Effects of Media and Growth Regulators on the Growth and Saponin Production of Ginseng Root (인삼 Root의 생장 및 사포닌 생성에 미치는 배지와 생장조절물질의 영향)

  • Effects of media and growth regulators on the growth and saponin accumulation of ginseng root were investigated to develop the ginseng root culture system. When Panax ginseng C. A. Meyer roots were induced and cultured in various liquid media, the maximum root growth and saponin production were obtained in SH medium and an initial doubleing time of ginseng root was approximately 10 days. The patterns and contents of ginsenosides of cultured ginseng root in various media were different from each other. SH and White media resulted in higher total ginsenosides contents than the other media. Among different combinations and concentrations of growth regulators, SH medium containing 4.0mg/ L NAA gave best growth of ginseng roots, while saponin content was highest in SH medium containing 0.5mg/L BAP. These results suggested that the rapid production of ginseng saponin is possible by root culture of Panax ginseng.

The Comparison of Ginseng Saponin Composition and Contents in Dried Ginseng Radices (건삼류 생약의 인삼사포닌 성분 비교)

  • This study was conducted to provide basic information on ginseng saponin of dried ginseng radices. In order to achieve the proposed objective ginsenoside compositions of dried ginseng radices extract with 70% ethyl alcohol were examined by HPLC. The total saponin content, the sum of all ginsenosides, showed that Wild simulated ginseng (WSG), White fine ginseng (WFG), Skin White ginseng (SWG), and White ginseng (WG) stood at 2.510%, 1.643%, 0.587, and 0.429%, respectively. WSG in PPD/PPT ratio was highest at 3.190, WFG (1.934), WG (1.600), SWG (1.386) in order. In the content of ginsenoside Rb1, one of the marker compounds of ginseng, WSG (1.095%) showed the highest content, and WFG (0.527%), SWG (0.246%), WG (0.133%) in this order. The content of ginsenoside Rb1 of WSG (1.095%) was 4.5 times higher than SWG (0.246%). WSG (0.230%) showed the highest content in ginsenoside Rg1, a marker compounds of ginseng, followed by WFG (0.180%), SWG (0.141%) and WG (0.086%). The content of ginsenoside Rg1 of WSG (0.230%) was 1.6 times higher than SWG (0.141%).

Identification of Saponin and Sapogenin in Root, Leaf and Stem of Ginseng by Thin Layer Chromatography (얇은막 크로마토그래피에 의한 인삼(人蔘)의 근(根) 엽(葉) 및 경(莖)의 saponin 및 sapogenin화합물(化合物) 동정(同定))

  • Saponins of ginseng root, leaf and stem were identified by TLC. Eleven unknown spots were detected in ginseng leaf and ten unknown spots in ginseng stem on TLC besides seven ginsenosides such as $ginsenoside-Rg_1,\;-Rf,\;-Re,\;-Rd,\;-Rc,\;-Rb_2,\;and\;-Rb_1$ which are contained in ginseng root. $Ginsenoside-Rg_3\;and\;-Rg_2$ were identified on TLC from mild hydrolysates with 50% acetic acid of total saponins from ginseng root, leaf and stem. Meanwhile, panaxadiol, panaxatriol and oleanolic acid were identified from hydrolysates with 7% ethanolic sulfuric acid of total saponin of ginseng root, while panaxadiol and panaxatriol from those of total saponins of ginseng leaf and stem.

  • To determine harvesting time of ginseng, the changes of saponin contents and PT/PD ratios of triol saponin vs. diol saponin in relation to root age of Panax ginseng. Total saponin contents were increased in relation to root age, 5-years was higher than 6-years for saponin contents. Therefore, it considered that can be obtained the desired results to harvest at 5-years. Also, PT/PD ratios were increased in relation to root age, 6-years was highest. Considering PT/PD ratios in relation with pharmaceutical action, it must be harvested at 6-years.

  • The present study was undertaken to elucidate the behavioral characteristics of nootropic candidates, entrophenoxine, N-methyl-D-glucamine, piracetam and red ginseng saponin components on stereotyped sniffing behavior induced by apomorphine in rats. Apomorphine, a direct dopaminergic receptor agonist, induced stereotyped behaviors including sniffing licking growing and biting in a dosedependent manner, and that behaviors were completely inhibited when measured at 1 week after 6-ydroxydopamine(6-HDA) treatment. Centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin(TS), panaxatriol (PT), and Rg1 enhanced but panaxadiol (PD) inhibited, whereas piracetam and Rb1 were not effective of the sterotyped sniffing behavior induced by apomorphine(1mg/kg). The enhanced stereotyped behavior by centrophenoxine, N-methyl-D-glucamine, red ginseng total saponin, panaxatriol(PT), and Rg1 seems to have a similarity to entrophenoxine, N-methyl-D-glucamate in modulating of dopaminergic neuroal activity and also my be useful for the nootropic candidates.

Effects of Ginseng Saponin and Its Related Materials on Aflatoxin Production by Aspergillus parasiticus NRRL2999 in semi-Synthetic Media (반합성 배지에서 Aspergillus parasiticus의 Aflatoxin생성에 미치는 인삼 Saponin과 그 관련물질의 영향)

  • The effects of ginseng saponin and its related materials on aflatoxin production by Aspergillus parasiticus NRRL2999 in yeast extract sucrose (YES) medium were studied. Maximal production of aflatoxins by the mold in the medium occurred after 9 days at $28^{\circ}C$. When various concentrations of ginseng saponin were added to the medium aflatoxin productions were significantly reduced (p<0.05) compared to the control after 9 days at $28^{\circ}C$. 0.05% of saponin in the medium greatly decreased aflatoxin synthesis, and no aflatoxins were synthesized by the mold in the medium contained 5.0% of saponin. When various concentrations of saponin diol and triol were added to the medium both ingibitory and sitimulatory effects on alfatoxin production were resulted. Saponin fraction numbers of 1, 2, 4, 5 and 6 decreased aflatoxin production, however the numbers of 3 and 7 stimulated the toxin production. 0.05% of adenosine, guanosine, caffeine and xanthosine in the media inhibited aflatoxin production (p<0.05), but adenine and cytosine increased the production. When 5.0% of saponin was added to the medium aflatoxins were not synthesized at all, but total lipid synthesis and mold growth were highly stimulated. Both the synthesis of total lipid and mold growth were reduced in case of aflatoxin synthesis stimulated.

Anxiolytic Effects of Total Saponin Fraction from Ginseng Radix Rubra on the Elevated Plus-Maze Model in Mice

  • This study was performed to investigate the anxiolytic effects of total sponin fraction from Ginseng Radix Rubra (KRG) in mice using the elevated plus-maze model. The water extract of KRG and ginseng total saponins (GTS) purified from the water extract of KRG were administered orally to mice. One hour after administration of KRG water extract and GTS, mice were tested on the elevated plus-maze. The water extract of KRG 100 mg/kg, and GTS 25 and 50 mg/kg did not increase open arm entries and time spent on open arm. However, GTS 100 mg/kg increased the number of open arm entries and time spent on open arm. On the other hand, as the plus-maze test was affected by changes in locomotor activity, an additional test was carried out with the specific aim of monitoring locomotor activity. The water extract of KRG 100 mg/kg, and GTS 25 and 50 mg/kg did not affect the locomotor activity. However, GTS 100 mg/kg significantly decreased locomotor activity. From this study, we suggest that GTS may play an imponant role on the anxiolytic effects in the plus-maze model.

Distribution of Saponin in Various Sections of panax ginseng root and Change of Its Contents According to Root Age (고려인삼근의 부위별 년근별 사포닌 함량 및 분포)

  • This study was carried out to get basic information that can be used in quality control for processing ginseng products and also in separation of pure ginsenosides for experimental purpose. The composition of various parts of 6 year-old ginseng was 4.1% of rhizome (node), 47.7% of main root, 34.1% of lateral root and 14.1% of fine Toot on dried weight basis. The weight ratios of epidermis-cortex and xylem were about 1 : 1 in main root and about 2 : 1 in lateral root. The distribution of total saponin content shows 29.2% in main root, 34.6% in lateral root, 29.1% in fine root and 7% in rhizome, but the order of the content per unit weight was fine root > rhizome > lateral root > main root. Total saponin content according to age of root was increased gradually within 3% for 6 years, as compared with two year old root. In view of the increase of root weight owing to the net amount of saponin in root increased continuously. The increase rates of total saponins per year were 3.1,12.3,19.8,43.8 and 21.1% in 2, 3, 4, 5 and 6 years-old ginseng root, respectively.

  • In conclusion, biochemical analysis revealed that GTS inhibited the methamphetamine-induced DA increase and the methamphetamine-induced DOPAC decrease in the mouse striatum. These results indicate that GTS could modulate the methamphetamine-induced striatal dopaminergic neuronal systems.

  • In order to study the effect of honey concentration on the quality of honeyed ginseng in the process of manufacturing honeyed ginseng, honeyed ginseng was manufactured under the various condition of honey concentration, 40 brix, 50 brix, 60 brix and 70 brix and the approximate composition, the yield and organoleptic test of them were investigated. The results obtained were as follows. 1. The higher honey concentration was used, the yield of honeyed ginseng was increased. 2. The higher honey concentration was used, the amount of total sugar was increased while the amount of crude saponin, crude protein. crude fat, crude fiber and crude ash was decreased. 3. Free sugars of honeyed ginseng consist of rhamnose, xylose, fructose, glucose, sucrose and maltose, 4. The amount of saponin lost in the process of manufacturing honeyed ginseng was 2.5%-5.0%, therefore honey concentration was not so effective on the loss of saponin. 5. According to organoleptic test, H-3 and H-2 treating honey concentration at 60 brix and 50 brix was the best and better of all.
