Prospect and Situation of Quality Improvement in Oilseed rape (유채 품질 평가 현황과 전망)

  • Rapeseed(Brassica napus L.) is an important oil crop as a vegetable oil, concentrated feed and industrial materials. The name "canola" was registered in 1979 by the Western Canadian Oilseed Crushers Association to describe "double-low" varieties. Double low indicates that the processed oil contains less than 2% erucic-acid and the meal less than 3mg/g of glucosinolates. Today annual worldwide production of rapeseed is approximately 35 million tons on 24 million hectares. China accounts for 33% of the world production and the European Economic Community for nearly 32%. Canola ranks 3rd in production among the world's oilseed crops following soybeans, sunflowers, peanuts and cottonseed. The recent advances in genomics and in gene function studies has allowed us to understand the detailed genetic basis of many complex traits, such as flowering time, height, and disease resistance. The manipulation of seed oil content via transgene insertion has been one of the earliest successful applications of modern biotechnology in agriculture. For example, the first transgenic crop with a modified seed composition to be approved for unrestricted commercial cultivation in the US was a lauric oil, rape-seed, grown in 1995. There were also some significant early successes, mostly notably the achievement of 40% to 60% lauric acid content in rapeseed oil, which normally accumulates little or no lauric acid. The name "$\textrm{Laurical}^{TM}$" was registered in 1995 by Calgene Inc. Nevertheless, attempts to achieve high levels of other novel fatty acids in seed oils have met with much less success and there have been several reports that the presence of novel fatty acids in transgenic plants can sometimes lead to the induction of catabolic pathways which break down the novel fatty acid, i.e. the plant recognizes the "strange" fatty acid and, far from tolerating it, may even actively eliminate it from the seed oil. It is likely that, in the future, transgenic oil crops and newly domesticated oil crops will both be developed in order to provide the increased amount and diversity of oils which will be required for both edible and industrial use. It is important that we recognize that both approaches have both positive and negative points. It will be a combination of these two strategies that is most likely to supply the increasing demands for plant oils in the 21st century and beyond.ant oils in the 21st century and beyond.

  • Seedling vigor of introgressive hybrids between birdsfoot refoil 'MO-20"(Lotus corniculatus) used as the maternal parent and tetraploid(2n=4x=24)accession PI302921 narrowleaf trefoil(Lotus corniculatus L.), and parents were compared in terms of dry matter accumulation , net photosynthetic rate, dark respiration rate, and growth analysis. The hybrids showed better seedling vigor than PI302921 as they had lower dark respiration rate and higher relative growth rate (RGR) than PI302921. Hybrids grew faster than MO-20, but there was not significant difference between them. Net photosynthesis per unit area increased during three weeks after emergence without difference among the entries, then decreased sharply with leaf age and shading marking much differences among entries Dark respiration rate was very high just after emergence, after which it decreased rapidly until it reached a stable level at 1 week through 3 weeks of age, and then dropped again. In general, dark respiration, photosynthesis, and seedling growth of the hybrid was similar to those if MO-20. Although there are some reports about interspecific hybrids of Lotus species, it still remains as a new area for genetic improvement of seedling vigor of L. corniculatus.atus.

  • Many artificial intelligence (AI) techniques have been embedded into various engineering technologies to assist them in achieving different goals. The integration of modern technologies with energy consumption management system and occupant's comfort inside buildings results in the introduction of intelligent building concept. The major aim of this integration is to manage the energy consumption effectively and keeping the occupant satisfied with the internal environment of the building. The last few couple of years have seen many applications of AI techniques for optimizing the energy consumption with maximizing the user comfort in smart buildings but still there is much room for improvement in this area. In this paper, a hybrid of two AI algorithms called firefly algorithm (FA) and genetic algorithm (GA) has been used for user comfort maximization with minimum energy consumption inside smart building. A complete user friendly system with data from various sensors, user, processes, power control system and different actuators is developed in this work for reducing power consumption and increase the user comfort. The inputs of optimization algorithms are illumination, temperature and air quality sensors' data and the user set parameters whereas the outputs of the optimization algorithms are optimized parameters. These optimized parameters are the inputs of different fuzzy controllers which change the status of different actuators according to user satisfaction.

Identification of Effective Microorganisms Isolated from Fermented Stevia Extract and Their Antimicrobial Activity (스테비아 추출물 발효액에서 분리된 유효 미생물들의 동정 및 항미생물 활성)

  • Stevia rebaudiana Bertoni is a sweet herb of the Asteraceae family originally derived from South America. Twenty three bacterial strains and ten yeast strains were isolated from fermented Stevia extract and identified by general taxonomic methods and molecular genetic method. Isolated strains from fermented Stevia extract include ten species of bacteria which belong to five genus and one species of yeast. Based on 16S and 18S rDNA sequence analysis, phylogenetic trees were constructed. Antimicrobial activity of the isolated strains was examined against various bacteria and plant-pathogenic fungi. Among them, Lactobacillus paracasei SB13 showed strong antibacterial activity towards a wide range of bacteria. These results may be useful to develop environmentally friendly microbial agent for soil improvement.

  • 1. Objectives As a basic trial for identification of Sasang constitutional gene marker, we genotyped and analysed statistical relationships of STR(short tandem repeat) alleles and its distribution in each constitution. 2. Methods After obtaining basic constitutional data with questionnaire (QSCC II), decision of constitution was made by 3 different constitution specialists' diagnosis, and only the samples of specialists' agreement of each constitution by discussion were taken into this research. Using multiplex PCR kit, total 146 constitutional samples were amplified in 16 autosomal STR marker, genotyped, and analysed statistically. Among 16 markers, 15 were analysed in this study excluding the amelogenin marker is used for in gender identification. 3. Results and Conclusions It is difficult to determine the relationship between constitution and STR marker as the sample size is small, however, Penta D and vWA were shown to be related statistically with constitution. It has been know that STRs has no genetic informations, however there are some recent research results showing STRs as a regulatory element, relationship between microsatellite instability and repeat number and size, and post-transcriptional sigualing. STRs which is not known about its function currently, are proposed to have function and/or regulatory activities anyhow with Sasang constitution. It is believed that the results of this study can halp determine and deatify the markers related to Sasang Constitutional Medicien.

  • Most current BIM systems are focused on the visual information of work status in the design and construction stages. In a passive BIM system, 3D CAD tool can visualize the interference elements of design drawings, however, it cannot suggest a solution to solve the interference status. And 4D CAD tool in the construction stage can simulate the appearance of each activity by construction schedule, however, it cannot suggest an optimized schedule plan considering specified schedule condition of the project. Recently, many organizations need BIM solutions that can improve the work status beyond the level of simple visual information from BIM system. Active BIM system can provide the solutions to the project manager. This study suggests active BIM functions for the solutions and attempts to develop a 4D CAD engine to validate the usability of the functions.

  • To improve the starch fermentation ability of yeast, hybrids were introduced by protoplast fusion of S. cerevisiae and S. diastaticus. The protoplasts of parental auxotrophic cells were fused in the presence of 10 mM CaCl$_2$and 30% of polyethyleneglycol (M.W 4, 000). The frequencies of fusant formation varied depending upon the strains used and were 3.51$\times$10$^{-4}$ to 5.04$\times$10$^{-4}$ for the regenerated protoplasts. The strains capable of extensive starch hydrolysis produce only 10% to total fusants. The 4 strains were finally selected by the results of starch fermentation and genetic stability test. The DNA content and cell volume of the fusants were greater than those of the parental strains.

  • Axenic tissue culture of a marine red algae Porphyra yezoensis foma narawaensis was established for the vegetative propagation of tissues as a seed stock and for the development of a low salinity-tolerant cell line. Callus tissues have been induced from the vegetative area of blade away from the hold fast when grown on PES-agar medium. The brownish red fragile callus was maintained under fluorescent light of ca. 2000 lux with 12 : 12 hr L : D at $16^{\circ}C$. Amounts of carbohydrate and protein was determined against the weight of callus. Optimum temperature of the callus growth was $14^{\circ}C\~18^{\circ}C$. Optimun concentration of sodium chloride was $2.0\%$ for the callus growth in PES-agar medium.

  • The World Wide Web has become the largest virtual system that is almost universal in scope. In recent research, it has become effective to utilize idle hosts existing in the World Wide Web for running applications that require a substantial amount of computation. This novel computing paradigm has been referred to as the advent of global computing. In this paper, we implement and propose a mobile object-based global computing framework called Tiger, whose primary goal is to present novel object-oriented programming libraries that support distribution, dispatching, migration of objects and concurrency among computational activities. The programming libraries provide programmers with access, location and migration transparency for distributed and mobile objects. Tiger's second goal is to provide a system supporting requisites for a global computing environment - scalability, resource and location management. The Tiger system and the programming libraries provided allow a programmer to easily develop an objectoriented parallel and distributed application using globally extended computing resources. We also present the improvement in performance gained by conducting the experiment with highly intensive computations such as parallel fractal image processing and genetic-neuro-fuzzy algorithms.

Estrus Synchronization and Pregnancy Rate Using Ovsynch Method in Uganda Dairy Farms (우간다 낙농가에서 Ovsynch 방법에 의한 발정동기화 및 수태율)

  • The artificial insemination (AI) is one of the best assisted reproductive technologies for increasing reproductive capacity and facilitating the genetic improvement in farm animals. AI has been used in Uganda for over 60 years, but a small population of the total herd has been used. This study was conducted to investigate the efficacy of AI with estrus synchronization technique and to propose ways of improving the productivity of dairy farms through AI services in Uganda. In total, 78 cows from 11 dairy farms were selected for timed-AI. Synchronization was performed according to the ovsynch programs followed by AI using frozen semen from Korean Holstein (0.5 ml straws). Pregnancy rate was varying among farms (0-50%) and the overall pregnancy rate was 28.2%. Cows in luteal phase at the time of treatment was 40.0% whereas that in follicular phase was 20.8%. After treatment, cows that showed normal estrus signal were 45.5% (25/55). Abnormal estrus was categorized into pre-estrus (9.1%), cystic ovaries (21.8%), anestrus (18.2%) and delayed ovulation (5.5%), respectively. These results imply that an assured protocol for timed-AI should be developed to improve the productivity of dairy farms through AI services in Uganda.
