바위채송화(돌나물과)집단의 유전적 구조: 유전자 이동과 물리적 장벽에 관한 통찰 (Population genetic structure of Sedum polytrichoides (Crassulaceae): Insights into barriers to gene flow)

  • 한반도 남동부에 위치한 주왕산 국립공원과 그 인접산지는 식물 개체군의 유전자 이동에 대한 물리적 장벽의 영향을 시험하기 위한 훌륭한 모델 시스템이다. 우리는 식물종의 경우, 격리된 집단이 연속적인 분포를 보이는 집단보다 유전적인 분화 정도가 더 클 것으로 예측했다. 바위채송화 대부분의 집단은 4곳의 고립된 계곡에서 생육하며, 10개 집단에서 12 종류의 알로자임 유전좌위를 사용하여 유전적 다양성과 구조를 평가했다. 저자들은 이 연구와 기존 연구된 둥근잎꿩의비름(4곳 계곡에서 격리되어 생육)과 기린초(상대적으로 연속적으로 분포) 결과와 비교했다. 우리는 기린초 집단내 유전적 변이가 중간 수준임을 발견했다($H_e=0.112$). 바위채송화 집단간 분기 수준도 중간 수준($F_{ST}=0.250$)이었고 예상대로 둥근잎꿩의비름(0.261)과 유사했지만 기린초(0.165)보다 상당히 높았다. 분자분산분석(AMOVA) 결과 바위채송화와 둥근잎꿩의비름은 기린초(4%)보다 계곡간 변이(각각 19%) 비율이 높았다. STRUCTURE 프로그램 분석에 의하면 대부분의 이런 변이는 중간에 있는 두 계곡간의 유전적 조성 차이 때문이다. 저자들은 종간에 관찰된 분화 수준의 차이(즉, 기린초 대 바위채송화와 둥근잎꿩의비름)는 연구 지역 내의 그들의 분포 차이에 기인한다고 결론지었다.

Genetic Phylogeny among Three Species Red Seabream, Black Seabream and Rock Bream Based on Mitochondrial DNA Sequences

  • The Perciformes include approximately 40% of all bony fishes and are the largest order of vertebrates. This order includes some of the most economically relevant marine fishes, particularly the red seabream, black seabream and rock bream. A 409 bp fragment of the cytochrome b (cyt b) gene and 403 bp and 518 bp fragments of ribosomal RNA (12S and 16S rRNA, respectively) were sequenced from five populations of natural and cultured red seabreams, natural black seabream, and natural and cultured rock breams. The mitochondrial DNA sequences were utilized for the genetic identification and population structural analyses of these three species. Phylogenetic relationships of intra- and inter-species were elucidated using three types of molecular genetic markers from three species of the order Perciformes in Korea. We noted no significant differences in the intra-specific variation of the cyt b and rRNA genes in each population however, inter-specific divergences were greater than intra-specific variation. Inter-specific variation was induced more by transition than transversion type in the cyt b and rRNA genes. The cyt b gene and rRNA genes make it possible to determine the inter-species divergence. The rRNA genes have more conserved sequences than the cyt b gene. Therefore, these genes are expected to prove useful among species belonging to the different genera or families.

  • 한국산 쉬리, Coreuleuciscus splendidus의 종내 집단간 유전자 다양성을 알기 위해 6개 주요강(북한강, 남한강, 금강, 오십천, 낙동강, 섬진강)으로부터 채집된 개체를 대상으로 16S rRNA 유전자와 미트콘드리아 cytochrome b 유전자에 근거하여 비교 분석하였다. 미트콘드리아 cytochrome b 유전자의 657 bp 길이의 염기서열 분석결과, 6개 집단간에 차이는 98.2~99.9%로 나타났으며 지리적으로 격리된 집단간에 높은 유전적 다양성을 보였다. 16S rRNA 유전자는 697 bp의 염기서열을 얻었으며, 종내 변이는 큰 차이가 없이 거의 동일하였다. 16S rRNA 유전자의 6개 집단간에는 97.7%에서 99.7%의 높은 유사성을 보였다.

Genetic Variation of the Mitochondrial Cytochrome b Sequence in Korean Rana rugosa (Amphibia; Ranidae)

  • Nucleotide sequences of a 501 base-pair (bp) fragment in the mitochondrial cytochrome b (cyt b) gene were analyzed for 12 populations of Rana rugosa from Korea and Japan using a polymerase chain reaction (PCR) and direct silver sequencing. Two genetically distinct groups (type-A and type-B) were found in Korea. Type-A was found throughout most of South Korea and type-B was restricted to the mid-southeastern regions (Samchok, Yongdok, Chongsong and Pohang). But in the Tonghae population, both types were found. The level of mitochondrial DNA (mtDNA) sequence differences ranged from 0% to .0.8% among six populations of type-A, and 0 to 1.0% among 4 populations of type-B. However, sequence differences between type-A and type-B ranged from 5.4% to 6.6%, Using Kimura's two-parameter distance, the level of genetic sequence divergence between type-A and type-B was 6.7%. The Japanese R. rugosa was clustered very far from the Korean R. rugosa with 14.7%. In the neighbor-joining and UPGMA tree, all Korean samples were grouped, but subdivided into two types in 99% of the bootstrap iteration.

버들치속(잉어과) 어류의 유전적 변이 및 종분화 (Genic Vadadon and Speciation of Fishes of the Genus Moroco(Cyprinidae))

  • 한국 및 일본산 버들치속 어류의 유전적 변이, 종의 분류학적 위치 및 계통변화를 구명하기 위하여 전기영동법을 이용, 26개 유전자를 검출 분석하였다. 버들치속 어류의 평균 유전적 변이 정도는 타 어류군에 비하여 낮은 편이었다. 분류상 문제시 되어 오던 한국산 M. lagowskii와 M. oxycephalus 및 일본산 M. steindachneri와 M. jouyi는 각 종 특유의 genetic markers를 갖고 있어 각기 독립된 별종으로 확인되었다. M. lagowskii의 분포지역인 경포호집단은 유전적으로 뚜렷한 차이가 있고 형태적 특징으로 보아 M. percnurus로 추정된다. 각 종의 분화연대를 추정한 결과 이들은 선신세후기에서 홍종세 초기에 걸처 종분화가 되었고, 고 Amur 하 수계를 통하여 이주, 분포한 것으로 추측된다.

Population genetic analysis of Salurnis marginella (Hemiptera: Flatidae)

  • Salurnis marginella Guérin-Méneville, 1829 (Hemiptera: Flatidae) is an invasive species first reported in 2003 in Iksan, which is located in the mid-western region of South Korea, and subsequently found in the nearby regions in 2005. However, molecular-perspective reports on their invasive characteristics are not yet available. In this study, population genetic characteristics of Korean S. marginella were evaluated using the mitochondrial COI region and sequencing 124 individual samples collected in 11 Korean localities. A total of 12 haplotypes were identified with a maximum sequence divergence of 1.368% (9 bp). Haplotype diversity was relatively higher than that of other insect species invaded into Korea, providing 2-6 haplotypes per populations, indicating that introduction to Korea may have happened rather extensively and consistently. Nucleotide diversity (π) was the highest in Iksan but owing to the limited sample size (three individuals) from this locality, additional studies are required for drawing conclusive inference regarding the place of entry to Korea. Ulsan, the easternmost population in the present study, revealed nearly the lowest diversity estimates, such as the lowest H and the second-lowest π; a unique haplogroup with a higher frequency; and an independent genetic cluster, suggesting that the introduction of S. marginella to Ulsan was an independent event. Further collection in Korea and neighboring countries, including the original distributional range is necessary to elucidate the invasive dynamics of S. marginella

갈겨니(Zacco temminki)의 진화에 관한 연구 VII.갈겨니 2 Type의 Mitochondrial DNA변이 (A Study on the Speciadon of a Fresh Water Fish Zacco temminckL VII. Vadation of Mitochondrial DNA between 2 Types of Zacco temmincki)

  • 한국산 담수 어류인 갈겨니(Zacco temminki)는 형태적으로 동일하거나 전기영동법에 의한 s-Mdh에 2type(MM과 MS type)이 있음을 (1987)이 주장하였으며 2type간 유전적 차이 정도를 분석한 결과 자매종으로 밝힌바 있다. 본 연구에서는 상기 결과를 토대로 2type의 mtDNA를 4개 집단에서 추출하여 11가지 제한 요소로 처리한 다음 fragment양상을 비교 분석하였다. 갈겨니 mtDNA의 전 genome크기는 약 16.7Kb였으며 frabment homolgy(F)에서 MS type 간 F값은 0.464,MM type간 F값은 0.762로 높은 값을 나타냈다. 또한 MM과 MS 이형간 평균 F값은 0.312(0.258-0.345범위)로 동형간보다 낮은 유사성을 나타내고 있다. 집단 또는 type간 nucleotide sequence divergence(p)를 산출한 결과 MS type 간의 P=0.128에 비해 동형간의 P값은 0.045로 매우 낮은 mtDNA sequece 변이를 나타내었다. 그러나 이형간인 2type평균 P값은 0.195(0.177-0.226범위)로 차이를 나타냈다. 이와 같은 2type간의 차이는 isozyme연구 분석 결과와 일치하고 있어 갈겨니 2type간의 종분화를 확신시키는 새로운 자료가 되었다.

Intraspecific diversity and phylogeography of bony lip barb, Osteochilus vittatus, in Sundaland, as revealed by mitochondrial cytochrome oxidase I (mtCOI)

  • Life history characteristics, habitat landscape, and historical events are believed to have shaped the patterns of genetic variation in many taxa. The bony lip barb, Osteohilus vittatus, represent a potamodromous fish that complete all life cycle in freshwater and is widely distributed in Southeast Asia. It usually lives in small rivers and other freshwater habitats, and movement between habitats for either food or reproduction has been typical. These life history characteristics may promote gene flow, leading to less structured populations. However, many freshwater habitats are fragmented, which restricts gene flow. We investigate how this interplay has shaped patterns of genetic variation and phylogeographic structure within this species in the Sundaland, a biodiversity hotspot with a complex geological history, using mitochondrial cytochrome oxidase I (mtCOI) as a genetic marker. Forty-six mtCOI sequences of 506 bp long were collected from ten localities, eight geographically isolated and two connected. The sequences were used for population genetic and phylogeographic analyses. Our results showed a low genetic diversity within populations but high between populations. There was a deep phylogeographic structure among geographically isolated populations but a lack of such structure in the connected habitats. Among geographically isolated populations, sequence divergence was revealed, ranging from 1.8% between Java and Sumatra populations to 12.2% between Malaysia and Vietnam. An indication of structuring was also observed among localities that are geographically closer but without connectivity. We conclude that despite high dispersal capacity, the joint effects of historical events, long-term geographic isolation associated with sea level oscillation during the Pleistocene, and restricted gene flow related to lack of habitat connectivity have shaped the phylogeographic structure within the O. vittatus over the Sundaland.

Comparison of the Genetic Relationships and Osteological Aspects in Six Branchiostegid Fish Species (Perciformes)

  • We analyzed partial sequences of cytochrome b (cyt-b), a mitochondrial DNA (mtDNA) gene, to determine the genetic relationships between six horsehead fish species: Branchiostegus japonicus, Branchiostegus albus, Branchiostegus auratus, Branchiostegus argentatus, Branchiostegus wardi, and an unidentified Branchiostegus species. The specimens were collected in Korea, China, Japan, and Vietnam. We compared their molecular phylogenetic relationships inferred from mtDNA cyt-b sequences with an osteological analysis. The unidentified species, B. sp., was similar to B. albus in terms of the lack of triangular silver-white dot at the posterior region of eyes (vs. large one present in B. japonicus), but was also similar to B. japonicus in terms of the presence of a straight-shaped first hemal spine (vs. a curve-shaped hemal spine in B. albus). Analysis of the mtDNA cyt-b sequences indicated that the smallest estimated sequence divergence was between the B. japonicus and B. sp. (0.70-0.94%), whereas the largest difference was between B. auratus and B. argentatus (23.06-23.36%). Both the maximum parsimony and maximum likelihood trees showed that the B. sp. was closely clustered with B. japonicus, and that B. auratus was most distant from the other species. When comparing the osteological characters, UPGMA tree showed that the B. japonicus and B. sp. were the most closely clustered species, and B. auratus was the most distantly clustered fish relative to the other species. The shape of the nasal, otolith and first hemal spine was informative for distinguishing B. auratus from the other species. These osteological differences were consistent with the differences in mtDNA.

Insights into evolution and speciation in the red alga Bostrychia: 15 years of research

  • Studies of the red algal genus Bostrychia over the last 15 years have made it a model system for many evolutionary processes within red algal species. The combination of newly developed, or first employed methods, in red algal species studies has made Bostrychia a pioneer genus in intraspecific studies. Bostrychia was the first genus in which a mitochondrial marker was used for intraspecific red algal phylogeny, and the first for which a 3-genome phylogeny was undertaken. The genus was the first red alga used to genetically show maternal plastid and mitochondria inheritance, and also to show correlation between cryptic species (genetically divergent intraspecific lineages) and reproductive incompatibility. The chemotaxonomic use, and physiological function of osmolytes, has also been extensively studied in Bostrychia. Our continuous studies of Bostrychia also highlight important aspects in algal species studies. Our worldwide sampling, and resampling in certain areas, show that intensive sampling is needed to accurately assess the genetic diversity and therefore phylogeographic history of algal species, with increased sampling altering evolutionary hypotheses. Our studies have also shown that long-term morphological character stability (stasis) and character convergence can only be correctly assessed with wide geographic sampling of morphological species. While reproductive incompatibility of divergent lineages supports the biological species nature of these lineages, reproductive incompatibility is also seen between isolates with little genetic divergence. It seems that reproductive incompatibility may evolve quickly in red algae and the unique early stages of fertilization (e.g., gametes covered by walls, active movement of spermatium nuclei to the distant egg nucleus), also well investigated in Bostrychia,. may be key to our understanding of this process.
