A blue biliprotein, insecticyanin (INS), has been purified from the last instar larval hemolymph of Agrius convolvuli by ultracentrifugation, Sephadex G-100 gel permeation chromatography, and preparative electrophoresis. The molecular mass of INS was estimated to be 26 kDa and the N-terminal sequence of INS revealed high similarity to that of Manduca sexta. Results of Western blotting and autoradiography indicated that INS is synthesized by the epidermis and released into the hemolymph. In contrast to the INS reported in other insects, Agrius convolvuli INS contained a small amount of lipid, predominately consisting of triacylglycerol. Subcellular localization of INS was determined using protein-A gold particles linked to secondary antibodies (anti-rabbit Ig). INS was heavily accumulated in the cytoplasmic inclusion body (CIB). CIBs showed a variety of shapes from rod to globule and generally surrounded the nucleus. They were mostly located near the basement membrane and especially abundant in the intersegmental membrane.
A novel tetrathiafulvalene (TTF)-based main-chain polymer (6TTF-polymer) was successfully synthesized via a condensation polymerization between a newly synthesized dihydroxy TTF derivative and a malonyl chloride, and its chemical structure was characterized by spectroscopic techniques. Molecular weight of the 6TTF-polymer (9,030 g/mol by gel permeation chromatography) was large enough to form the ductile film. The electrochemical and optical properties of the 6TTF-polymer were further estimated by cyclic voltammetry, ultraviolet and photoluminescence spectroscopes. The highest occupied molecular orbital level ($E_{HOMO}$=-4.79 eV) and band-gap energy ($E_g$=1.91 eV) of the 6TTF-polymer suggested that TTF-based polymer could act as a good electron donating material for the optoelectronic applications.
A new methacrylate copolymer that includes chalcone as a side group, poly(4-methacryloyloxyphenyl-4'-methoxystyryl ketone-co-styrene) was synthesized by free radical copolymerization. FTIR and $^1H$ NMR spectroscopic techniques were used to characterize the homopolymers and copolymers. The copolymerizations were carried out to high conversions. Copolymer compositions were established by $^1H$ NMR spectra analysis. The monomer reactivity ratios for copolymer system were determined by the linearized Kelen $T{\ddot{u}}d{\ddot{o}}s$, and Extended Kelen $T{\ddot{u}}d{\ddot{o}}s$ methods and a non-linear least squares method. The molecular weights and polydispersity index of copolymers were measured by using the gel permeation chromatography (GPC). The effect of copolymer compositions on their thermal behavior were studied by differential scanning calorimetry and thermogravimetric analysis methods. The optical properties of the resulting copolymer were also investigated.
다양한 폴리디메틸실록산계 공중합체를 안정화제로 이용하여 초임계 이산화탄소 내에서의 개환중합을 통해 PLLA를 제조하였다. 초임계 개환중합에 사용된 폴리디메틸실록산 공중합체는 폴리디메틸실록산 거대개시제를 사용하여 그룹전이중합법(GTP법)을 이용하여 제조하였다. $\small{L}$-락타이드의 초임계 현탁중합을 통해 수미크론크기의 입자가 형성되었음을 전자 주사 현미경을 통해 확인하였고, 또한 겔 투과 크로마토 그라피 분석을 통해 상당히 좁은 분자량 분포의 PLLA가 제조되었음을 확인하였다.
To produce biopolymer, Metarrhizium anisopliae (Metschn.) Sorok was cultured in a medium containing glucose 1.0%, sucrose 2.0% , soluble starch 1.0%, yeast extract 0.5%, polypeptone 0.05%, K$_{2}$HPO$_{4}$ 0.1% MgSO$_{4}$ $\CDOT $ 7H$_{2}$O 0.02%. The culture broth was centrifuged and the polymer was harvested by adding methanol to the culture supermatant. When three times of methanol was added, the polymer was coagulated and precipitated. Then it was further purified through successive SK-1B, SA-20P, HP-20 column chromatographies. This polymer was designated as Biopolymer YU-122.C:H ratio of this Biopolmer YU-122 was 1:2 and small amount of N is detected by CHN analyzer. Glucose and glactose are main components of this polymer. Average molecular weight of this biopolymer was 1.7%$\times $10$^{6}$ by Sepharose 4B gel permeation chromatography. Optimal condition for biopolymer production was investigated. When 5% of mannitol was used as a carbon source, and polypepton as a N source, highest productivity of biopolymer was achieved. C/N ratio as nutrient was also a major factor in polymer production and its optimal ratio was 3.
Glucose syrup and lactose, mixed with citric acid as a polymerizing catalyst, was processed using twin screw extruder, in which 40 of L/D(length/diameter) ratio was designed to provide sufficient retention in extruder for polymerization of sugars. The polymerization yields of glucose syrup were 36.90%, 55.44% and 77.10% at 160, 180 and 20$0^{\circ}C$, respectively, while those of lactose were 26.45%, 38.16% and 45.86% at the same temperatures. Gel permeation chromatography exhibited that the higher molecular weight fractions were increased with extrusion temperature, which also led to increasing hydrodynamic intrinsic viscosity. Both uco-oligosaccharides and lacto-oligosaccharides produced by extrusion of glucose syrup and lactose were stable for thermal treatments over a wide range of pH3.0~11.0. In addition, $\alpha$-amylase and amyloglucosidase treatment of gluco-oligosaccharides did not affect the solution viscosity, indicating the random linkage rather than $\alpha$-1, 4 linkages of glucose and thus the potential applications as a dietary fiber. In this research it was clearly observed that twin screw extrusion can be successfully utilized to produce gluco-oligosaccharides and lacto-oligosaccharides rapidly and continuously in conjunction with selective control of polymerized composition.
A polyrotaxane-biotin conjugate was synthesized and its interaction with streptavidin measured using surface plasmon resonance(SPR) detection. A biodegradable polyrotaxane in which ca, 22 molecules of ${\alpha}$-cyclodextrina(${\alpha}$-CDs) were threaded onto a poly(ethylene oxide) chain(M$\sub$n:4,000) capped with benzyloxycarbonyl-L-phenylalanine was conjugated with a biotin hydorazide and 2-aminoethanol after activing the hydroxyl groups of ${\alpha}$-CDs in the polyrotaxane using N, N'-carbonyldiimidazole. The results of the high-resolution $^1$H-nyclear lmagnetic resonance($^1$H-NMR)spectra and gel permeation chromatography of the conjugate showed that ca, 11 biotin molecules were actually introduced to the polyrotaxane scaffold. An SPR analysis showed that the binding curves of the biotin molecules in the conjugate on the streptavidin-deposited surface changed in a concentration dependent manner, indicating that the biotin in the conjugate was ac-tually recognized by streptavidin. The association equilibrium constant(K$\sub$a/) of the interaction be-tween the conjugate and steptavidin tetramer was of the order 10$\^$7/. These results suggest that polyrotaxane is useful for scaffolds as a polymeric ligand in biomedical fields.
The thermal degradation of polymethyl methacrylate(PMMA) blend namely polymethyl methacrylate-polycarbonate(PMMA-PC) blend and polymethyl methacrylate-polystyrene(PMMA-PS) blend were carried out by isothermal method under air at several heating temperature from 220 to $270^{\circ}C$. Molecular weight changes during the thermal decomposition were monitored by means of the viscosity average molecular weight($\bar{M}v$). The viscosity average molecular weight was determined by Gel Permeation Chromatography(GPC). The dominant process in the degradation of PMMA-PC and PMMA-PS blend were main chain scission randomly due to weak links that may be distributed along the polymer backbone and the initial rate which the bonds are broken is not sustained. The infra-red spectra of degraded PMMA-PS blend show that the presence of aromatic ketone band at $1685cm^{-1}$. However, the infra-red spectra of degraded PMMA-PC blend show that the presence of hydroperoxide band at $3450cm^{-1}$. Thus indicating that the weak links are attacked by oxygen from the air and produce hydroperoxide or ketone. The activation energies of PMMA-PC blend and PMMA-PS blend were 18.2 and 17.9 Kcal/mol, respectively.
Two types of chitosanases produced from Aspergillus fumigatus KH-94 were purified by ion exchange and gel permeation chromatography. Molecular weights of the enzymes are 22.5 kDa (chitosanase I) and 108 kDa (chitosanase II). pI, optimum pH, and temperature of chitosanase I are 7.3, 5.5, and 70-$80^{\circ}C$, respectively, and those of chitosanase II are 4.8, 4.5~5.5, and 50~$60^{\circ}C$, respectively. Activities of both chitosanases were increased by $Mn^{2+}$ but inhibited by $Cu^{2+}$ and $Hg^{2+}$ . Chitosanase I has endo-splitting activity that hydrolyzes chitopentaose, chitohexaose, and chitosan to chitobiose, chitotriose, and chitotetraose, whereas chitosanase II has exo-splitting activity that hydrolyzes chitobiose and chitosan to glucosamine. Chitosanase II was found to have transglycosylation activity also in the reaction of 2% more chitooligosaccharides as a substrate and at the initial reaction. The higher degree of deacetylation, the stronger activities of chitosanase Iand II toward chitosans. Both chitosanases could hydrolyze chitosan and glycol chitosan but not chitin, cellulose, and carboxymethyl cellulose. To produce higher degree of polymerization of chitooligosaccharides, chitosanase I was used and yielded 80% of recovery.
Bacillus sp. A56 was studied, because of its high flocculating activity. The flocculating substance produced by this strain was purified by ethanol precipitation, cetylpyridinium chloride (CPC) precipitation, and gel permeation chromatography (GPC). The FT-IR spectrum of the purified bioflocculant, designated as BF-56, showed typical characteristics of polysaccharides. The non-sugar substituents, and sugar components of BF-56 containing glucose, fucose, glucuronic acid, and galactose in an approximate molar ratio of 2.76:1.10:1:0.12, suggested that it was a novel bioflocculant with an estimated molecular mass of over $7{\times}10^3$ kDa. Rheological analysis of BF-56 revealed that it was a pseudoplastic that had higher apparent viscosity rate at dilute concentrations than those of zooglan. The solution of bioflocculant BF-56 exhibited non-Newtonian characteristics and it was compatible to high concentrations of salts such as KCl, NaCl, $CaCl_2,\;or\;FeCl_3.$ The present results suggested strong possibility of bioflocculant BF-56 to be fully applicable to industries such as wastewater treatment.
