한국의 두 강으로부터 재첩의 유전적 종다양성과 열안정성 변이체 (Genetic Diversity and Thermostabilitical Variants of Corbicula japonica from the Two Main Rivers in Korea)

  • We examined the genetic variation within the species, the patterns of genetic diversty between populations, thermostability variations of enzymes and temperature tolerances of Corbicula japonica from the two main rivers In Korea. Starch gel electrophoresis was used to examine the genetic variation of 22 locl. Henting experiments of electrophoresis under the condition of 40$\pm$5$^{\circ}$ for 15$\pm$5 min disclose thermostabllity differences, called heat-sensitive and heat-resistant types, within each 디ectrophoretic allozyme. Genetic diversity at the natural species level was high (77.3%), whereas the extent of heat-treat groups was relatively low (52.6%). The genetic diversity trends to decrease from the source of two main siderable high genetic diversity compared with a mean value of C. japonica species, It is recommended that several populations of the species in Korea should be preserved.

Mitochondrial DNA Sequence Variations and Genetic Relationships among Korean Thais Species (Muricidae: Gastropoda)

  • Thais Roding, 1798, commonly known as rock-shell, is among the most frequently found gastropod genera worldwide on intertidal rocky shores including those of Japan, China, Taiwan and Korea. This group contains important species in many marine environmental studies but species-level taxonomy of the group is quite complicated due to the morphological variations in shell characters. This study examined the genetic variations and relationships among three Korean Thais species based on the partial nucleotide sequences of mitochondrial cox1 gene fragments. Phylogenetic trees from different analytic methods (maximum parsimony, neighbor-joining, and maximum likelihood) showed that T. bronni and T. luteostoma are closely related, indicating the most recent common ancestry. The low sequence divergence found between T. luteostoma and T. bronni, ranging from 1.53% to 3.19%, also corroborates this idea. Further molecular survey using different molecular marker is required to fully understand a detailed picture of the origin for their low level of interspecific sequence divergence. Sequence comparisons among conspecific individuals revealed extensive sequence variations within the three species with maximum values of 2.43% in T. clavigera and 1.37% in both T. bronni and T. luteostoma. In addition, there is an unexpectedly high level of mitochondrial genotypic diversity within each of the three Korean Thais species. The high genetic diversity revealed in Korean Thais species is likely to reflect genetic diversity introduced from potential source populations with diverse geographic origins, such as Taiwan, Hong Kong, and a variety of different coastal regions in South China and Japan. Additional sequence analysis with comprehensive taxon sampling from unstudied potential source populations will be also needed to address the origin and key factors for the high level of genetic diversity discovered within the three Korean Thais species studied.

Genetic Variations of Aspergillus fumigatus Clinical Isolates from Korea

  • Fungal infections by human pathogenic fungi are increasing globally in elderly, children and immune suppressed or deficient patients. Aspergillus fumigatus is one of the well-known pathogenic fungi and causes aspergilloses in human world widely. However, current identification and classification methods based on its phenotypic characteristics still have limitations. Therefore, currently, molecular biological tools using their DNA sequences are used for genotype identification and classification. In the present study, in order to analyze genetic variations of A. fumigatus clinical isolates, a total of six housekeeping genes were amplified by PCR using specific primer pairs and multi-locus sequence typing (MLST) assay. Results from phylogenetic tree analysis showed that most A. fumigatus strains (88.9%) from respiratory specimens were classified into cluster A and B, and approximately half of A. fumigatus strains (46%) from non-respiratory specimens were classified into cluster C and D. Although the sample size was limited, genetic characteristics of A. fumigatus clinical isolates according to their origins were very similar and well-correlated with other clinical data.

Evaluation of Genetic Heterogeniety among the Corn Landraces Collected from Farmer's Field

  • This paper describes the variations in eight agronomic traits in three unadapted local landraces and an inbred cultivar of corn. To compare the agronomic traits in field evaluation with molecular marker evaluation the genotypes of the plant introduction were also checked by 4 microsatellite-SSR loci. The variations of the eight agronomic traits were higher in the local landrades than in the inbred line. which was substantiated by the high genetic variation in the landrades with microsatellite-SSR loci. The level of genetic variation was also different between landraces. Since the genetic evaluation can be easily quantified by the analysis of microsatellite-SSR loci. the threshold level of genetic homogeneity in the population for parental lines in breeding program can be determined and the effort of maintaining the landrace population would be alleviated. As an example in our analysis. the entry from Whachon should not need the same number of selfing generations as the other two landraces to get the level of inbred state. Since this line showed lowest intra-genetic variation within the population.

Genetic Analysis of SCN5A in Korean Patients Associated with Atrioventricular Conduction Block

  • Recent several studies have shown that the genetic variation of SCN5A is related with atrioventricular conduction block (AVB); no study has yet been published in Koreans. Therefore, to determine the AVB-associated genetic variation in Korean patients, we investigated the genetic variation of SCN5A in Korean patients with AVB and compared with normal control subjects. We enrolled 113 patients with AVB and 80 normal controls with no cardiac symptoms. DNA was isolated from the peripheral blood, and all exons (exon 2-exon 28) except the untranslated region and exon-intron boundaries of the SCN5A gene were amplified by multiplex PCR and directly sequenced using an ABI PRISM 3100 Genetic Analyzer. When a variation was discovered in genomic DNA from AVB patients, we confirmed whether the same variation existed in the control genomic DNA. In the present study, a total of 7 genetic variations were detected in 113 AVB patients. Of the 7 variations, 5 (G87A-A29A, intervening sequence 9-3C>A, A1673G-H558R, G3578A-R1193Q, and T5457C-D1819D) have been reported in previous studies, and 2 (C48G-F16L and G3048A-T1016T) were novel variations that have not been reported. The 2 newly discovered variations were not found in the 80 normal controls. In addition, G298S, G514C, P1008S, G1406R, and D1595N, identified in other ethnic populations, were not detected in this study. We found 2 novel genetic variations in the SCN5A gene in Korean patients with AVB. However, further functional study might be needed.

Genetic Diversity and Relationship in Soybean MDP (Mutant Diversity Pool) Revealed by TRAP and TE-TRAP Markers

  • Mutation breeding is the useful tool to improve agronomic traits in various crop species. Soybean is most important crop and is rich in protein and oil contents. Despite of the importance as economic value and various genetic resource of soybean, there have been limited studies of genetic relationship among mutant resources through radiation breeding. In this study, the agronomical phenotype for selecting various genetic resources was evaluated in 528 soybean mutant lines. As a result, 210 soybean mutants with their original cultivars were selected with various traits. We named 210 selected lines as Mutant Diversity Pool (MDP). The genetic diversity and the relationship of the MDP were investigated using TRAP and TE-TRAP markers. In TRAP analysis, sixteen primer combination (PC)s were used and a total of 551 fragments were amplified. The highest (84.00%) and the lowest (32.35%) polymorphism levels were showed in PC MIR157B+Ga5 and B14G14B+Ga3, respectively. The mean of PIC values was 0.15 ranging from 0.07 in B14G14B+Sa12 to 0.23 in MIR157B+Sa4. Phylogenetic and population structure analysis indicated that the 210 MDP lines dispersed to four groups among the wild types and their mutants. The highest genetic diversity among populations was observed between lines Paldal and 523-7 (Fst=0.409), whereas the lowest genetic diversity was between population KAS360-22 and 94seori (Fst=0.065). AMOVA showed 11.583 (21.0%) and 43.532 (79.0%) variations in inter and intra mutant population, respectively. Overall, the genetic similarity of each intra mutant populations was closer than that of inter mutant population. A total of 408 fragments were amplified in the 210 MDP using twelve PCs of TE-TRAP markers that were obtained from a combination of three TIR sequence of transposable elements (MITE-stowaway; M-s, MITE-tourist; M-t, PONG). The highest (77.42%) and the lowest (56.00%) polymorphism levels were showed in PONG+Sa4 and PONG+Sa12, respectively. The mean of PIC values was 0.15 ranging from 0.09 in M-s+Sa4 and M-s+Ga5 to 0.21 in M-t+Ga5. AMOVA of M-s showed 2.209 (20%) and 8.957 (80%) variations in inter and intra mutant population, respectively. AMOVA of M-t showed 2.766 (18%) and 12.385 (82%) variations in inter and intra mutant population, respectively. AMOVA of PONG showed 3.151 (29%) and 7.646 (71%) variations in inter and intra mutant population, respectively. According to our study, the PONG had higher inter mutant population and lower intra mutant population. This mean was that for aspect of radiation sensitivity, M-s and M-t showed higher mobility than that of PONG. Our results suggest that the TRAP and the TE-TRAP markers may be useful for assessing the genetic diversity and relationship among soybean MDP and help to improve our knowledge of soybean mutation/radiation breeding.

엽록체 DNA trnL-F 및 핵리보조옴 DNA ITS, ETS 염기서열에 의한 고려인삼의 유전적 변이와 계통학적 유연관계 (Genetic Variation and Phylogenetic Relationship of Korean Ginseng based on cpDNA trnL-F, nrDNA ITS and ETS Sequences)

  • Genetic variations and relationship based on the sequences of cpDNA trnL-F gene, nrDNA ITS and ETS region among the twenty four taxa including Panax ginseng C.A. Meyer and its related species were investigated. And taxonomic status and molecular phylogenetic relationship between P. ginseng and related groups were discussed. Molecular systematic data from cpDNA and nrDNA sequences were very useful to elucidate the genetic variations and relationships among the treated taxa. It was found that P. ginseng is the independent unique species with distinct genetic limitation from the related species such as P. quinquefolius, P. japonicum, P. notoginseng and P. pseudoginseng. P. ginseng including cultivated types as well as wild ones formed monophyletic group with high genetic similarities. P. quinquefolius and P. japonicum were the most related sister groups of P. ginseng based on the molecular phylogenetic results in this study.

계방산, 오대산 및 지리산 야생 표고균주의 유전적 변이 (Genetic Variation of the Wild Strains of Lentinula edodes in Three Mountains of Korea)

  • RAPD 검정법을 이용하여, 우리 나라에 자생하고 있는 야생 표고균주의 지역간 유전변이에 관해 분석하였다. 사용된 야생 표고균주는 계방산 수집 10개, 오대산 수집 11개, 지리산 수집 11개의 야생 표고 총 32균주를 사용하였다. Genomic DNA를 추출하여 10개의 random primer를 사용하여 PCR 증폭시킨 후 전기영동 한 결과 170개의 밴드가 관찰되었으며, 그중 다형성 밴드는 161개가 검출되었다. 이들 전기영동 상에 나타난 밴드의 유무(1,0)로 cluster 분석을 한 결과, 계방산과 오대산 표고균주들이 하나의 그룹으로, 나머지 지리산 표고균주들이 다른 하나의 그룹으로 형성되었다. 또한 AMOVA 분석결과 세 지역의 집단간 유전적인 차이는 12.5%를 나타내었고, 각 균주들 간에는 87.5%임을 알 수 있었다. 이러한 결과는 계방산과 오대산 균주들은 그룹 내 분화가 큰 것이 주요 원인일 것으로 생각되며, 반면, 지리산 균주들은 계방산과 오대산과는 유전적 격리가 존재함을 알 수 있었다.

Genetic structure of Larimichthys polyactis (Pisces: Sciaenidae) in the Yellow and East China Seas inferred from microsatellite and mitochondrial DNA analyses

  • Genetic variation was surveyed at four microsatellite loci and 1416 base pairs (bp) of the mitochondrial DNA (mtDNA) cytochrome c oxidase I gene (COI) to clarify the genetic structure of the small yellow croaker, Larimichthys polyactis, in the Yellow and East China Seas, especially regarding four provisional populations, (one Korean and three Chinese populations). Based on microsatellite DNA variations, the estimated expected heterozygosity ($H_E$) in each population ranged from 0.776 to 0.947. The microsatellite pairwise $F_{ST}$ estimates showed no significant genetic differentiation between the populations. MtDNA variations also indicated no genetic structure in L. polyactis, but very high variability. The absence of genetic differentiation among and within populations of L. polyactis may either result from the random migration of the adult or the passive dispersal of the eggs and larvae.

Geographic Variations of Three Fulvia mutica Populations

  • In the present study, the seven primers BION-33, BION-34, BION-37, BION-41, BION-44, BION-45 and BION-42 generated the total number of loci, average number of loci per lane and specific loci in Hongseong, Yeosu and Goheung population of F. mutica, respectively. 7 primers generated 19 specific loci in the Hongseong population, 29.3 in the Yeosu population and 23.1 in the Goheung population, respectively. Especially, the decamer primer BION-37 generated 7 unique loci to each population, which were identifying each population, approximately 700 bp in Hongseong population. In this study, the dendrogram obtained by the seven primers indicates three genetic clusters: cluster 1 (HONGSEONG 01-HONGSEONG 07), cluster 2 (YEOSU 08-YEOSU 14) and cluster 3 (GOHEUNG 15-GOHEUNG 21). Among the twenty one cockles, the shortest genetic distance that displayed significant molecular differences was between individuals 17 and 19 from the Goheung population (genetic distance = 0.051), while the longest genetic distance among the twenty-one cockle individuals that displayed significant molecular differences was between individuals HONGSEONG no. 03 and YEOSU no. 12 (genetic distance = 0.616). Relatively, individuals of YEOSU population were fairly closely related to that of GOHEUNG population. Ultimately, PCR fragments revealed of in this study may be useful as a DNA marker the three geographic populations to distinguish.
