유전자변형 배추를 섭취한 마우스 장기에서의 Housekeeping Gene의 발현 분석 (Analysis of Housekeeping Gene Expression in Mice Administered to GM and non-GM Cabbage)

  • 유전자 변형 작물은 생산성 측면에서 많은 장점이 있지만 이를 섭취할 경우 잠재적인 위험 요소들에 의해 많은 문제가 대두대고 있다. 본 연구는 저항성유전자를 이입한 배추에서 Profillin, Tubulin-${\alpha}$ (Tub-${\alpha}1$), Heat-shock protein (Bchsp 17.6) and Ubiquitin conjugating enzyme (UBE)의 발현과 이를 30일간 섭취한 마우스에서 ${\beta}$-actin(${\beta}$-act), ${\beta}$-2-microglobulin (B2m), Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and ${\beta}$-glucuronidase (Gus)의 발현 정도를 RT-PCR을 통해 알아보았다. 실험 결과 저항성유전자를 이입한 배추와 그렇지 않은 배추의 유전자 발현 패턴은 큰 차이를 보이지 앓았으며, 이를 섭취한 마우스 장기에서도 발현에 따른 큰 차이는 나타나지 않았다.

Molecular Cloning, Bioinformatics Analysis and Expression Profiling of a Gene Encoding Vacuolar-type $H^+-ATP$ Synthetase (V-ATPase) c Subunit from Bombyx mori

  • As the genome of B.mori is available in GenBank and the EST database of B.mori is expanding, identification of novel genes of B.mori is conceivable by data-mining techniques. We used the in silico cloning method to get the vacuolar-type $H^+-ATP$ synthetase (V-ATPase) c subunit (16 kDa proteolipid subunit) gene of B.mori and analysed with bioinformatics tools. The result was confirmed by RT-PCR and sequencing. The V-ATPase c subunit cDNA contains a 468 bp ORF. The ORF encoded a 155-residue protein that showed extensive homology with V-ATPase c subunits from other 15 species and contained four membrane-spanning helices. Tissue expression pattern analysis revealed that V-ATPase c expressed strongly in Malpighian tubules, not in fat body. This gene has been registered in GenBank under the accession number EU082222.

  • Genome-scale expression data provides us with valuable insights about organisms, but the biological validation of in-silico analysis is difficult and often controversial. Here we present a new approach for integrating previously established knowledge with computational analysis. Based on the known biological evidences, IGAM (Integrated Gene Association Matrix) automatically estimates the relatedness between a pair of genes. We combined this association knowledge to the regulatory network modeling and fuzzy clustering in yeast 5. Cerevisiae. The result was found to be more effective for extracting biological meanings from in-silico inferences for gene expression data.

  • Objectives : he purpose of this study was to examine the genetic variations and changes of gene expression in the human constitutions. Methods : To analysis variations of individual gene expression, we had selected three groups of volunteers. In each group have a typical constitutional characteristics. By this rime we are analyzed their gene expression patterns by using DNA chip. Results : we can acquire a new information of standard for human constitutions. 1. The 21 genes under express and 3 genes over express in So-Yang constitution 2. The 18 genes under express and 18 genes over express in So-Eum constitution 3. The 16 genes under express and 2 genes over express in Tae-Eum constitution Conclusions : Constitution, QSCCII, Character, Genome, DNA chip.

Expression Profiles of Apoptosis Genes in Mammary Epithelial Cells

  • To investigate apoptosis in HC11 mammary epithelial cells, we compared the gene expression profiles of actively growing and serum-starved apoptotic cells using a mouse apoptosis gene array and $^{33}P$-labeled cDNA prepared from the RNA of the two cultures. Analysis of the arrays showed that expression of several genes such as clusterin, secreted frizzled related protein mRNA (sFRP-1), CREB-binding protein (CBP), and others was higher in the apoptotic cells whereas expression of certain genes including survivin, cell division cycle 2 homolog A (CDC2), and cyclin A was lower. These expression patterns were confirmed by RT-PCR and/or Northern analyses. We compared the expression of some of these genes in the mouse mammary gland under various physiological conditions. The expression levels of genes (clusterin, CBP, and M6P-R) up-regulated in apoptotic conditions were higher at involution than during lactation. On the other hand, genes (Pin, CDC2) downregulated in apoptotic conditions were relatively highly expressed in virgin and pregnant mice. We conclude that certain genes such as clusterin, sFRP-1, GAS1 and CBP are induced in apoptotic mammary epithelial cells, and others are repressed. Moreover, the apoptosis array is an efficient technique for comparing gene expression profiles in different states of the same cell type.

Introduction of Hog Cholera Virus Gene into Potato Plants by Agrobacterium-mediated Transformation and the Analysis of Its Expression

  • The HCV gene was expressed in potato plants under the control of the constitutive CaMV 355 promoter or tuber-specific patatin promoter. Solanum tuberosum plants carrying a plant expression vector harboring the encoding region of HCV gene were generated by Agrobacterium tumefaciens-mediated in vitro transformation methods. The presence of HCV gene in the plant genome was detected by PCR and DNA hybridization experiments. We obtained the 5 lines of transgenic potato with the pMBPHCV construct and 4 lines of transgenic potato with the pATHCV construct. The HCV transgenic stably integrated into the potato genome, as well as their transcription. HCV mRNA was identified in leaf and tuber tissues of transgenic plants by Northern blot analysis. The transgenic potato plants produced the expected transcript, and the corresponding HCV protein accumulated in individual transgenic plants.

조류 로타바이러스의 NSP4 유전자 염기서열분석 및 발현 (Nucleotide sequence analysis and expression of NSP4 gene of avian rotavirus)

  • The nonstructural protein 4 (NSP4) of rotavirus encoded by gene 10, plays an important role in rotavirus pathogenicity. In this study, NSP4 gene of avian rotavirus (AvRV-1, AvRV-2) was analyzed and expressed using baculovirus expression system. The sequence data indicated that the NSP4 gene of AvRV-1 and AvRV-2 were 727 bases in length, encoded one open reading frame of 169 amino acids beginning at base 41 and terminating at base 550, and had two glycosylation sites. Nucleotide sequences of NSP4 gene of AvRV-1 and AvRV-2 exhibited a high degree of homology ($88.1{\pm}7.6%$) with avian rotaviruses, namely Ty1, Ty3 and PO-13. Phylogenetic analysis showed that AvRV-1 and AvRV-2 belonged to genotype NSP4[E], which is widely found in group A avian rotaviruses. The baculovirus-expressed NSP4 migrated at 20-28 kDa and reacted with NSP4-specific antiserum by FA and Western blot. Furthermore, it was found to be a glycoprotein by using tunicamycin, which is a specific inhibitor of N-linked glycosylation.

Comparison of Invariant NKT Cells with Conventional T Cells by Using Gene Set Enrichment Analysis (GSEA)

  • Background: Invariant Natural killer T (iNKT) cells, a distinct subset of CD1d-restricted T cells with invariant $V{\alpha}{\beta}$ TCR, functionally bridge innate and adaptive immunity. While iNKT cells share features with conventional T cells in some functional aspects, they simultaneously produce large amount of Th1 and Th2 cytokines upon T-cell receptor (TCR) ligation. However, gene expression pattern in two types of cells has not been well characterized. Methods: we performed comparative microarray analyses of gene expression in murine iNKT cells and conventional $CD4^+CD25^-$ ${\gamma}{\delta}TCR^-$ T cells by using Gene Set Enrichment Analysis (GSEA) method. Results: Here, we describe profound differences in gene expression pattern between iNKT cells and conventional $CD4^+CD25^-$ ${\gamma}{\delta}TCR^-$ T cells. Conclusion: Our results provide new insights into the functional competence of iNKT cells and a better understanding of their various roles during immune responses.

Isolation and Characterization of Parvalbumin Beta Gene from Channel Catfish (Ictalurus punctatus)

  • Our previous studies of both microarray analysis in channel catfish muscle gene expression of 2 different ages and channel catfish muscle expressed sequence tag profiles demonstrated parvalbumin beta is one of the highly expressed muscle transcriptome. We have cloned and sequenced complementary DNA encoding the channel catfish parvalbumin which encode 109 amino acids. The deduced amino acid sequences of the catfish parvalbumin are highly conserved with those cloned from other teleosts. The availability of the catfish parvalbumin provides the opportunity of studying fish epitopes.

Expression of anoctamin 7 (ANO7) is associated with poor prognosis and mucin 2 (MUC2) in colon adenocarcinoma: a study based on TCGA data

  • Colon adenocarcinoma (COAD) is the predominant type of colorectal cancer. Early diagnosis and treatment can significantly improve the prognosis of COAD patients. Anoctamin 7 (ANO7), an anion channel protein, has been implicated in prostate cancer and other types of cancer. In this study, we analyzed the expression of ANO7 and its correlation with clinicopathological characteristics among COAD patients using the Gene Expression Profiling Interactive Analysis 2 (GEPIA2) and the University of Alabama at Birmingham CANcer (UALCAN) databases. The GEPIA2, Kaplan-Meier plotter, and the Survival Genie platform were employed for survival analysis. The co-expression network and potential function of ANO7 in COAD were analyzed using GeneFriends, the Database for Annotation, Visualization and Integrated Discovery (DAVID), GeneMANIA, and Pathway Studio. Our data analysis revealed a significant reduction in ANO7 expression levels within COAD tissues compared to normal tissues. Additionally, ANO7 expression was found to be associated with race and histological subtype. The COAD patients exhibiting low ANO7 expression had lower survival rates compared to those with high ANO7 expression. The genes correlated with ANO7 were significantly enriched in proteolysis and mucin type O-glycan biosynthesis pathway. Furthermore, ANO7 demonstrated a direct interaction and a positive co-expression correlation with mucin 2 (MUC2). In conclusion, our findings suggest that ANO7 might serve as a potential prognostic biomarker and potentially plays a role in proteolysis and mucin biosynthesis in the context of COAD.
