  • To elucidate the toxic effect of oxygen free radicals on cultured mouse spinal motor neurons damaged by hydrogen peroxide(H₂O₂)-induced neurotoxicity, we examined the neurotoxicity induced by oxygen radicals by NR assay when cultured spinal motor neurons were grown in the medium containing various concentrations of H₂O₂ for 6 hours. In addition, neuroprotective effects of herb extracts such Rhizoma Gastrodiae(RG), on H₂O₂-induced neurotoxicity in cultured spinal motor neurons were evaluated after cultured spinal motor neurons were preincubated with various concentrations of herb extract, RG for 2 hours before 50uM H₂O₂ for 6 hours. H₂O₂ decreased remarkably cell viability in dose-and time-dependent manner in these cultures, and also herb extract, RG increased cell viability of spinal motor neurons damaged by H₂O₂ in these cultures. From the above results, it is suggested that H₂O₂ was toxic in cultured spinal motor neurons derived from mouse, and RG was effective in blocking the neurotoxicity induced by oxidative stress in these cultures.

  • We had studied about the therapy of pinggan with apoplexy and the rusult were obtaind as follows. 1. The therapy of Pinggan is based on Treatise on Exogenous Febrile Diseases(傷寒論)'s ShaoyaoGancaoTang(芍藥甘草湯) on the ground of The Yellow Emperor's Internal Classic(黃帝內經). 2. Ye Tianshi(葉天士) theorized about the therapy of Pinggan. It was that the internal wind theory combind the theory of that liver and kidney having the same origin. He said that 'Through mild the liver, we calm down the liver to stop the wind, and nourish the kidney and then removed heat'. 3. The prodomal stage was treated by prescription of Ziyin-Huoxue-Xifeng(滋陰活血熄風方), WendanTang(溫膽湯), TianmaGoutongYin(天麻鉤藤飮) and ZhenganXifengTang(鎭肝熄風湯). and The therapy has been used to the symptoms of deficiendy of liver-yin and kidney-yin and psycotic symptoms In convalescence. The prescription of ZhenganXifengTang(鎭肝熄風湯), TianmaGoutongYin(天麻鉤藤飮), and DighuangYinzi(地黃飮子) waked well. 4. Internal cerebral hemorrhage, cerebral infarction, movement disorder and senile dementia were treated by the therapy of pinggan, for the action of hypotensor, lipolysis, ataralgesia, defervesence. 5. • Objective:Gagam-Gongjin-dan (GGD) is an oriental medicinal prescription composited with Cervi parvum Cornu, Corni Fructus, Angelica Gigantis Radix, Lycii Fructus, Dioscoreae Rhizoma, Citri Pericarpium, Gastrodiae Rihzoma, Agastachis Herba, Cassiae cortex, Scutellariae Radix and Schisandrae Fructus. The purpose of this study was to investigate the effects of GGD extract against acetaminophen (APAP)-induced liver injury in mice. Methods:GGD extract was prepared by extracting with methanol for 7 days. The extract was freeze-dried following filtration through vacuum distillation system. The first, we investigated the antioxidant effects of GGD extract on electronic donating ability (DPPH), nitrite (NO) scavenging and superoxide dismutase (SOD)-like activity. The next, we investigated the possible hepatoprotective effect of GGD extract administration against acetaminophen-induced liver injury in mice. Mice were orally administrated with or without GGD extract of different doses (25-100 mg/kg/day) one times per day for 6 days. After 3 days, APAP was orally applied with a single dose (400 mg/kg). Results:GGD extract increased DPPH, NO and SOD-like activities in dose dependant. APAP treatment significantly increased aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities in plasma. Also, APAP treatment significantly evaluated lipid peroxidation product thiobarbituric reacting substances (TBARS) and depleted some antioxidant enzymes (superoxide dismutase, catalase, d-aminolevulinate dehydratase and gluthathione peroxidase activities) in liver homogenates compared to the control group. However, the orally administration of GGD extract was able to counteract these effects. Histological studies provided supportive evidence for biochemical analysis Conclusions:These results suggest that GGD extract has a potential antioxidant and hepatoprotective effect against APAP-induced liver injury, these properties may contribute to liver disease care.

  • We recently reported that Gastrodia elata extracts (GEE) had an effects to protect against lipopolysaccharide-induced cognitive impairment in vivo model. In this study, we investigated the neuroprotective effects and the mechanism of action of GEE in hydrogen peroxide (H2O2)-induced cell death of SH-SY5Y human neuroblastoma cell. The SH-SY5Y cells were divided into five groups, including control(non-treated group), 100 μM H2O2, 100, 200, 500 ㎍/㎖ GEE+ 100 μM H2O2 groups. Pre- and co-treatment with GEE prevented cell death induced by 100 μM H2O2 for 24 h in SH-SY5Y cells. Our findings also showed that anti-oxidants enzymes (Cu/Zn superoxide dismutase, Mn superoxide dismutase, catalase) were up-regulated by 100 μM H2O2. But GEE suppressed H2O2-induced anti-oxidants enzymes decrease in a dose-dependent manner. Treatment with GEE also inhibited phosphorylation of eukaryotic initiation factor-2α (eIF-2α) and p38 by H2O2. Taken together, the neuroprotective effects of GEE in terms of recovery of antioxidant enzymes expression, down-regulation of eIF-2α and p38 phosphorylation, and inhibition of cell death are associated with reduced oxidative stress in SH-SY5Y cells.

  • Objectives : Gagam-Gongjin-dan (GGD) composited with Cervi parvum Cornu, Corni Fructus, Angelica gigantis Radix, Lycii Fructus, Dioscoreae Rhizoma, Citri Pericarpium, Gastrodiae Rihzoma, Agastachis Herba, Cassiae cortkex, Scutellariae Radix, Schisandrae Fructus has been traditionally used for chronic diseases or weakness after illness in oriental countries. However, little is known about the effects of methanol extract of GGD on immune responses to ovalbumin (OVA) plus alum. Therefore, the purpose of this study was to investigate the effects of GGD on immune responses to ovalbumin plus alum in Balb/c mice. Methods : In this study, the extract of GGD was prepared by extracting with methanol for 7 days. The extract was freeze-dried following filtration through vacuum distillation system. Mice were orally administrated with or without GGD extract of different doses (50-200 mg/kg/day) for 30 days. We examined the effects of GGD extract on the serum levels of total IgE, OVA-specific IgE, IgG1, IgG2a, and CTACK/CCL27 production and CCR10 expression in lymph node cells and body weight change and foot pad swelling responses in ovalbumin treated Balb/c. Results : The oral administration of GGD dose-dependently reduced the serum levels of total IgE, OVA-specific immunoglobulin (IgE, IgG1 and IgG2b) and CTACK/CCL27 production in ovalbumin treated BALB/c mice. The levels of CCR10 expression from lymph node cells of OVA treated mice were markedly suppressed by treatment with GGD in a concentration dependent manner. Furthermore, foot pad swelling responses were also markedly suppressed by GGD. However, body weight were significantly increased dose dependently by GGD treatment. Conclusions : These results suggest that GGD treatment suppresses immune responses to ovalbumin, and these properties may contribute to allergic disease care.
