. GPY-1 was assessed at 30 ℃, and Higher than 70% of the enzyme activity was maintained at the temperature range of 20-40℃. Although the optimum pH for the highest enzyme activity was pH 5.0, the enzyme was stable throughout the pH range of 5.0-8.0. In addition, Rhodotorula sp. demonstrated antifungal activity against the ginseng root rot disease caused by Botrytis.

Chemical Components, DPPH Radical Scavenging Activity and Inhibitory Effects on Nitric Oxide Production in Allium hookeri Cultivated under Open Field and Greenhouse Conditions (노지 및 시설재배 삼채 뿌리 및 잎의 이화학 성분, DPPH 라디칼 소거능 및 Nitric Oxide 생성 억제효과)

  • To enhance the utilization of Allium hookeri (AH) as a food, characteristics of AH roots and leaves cultivated under open field and greenhouse conditions were investigated. The moisture content of the roots and leaves were 81.05 to 84.18% and 88.85 to 90.12%, respectively. The moisture content of AH cultivated in the open field was 2 to 3% lower than the moisture content of AH cultivated in the greenhouse for both roots and leaves. The content of nitrogen-free extract, carbohydrates, was 13.49 to 16.20% in the roots and 7.08 to 7.79% in the leaves. The main mineral generated from both open field and greenhouse cultivation was potassium, at 503.98 to 512.08 mg% in leaves. The free sugar content of roots cultivated in the open field was four times higher than the content in the leaves, and roots cultivated in the greenhouse contained three times lower free sugar than the leaves. In particular, the fructose content of roots cultivated in the open field was about 12 times higher than roots cultivated in the greenhouse. The crude saponin and total polyphenol content was higher in leaves than roots, and was higher in the open field than the greenhouse. The $IC_{50}$ for DPPH radical scavenging activity was highest, 2.74 mg/mL, in 70% MeOH extracts of AH leaves cultivated in the greenhouse. Water and 70% MeOH extracts of AH leaves cultivated in the greenhouse showed no cytotoxicity to RAW 264.7 cells. Water extracts of AH leaves cultivated in the open field markedly inhibited the production of the inflammatory mediator nitric oxide. These results suggest that AH may be used as the material of health functional food.

Radioprotective Effect of Red Ginseng in Irradiated Mice with ${\gamma}$-ray (생쥐에서 홍삼의 감마선조사에 의한 방어효과)

  • Recently, the incidents of direct or indirect radiation exposure due to increase of use of radiation or radioisotope are on the increase in medical and industrial circles. If cells are irradiated, free radicals are created through biological process, and cells are directly or indirectly damaged. This research intends to explore into the effect of saponin at the level of cell (in vitro) and entity (in vivo), using red ginseng extract "saponin", as radioprotective agent. In the experiment implemented at the level of cell (in vitro), degree of cell activity was measures by adding mouse mesenchymal stem cells "C3H/10T1/2 cells" into red ginseng extract "saponin(0, 0.05, 0.2, and 0.4 g/L)", and then the optimal concentration of saponin influencing cells was calculated, in 24, 48, 72, and 96 hours after gamma irradiation at the optimal concentration of saponin, each cell survival rate was observed through XTT assay. The best time period of cultivation for the optimal activity of C3H/10T1/2 cells was as 48 hours, and the degree of optimal activity was shown at 0.05 g/L. In 48 hours after irradiation of 5 Gy to C3H/10T1/2 cells at 0.05 g/L, the degree of activity of cells increased by 10%. In the experiment implemented at the level of entity (in vivo), red ginseng extract "saponin" at a dose of 100 mg/kg/day was injected into the abdominal cavity of six-week immature mouse for two weeks. Right after the last abdominal injection, total body irradiation of gamma rays was carried out at a dose of 5 Gy and 10 Gy. And after irradiation, the blood sample was taken, and then the number of red corpuscles was counted. In result, the decrement of experimental group treated with red ginseng extract "saponin" was 2.3 times larger than that of control group. In view of the results so far achieved, it was revealed that red ginseng extract "saponin" has a radiation exposure protection effect in the experiment implemented at the level of cell (in vitro). In case of animal experiment, the decrement of number of red corpuscles decreased. Finally, it is necessary to carry out more various researches continuously.

Effect of Different Media on Growth and Yield in Hydroponic culture of Angelica keiskei Koidzumi (신선초(神仙草) 양액재배시(養夜載培時) 고형배지종류(固形培地種類)에 따른 생육(生育) 및 수량(收量))

  • This experiment was conducted to find appropriate media in hydroponic Culture of Angelica keiskei. The mediaused were carbonized rice hull, perlite, vermiculite, mixture(carbonized rice hull (1) : perlite (1) : peatmoss (1)), mixture+peatmoss20%, mixture+peatmoss40%, mixtrue+peatmoss60%, perlite 40%+peatmoss40%+vermiculite20% and open field cultivation. The highest yield of Angeli ca keiskei was shown 4,428ka/10a at vermiculite. The yield of vermiculite was increased about 133 percent compared to that of open field cultivation. The highest germanium contents was shown 0.52ppm at mixture+peatmoss 40%. The contents of germanium increased about 108 percent at mixture+peatmoss40% compared to that of open field cultivation.
