Current Insights into Research on Rice stripe virus

  • Rice stripe virus (RSV) is one of the most destructive viruses of rice, and greatly reduces rice production in China, Japan, and Korea, where mostly japonica cultivars of rice are grown. RSV is transmitted by the small brown plant-hopper (SBPH) in a persistent and circulative-propagative manner. Several methods have been developed for detection of RSV, which is composed of four single-stranded RNAs that encode seven proteins. Genome sequence data and comparative phylogenetic analysis have been used to identify the origin and diversity of RSV isolates. Several rice varieties resistant to RSV have been selected and QTL analysis and fine mapping have been intensively performed to map RSV resistance loci or genes. RSV genes have been used to generate several genetically modified transgenic rice plants with RSV resistance. Recently, genome-wide transcriptome analyses and deep sequencing have been used to identify mRNAs and small RNAs involved in RSV infection; several rice host factors that interact with RSV proteins have also been identified. In this article, we review the current statues of RSV research and propose integrated approaches for the study of interactions among RSV, rice, and the SBPH.

  • According to the accident statistics published by the National Police Agency in 2007, the number of commercial vehicle accidents explains 3.5 percent of the total number of traffic accidents of the year. Compared to other types of vehicles commercial vehicles may provide more serious damages to both driver himself and passengers. Thus, they generate more serious social and economic problems. There have been various forms of systems such as a digital speedometer or a black box to meet the social requirement for reducing traffic accidents and improving safe driving. However, since the current systems are based on the data often accidents happened, there are lots of limitations to control drivers in real-time. Also, the current speedometers provide drivers with only speeds of vehicles and RPM information regardless of actual dangerous drive behaviors. Therefor, they lack of the effectiveness in terms of safety. In this research, real-time information systems for improving driver safety based on automatic risky driving behaviors, and thresholds to determine risky driving patterns were studied.

Two-Cell Spheroid Angiogenesis Assay System Using Both Endothelial Colony Forming Cells and Mesenchymal Stem Cells

  • Most angiogenesis assays are performed using endothelial cells. However, blood vessels are composed of two cell types: endothelial cells and pericytes. Thus, co-culture of two vascular cells should be employed to evaluate angiogenic properties. Here, we developed an in vitro 3-dimensional angiogenesis assay system using spheroids formed by two human vascular precursors: endothelial colony forming cells (ECFCs) and mesenchymal stem cells (MSCs). ECFCs, MSCs, or ECFCs+MSCs were cultured to form spheroids. Sprout formation from each spheroid was observed for 24 h by real-time cell recorder. Sprout number and length were higher in ECFC+MSC spheroids than ECFC-only spheroids. No sprouts were observed in MSC-only spheroids. Sprout formation by ECFC spheroids was increased by treatment with vascular endothelial growth factor (VEGF) or combination of VEGF and fibroblast growth factor-2 (FGF-2). Interestingly, there was no further increase in sprout formation by ECFC+MSC spheroids in response to VEGF or VEGF+FGF-2, suggesting that MSCs stimulate sprout formation by ECFCs. Immuno-fluorescent labeling technique revealed that MSCs surrounded ECFC-mediated sprout structures. We tested vatalanib, VEGF inhibitor, using ECFC and ECFC+MSC spheroids. Vatalanib significantly inhibited sprout formation in both spheroids. Of note, the $IC_{50}$ of vatalanib in ECFC+MSC spheroids at 24 h was $4.0{\pm}0.40{\mu}M$, which are more correlated with the data of previous animal studies when compared with ECFC spheroids ($0.2{\pm}0.03{\mu}M$). These results suggest that ECFC+MSC spheroids generate physiologically relevant sprout structures composed of two types of vascular cells, and will be an effective pre-clinical in vitro assay model to evaluate pro- or anti-angiogenic property.

A Study on the Determination for Stochastic Reservoir Capacity (추계학적 저수용량 결정에 관한 연구)

  • The generated sequences of monthly flows were analyzed based on the range concept. With the optimum operation rule of the reservoirs as the one which maximizes the wateruse downstream the waterrelease from the reservoir was determined and with \ulcorner consideration to the mean inflows and the range of monthly flows the required reservoirs capacity was stochastically determind. It is suggested that the result obtained in this study would be applied to approximately estimate, in the stage of preliminary design, the required capacity of a reservoir in question with the limited information such as the mean monthly inflow and the period of reservoir operation. For the determination of a reservoir capacity Rippl's mass-curve method has been long used with the past river flow data assuming the same flow records will be repeated in the future. This study aims to find out a better method for determining the reservoir capacity by employing the analytical theory based on the stochastic process. For the present study the synthetic generation methods of Thomas-Fiering type was used to synthetically generate 50 years of monthly river inflows to three single-purpose reservoirs and three multi-purpose reservoirs.

Ensemble Daily Streamflow Forecast Using Two-step Daily Precipitation Interpolation (일강우 내삽을 이용한 일유량 시뮬레이션 및 앙상블 유량 발생)

  • Input uncertainty is one of the major sources of uncertainty in hydrologic modeling. In this paper, first, three alternate rainfall inputs generated by different interpolation schemes were used to see the impact on a distributed watershed model. Later, the residuals of precipitation interpolations were tested as a source of ensemble streamflow generation in two river basins in the U.S. Using the Monte Carlo parameter search, the relationship between input and parameter uncertainty was also categorized to see sensitivity of the parameters to input differences. This analysis is useful not only to find the parameters that need more attention but also to transfer parameters calibrated for station measurement to the simulation using different inputs such as downscaled data from weather generator outputs. Input ensembles that preserves local statistical characteristics are used to generate streamflow ensembles hindcast, and showed that the ensemble sets are capturing the observed steamflow properly. This procedure is especially important to consider input uncertainties in the simulation of streamflow forecast.

Valuing Recreational Benefits of Dam Lakes Using a Choice Experiment Approah (선택실험법을 이용한 댐호수의 특성별 휴양가치 분석)

  • This study conducts a choice experiment to estimate the recreational values of major dam reservoirs in Korea. A discrete-choice econometric model is estimated with the data of experimented choices. Not only the recreation value of each dam lake but also the values of the characteristics of the lakes are estimated. It is shown that the value function of the amount of reserved water is not monotonically increasing. Water quality is the most important characteristic that affects the recreation value. Various other characteristics of the dams such as the availability of education facility, restaurants, hotels and parking lots also generate substantial amounts of recreational benefits.

Salinity of Representative Korean Foods High in Sodium from Home Meals, Foodservices, and Restaurants (가정식, 급식, 외식 고나트륨 한식 대표 음식의 염도 분석)

  • Objectives: This study was conducted to analyze the salinity of representative Korean foods high in sodium to generate data for use as a fundamental resource for setting salinity standards in foods. Methods: A total of 480 foods from 16 representative Korean foods high in sodium were collected from 10 households, 10 industry foodservice establishments, and 10 Korean restaurants in four regions (Capital area, Chungcheong Province, Gyeongsang Province, and Jeolla Province) and analyzed for salinity. Results: Among the foods, stir-fried anchovies (4.07~4.45%) showed the highest salinity, followed by pickled onion (1.86~2.62%), cabbage kimchi (1.83~2.2%), braised burdock and lotus root (1.79~2.17%), and sliced radish kimchi (1.78~1.89%) (p<0.001). The salinity of kimchi from home meals (2.2%) was significantly higher than that of foodservice (1.83%) and restaurant (1.93%) kimchi (p<0.05). Salinity in each group of food was highest in kimchi (1.83~2.04%), followed by braised dishes (1.54~1.78%), steamed dishes (1.0~1.22%), stir-fried dishes (1.02~1.18%), and soup or stew (0.74~1.02%) (p<0.001). The salinity of soup and stew from restaurants (1.02%) was significantly higher than that of home meal (0.84%) and foodservice (0.74%) soup and stew. Conclusions: Determination of the salinity of representative Korean foods known to be high in sodium by eating place is expected to be useful to establishing guidelines for reduction of salinity.

Genetic Distances and Variations of Three Clupeid Species Determined by PCR Technique

  • In this study, seven oligonucleotides primers were shown to generate the shared loci, specific loci, unique shared loci to each species and shared loci by the three species which could be obviously calculated. Euclidean genetic distances within- and between-species were also calculated by complete linkage method with the sustenance of the hierarchical dendrogram program Systat version 13. The genomic DNA isolated from herring (Clupea pallasii), Korean anchovy (Coilia nasus) and large-eyed herring (Harengula zunashi), respectively, in the Yellow Sea, were amplified several times by PCR reaction. The hierarchical dendrogram shows three chief branches: cluster 1 (PALLASII 01, 02, 03, 04, 06 and 07), cluster 2 (NASUS 08, 09, 10, 11, 12, 13 and 14), and cluster 3 (ZUNASHI 15, 16, 17, 18, 19, 20, 21 and PALLASII 05). In three clupeid species, the shortest genetic distance displaying significant molecular difference was between individual PALLASII no. 03 and PALLASII no. 02 (0.018). Individual no. 06 of PALLASII was most distantly related to NASUS no. 11 (genetic distance = 0.318). Individuals from herring (C. pallasii) species (0.920) exhibited higher bandsharing values than did individuals from Korean anchovy (C. nasus) species (0.872) (P<0.05). As a result, this PCR analysis generated on the genetic data displayed that the herring (C. pallasii) species was widely separated from Korean anchovy (C. nasus) species. Reversely, individuals of Korean anchovy (C. nasus) species were a little closely related to those of large-eyed herring (H. zunashi) species.

A Case Study on a Large Scale Borehole Test Blasting to Generate Man-made Earthquake (인공지진 발생을 위한 대규모 시추공 시험발파 사례연구)

  • In the process of identifying the earth's crust structures to accurately locate the seismic epicenter, man-made earthquakes need to be generated. Such a large-scale ground vibration can be generated by a deep borehole blasting, but it can also accompany some environmental impacts on the surroundings. In this respect, a borehole test blasting was carried out to determine the maximum charge weight that could be used without affecting the various structures around the blast site. Total 400kg of gelatine-type dynamites was used in the test blast. As a result, a prediction equation for ground vibrations was derived from the measured data. With the allowable level of 3.0 mm/s for residential structures, the maximum charge weight was determined to be 677kg if military structures near the site were considered. But if the military structures were not considered, it was found that up to 2100kg of explosives could be used without affecting old houses in the nearby village.

Improving the Performance of AODV(-PGB) based on Position-based Routing Repair Algorithm in VANET

  • Vehicle ad hoc networks (VANET) are one of the most important technologies to provide various ITS services. While VANET requires rapid and reliable transmission, packet transmission in VANET is unstable because of high mobility. Many routing protocols have been proposed and assessed to improve the efficiency of VANET. However, topology-based routing protocols generate heavy overhead and long delay, and position-based routing protocols have frequent packet loss due to inaccurate node position. In this paper, we propose a position-based routing repair algorithm to improve the efficiency of VANET. This algorithm is proposed based on the premise that AODV (-PGB) can be used effectively in VANET, if the discovery, maintenance and repair mechanism of AODV is optimized for the features of VANET. The main focus of this algorithm is that the relay node can determine whether its alternative node exits and judge whether the routing path is disconnected. If the relay node is about to swerve from the routing path in a multi-hop network, the node recognizes the possibility of path loss based on a defined critical domain. The node then transmits a handover packet to the next hop node, alternative nodes and previous node. The next node repairs the alternative path before path loss occurs to maintain connectivity and provide seamless service. We simulated protocols using both the ideal traffic model and the realistic traffic model to assess the proposed algorithm. The result shows that the protocols that include the proposed algorithm have fewer path losses, lower overhead, shorter delay and higher data throughput compared with other protocols in VANET.
