Cucumber mosaic virus (CMV) is one of the most important viral diseases in pepper (Capsicum annuum L.), and several genes for resistance were reported in Capsicum spp. In Korea, a single dominant gene that is resistant to $CMV_{Fny}$ and $CMV_{P0}$ has been used for breeding. Recently, a new strain ($CMV_{P1}$) was reported that could infect cultivars resistant to both $CMV_{Fny}$ and $CMV_{P0}$. Therefore, breeding of more robust CMV-resistant cultivars is required. In this study, we surveyed the inheritance of $CMV_{P1}$ resistance and analyzed the location of the resistance loci. After $CMV_{P1}$ inoculation of various germplasms and breeding lines, one accession (ICPN18-8) showed no visual symptoms at 15 dpi (days post inoculation) but was susceptible after 45 dpi, and one resistant line (I7339) showed resistance until at 45 dpi. The latter line was used for tests of resistance inheritance. A total of 189 $F_2$ plants were examined, with 42 individuals showing resistance at 15 dpi and a phenotype segregation ratio close to 1:3 (resistant:susceptible plants). In a lateral ELISA test at 45 dpi, 11 plants showed resistance, and the segregation ratio was changed to 1:15. These results indicate that resistance in C. annuum 'I7339' is controlled by two different recessive genes; we named these resistance genes 'cmr3E' and 'cmr3L,' respectively. To locate these two resistant loci in the pepper linkage map, various RAPD, SSR, and STS markers were screened; only nine markers were grouped into one linkage group (LG). Only one RAPD primer (OPAT16) was distantly linked with cmr3E (22.3 cM) and cmr3L (20.7 cM). To develop more accurate markers for marker-assisted breeding, enriching for molecular markers spanning two loci will be required.

The responses to chilling temperature of 12 Korean cucumber varieties was compared to those of two U.S.A. (previously determined cold tolerant NC76 and 'Chipper'), and Chinese and Japanese germplasms. Seedlings of each entry were exposed to $4^{\circ}C$ (Experiment 1) and $1^{\circ}C$ (Experiments 2 and 3) at the first-true leaf stage for eight and nine hours, respectively, under 80% relative humidity (RH) and $149{\mu}moles{\cdot}m^{-2}{\cdot}s^{-1}$ photosynthetic photon flux (PPF). The chilling response [damage rating (DR)] of each accession was based on visual ratings (1 to 5) after treatment, where 1 = no damage, 2 = slight, 3 = moderate, 4 = advanced, and 5 = severe damage. Predictably the cumulative average DR of chilling tolerant line NC76 and 'Chipper' after chilling w as 1 and 1.1, respectively. Korean 'Nacdongchungjang' was most sensitive to chilling temperatures [DR = 2.3] when compared to the other entries examined. The sensitivity to chilling of 'Nacdongchungjang' was followed by Chinese 'Dongguan' [DR = 1.7]. In contrast, 'Saeronchungjang' (DR = 1) and 'Janghyungnachap' (DR = 1) were the most chilling tolerant of the Korean accessions examined and equivalent to the response of line NC76 and 'Chipper'. Nevertheless, chloroplast type genotyping of these accessions with known chilling-linked sdCAPS genomic markers revealed genotypic differences between chilling tolerant lines (NC76 and 'Chipper') and all Korean lines examined.
The objectives of this study were to evaluate total phenolic content (TPC) and individual phenolic compounds in leaves of perilla genetic resources, assess whether they could be used as distinguishing factor among germplasms, and evaluate their relationship with some quantitative and qualitative morphological characters. TPC and individual phenolic compounds were determined using Folin-Ciocalteu method and UPLC-PDA system, respectively. Wide variations in TPC (7.99 to 133.70 mgGAE/g DE), rosmarinic acid (ND to 21.05 mg/g DE), caffeic acid (ND to 1.17 mg/g DE), apigenin-7-O-diglucuronide (ND to 2.21 mg luteolin equivalent (mgLUE)/g DE), scutellarein-7-O-glucuronide (ND to 5.25 mg LUE/g DE), and apigenin-7-O-glucuronide (ND to 2.81 mg LUE/g DE) were observed. Intensities of green pigment at abaxial and adaxial leaf surfaces were positively correlated with phenolic compounds whereas leaf length and width had negative correlation. Purple pigmented accessions were shorter in leaf length and width but exhibited higher amount of phenolic compounds compared to green pigmented accessions in most cases. Leaf shape was not related with content of phenolic compounds, color of leaves, and length/width of leaves. TPC and individual phenolic compounds along with morphological characters could be useful distinguishing factors for perilla genetic resources.
This study was carried out to get the information of Plum tree (192 germplasm) collected in Korea, and to evaluate the organic acid contents and fermentation solution. The organic acid content of fruit was 50.9 ± 6.0 mg/g, and which was composed of 55.5% of citric acid, 43.4% of malic acid and 1.1% of oxalic acid, and showed large difference among germplasms. Oxalic acid and malic acid made no differences in organic acid content according to flesh color, whereas citric acid and total organic acid contents were highest in orange color and lowest in whitish green. Malic acid, citric acid and total organic acid contents did not show differences among fruit weight groups, but oxalic acid content was highest at fruit weight of 5.1 ~ 10.0 g and lowest at more than 20.1 g. The sugar content of fermentation solution of fruit was 55.7 ± 1.6 °Brix and the harvest rate was 116.7 ± 8.7%. The correlation coefficients among fruit weight, the sugar content (°Brix) and harvest rate of fermentation solution were very low, and there were correlations of r=-0.551⁎⁎ between fruit weight and oxalic acid, r=-0.767⁎⁎ between malic acid and citric acid, and r=0.834⁎⁎ between citric acid and total organic acid content.
The preservation of pear germplasm, like that of other clonal germplasms, is difficult because it requires conservation of whole plants or their tissues. Among the currently available methods for long-term conservation of clonal germplasm, cryopreservation of shoot tips is the most reliable and cost- and space-effective option. Alginate-coated axillary shoot tips from in vitro-grown pear were conserved successfully in liquid nitrogen (LN) following dehydration. Shoot recovery from cryopreserved shoot tips was improved greatly after 8 weeks of cold acclimation, but recovery decreased slightly after then. The highest regeneration rate was observed when in vitro shoot tips were preincubated in MS (Murashige and Skoog) medium with 0.3 M sucrose for 48 h, and when alginate-coated shoot tips were precultured in MS medium with increasing sucrose concentrations (0.5 M and 0.7 M) for 8 and 16 h, respectively. When the encapsulated beads were dehydrated for up to 7 h [25% water content (fresh weight basis)] under laminar flow, the highest regeneration rate was observed in "BaeYun No. 3" (55.7%) and "Whanggeum" (43.3%) after warming from LN. This technique is useful as a practical procedure to cryopreserve plant material that is sensitive to freezing of the surrounding cryoprotectant medium. Therefore, this technique appears to be promising for the cryopreservation of shoot tips from in vitro-grown plantlets of pear germplasm.

The production of spring wheat, the major crop in Mongolia, is accounting for 98% of the cultivated area. Collection, conservation and utilization of wheat germplasm resources play an important role in wheat breeding and production in Mongolia. Understanding genetic variability in the existing genebank accessions is important for collection and conservation of wheat germplasms. To determine the genetic diversity and population structure among a representative collection of Mongolian local wheat cultivars and lines, 200 wheat accessions were analyzed with 15 SSR markers distributed throughout the wheat genome. A total of 85 alleles were detected, with 3 to 5 alleles per locus and a mean genetic diversity value of 5.66. The average genetic diversity index was 0.68, with values ranging from 0.37 to 0.80. The 200 Mongolian wheat accessions were divided into two subgroups based on STRUCTURE, un-rooted NJ cluster and principal coordinate analyses. The results from this study will provide important information for future wheat germplasm conservation and improvement programs with Mongolian genebank.
It is important to understand pedigree of rice cultivars commonly used for breeding. In this paper, pedigree tables for tracking the pedigree of 17 representative rice cultivars recommended by Rural development Adminstration (RDA) were completed and analyzed using two kinds of web database system; 'IRIS' and 'RRDB'. Seven cultivars, namely, 'Sangmibyeo', 'Ilpumbyeo', 'Saegewhabyeo', 'Surabyeo', 'Shindongjinbyeo', 'Ilmibyeo' and 'Jungwhabyeo' had 'Koshihikari' on the pedigree of their ancestor. Besides 'Koshihikari', the most feguently used ancestral germplasms among the high quality rice cultivars were 'Fujisaka 5', 'Kameno o' and 'Asahi', 'Fujisaka 5' was used as ancestral parent in 12 out of 17 cultivars. Interestingly, 'Kameno o' was used in pedigree of 16 out of 17 high quality varieties and 'Asahi' was used in the ancestral pedigree of all 17 varieties. 'Hwayeongbyeo' was used as one of parent in the breeding of 'Dongjin 1', 'Hwabongbyeo', 'Saegewhabyeo' and 'Junambyeo'. 'Ilpumbyeo' was used in the breeding pathway of 'Junambyeo' and 'Saegewhabyeo', 'Mangeumbyeo' itself was not enlisted as one of high quality rice cultivars, but was used as a breeding parent of three high quality varieties, namely, 'Saegewhabyeo', 'Hwabongbyeo' and 'Nampyeongbyeo'. Incorporated with evaluation data, pedigree will provide a valuable chance to genealogical tracking of agronomic traits such as disease resistance, grain quality and etc.

More than 2,500 accessions of squash germplasm are preserved in the National Agrobidiversity Center of RDA, Korea. We investigated the ecological and morphological characteristics on 100 accessions of the squash germplasms with a low viability. Two typical types of oriental squash (Cucurbita moschata) and western squash (Cucurbita maxima) are possessed 89 and 11 accessions, respectively. The origin of 100 accessions was divided into three groups, group I (Korea landraces with 81 accessions), group II (Nepal with 15 accessions) and group III (Russia, Uzbekistan, Laos, and Mexico with each one accession). Seventy eight accessions germinated at $27^{\circ}C$ in growth chamber. Days to first flowering of the female and male flower ranged from 32 days to 67 days and 24 days to 55 days after planting, respectively. Vine length showed the largest variation ranging from 357.8 cm to 1,465 cm, also the average length of ten stem node range has a variable ranges from 92 cm to 217.5 cm. All regenerated squash accessions have an only collided tendril and have three to seven lateral shoots. The flattened globe-shaped squash is the most predominant and exhibited ordinarily round shaped, ball shaped, long oblong-shaped, elongated shape, and pear shape. The average weight of fruit ranged from 1.33 kg to 7.95 kg. Sugar contents showed 5.4~12.6 $Brix^{\circ}$ without ripening period.

Potatoes have been recognized for a long time as one of the major food crops as well as horticultural crops. Potato production as a table food has been decreased in developed countries, while it has been steadily increased in the third world countries for it importance as food source. It is a new trend to look for the food, not only as a feeding crop but also healthy food. It is also time for the potato producers to look for the potato having high economic value as found in medicinal plants. There are great diversities in potato species, indicating that valuable compounds can be found in different amounts, depending on potato species. We screened the cultivars, breeding clones, and germplasms based on the vitamin C, Vitamin E, antioxidant compounds, diverse sugar types, important amino acids, and other valuable compounds. We could select the breeding clones KC003, 98Wl17, 99j717, and Vally 8 (A group) due to their high levels of antioxidant compounds, and it can be said that most of the red and purple colored potato clones belong to the A group. In the contents of essential amino acids, ‘Taebook Valley’,‘Summer Valley’ and other breeding clones were found to be high in amount. We also made crosses between breeding clones with high biological function and low agronomic traits and low biological function with high quality in agronomic characteristics . The patterns of genetic trends of these offsprings in comparison with their parents will be reported as well. And the potential of using potato as antibody production of anti-cancer will be discussed.
In woody plant germplasms, using prefrozen dormant buds for materials is one way to achieve successful cryopreservation. The protocol of cryopreservation for White birch (Betula platyphylla var. japonica) winter vegetative buds is the following. First, the branches of White birch were collected in January 20, when the vegetative buds were still in a state of quiescence. The winter buds with about 5㎜ of xylem tissue were removed from the branches. They were dehydrated to moisture contents about 44% by air dry treatment. The buds were prefrozen, with the temperature being decreased by 5∼-20$\^{C}$ and then transfered to the LN(liquid nitrogen) maintained below -l96$\^{C}$. After cryopreservation, the vegetative buds were rapidly thawed in a water bath at 40$\pm$5$\^{C}$. In this case, the cell survival rate of samples was about 86%. After sterilization, buds were then cultured on MS medium. These results demonstrate the feasibility for cryopreservation of winter vegetation buds of Betula platyphylla var. japonica.
