Development of Drought Assessment Scheme using Root Zone Soil Moisture (토양수분을 이용한 가뭄평가기법 개발)

  • 최근 원격탐사기법을 이용한 많은 가뭄평가기법들이 개발되었으나 산림과 함께 산악지형이 우세한 우리나라의 경우 지형특성으로 인하여 가뭄평가시 불확실성이 증가하게 된다. 특히, 농업가뭄은 기후와 지표특성에 큰 영향을 받기 때문에 기후특성만을 고려한 가뭄지수는 실제 필요한 농업가뭄의 특성을 반영하는데 있어서 한계가 있다. 따라서 본 연구에서는 기후와 지표특성을 함께 고려할 수 있는 토양수분을 이용한 가뭄평가기법(Drought Assessment Scheme)을 개발하였다. 가뭄평가기법을 위하여 역추적기법(Inverse Modeling-IM) 기반의 자료동화기법(Data Assimilation, DA)을 이용하였다. 자료동화기법은 1-Dimensional (1-D) 기반의 토양의 물리적 특성을 고려하는 SWMI_ST 모형과 최적화 알고리즘(유전자 알고리즘, Genetic Algorithm-GA)을 연계하여 실측 및 위성기반의 토양수분자료로부터 토양의 수리학적 매개변수(${\alpha}$, n, ${\Theta}_{res}$, ${\Theta}_{sat}$, $K_{sat}$)를 추출한다. 본 연구에서는 LANDSAT(30 m X 30 m) 및 MODerate Resolution Imaging Spectroradiometer(MODIS, 500 m X 500 m) 이미지자료를 이용하여 시 공간적으로 분포되어 있는 토양수분을 산정하였으며, 이후 자료동화기법을 이용하여 LANDSAT/MODIS 토양수분자료로 부터 공간적으로 분포되어 있는 토양의 매개변수를 추출하였다. 추출된 매개변수, GIS 기반의 지표피복 및 기상자료를 이용하여 장기간의 토양수분을 산정 및 예측 할 수 있다. 고해상도의 이미지 자료를 사용하는 가뭄평가기법은 필지~시 군 단위까지 실제 우리나라 지형특성을 고려하여 효율적으로 가뭄을 모니터링 및 예측 할 수 있다.

Effect of Genetic Predisposition on Blood Lipid Traits Using Cumulative Risk Assessment in the Korean Population

  • Dyslipidemia, mainly characterized by high triglyceride (TG) and low high-density lipoprotein cholesterol (HDL-C) levels, is an important etiological factor in the development of cardiovascular disease (CVD). Considering the relationship between childhood obesity and CVD risk, it would be worthwhile to evaluate whether previously identified lipid-related variants in adult subjects are associated with lipid variations in a childhood obesity study (n = 482). In an association analysis for 16 genome-wide association study (GWAS)-based candidate loci, we confirmed significant associations of a genetic predisposition to lipoprotein concentrations in a childhood obesity study. Having two loci (rs10503669 at LPL and rs16940212 at LIPC) that showed the strongest association with blood levels of TG and HDL-C, we calculated a genetic risk score (GRS), representing the sum of the risk alleles. It has been observed that increasing GRS is significantly associated with decreased HDL-C (effect size, $-1.13{\pm}0.07$) compared to single nucleotide polymorphism combinations without two risk variants. In addition, a positive correlation was observed between allelic dosage score and risk allele (rs10503669 at LPL) on high TG levels (effect size, $10.89{\pm}0.84$). These two loci yielded consistent associations in our previous meta-analysis. Taken together, our findings demonstrate that the genetic architecture of circulating lipid levels (TG and HDL-C) overlap to a large extent in childhood as well as in adulthood. Post-GWAS functional characterization of these variants is further required to elucidate their pathophysiological roles and biological mechanisms.

Genetic Difference Analysis and Environmental Assessment of Miscanthus sinensis and Phragmites australis to Apply Regional Seed for Restoration in Korea (복원 소재로서 지역 종자 적용을 위한 억새와 갈대의 유전적 변이분석)

  • Restoration ecology is the practical study of renewing and restoring a spoilt, degraded, or devastated ecosystems in the environment. Because the Korean industry has been drastically developed for the past few decades, the Korean ecosystem requires restoration using regional seed. In this study, we identified the variation of phylogenic relationship of Miscanthus sinensis or Phragmites australis by locations in Korea. Chloroplast DNA atpF-H and psbA-trnH interspace region were used as a molecular marker to resolve the phylogenic relationship in 10 different locations. We performed the molecular phylogenetic analysis with 10 chloroplast DNAs from each location using Kimura 2-parameter. The analysis of Miscanthus showed that all atpF-H genes were exact matches except for Ose san. In contrast to Mischanthus, the atpF-H genes from Phragmites were observed to have more variation. A total of 7 locations revealed the variation in chloroplast gene. According to the phylogenic tree in Phragmites, 2 of 10 samples in 6 locations and 3 of 10 in 1 location showed variation with 0.160-0.181 genetic distance. According to the genetic distance of the Miscanthus sinensis, there were no mutations in all regions except the Hongsung. These results support regional differences and show the necessity for seed collection by region. In the case of Phragmites australis, genetic variation occurred in all regions.

An assessment of hair loss ameliorating effect of medicinal plant extracts, amino acid and vitamin complex in human subject (약용 식물 추출물, 아미노산, 비타민 복합제의 탈모 개선 효과 평가를 위한 인체적용시험)

  • Alopecia is usually presented as patchy, no scarring hair loss. It seems to be a hormonal imbalance or an immune mediated disease, whereas genetic predisposition, environmental and psychological factors may be involved in its etiology. Many treatments of alopecia disease have been widely applied, but their side effects are also well known. The purpose of this study is to demonstrate efficacy and safety of a food supplement containing medicinal plant extracts, amino acid and vitamin complex on the prevention of hair loss in human. A total of 20 subjects aged between 29-50 years with subjective hair loss complaint were recruited for this study. Each subject took 1 g/day supplement for 3 months. 5-Scale self-assessment test, hair loss count and measurement of hair thickness were conducted to evaluate the effect of supplement on the improvement of hair loss. Subjective hair loss was decreased and hair damage and thickness were improved in self-assessment test. After 3 months of supplementation, hair loss counts were decreased (P<0.05) while hair thickness was increased(P<0.001) significantly. There was no side effect observed during the study. These results suggest that the supplement consist of medicinal plant extracts, amino acid and vitamin complex might be useful for the improvement of hair loss with no significant side effects.

Risk Assessment of Genetically Modified Organisms (유전자변형 생물체의 위해성평가)

  • New breeding method by genetic engineering is expected as a key technology to solve food shortage due to the growing world population in the year 2000s. Many genetically modified organisms (GMOs) were already developed and the commercial cultivation had started. The first GMO, Flavr Savr tomato, which rotted at a much slower pace than ordinary ones, was developed in US in 1994. Since then, over than 70 different agricultural products including corn, cotton, soybean, papaya, potato, and squash made with genetically modified plants are reportedly on sale worldwide. Supporters favor the GMOs because they have greater yields, longer shelf lives and stronger resistance to disease and insects. On the other hand, opponents say that the supporters ignore a potential danger that they may damage the environment as well as human beings. To assure the safe development and use of GMOs as food and other biotech products, the possible risks on biological environment and human health should be throughly examined and regulated by developer and government. Because the biosafety problem is a global, environmental, and trade issue, a new international treaty is under development. The Cartagena Protocol on Biosafety was adopted at the 1 st Extraordinary Conference of Parties of the Convention on Biological Diversity which was held at Mont-real, Canada, Jan. 29th, 2000. The adoption of the Protocol is seen as a breakthrough in that it is based on the" Precautionary Principle" despite scientific uncertainties surrounding potential risks that GMOs may inflict on human health and the environment and that it has laid the ground for introduction of specific steps to handle international trading of GMOs. In this paper, the authors would like to introduce the current status and perspective of environmental and human risk assessment of GMOs.t of GMOs.

Hybridization between Marine Medaka Oryzias dancena and Javanese Medaka Oryzias javanicus (바다송사리 Oryzias dancena와 자바송사리 Oryzias javanicus 간 잡종 유도)

  • Inductions of hybrids and reciprocal hybrids between Oryzias dancena and O. javanicus (ODJ and OJD) were conducted and backcross hybrids between female O. dancena and male ODJ were also produced for biological and cytogenetic analysis. Embryonic development of ODJ and OJD were compared with those of their parents. Developmental time was fastest in O. dancena and ODJ, followed by O. javanicus and OJD. Oryzias dancena hatched 11 days (d) after fertilization, ODJ at 13 d, O. javanicus at 14 d and OJD at 15 d. The abnormality of external morphology rate in ODJ was 10.6%; however, OJD showed a high degree of abnormality, over 90%. The proportion of males was 90.0% and 31.3% for ODJ and OJD, respectively. Cytogenetic analysis was conducted to obtain basic information for genetic identification of O. dancena, O. javanicus and their hybrids. The karyotypes of all experimental groups showed 2n=48 chromosomes and the fundamental number (FN) was 48. The first pair carried secondary constrictions near the centromeric regions. Erythrocyte area and volume were $9.8\;{\pm}\;0.5\;{\mu}m^2$ and $18.2\;{\pm}\;1.0\;{\mu}m^3$, respectively, for O. dancena, $8.3\;{\pm}\;0.5\;{\mu}m^2$ and $15.8\;{\pm}\;1.5\;{\mu}m^3$ in O. javanicus, and $18.3\;{\pm}\;0.5\;{\mu}m^2$ and $15.7\;{\pm}\;1.3\;{\mu}m^3$ in ODJ. Erythrocyte area and volume in ODJ were similar to those of O. javanicus. In backcross hybrids between female O. dancena and male ODJ, all embryos failed to develop and died in the late gastrula stage.

RNA-Seq De Novo Assembly and Differential Transcriptome Analysis of Korean Medicinal Herb Cirsium japonicum var. spinossimum

  • Cirsium japonicum belongs to the Asteraceae or Compositae family and is a medicinal plant in Asia that has a variety of effects, including tumour inhibition, improved immunity with flavones, and antidiabetic and hepatoprotective effects. Silymarin is synthesized by 4-coumaroyl-CoA via both the flavonoid and phenylpropanoid pathways to produce the immediate precursors taxifolin and coniferyl alcohol. Then, the oxidative radicalization of taxifolin and coniferyl alcohol produces silymarin. We identified the expression of genes related to the synthesis of silymarin in C. japonicum in three different tissues, namely, flowers, leaves, and roots, through RNA sequencing. We obtained 51,133 unigenes from transcriptome sequencing by de novo assembly using Trinity v2.1.1, TransDecoder v2.0.1, and CD-HIT v4.6 software. The differentially expressed gene analysis revealed that the expression of genes related to the flavonoid pathway was higher in the flowers, whereas the phenylpropanoid pathway was more highly expressed in the roots. In this study, we established a global transcriptome dataset for C. japonicum. The data shall not only be useful to focus more deeply on the genes related to product medicinal metabolite including flavolignan but also to study the functional genomics for genetic engineering of C. japonicum.

  • In weapon assignment studies to defend against threats such as ballistic missiles and long range artillery, threat assessment was partially lacking in analysis of various threat attributes, and considering the threat characteristics of warheads, which are difficult to judge in the early flight stages, it is very important to apply more reliable optimal solutions than approximate solution using LP model, Meta heuristics Genetic Algorithm, Tabu search and Particle swarm optimization etc. Our studies suggest Generic Rule based threat evaluation and weapon assignment algorithm in the basis of various attributes of threats. First job of studies analyzes information on Various attributes such as the type of target, Flight trajectory and flight time, range and intercept altitude of the intercept system, etc. Second job of studies propose Rule based threat evaluation and weapon assignment algorithm were applied to obtain a more reliable solution by reflection the importance of the interception system. It analyzes ballistic missiles and long-range artillery was assigned to multiple intercept system by real time threat assessment reflecting various threat information. The results of this study are provided reliable solution for Weapon Assignment problem as well as considered to be applicable to establishing a missile and long range artillery defense system.

A numerical application of Bayesian optimization to the condition assessment of bridge hangers

  • Bridge hangers, such as those in suspension and cable-stayed bridges, suffer from cumulative fatigue damage caused by dynamic loads (e.g., cyclic traffic and wind loads) in their service condition. Thus, the identification of damage to hangers is important in preserving the service life of the bridge structure. This study develops a new method for condition assessment of bridge hangers. The tension force of the bridge and the damages in the element level can be identified using the Bayesian optimization method. To improve the number of observed data, the additional mass method is combined the Bayesian optimization method. Numerical studies are presented to verify the accuracy and efficiency of the proposed method. The influence of different acquisition functions, which include expected improvement (EI), probability-of-improvement (PI), lower confidence bound (LCB), and expected improvement per second (EIPC), on the identification of damage to the bridge hanger is studied. Results show that the errors identified by the EI acquisition function are smaller than those identified by the other acquisition functions. The identification of the damage to the bridge hanger with various types of boundary conditions and different levels of measurement noise are also studied. Results show that both the severity of the damage and the tension force can be identified via the proposed method, thereby verifying the robustness of the proposed method. Compared to the genetic algorithm (GA), particle swarm optimization (PSO), and nonlinear least-square method (NLS), the Bayesian optimization (BO) performs best in identifying the structural damage and tension force.

Genetic Variation of Parental Inbred Lines for Korean Waxy Corn Hybrid Varieties revealed by SSR markers (우리나라 찰옥수수 품종들의 교배친 자식계통들에 대한 유전적 변이성)

  • In maize, knowledge of genetic diversity and genetic relationships among elite inbred lines is an significant impact on the selection of parental lines for hybrid varieties. Genetic diversity and genetic relationships among 11 parental inbred lines of Korean waxy and normal corn varieties were analyzed using 50 SSR markers distributed over the whole genome. A total of 171 allele bands were detected with an average of 3.4 alleles per locus. Number of allele bands per locus ranged from two to six and gene diversity varied from 0.165 to 0.900 with an average of 0.596 depending on the SSR loci. The cluster tree recognized three major groups with 61.6% genetic similarity. Group I includes 7 inbred lines (KL103, HW1, HW4, HW6, HW7, HW8, HW9), with similarity coefficients of between 0.616 and 0.730. Group II includes 2 inbred lines (HF1, HF2), with similarity coefficients of 0.959. Group III includes 2 inbred lines (HW3, HW5), with similarity coefficients of 0.713. The present study indicates that the SSR markers chosen for this analysis are effective for the assessment of genetic diversity and genetic relationships among 11 parental inbred lines.
