Anticholinesterase activity of Cinnamomum zeylanicum L. leaf extract

  • Cinnamomum zeylanicum (C. zeylanicum) is a tropical evergreen tree of Lauraceae family. It is one of the oldest culinary spices known and used traditionally in many cultures for centuries. In addition to its culinary uses, cinnamon also possesses as a folk remedy of many health disease condition including analgesic, antiseptic, antispasmodic, aphrodisiac, astringent, carminative, haemostatic, insecticidal, and parasiticide and memory enhancing property. This study was aimed to assess the acetylcholinesterase and butyrylcholinesterase inhibitory activity of standardized methanol extract of the C. zeylanicum. Gas chromatography - mass spectrometry (GC-MS) and high performance liquid chromatography (HPLC) analysis were done to identify the presence of eugenol as chemical component and support the neuroprotective activity in the extract. Anticholinesterase inhibitory activity of crude methanol extract of C. zeylanicum leaves and cinnamon oil were evaluated by 96-well microtiter plate assay and thin layer chromatography bioassay detection methods. This study revealed that cinnamon oil ($IC_{50}:45.88{\pm}1.94{\mu}g/ml$) has better anticholinesterase activity than methanol extract ($IC_{50}:77.78{\pm}0.03{\mu}g/ml$). In HPLC analysis, retention time of eugenol in cinnamon oil was found to be 15.81 min which was comparable with the retention time (15.99 min) of the reference standard, eugenol. Seven chemical compounds were identified by GC-MS analysis, in which eugenol as an important phytoconstituents. Thus the phytochemicals from C. zeylanicum methanol leaves extract could be developed as potential source of anticholinesterase activity, with particular benefit in the symptomatic treatment of Alzheimer's disease.

  • Gas chromatography (GC) separation technology and metal oxide semiconductor (MOS) gas sensors have been integrated for the effective analysis of volatile sulfur compounds (VSCs) such as H2S, CH3SH, (CH3)2S, and (CH3)2S2. The separation and detection characteristics of the GC/MOS system using diluted standard gases were investigated for the qualitative and quantitative analysis of VSCs. The typical concentrations of the standard gases were 0.1, 0.5, 1.0, 5.0, and 10.0 ppm. The GC/MOS system successfully separated H2S, CH3SH, (CH3)2S, and (CH3)2S2 using a celite-filled column. The reproducibility of the retention time measurements was at a 3% relative standard deviation level, and the correlation coefficient (R2) for the VSC concentration was greater than 0.99. In addition, the chromatograms of single and mixed gases were almost identical.

  • This study was to decompose carbon tetrachloride and CFC with pyrolytic incineration unit because of prohibition of their usage sooner or later. We have investigated heating value and temperature versus decomposing rate, removal of $Cl_2$ and dust in the flue gas, The results obtained were as follows; 1. In combustion condition to decompose $CCl_4$ heating value was 3,300Kcal/Kg, retention time was 2,0 sec. incinerator exit temperature was $950^{\circ}C$. 2.The removal of HCI and $Cl_2$ in flue gas used NaOH as reagent, then molar ratio o of $Na^+/Cl^-$ was 1.07. 3. NaCI of dust component was more than 90 %, 2 stage venturi scrubber was used to remove dust, then removal rate of dust was 99% over at L/G of $1.7Vm^3$

Effects of Operating Parameters on Toluene Removal in Dielectric Barrier Discharge Process (무성방전내에서 톨루엔 제거에 미치는 운전변수의 영향)

  • We investigated the effects of operating variables, such as electrical. reactor and gas parameters on toluene removal and discharge property in the dielectric barrier discharge (DBD) process. The toluene removal was initiated with the energy transfer to the reactor by loading of voltages higher than the discharge onset value. The energy transfer and toluene removal increased with the applied voltage. Higher removal rate was observed with smooth surface electrode despite of lower energy transfer compared with the coarse electrode, because more uniform discharge can be obtained on smooth surface state. The decrease of dielectric material thickness enhanced the removal efficiency by increasing the discharge potential. The toluene removal efficiency decreased with the increase of the inlet concentration. The increase of gas retention time enhanced the removal efficiency by the increase of energy density. The oxygen and humidity contents seem to exert significant influences on the toluene removal by dominating the generation of electrons, ions, and radicals which are key factors in the removal mechanism.

A Study on Evaluation of Accuracy and Precision on B, T, X Analysis Using Thermal Desorption/Gas Chromatograph/Flams Ionization Detector (열탈착/GC/FID를 이용한 B, T, X 분석의 정확도 및 정밀도 평가)

  • By using thermal desorption/gas chromatograph/flame ionization detector(TD/GC/FID), this study was carried out to evalute an accuracy and a precision on Benzene(B), Toluene(T), o-Xylene(X) analysis in an industrial hygiene laboratory. Limits of detection of TD/GC/FID on B, T, X were showed 13.75ng/sample or less. For the accuracy of the method by concentration levels, overall bias was showed 7.7% as an absolute value, and the pooled coefficient of variation showed 3.51%. For the precision on repeatability of peak area and retention time between within-run and between-run of analytical system, it is showed the results of within-run gave better than those of between-run. Also the accuracy by sorbents(Tenax TA and Chromosorb 106)was evaluated, and the precision on reproducibility between MDHS72 and this study was compared. It is showed it is possible for TD/GC/FID to evaluate accurately B, T, X concentration levels of less than 1ppm at indoor or outdoor of workplaces in Korea.

  • A novel process for $H_2S$ gas treatment has been introduced, based on the combined action of a chemical absorption step and a biological step involving the biocatalytic activity of the bacterium Thiobacillus ferrooxidans. The aim of this study is the development of a process for $H_2S$ elimination from gas streams based on that chemical/biological method. The immobilized biomass reactor/chemical adsorption system is suitable for application of the removal of $H_2S$. A double stage reactor was used for the experimental work. The removal efficiencies of over 99% were observed in the range of inlet $H_2S$ concentration from 200 to 1,000ppm. The novel process showed the stable elimination efficiencies of over 95% under the retention time range from 20 to 40sec at the 1,000ppm of $H_2S$ inlet concentration.

Determination of panthenol, cholecalciferol and tocopherol in cosmetic products by gas chromatography-mass spectrometry in SIM mode

  • A novel simple method to detect vitamins in cosmetic products by gas chromatography-mass spectrometry(GC-MS) has been developed. Three vitamins(panthenol, cholecalciferol and tocopherol) were used for this study. Vitamins were prepared by dissolving in tetrahydrofuran(THF), and silylated with bis-trimethylsilyltrifluoroacetamide-trichloromethylsilane(BSTFA). Silated vitamins were separated on a fuses-silica capillary column coated with DB-5. The identification of each vitamin was accomplished by retention time and mass spectrum library search with a computer, and the quantitation was made in the selected-ion monitoring(SIM) mode of GC-MS. SIM mode had given sensitivity to determine 50pg of panthenol, 285pg of cholecalciferol and 130pg of tocopherol. Linearity was maintained over the range 0.005-0.20% for each vitamin. Each cosmetic product(i.e. hair tonic and lotion) was found to contain amounts of the vitamins. This method was sensitive and gave 77.5-99.9% recovery of each vitamin from these cosmetic products. From these results, we concluded that silylation with BSTFA followed by GC-MS analysis allows the simple, covenient and exact determination of panthenol, cholecalciferol and tocopherol.

  • An experimental research was performed employing the two stage anaerobic digestion of the mixture of the nightsoil and septic tank sludge to determine the effects of various hydraulic retention time of the digestion on chemical characteristics and treatment effeciency, thus determining the proper retention time. Results of the research are as follows, 1. Volatile-acid decreased as HRT increased. 2. Alklinity and ammonia-N tended to increase as HRT increased as did pH values, however, were observed to be constant at higher HRT values than 15 days. 3. The removal efficiencies of TBOD, TCOD and VS increased as HRT increased. 4. The removal efficiency of volatile solid decreased as VS loading increased. 5. It was observed that the rates of gas production were: 0.33 with HRT of 5 days, 0.58 with HRT of 15 days and $0.57m^3/kg$ VS fed/day with HRT of 25 days respectively. It is believed that the highest rate of gas production was at HRT of 15 days. 6. The sludge settling experiment showed that the minimum settling time required to ensure the desired underflow concentraton was estimated to be 8.6 days.

Headspace gas chromatographic analysis as an objective method for measuring rancidity in soybean oil (Headspace 가스크로마토그라피를 이용한 대두유 산화진행도의 객관적 평가방법)

  • An objective method for measuring rancidity in soybean oil was developed based on the volatile contents obtained by headspace gas chromatographic analysis. Apparatus for headspace volatile cold trapping-thermal desorption was described, designed to collect and analyze volatiles of soybean oil. Samples of commercially processed soybean oil were stored under controlled condition and then evaluated for flavor by a eight member trained panel and for volatile contents by headspace gas chromatographic analysis. Twenty-three GC peaks were identified on the basis of relative retention time of reference compounds and gas chromatography-mass spectrometry. The volatile compounds identified were autoxidation products of principal unsaturated fatty acids of soybean oil. The simple linear regressions of flavor score with the pentanal or hexanal contents in aged soybean oil gave correlation coefficient of 0.990 and 0.994. respectively. Pentanal and hexanal contents could be used as a potentially useful index for predicting flavor scores of soybean oil.

  • Several commercial cottonseed, corn and rapeseed oils were stored at $60^{\circ}C\;and\;70^{\circ}C$ with daily exposure of fluorescent light for 12 hours and evaluated their rancidity by headspace gas chromatographic analysis of pentanal and hexanal. The data of gas chromatographic analysis was compared with organoleptic flavor evaluation. For headspace gas chromatographic analysis, the volatile compounds were recovered by porous polymer trap and flushed into a fused silica capillary column at $250^{\circ}C$. Twenty-three GC peaks were identified on the basis of relative retention time of reference compounds and gas chromatography-mass spectrometry. The results showed that the contents of pentanal and hexanal were linearly increased during storage. A very simple linear relationship was found between organoleptic flavor scores and amounts of two volatile compounds with very high correlation coefficient. This results suggested the possible implication of pentanal and hexanal as an quality index for rancidity evaluation of cottonseed, corn and rapeseed oils.
