구름버섯과 영지버섯의 항암효과에 관한 연구 (Studies on Antitumor Activity and Antimicrobial Activity of Coriolus Versicolor(Fr.)Quel and Ganoderma Lucidum (Fr.)Karst)

  • The purpose of the study was to verify if or not as funtional food by estimating antitumor activity and antimicrobial activity of Coriolus versicolor(Fr.)Quel and Ganoderma Lucidum (Fr.)Karst. 1. The contents of moisture. crude fiber and crude protein of GL were higher $(18.28\%,\;10.3\%,\;78.4\%)$ than that of CV, but the content of crude lipid GL was higher than that of CV. 2. Inhibitory effects of on the growth of AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cell were increased by increasing of concentration in added methanol extracts of CV and GL. 3. Antimicrobial activity on Escherichia col1; Staphylococcus aureus, Salmonella typhimurium existed methanol extracts of CV. Therefore. it is suggested that CV is worth as functional food.

  • The purpose of this study was to investigate the physiological functionality of Coriolus versicolor Quel (CV) and Ganoderma IUCIdum (Fr.)Karst (GL) by estimate the proximate composition and content of antioxidant components. 1. In the physicochemical property. the content of proximate composition of GL was higher 08.28% of moisture, 10.3% of crude protein, 78.4% of crude fiber) than that of CV, but the content of crude lipid and ashes of GL was higher than that of CV. 2. CV had relatively higher content of antioxidant components such as total phenol, carotenoids and v itamin C. than GL did. Antioxidant mineral components such as magnesium and zinc also had relatively higher in CV than in GL. 3. TBA value, conjugated diene production, LDL oxidation, DPPH of methanol extracts in CV and GL were as high as a-Tocopherol.

  • This study was performed to investigate the effects of cell-wall broken spores of Ganoderma lucidum on the lipid accumulation and body weight reduction in C57BL/6J mice. Six-week-old C57BL/6J mice were randomly divided into 5 groups and assigned to one of these groups; normal chew diet(Nor) group, high-fat diet(HFD) group, HFD plus spores of Ganoderma lucidum 800 mg/kg/day (HFD + GS/B) group, HFD plus cell-wall broken spores of Ganoderma lucidum 400 mg/kg/day (HFD + BGS/A) group and finally HFD plus cell-wall broken spores of Ganoderma lucidum 800mg/kg/day (HFD + BGS/B). The experimental groups which were treated oral co-administration with cell-wall broken(or original) spores of Ganoderma lucidum and HFD significantly attenuated accumulative body weight gain, compared with HFD group. Administration of these experimental materials also resulted in significant reduction not only the serum levels of total cholesterol, homocysteine but also the lipid accumulation in liver tissue. But in the almost of results the cell-wall broken spores of Ganoderma lucidum were evaluated superior than the original one. These results indicate that cell-wall broken spores of Ganoderma lucidum may inhibit the lipid accumulation in blood as well as liver tissue. Therefore it may be a valuable candidate for the therapy preventing obese induced hyperlipidemic symptoms.

Favorable Culture Conditions for Mycelial Growth of Korean Wild Strains in Ganoderma lucidum

  • Ganoderma lucidum (Fr.) Karst (Polyporaceae), belonging to basidiomycota, is one of the most famous medicinal mushrooms. This study was carried out to investigate favorable mycelial growth conditions, such as pH, temperature, growth media, carbon sources and nitrogen sources of Korean strains in G. lucidum. The most suitable temperature for the mycelial growth was obtained at $30^{\circ}C$. In general, optimal temperature range for the mycelial growth was found at $25{\sim}30^{\circ}C$. This Mushroom has a broad pH range ($5{\sim}9$) for its mycelial growth and mostly favorable growth was found at pH 5. Generally, Hamada, Glucose peptone, YM, Mushroom complete and Lilly media were the most suitable for the mycelial growth of G. lucidum. Among 10 different carbon sources, dextrin, galactose and fructose were best but the rest of other carbon sources also facilitated the growth of mycelia. The most suitable nitrogen sources were ammonium acetate, glycine, arginine and calcium nitrate, but to a certain extent, all of the supplemented nitrogen sources also stimulated the mycelial growth.

영지버섯중의 중금속 함량 (Investigation of Heavy Metal Contents in Ganoderma lucidum(Fr.) Karst)

  • Ganoderma lucidum has been widely used not only as ingredients in herbal medicine but also in pharmacological soft drinks. The author collected for analysis of content of 8 kinds of heavy metal(Cd, pb, Hg, Cu, Mn, Fe, As) in soil and cluture soil in an around the Taegu area including Sang Ju, Non Gong, Keum Ho, and Weol Bae. THe toxic content in Gaoderma lucidum showed relatively low level as in cadmium lead mercury arsenic : 0.8-0.13ppm 0.17-1.43ppm 0.02-0.32ppm 0.01-0.19ppm respectively : in copper mangenese zinc and iron : 0.93-4.29ppm, 0.37-2.18ppm 1.02-1.65ppm, 4.57-11.04ppm those grown in soil showed higher percentages of content than those grown on logs in lead copper zinc and iron by 43.2% 68.6%, 20.3% and 43.2% respectively. The content of heavy metals in those grown in soil and culture soil tended to be higher in the areas near factories of industrial complexes especially in manganese and iron. The content of heavy metals in soil and culture soil appeared lower than the mean values of Korean Soil. No interrelationship was found in the content of heavy metals between those of Ganoderma lucidum grown on logs and those grown in soil. In case of pot cultivation however the mushroom spawns are grown originally in soil which seems to influence the degree of content of heavy metals of media.

  • 한국산(韓國産) 영지(靈芝)버섯의 계통분류(系統分類) 및 유전형질(遺傳形質)을 탐색(探索)하기 위하여 영지균(靈芝菌)의 배양적(培養的) 특성(特性)을 조사(調査)한 결과(結果)는 다음과 같다. 1. 한국산(韓國産) 자생영지균(自生靈芝菌)은 Oat meal agar 및 Rice bran extract agar에서 균사생장(菌絲生長)이 양호(良好)하고 Mycelial sector, Mycelial strand, Concentric zone의 형성(形成)은 배지(培地)의 영향(影響)을 받으나 균주(菌株)에 따라서도 차이(差異)를 보였다. 2. 영지균(靈芝菌)은 온도(溫度) $30^{\circ}C$, pH 5.5~6.0일때 균사생장(菌絲生長)이 좋았으며 백색형광등(白色螢光燈) 조사(照射)는 균사생장(菌絲生長)을 억제(抑制)하였고 광처리(光處理)의 균사생장억제기간(菌絲生長抑制時間)은 4~8시간(時間)사이였다. 3. 균사생장중(菌絲生長中)의 Mycelial strand의 형성(形成)에는 광(光)의 조사(照射) 그리고 Mycelial sector의 형성(形成)에는 배지(培地)의 영양원(營養源)과 광(光)의 조사등(照射等) 제(諸) 요인(要因)이 관여(關與)하였고 균주간(菌株間)에도 명확(明確)한 차이(差異)를 보였다. 4. 균배양중(菌培養中) 및 균사생장후(菌絲生長後) 광조사(光照射)는 자실체형성(子實體形成)을 유도하였는데 그 영향(影響)은 균주(菌株)에 따라 뚜렷한 차이(差異)를 보였고 균사생장중(菌絲生長中) 후막포자(厚膜胞子)의 형성(形成)은 인공재배시(人工栽培時) 편각형(扁角型) 자실체(子實體)를 형성(形成)하는 균주(菌株)에서 많이 발견(發見)되었고 녹각형(鹿角型) 자실체(子實體)를 형성(形成)하는 균주(菌株)에서는 많지 않았다.

  • 영지(靈芝)버섯(Ganoderma lucidum (Fr.) Karst.)의 16개 균주간(菌株關) 특성을 전기영동법(電氣泳動法)을 사용(使用)하여 단백질(蛋白質) pattern과 esterase, LAP의 동립효소(同位酵素) pattern에 의해 비교(比較) 관찰하여 다음과 같은 결과를 얻었다. 1. 자실체(子實體)의 esterase의 band pattern은 각(各) 균주간(菌株間) 특정 band의 유 무(有 無)와 위치(位置) 등에 의해 분류(分類) 할 수 있었으며 자실체(子實體)의 형태(形態)가 유사한 것은 band pattern에서도 유사하였다. 2. 동일균주(同一菌株)에서 편각(扁角)과 녹각형태(鹿角形態)로 형성(形成)된 자실체(子實體)의 esterase band pattern은 서로 동일(同一)한 pattern을 보였다. 3. 균주간(菌株間) 유연관계인 유사도 지수는 12.5%에서 100%의 변이 폭을 보였다. 특히 6과 7번의 두 균주간(菌株間)과 13,14,15,16번의 4균주간(菌株間)은 유사도지수가 100%이므로 유전적(遺傳的)으로 동일(同一)한 균(菌)이라 사료(思料)된다. 4. 균사(菌絲)의 단백질(蛋白質)과 esterase의 band pattern은 자실체(子實體)의 pattern과는 달랐으며 자실체(子實體)에서 동일(同一)한 pattern을 보였던 균주간(菌株間)에서도 균주간(菌株間) 특성을 가장 잘 보여 주었으므로 영지(靈芝)버섯 분류(分類)에는 균사(菌絲)의 esterase pattern이 가장 적합하다.

한국산(韓國産) 담자균류(擔子菌類) 항암(抗癌) 성분(成分)에 관한 연구(硏究) (Studies on the Antineoplastic Components of Korean Basidiomycetes)

  • To investigate antitumor components of Korean higher fungi, the carpophores of Ganoderma lucidum (Fr.)Karst. were collected at Gal-mae-ri in Gyeong-gi Province and extracted with 0.1N sodium hydroxide solution. The extract was purified by dialyzing through Visking tube and a dark brownish power was obtained. The antitumor activity of the fraction was tested against sarcoma 180 implanted in mice. The tumor inhibition ratio of the fraction against the tumor was 87.6% in doses of 50 mg/kg/day for the period of ten days. The tumor in five of 15 mice was completely regressed. The antitumor fraction was chemically analyzed and found to be a complex of polysaccharide and protein, from which four monosaccharides and eighteen amino acids were identified.

원목(原木), 병재배(甁栽培) 및 야생(野生) 영지(靈芝)의 추출물이 Saccharomyces cerevisiae의 생리에 미치는 영향 (Effect of Ganoderma lucidum(Wood, Pot cultivated & Wild) Extract on the Physiological Characteristics of Saccharomyces cerevisiae)

  • 영지(靈芝(Ganoderma lucidum(Fr.) Karst)) 추출물이 효모(Saccharomyces cerevisiae)의 증식과 생리에 미치는 영향을 검토하기 위하여 재배별 영지추출물을 각각 0, 0.1, 0.5, 1.0% 첨가한 Hayduck 액 배지에 효모를 접종하여 5일간 $30^{\circ}C$에서 발효시키면서 $CO_2$ 발생량, 균체생산, alcohol 발효 등을 조사하였다. 발효과정중 효모의 $CO_2$ 발생량은 발효 120시간 경과후 영지추출물 첨가구에서 모두 대조구보다 증가하였으며 이들 중 병재배 영지추출물 1.0% 첨가구에서 $CO_2$발생량이 가장 많았다. 발효과정중 효모의 세포수는 영지추출물 첨가구의 경우 모두 대조구보다 증가하였으며 병재배 영지추출물 첨가구가 원목영지추출물 및 야생영지추출물보다 세포수가 많았다. 효모의 건조균체량은 pot 0.5%구(區)>Pot 1.0%구(區) wild 1.0%구(區)>wood 1.0%구(區)>wood 0.5%구(區)>wild 0.5% 구(區)>wild 0.1% 구(區)>Pot 0.1% 구(區)> wood 0. 1% 구(區)>대조구의 순서로 증가하였으며 병재배영지추출물 첨가구는 원목재배나 야생영지추출물 첨가구보다 많았다. 각 처리구의 alcohol 함량은 발효 72시간의 경우 영지추출물 첨가구는 모두 대조구보다 3배 이상으로 많았으나 120시간 발효 후에는 큰 차이가 없었다. 발효과정중 효모의 당소비율과 발효율은 발효초기에 영지추출물 첨가구가 대조구보다 현저히 높았으나, 그 후 시간이 경과함에 따라 비슷하게 나타났다.
