Relationship Between Enhancement of Electrostriction and Decrease of Activation Energy in Porcine Pancreatic Lipase Catalysis

  • The contribution of electrostriction of water molecules to the stabilization of the negatively charged tetrahedral transition state of a lipase-catalyzed reaction was examined by means of kinetic studies involving high-pressure and solvent dielectric constant. A good correlation was observed between the increased catalytic efficiency of lipase and the decreased solvent dielectric constant. When the dielectric constant of solvents was lowered by 5.00 units, the losses of activation energy and free energy of activation were 7.92 kJ/mol and 11.24 kJ/mol, respectively. The activation volume for $k_{cat}$ decreased significantly as the dielectric constant of solvent decreased, indicating that the degree of electrostriction of water molecules around the charged tetrahedral transition state has been enhanced. These observations demonstrate that the increase in the catalytic efficiency of the lipase reaction with decreasing dielectric constant resulted from the stabilization of electrostatic energy for the formation of an oxyanion hole, and that this stabilization was caused by the increase of electrostricted water around the charged tetrahedral transition state. Therefore, we conclude that the control of solvent dielectric constant can stabilize the tetrahedral transition state, thus lowering the activation energy.

Optimization, Purification, and Characterization of Haloalkaline Serine Protease from a Haloalkaliphilic Archaeon Natrialba hulunbeirensis Strain WNHS14

  • The present study addresses isolation, optimization, partial purification, and characterization of a haloalkaline serine protease from a newly isolated haloarchaeal strain isolated from Wadi El Natrun in Egypt. We expected that a two-step sequential statistical approach (one variable at a time, followed by response surface methodology) might maximize the production of the haloalkaline serine protease. The enzyme was partially purified using Hiprep 16/60 sephacryl S-100 HR gel filtration column. Molecular identification revealed the newly isolated haloarchaeon to be Natrialba hulunbeirensis strain WNHS14. Among several tested physicochemical determinants, casamino acids, KCl, and NaCl showed the most significant effects on enzyme production as determined from results of the One-Variable-At-A-time (OVAT) study. The BoxBehnken design localized the optimal levels of the three key determinants; casamino acids, KCl, and NaCl to be 0.5% (w/v), 0.02% (w/v), and 15% (w/v), respectively, obtaining 62.9 U/ml as the maximal amount of protease produced after treatment at 40℃, and pH 9 for 9 days with 6-fold enhancement in yield. The enzyme was partially purified after size exclusion chromatography with specific activity, purification fold, and yield of 1282.63 U/mg, 8.9, and 23%, respectively. The enzyme showed its maximal activity at pH, temperature, and NaCl concentration optima of 10, 75℃, and 2 M, respectively. Phenylmethylsulfonyl fluoride (PMSF, 5 mM) completely inhibited enzyme activity.

  • Machine learning methods diversely applied to the Internet of Things (IoT) field have been successful due to the enhancement of computer processing power. They offer an effective way of detecting malicious intrusions in IoT because of their high-level feature extraction capabilities. In this paper, we proposed a novel feature selection method for malicious intrusion detection in IoT by using an evolutionary technique - Genetic Algorithm (GA) and Machine Learning (ML) algorithms. The proposed model is performing the classification of BoT-IoT dataset to evaluate its quality through the training and testing with classifiers. The data is reduced and several preprocessing steps are applied such as: unnecessary information removal, null value checking, label encoding, standard scaling and data balancing. GA has applied over the preprocessed data, to select the most relevant features and maintain model optimization. The selected features from GA are given to ML classifiers such as Logistic Regression (LR) and Support Vector Machine (SVM) and the results are evaluated using performance evaluation measures including recall, precision and f1-score. Two sets of experiments are conducted, and it is concluded that hyperparameter tuning has a significant consequence on the performance of both ML classifiers. Overall, SVM still remained the best model in both cases and overall results increased.

  • This paper presents a novel objective function for distribution system reconfiguration for reliability enhancement. When islanding operations of distributed generators is prohibited, faults in the feeder interrupt the operation of distributed generators. For this reason, we include the customer interruption cost as well as the distributed generator interruption cost in the objective function in the network reconfiguration algorithm. The network reconfiguration in which genetic algorithms are used is implemented by MATLAB. The effect of the proposed objective function in the network reconfiguration is analyzed and compared with existing objective functions through case studies. The network reconfiguration considering the proposed objective function is suitable for a distribution system that has a high penetration of distributed generators.

Flat Speaker Design by Optimization of Plane Actuator (평판 작동기의 최적화를 통한 평면 스피커 설계)

  • In this study, a design method using plane actuator is developed to make new speaker system, whose shape is much thinner than that of conventional loudspeaker. Piezofilm(PVDF) is used as plane actuator of flat speaker. To avoid the distortion of sound radiated from flat speaker, the frequency response of radiated sound to be flat is taken as the design objective. The electrode pattern and orientation angle of piezofilm actuator is optimized to satisfy the design objective. The formulation is based on the coupled finite element and boundary element method. Genetic algorithm is used in the optimization process, which is useful in the optimization of discrete design variables. Frequency response with optimized piezofilm actuator is made flat enough to satisfy the design objective. For the enhancement of sound power, double-layered piezofilm actuators are also considered. The sound power with double-layered actuator becomes larger than that with single-layered actuator as expected.

  • Recent advancement in molocular biology enhanced further understanding of the carcinogenesis of oral cancer and its relation with various genetic backgrounds. Familial risk factors includes similar habits of the family and polymorphic variations of the genes. Recently, human papilloma virus has been suggested to be linked with oral cancer progression. Enhancement of understanding of the damage or alteration in molecular pathway in various cellular response of oral cancer progression would lead the targeted therapy or precise early diagnosis of the oral cancer.

  • DNA immunization induces B and T cell responses to various pathogens and tumors. However, these responses are known to be relatively weak and often transient. Thus, novel strategies are necessary for enhancing immune responses induced by DNA immunization. Here, we demonstrated that co-immunization of influenza virus nucleoprotein (NP) gene significantly enhances humoral and cell-mediated responses to codelivered antigens in mice. We also found that NP DNA coimmunization augments in vivo proliferation of adoptively transferred antigen-specific CD4 and CD8 T cells, which enhanced protective immunity against tumor challenge. Our results suggest that NP DNA can serve as a novel genetic adjuvant in cocktail DNA vaccination.

Validation of Heterodimeric TAT-NLS Peptide as a Gene Delivery Enhancer

  • Cationic liposomes have been actively used as gene delivery vehicles despite their unsatisfactory efficiencies because of their relatively low toxicity. In this study, we designed novel heterodimeric peptides as nonviral gene delivery systems from TAT and NLS peptides using cysteine-to-cysteine disulfide bonds between the two. Mixing these heterodimeric peptides with DNA before mixing with lipofectamine resulted in higher transfection efficiencies in MCF-7 breast cancer cells than mixing unmodified TAT, NLS, and a simple mixture of TAT and NLS with DNA, but did not show an adverse effect on cell viability. In gel retardation assays, the DNA binding affinities of heterodimeric peptides were stronger than NLS but weaker than TAT. However, this enhancement was only observed when heterodimeric peptides were premixed with DNA before being mixed with lipofectamine. The described novel transfection-enhancing peptide system produced by the heterodimerization of TAT and NLS peptides followed by simple mixing with DNA, increased the gene transfer efficiency of cationic lipids without enhancing cytotoxicity.
