The genetic variability of three poultry breeds namely Aseel, Miri and Nicobari taken from different geographical locations of India were evaluated using 15 microsatellite loci. No. of alleles varied from 3 to 9 in Aseel, 3 to 8 in Miri and 2 to 7 in Nicobari. Mean PIC values in Aseel, Miri and Nicobari breeds were 0.64, 0.66 and 0.63, respectively. Average unbiased heterozygosity and direct count heterozygosity were 0.65 and 0.59, 0.68 and 0.61, and 0.64 and 0.57 in Aseel, Miri and Nicobari breeds, respectively. High heterozygosity values revealed in this study are indicative of low level of inbreeding, large population size and no or low selection pressure for commercial trait in all three populations. The estimate of genetic distances using Nei's standard, Nei's minimum and Reynold's distance revealed Aseel and Nicobari to be more closely related than Miri breed of poultry.
The aim of this study was to investigate genetic development and genotype${\times}$environment interactions (GEI) in postweaning body weight of fattening bulls at the end of test period (WT-T) under various beef fattening environments. Data on a total of 24,247 fattening bulls obtained from the industrial farm, breeding farms and testing stations were used. Heritability estimates for WT-T in all environments were nearly similar. Significant genetic developments of sire, dam and progenies for WT-T were observed in all environments. However, many differences in annual genetic developments between the environments were significant. The genetic correlations for WT-T between industrial farm and breeding farms, industrial farm and testing stations and breeding farms and testing stations were respectively 0.004, 0.004 and 0.013. These low estimates of genetic correlations and significant differences in genetic developments among environments clearly show the existence of GEI for WT-T among various fattening environments. Results of this study indicate the need for environment-specific genetic evaluation and selection of beef bulls for commercial beef production.

Microsatellites (MS) are useful for quantifying genetic variation within and between populations and for describing the evolutionary relationships of closely related populations. The main objectives of this work were to estimate genetic parameters, measure genetic distances and reconstruct phylogenetic relationships between Australian Angora/Angora_Aus/ and Cashmere/Cashmere_Aus/ populations and three Mongolian Cashmere goat (Bayandelger/BD/, Zavkhan Buural/ZB/, and Gobi Gurvan Saikhan/GGS/) populations based on variation at fourteen MS loci. The level and pattern of observed and expected heterozygosity and polymorphic information content of the fourteen loci studied across the populations were quite similar and high. Except for SRCRSP07, all studied microsatellites were in Hardy-Weinberg Equilibrium (p<0.001). Moderate genetic variation (7.5%) was found between the five goat populations with 92.5% of total genetic variation attributable to diversity existing between the individuals within each population. The greatest Nei's genetic distances were found between the Angora and four Cashmere populations (0.201-0.276) and the lowest distances were between the Mongolian Cashmere goat populations (0.026-0.031). Compared with other Cashmere goat populations, the GGS (crossbred with Russian Don Goats) population had the smallest pairwise genetic distance from the Australian Angora population (0.192). According to a three-factorial correspondence analysis (CA), the three different Mongolian Cashmere populations could hardly be distinguished from each other.
India possesses a total buffalo population of 105 million out of which 26.1% inhabit Uttar Pradesh. The buffalo of Uttar Pradesh are described as nondescript or local buffaloes. Currently, there is no report about the genetic diversity, phylogenetic relationship and matrilineal genetic structure of these buffaloes. To determine the origin and genetic diversity of UP buffaloes, we sequenced and analysed the mitochondrial DNA D-loop sequences in 259 samples from entire Uttar Pradesh. One hundred nine haplotypes were identified in UP buffaloes that were defined by 96 polymorphic sites. We implemented neutrality tests to assess signatures of recent historical demographic events like Tajima's D test and Fu's Fs test. The phylogenetic studies revealed that there was no geographic differentiation and UP buffaloes had a single maternal lineage while buffaloes of Eastern UP were distinctive from rest of the UP buffaloes.
Cancer cell heterogeneity is a serious problem in the control of tumor progression because it can cause chemoresistance and metastasis. Heterogeneity can be generated by various mechanisms, including genetic evolution of cancer cells, cancer stem cells (CSCs), and niche heterogeneity. Because the genetic heterogeneity of CSCs has been poorly characterized, the genetic mutation status of CSCs was examined using Exome-Seq and RNA-Seq data of liver cancer. Here we show that different surface markers for liver cancer stem cells (LCSCs) showed a unique propensity for genetic mutations. Cluster of differentiation 133 (CD133)-positive cells showed frequent mutations in the IRF2, BAP1, and ERBB3 genes. However, leucine-rich repeat-containing G protein-coupled receptor 5-positive cells showed frequent mutations in the CTNNB1, RELN, and ROBO1 genes. In addition, some genetic mutations were frequently observed irrespective of the surface markers for LCSCs. BAP1 mutations was frequently observed in CD133-, CD24-, CD13-, CD90-, epithelial cell adhesion molecule-, or keratin 19-positive LCSCs. ASXL2, ERBB3, IRF2, TLX3, CPS1, and NFATC2 mutations were observed in more than three types of LCSCs, suggesting that common mechanisms for the development of these LCSCs. The present study provides genetic heterogeneity depending on the surface markers for LCSCs. The genetic heterogeneity of LCSCs should be considered in the development of LCSC-targeting therapeutics.
The genetic relationship of gestation length (GL) with birth and weaning weight (BW, WW) was investigated using data collected from the Hanwoo Experiment Station, National Institute of Animal Science, RDA, Republic of Korea. Analytical mixed models including birth year‐season, sex of calf, linear and quadratic covariates of age of dam (days) and linear covariate of age at weaning (days) as fixed effects were used. Corresponding restricted maximum likelihood (REML) and Bayesian estimates of variance components and heritability were obtained with two models; Model 1 included only direct genetic effect and Model 2 included direct genetic, maternal genetic and permanent environmental effect. All the genetic parameter estimates from REML were corresponding to the Bayesian estimates. Direct heritability estimates for GL, BW, and WW were 0.48, 0.33 and 0.25 by Model 1. From Model 2, direct and maternal heritability estimates were 0.38 and 0.03 for GL, 0.14 and 0.05 for BW, and 0.08 and 0.05 for WW. Genetic correlation estimates between direct and maternal effects were 0.05 for GL, 0.59 for BW, and 0.52 for WW. Estimates of direct genetic correlation between GL and BW (WW) were 0.44 (0.21). Positive genetic correlation of GL with BW and WW imply that selection for greater BW or WW would lead to prolonged gestation length.
Four Korean native cattle (KNC) breeds-Hanwoo, Chikso, Heugu, and Jeju black-are entered in the Domestic Animal Diversity Information System of the United Nations Food and Agriculture Organization (FAO). The objective of this study was to assess the genetic diversity, phylogenetic relationships and population structure of these KNC breeds (n = 120) and exotic breeds (Holstein and Charolais, n = 56). Thirty microsatellite loci recommended by the International Society for Animal Genetics/FAO were genotyped. These genotypes were used to determine the allele frequencies, allelic richness, heterozygosity and polymorphism information content per locus and breed. Genetic diversity was lower in Heugu and Jeju black breeds. Phylogenetic analysis, Factorial Correspondence Analysis and genetic clustering grouped each breed in its own cluster, which supported the genetic uniqueness of the KNC breeds. These results will be useful for conservation and management of KNC breeds as animal genetic resources.
Objective: Tibetan pigs, an excellent species unique to China, face serious threats, which in turn affects the development and utilization of the outstanding advantages of plateau hypoxia adaptability and reduces their genetic diversity. Therefore, a discussion of measures to conserve this genetic resource is necessary. The method, based on genetic diversity, genetic divergence and total genetic contribution rate of population, reflects the priority conservation order and varies depending on the three different purposes of conservation. Methods: We analyzed mitochondrial DNA control region (D-loop) variation in 1,201 individuals from nine Tibetan pig populations across five provinces and downloaded 564 mtDNA D-loop sequences from three indigenous pig breeds in Qinghai, Sichuan, and Yunnan Provinces distributed near the Tibetan pigs. Results: We analyzed three different aspects: Changdu Tibetan pigs have the highest genetic diversity, and from the perspective of genetic diversity, the priority conservation is Changdu Tibetan pigs. Hezuo Tibetan pigs have the highest genetic contribution, so the priority conservation is Hezuo Tibetan pigs in the genetic contribution aspect. Rkaze Tibetan pigs were severely affected by indigenous pig breeds, so if considering from the perspective of introgression, the priority conservation is Rkaze Tibetan pigs. Conclusion: This study evaluated genetic diversity and comprehensively assessed conservation priority from three different aspects in nine Tibetan pig populations.
