In Vivo Quantitative Analysis of PKA Subunit Interaction and cAMP Level by Dual Color Fluorescence Cross Correlation Spectroscopy

  • We employed dual color Fluorescence Cross Correlation Spectroscopy (FCCS) to measure the interaction between PKA regulatory (RII) and catalytic subunits (CAT) in living cells. Elevation of intracellular cAMP with forskolin decreased the cross-correlation amplitude between RFP-fused RII (RII -mRFP) and GFP-fused CAT (CAT-EGFP) by 50%, indicating that cAMP elevation leads to dissociation of RII-CAT complexes. Moreover, diffusion coefficient analysis showed that the diffusion rate of CAT-EGFP was significantly increased, suggesting that the decreased RII-CAT association caused by cAMP generated free CAT subunits. Our study demonstrates that in vivo FCCS measurements and their quantitative analysis permit one not only to directly quantify protein-protein interactions but also to estimate changes in the intracellular cAMP concentration.

On Performance Analysis of Position Based Routing Algorithms in Wireless Networks

  • This paper presents an overview of position-based routing algorithms. We analyze performances of routing algorithms such as Hybrid Opportunistic Forwarding (HOF), Opportunistic multi-hop routing (ExOR), Location based Geocasting and Forwarding (LGF), and Greedy Forwarding in nearest with forward Progress (GFP) routing algorithms to find the best one in terms of packet error rate and throughput efficiency over effects of fading and noise variance in wireless networks. The analyses in closed form expressions are confirmed by the simulation results, which fully agree to analysis results. Additionally, the simulation results indicate significant differences among algorithms when varying the average SNR or the number of relays.

  • 본 연구에서는 고분해능 및 고감도화된 시스템 개발을 위하여 AOTF를 이용하여 하이퍼스펙트럼 영상기법을 활용한 전임상 영상장비에 대한 연구를 수행하였다. 제작된 고감도 하이퍼스펙트럼 분자영상 시스템의 생물학적 적용을 위하여, AOTF의 파장 또는 진동수를 변화시키면서 GFP가 발현된 HEK 293 세포의 이미지를 촬영하였다. 또한, 제작된 실험 대상물 이미지화 시스템을 이용해서 실험용 쥐의 이미지를 촬영하였다. 실험용 쥐를 크세논 아크램프 사용 전 후 이미지를 촬영한 결과 크세논 아크 램프 사용 후에는 청색의 선명한 영상을 얻을 수 있었다.

  • It is noteworthy that IMT -2000 communication network based on All-HP/AIN(Advanced Intelligent Network) should accomodate current and future wire/wireless AIN service easily through integration and gearing AIN construction elements. In this paper. Intelligent Farmer model(I-Farmer Model) and methodology are suggested in order to solve the several problems including standardization on implementation of Q3 interface in Telecommunication Management Network(TMN) agents which is caused by heterogeneous platform environment and future maintenance. Also this paper proposes ITI algorithm transforming the system which is designed by I-Farmer model to Interface Specification Model(ISM) applying the I-Farmer model. In addition to ITI algorithm. we suggest NTS(Node to SIB) algorithm converting entity node and ILB/OLB component in agent system designed by the I-Farmer model to SIB of AIN GFP(Global Functional Plane) and to ASIB for application program.

Quantitative Evaluation of the Mode of Microtubule Transport in Xenopus Neurons

  • Tubulin is synthesized in the cell body and must be delivered to the axon to support axonal growth. However, the exact form in which these proteins, in particular tubulin, move within the axon remains contentious. According to the "polymer transport model", tubulin is transported in the form of microtubules. In an alternative hypothesis, the "short oligomer transport model", tubulin is added to existing, stationary microtubules along the axon. In this study, we measured the translocation of microtubule plus ends in soma segments, the middle of axonal shafts and the growth cone areas, by expressing GFP-EB3 in cultured Xenopus embryonic spinal neurons. We found that none of the microtubules in the three compartments were transported rapidly as would be expected from the polymer transport model. These results suggest that microtubules are stationary in most segments of the axon, thus supporting the model according to which tubulin is transported in nonpolymeric form in rapidly growing Xenopus neurons.

Anti-oxidative Effect of Piperine from Piper nigrum L. in Caenorhabditis elegans

  • Piper nigrum L. (Piperaceae), which is a well-known food seasoning, has been used as a traditional medicine for the treatment of vomiting, abdominal pain, diarrhea and anorexia in Korea, China and Japan. Methanol extract from the fruit of P. nigrum was successively partitioned as n-hexane, methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions the ethyl acetate soluble fraction showed the most potent DPPH radical scavenging activity, and piperine was isolated from the ethyl acetate fraction. To know the antioxidant activity of piperine, we tested the activities of superoxide dismutase (SOD) and catalase together with oxidative stress tolerance and intracellular ROS level in Caenorhabditis elegans. To investigate whether piperine-mediated increased stress tolerance was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain including CF1553. Consequently, piperine enhanced SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, piperine-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.

  • Transgenic Eleutherococcus senticosus plants were prepared by introducing the genes for squalene synthase (SQS), hygromycin phosphotransferase (HPT) and green fluorescent Protein (GFP) through Agrobacterium-mediated transformation. The enzyme, SQS, represents a putative branch point in the isoprenoid pathway capable of diverting carbon flow specifically to the biosynthesis of phytosterol and oleanolic acid. The full SQS gene was isolated from P. ginseng roots. Early globular embryo clusters developed from embryogenic callus were used as the explant source. (omitted)
