Expression of neurotransmitter receptors in oral keratinocytes and their response to agonists

  • This study aimed to investigate whether neurotransmitter receptors in the nervous system were also expressed in oral keratinocytes. Expressions of various neurotransmitter receptor genes in immortalized mouse oral keratinocyte (IMOK) cells were examined by reverse transcriptase polymerase chain reaction. IMOK cells expressed calcitonin gene-related peptide (CGRP) receptor subunit genes Ramp1 and Ramp3 and glutamate receptor subunit genes Grina, Gria3, Grin1, Grin2a, and Grin2d. Moreover, IMOK cells expressed Adrb2 and Chrna5 that encode beta 2 adrenergic receptor and cholinergic receptor nicotinic alpha 5 for sympathetic and parasympathetic neurotransmitters, respectively. The expression of Bdkrb1 and Ptger4, which encode receptors for bradykinin and prostaglandin E2 involved in inflammatory responses, was also observed at low levels. Expressions of Ramp1 and Grina in the mouse gingival epithelium were also confirmed by immunohistochemistry. When the function of neurotransmitter receptors expressed on IMOK cells was tested by intracellular calcium response, CGRP, glutamate, and cholinergic receptors did not respond to their agonists, but the bradykinin receptor responded to bradykinin. Collectively, oral keratinocytes express several neurotransmitter receptors, suggesting the potential regulation of oral epithelial homeostasis by the nervous system.

Expressions of Gonadotropin Subunit Genes in the Reproductively Inactive Golden Hamsters

  • Photoperiod has well been established to regulate testicular activities in golden hamsters. These animals breed actively around summer but become infertile in winter. In males, testicles are full of multistep germ cells including spermatozoa in summer. But in winter only fundamental cells consisting of the testicles are detected. The testicular degeneration is accompanied by the reduced levels of blood gonadotropins and testosterone. In this study, the expressions of gonadotropin subunit genes were investigated in the reproductive active and inactive testicles. And parts of sequences of the gonadotropin subunits were identified and compared with those of other rodents. As results, common gonadotropin alpha (CGa), follicle-stimulating hormone (FSH) β, and luteinizing hormone (LH) β genes were equivalently detected in pituitaries of both sexually active and inactive animals. In considering low concentrations of gonadotropin hormones determined in pituitary, the present findings imply that the processes involved in translation and/or formation of functional hormones could be impeded in the sexually inactive hamsters. All the nucleotide sequences of gonadotropin subunits identified in this study were same as those reported previously except for one base in CGa. An unsure amino acid deduced from the CGa sequence was confirmed from mRNA sequencing. The outcomes mentioned above suggest that animals with regressed testes prepare for the sexually active period forthcoming in the future.

Biological effect of recombinant IL-1$\beta$ on the expression of antiviral genes in the gill of rainbow trout, Oncorhynchus mykiss

  • We have investigated the biological effects of recombinant IL-1$\beta$ (rIL-1$\beta$) on the expression of antiviral genes such as Myxovirus-3 (MX-3) and Interferon regulating factor-1 (IRF-1), which are related to type I interferon. When ten micrograms of rIL-1$\beta$ were treated, we observed the stimulatory effect on the expression of these antiviral genes. Interestingly, at the early stage of stimulation, these genes were down-regulated and then up-regulated by the results obtained that the expressions of these genes were decreased at day 1 post-injection and gradually increased at day 3 post-injection. Thus, the stimulatory effect of rIL-1$\beta$ on the expression of MX-3 and IRF-3 gene might be an indirect stimulatory effect because significant up-regulation was delayed until day 3 post-injection.

Anti-obese related pharmacological effects of standard potato protein extracts on the 45%Kcal high fat diet supplied mice

  • Objectives : In present study, therefore, possible beneficial pharmacological activities of standard potato protein extracts (SPE) were observed on the mild diabetic obese mice. Methods : After end of 12 weeks of continuous oral administrations of three different dosages of SPE 400, 200 and 100 mg/kg, or metformin 250 mg/kg, analyzed the hepatoprotective, hypolipidemic, hypoglycemic, nephroprotective and anti-obesity effects, separately. In addition, liver antioxidant defense systems were additionally measured with lipid metabolism-related genes expressions and hepatic glucose-regulating enzyme activities for action mechanism. Results : All of diabetes and related complications including obesity were significantly inhibited by treatment of SPE 400, 200 and 100 mg/kg, dose-dependently, and they also dramatically normalized the hepatic lipid peroxidation and depletion of liver endogenous antioxidant defense system, the changes of the hepatic glucose-regulating enzyme activities, also changes of the lipid metabolism-related genes expressions including hepatic $AMPK{\alpha}1$ and $AMPK{\alpha}2$ mRNA expressions, dose-dependently. Especially, SPE 200 mg/kg constantly showed favorable inhibitory activities against type II diabetes and related complications as comparable to those of metformin 250 mg/kg in HFD mice, respectively. Conclusions : The present work demonstrated that SPE 400, 200 and 100 mg/kg showed favorable anti-diabetic and related complications including obesity refinement activities in HFD mice, through AMPK upregulation mediated hepatic glucose enzyme activity and lipid metabolism-related genes expression, antioxidant defense system and pancreatic lipid digestion enzyme modulatory activities.

발효대두추출물의 인간 유방암 MCF7 세포에서 전이 관련 ERBB2와 PLAU 발현 억제 효과 (Suppression of metastasis-related ERBB2 and PLAU expressions in human breast cancer MCF 7 cells by fermented soybean extract)

  • 발효대두 청국장에는 대두단백질이 발효에 의해 분해 형성된 다양한 펩타이드류가 들어 있다. 청국장 추출물이 처리된 유방암세포의 microarray data와 잘 알려진 유방암 전이 마커를 합쳐서 새로운 연결망이 제조되었으며 이를 이용해 전이 마커와 발현 차이가 있는 단백질 사이의 상호작용을 체크하였다. 연결망 분석을 통해 PLAU (plasminogen activator, urokinase, uPA)와 ERBB2 (epidermal growth factor receptor 2)를 실제 전이 가능성을 보여주는 유전자로 선택하였다. MCF7 암세포를 청국장추출물로 처리하고 PLAU와 ERBB2 발현정도를 측정하였다. 청국장 추출물은 PLAU와 ERBB2 발현을 상당히 억제하였다. 청국장 추출물을 처리한 암세포에서 염증 마커인 NO의 생산이 감소하였다. 인간 유방암세포에서 PLAU와 ERBB2 발현을 특이적으로 감소시키는 펩타이드를 찾아내는 것은 흥미로운 일일 것이다.

Expression of Auxin Response Genes SlIAA1 and SlIAA9 in Solanum lycopersicum During Interaction with Acinetobacter guillouiae SW5

  • Indole-3-acetic acid (IAA) production is a typical mechanism of plant growth promotion by some rhizobacteria. However, a functional genomic study is necessary to unravel the function and mechanism of IAA signaling during rhizobacteria-plant interactions. In this study, the expression of SlIAA1 and SlIAA9 among the auxin response genes in tomato was examined during the interaction between IAA-producing Acinetobacter guillouiae SW5 and tomato plants. When 3-day grown tomato seedlings were treated for 30 min with 10~100 µM of IAA produced by bacteria from tryptophan, the relative mRNA levels of SlIAA1 and SlIAA9 increased significantly compared with those of the control, demonstrating that IAA produced by this bacterium can induce the expressions of both genes. Inoculation of live A. guillouiae SW5 to tomato seedlings also increased the expressions of SlIAA1 and SlIAA9, with more mRNA produced at higher bacterial density. In contrast, treatment of tomato seedlings with dead A. guillouiae SW5 did not significantly affect the expression of SlIAA1and SlIAA9. When 3-day bacterial culture in tomato root exudates was administered to tomato seedlings, the relative mRNA level of SlIAA1 increased. This result indicated that the plant may take up IAA produced by bacteria in plant root exudates, which may increase the expression of the auxin response genes, with resulting promotion of plant growth.

감마선 변이체 스프레이 국화 'ARTI-purple'의 화색 관련 유전자 발현 분석 (Expression Analysis of Flower Color Related Genes in Spray-type 'ARTI-purple' Developed by Gamma-ray Mutagenesis)

  • Anthocyanins are major plant pigment and produced through phenylpropanoid pathway. In this study, anthocyanin biosynthesis mechanisms of chrysanthemum flowers were studied using 'Argus' and flower color mutant 'ARTI-purple' which were induced by 40 Gy gamma irradiation ($Co^{60}$). And, three chrysanthemums, 'Ford', 'Yeonja' and 'Orando' were additionally used as the check varieties to understand the relationship between flower color and expression patterns of genes. The expression patterns of the anthocyanin biosynthetic genes were matched with the flower color of the check varieties. High anthocyanin concentration of 'Orando' showed the high expression of anthocyanin biosynthetic genes. In the white flower of 'Ford', expressions of CHI, DFR and ANS were not identified. Despite different flower color, 'Argus' and 'ARTI-purple' showed different expression patterns compared with the check varieties. From the dot blot analysis, we screened the seven genes showing the different expressions between 'Argus' and 'ARTI-purple'.

Conjugated Linoleic Acid 이성체가 3T3-L1 지방전구세포 분화중 세포증식, 세포사멸 및 Ucp 유전자 발현에 미치는 영향 (The Effect of Conjugated Linoleic Acid Isomers on the Cell Proliferation, Apotosis and Expressions of Uncoupling Protein (Ucp) Genes during Differentiation of 3T3-L1 Preadipocytes)

  • It has been reported that CLA decreases fat deposition in vivo and in vitro experiments. Among CLA isomers, c9t11 and t10c12 have been shown to exert active biological activities. For example, t10c12 reduces body weight and increases lean body mass, whereas, c9t11 has little effect on body fattness. However, the underlying molecular mechanism for the anti-obesity action of CLA isomers are not well understood. The purpose of this study was to examine the effects of t10c12 and c9t11 on lipid accumulation, cell proliferation, cell death and the expression levels of Ucp genes which are proposed as targets for anti-obesity in 3T3-L1 preadipocytes. Isomers of CLA at 50$\mu$M were added into preadipocyte differentiation medium for 3, 6 and 9days. Control cells received only the vehicle in the differentiation medium. Cytochemical analyses for lipid accumulation, cell proliferation and apotosis were carried out to compare lipidogenesis and cellular activity. RT-PCR analysis of GAPDH, Ucp 2,3 and 4 were also performed to find any modulatory effects of CLA isomers on the metabolic genes. Lipid accumulation indicated by Oil Red-O staining was inhibited in CLA isomers as compared to the control. T10c12 isomer showed less lipidogenesis than c9t11 did. A decrease occurred in CLA isomers as shown by BrdU incorporation. Apotosis has occured at higher level in t10c12 when compared to that of t9c11. Ucp 2, 3 and 4 genes were also upregulated in CLA isomers. T10c12 showed higher level of Ucp gene expressions than the c9t11 did. The biological activities of CLA isomers were also found to be different during differentiation of 3T3-L1 preadipocytes, suggesting that different isomers may be active in certain stage of lipidogenesis. The results indicate that both c9t11 and t10c12 CLA isomers decrease lipidogenesis, inhibit cell proliferation, increase cell death and upregulate in Ucp gene expressions during 3T3-L1 preadipocyte differentiation. T10c12 isomer was more effective than c9t11 in overall anti-obesity activity.

  • To enhance our understanding of toxicity mediated through the pathway by which TCDD stimulates gene expression, we have investigated genes whose expressions are changed after treatment with TCDD and/or MNNG in human Chang liver cell. First, we treated with MNNG and TCDD for two weeks to transform human Chang liver cell. We obtained cell looks like to be transformed and compared the differential gene expression by using cDNA chip (Macrogen) which carrys genes related with signal transduction pathways, oncogenes and tumor suppressor genes, etc. We found that TCDD up- or down-regulated 203 and 111 genes including oncogenes and tumor suppressor genes in human Chang liver cell two fold or more, respectively. Second, we compared the differential gene expression after treatment with TCDD only by using cDNA chip (Superarray) which carrys genes related with cell cycle regulations, and found that TCDD up regulated genes related with cell proliferation as well as cell growth inhibition in human Chang liver cell two fold or more, respectively. These results suggest that toxicity induced by TCDD may reflect sustained alterations in the expression of many genes and that the changes reflect both direct and indirect effects of TCDD.

Possibility of the Use of Public Microarray Database for Identifying Significant Genes Associated with Oral Squamous Cell Carcinoma

  • There are lots of studies attempting to identify the expression changes in oral squamous cell carcinoma. Most studies include insufficient samples to apply statistical methods for detecting significant gene sets. This study combined two small microarray datasets from a public database and identified significant genes associated with the progress of oral squamous cell carcinoma. There were different expression scales between the two datasets, even though these datasets were generated under the same platforms - Affymetrix U133A gene chips. We discretized gene expressions of the two datasets by adjusting the differences between the datasets for detecting the more reliable information. From the combination of the two datasets, we detected 51 significant genes that were upregulated in oral squamous cell carcinoma. Most of them were published in previous studies as cancer-related genes. From these selected genes, significant genetic pathways associated with expression changes were identified. By combining several datasets from the public database, sufficient samples can be obtained for detecting reliable information. Most of the selected genes were known as cancer-related genes, including oral squamous cell carcinoma. Several unknown genes can be biologically evaluated in further studies.
