Systematic study on the genus Corbicula (Bivalvia : Corbiculidae) in Korea (한국산 재첩속(Corbicula) 이매패류의 계통분류학적 연구)

  • A genetic analysis using starch gel electrophoresis was performed to clarify the degrees of genetic differentiation and the phylogenetic relationships among four species of the genus Corbicula (Bivalvia, Corbiculidae). The average genetic similarity coefficients among the populations of each species on C. fluminea, C. leana and C. colorata were very closed (Rogers' S$\geq$0.970), whereas the other C. japonica was separated into two genetic groups (S=0.873) with distinct allelic difference at the Gpl locus. Thuefore additional taxonomic studies are needed on this species. On the other hand, these four species were well distinguished from each other genetically with distinct allelic differences at 4 to 9 loci. The genetic similarity coefficients (S) between C. leana and C. colorata, C. fluminea and C. leana, and C. fluminea and C. colorata were 0.737, 0.689, and 0.594, respectively. In particular, the average genetic similarity coefficients between these three species (inhabiting in freshwater) and the other C. japonica (inhabiting in brackishwater) were notably lower (S=0.370).

Genetic Diversity Among Waxy Corn Accessions in Korea Revealed by Microsatellite Markers

  • Knowledge of genetic diversity and of the genetic relationships among elite breeding materials has had a significant impact on the improvement of crops. In maize, this information is particularly useful in i) planning crosses for hybrid and line development, ii) in assigning lines to heterotic groups and iii) in plant variety protection. We have used the SSR technique to study the genetic diversity and genetic relationships among 76 Korean waxy corn accessions, representing a diverse collection from throughout Korea. Assessment of genetic diversity among members of this group was conducted using 30 microsatellite markers. Among these 30 microsatellite markers, we identified a total of 127 alleles (with an average of 4.2 and a range of between 2 and 9 alleles per locus). Gene diversity at these 30 microsatellite loci varied from 0.125 to 0.795 with an average of 0.507. The cluster tree generated with the described microsatellite markers recognized two major groups with 36.5% genetic similarity. Group I includes 63 inbred lines, with similarity coefficients of between 0.365 and 0.99. Group II includes 13 inbred lines, with similarity coefficients of between 0.45 and 0.85. The present study indicates that the 30 microsatellite loci chosen for this analysis are effective molecular markers for the assessment of genetic diversity and genetic relationships between Korean waxy corn accessions. Specifically, this study's assessment of genetic diversity and relationships between a set of 76 Korean waxy corn inbred lines will be helpful for such activities as planning crosses for hybrid and line development and association mapping analyses of maize breeding programs in Korea.

Genetic Relationahips of the Two Morphorogical Types of Myzus persicae(Homoptera:Aphididae) Collected from Tobacco Plants Based on Random Amplified Polymorphic DNA(RAPD) (연초에서 발생하는 복숭아혹진딧물(Myzus persicae)형태형 2종의 Random Amplified Polymorphic DNA(RAPD)을 이용한 유전적 유연관계 분석)

  • Random amplified polymorphic DNA (RAPD) markers were used to analyze genetic similarity among 8 clones of apierous green peach aphid, two types (M. persicae Sulzer and M. nicotianae lack man) classified by their mo~hologi~cahla raters and host preference (Blackman, 1987), collected from tobacco plants. The genetic variation among these clones was evaluated by polymerase chain reaction amplification with 20 random primers. The higher GC contents of primers, the better in amplification efficiency of PCR reaction in general. The genetic similarities among eight aphid clones were analyzed from UPGMA (unweighted pair group average method) cluster analysis based on simple matching coefficient. The range of genetic similarity coefficients was 0.414 to 0.808. The most close relationship among the clones was similarity coefficient of 0.808 between the PG2 and the PG3 clone. The eight aphid clones analyzed were clustered into three groups by the genetic similarity coefficient. The first group, PG1, PG2, PG3 clone including in M. persicae type by their morphological characters and RED clone in M. nicotianae type was clustered at the genetic similarity coefficient of 0.643. The second group, GR1, GR2, BRN in M. nicotianae type was at the 0.636;and the third group was DBR clone in M. persicae type. The results did not indicate any correlation between m&-phological types (M. persicae and M. nicotianae) and RAPD polymorphism. We could not detect any obvious genetic relationships of the two morphological types of the green peach aphid collected from tobacco plants.

Genetic Polymorphisms and phylogenetic Relationships of Italian Ryegrass Cultivars Based on Random Amplified Polymorphic DNA ( RAPD ) Markers (RAPD 표지인자를 이용한 이탈리안 라이그라스 품종의 유전적 변이 및 유연관계 분석)

  • Eleven Italian ryegrass cultivars were examined for their genetic polymorphisms and phylogenetic relationships using randomly amplified polymorphic DNA (RAPD) markers. In RAPD analysis of 34 random primers, 96 of total 162 bands obtained from 16 primers were polymorphic and sizes of polymorphic band ranged between 0.5 and 1.5kb. Number of bands amplified per primer was varied from 3 to 16 and average number was 14.8. Phylogenetic relationship among cultivars based on the RAPD analysis was examined using UPGMA computer program. In pairwise genetic similarity test of 11 Italian ryegrass cultivars, Grazer and Orlando showed highest coefficient of genetic similarity as 0.740, whereas Marshall and Orlando was lowest as 0.438. Eleven Italian ryegrass cultivars were grouped into 3 major clusters and genetic distance of clusters ranged between 0.567 and 0.646, indicating low level of genetic variation.

  • A genetic analysis using starch gel electrophoresis was peformed to clarify the degrees of genetic differentiation among two species of the genus Koreanomelania and Koreoleptoxis (Gastropoda, Pleuroceridae). The average genetic similarity coefficients (S) among the populations of K. nodifila was S=0.848 (0.805-0.905) and K. globus ovalis was S : 0.755 (0.666-0.860). The genetic similarity coefficients between Semisulcospira gottschei and K. nodifila, S. gottschei and K. globus ovalis, and K. nodifila and K. giobus ovalis were 0.194, 0.301, and 0.301, respectively. So between K. nodifila and K. globus ovalis seems to be genetically two distinct taxa.

  • Genetic diversity among selected isolates of Macrophomina phaseolina, a causal agent of charcoal rot (stalk rot) disease in sorghum was studied using PCR-RAPD markers. A set of ten isolates, from ten different rabi sorghum genotypes representing two traditional sorghum growing situations viz., Dharwad- a transitional high rainfall region and Bijapur- a semi-arid low rainfall region in South India. From a set of 40 random primers tested, amplicon profiles of 15 were reproducible. A total of 149 amplicon levels, with an average of 9.9 bands per primer, were available for analysis, of which 148 were polymorphic (99.3%). It was possible to discriminate all the isolates with any of the 15 primers employed. UPGMA clustering of data indicated that the isolates shared varied levels of genetic similarity within a range of 0.14 to 0.72 similarity coefficient index and it was suggestive that grouping of isolates was not related to sampling location in anyway. A high level of genetic heterogeneity of 0.28 was recorded among the isolates.

  • Genetic diversity of 45 tea accessions from Korea, Japan, China and Taiwan was investigated by using RAPD analysis. Out of the eighty primers screened, twenty primers generated 99 polymorphic bands with a polymorphic rate 87.0%. The size of the amplified fragments ranged from about 3,138 bp to 520 bp. By cluster analysis, all of the 45 accessions can be grouped into five groups. Over 90% of the 32 Korean accessions belonged to group II, III, IV and V. Moreover, newly developed Korean cultivars (accession no. 13, 14 and 15) belonged to very different group compared with any other Korean accessions. Among the Korean accessions, the minimum genetic similarity 0.500 was obtained between accession no. 17 and 37 and the largest genetic similarity 0.912 between no. 20 and 21.

  • The genetic diversity and the genetic relationship among 30 genetic resources of T. officinale and T. coreanum collected from 20 regions in Korea were evaluated by using ISSR markers. Out of 127 loci detected overall, 122 were identified to be polymorphic with a rate of 96.0% at the 30 individuals. The intraspecific polymorphism between T. officinale and T. coreanum was 92.6% and 88.2%, respectively. The genetic similarity matrix (GSM) revealed a wide range of variablility among the 30 accessions, spanning from 0.179 to 922. According to the clustering analysis, different species T. officinale and T. coreanum, were divided into independent groups and all of the accessions could be classified into 7 categories. Especially, all of the mountain collected accessions belonged to independent groups. The study findings indicate that T. officinale and T. coreanum accessions have a high genetic diversity and accordingly carry a germ-plasm qualifying as good genetic resources for breeding.

  • Broadening the genetic base of Platycodon grandiflorum DC. cultivar to sustain improvement requires assessment of genetic diversity available in P. grandiflorum DC.. The objective of this study was to analyze the genetic variation, genetic relationship among 48 samples collected from East-Asian Area by means of RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction) markers. From the 18 primers tested, produced total 211 bands with an average of 11.7 bands per primer and obtained 103 polymorphic band with an average of 5.7 bands per primer,s revealed relatively high percentage of polymorphic bands (48.8%). The genetic similarities calculated from RAPD data varied from 0.688 to 0.994 and were clustered to six major groups on a criterion of 0.78 similarity coefficient. The present study has revealed the significant genetic similarity among the samples tested. The analysis of genetic relationships in P. grandiflorum using RAPD-PCR banding data can be useful for the breed improvement.

  • Electrophoretic analysis was carried out to elucidate genetic relationships of four Korean scallops, Patinopecten yessoensis, chlamys ferreri ferreri, Chlamys swifti and Amusium japonicum japonicum, and of a Chinese population of C. ferreri ferreri purchased form a market. Glucose phosphate isomerase banding pattern was highly varied among eight loci. Three populations of C. ferreri ferreri were more closely clustered in a dendrogram within the range of Nei's genetic similarity values of 0.730-0.830. P. yessoenensis and Chlamys swifti were clustered with genetic similarity value of 0.647. These two clusters were lineated at the value of 0.598. A. japonicum japonicum was clustered with other three species at value of 0.541.
