In this study, four plants grown naturally in Egypt that are usually consumed by pastoral nomads were screened for their total phenolic contents, antioxidative, and anti-$\alpha$-amylase activities. Dried powder of plant's part was extracted in absolute or 70% ethanol. A polar extract of Panicum turgidum (PTPE) had the highest total polyphenol content {92.5 mg gallic acid equivalents (GAE)/g}, followed by an alkaloid extract of Withania somnifera (WSAlk; 77.5 mg GAE/g), and an ethanol extract of Leptadenia pyrotechnica (LPEE; 59.1 mg GAE/g). By employing different assays such as DPPH radical scavenging, reducing power, $Fe^+$ chelating, $H_2O_2$ scavenging and total antioxidant capacity, it was shown that PTPE, WSEE (ethanol extract of W. somnifera), WSAlk and LPEE had promising antioxidant activity, though, their potency varied according to the different tests. WSAlk had the highest level of $\alpha$-amylase inhibition (40.2%) in vitro, followed by WSEE (30.5%). Therefore, it can be concluded that these plants, especially extracts of PTPE, and WSAlk are beneficial to physiological health, and could be used in food and pharmaceutical industries to prepare dietary supplements, functional foods or food preservatives.
Objectives : This research aimed to evaluate the effects of Rubus coreanus $M_{IQ}$. Methods: These extracts from fresh R. coreanus $M_{IQ}$. were investigated with radical scavenging activity, the content of total phenolics, ferric thiocyanate method and flavonoid contents Results: These results revealed that the ethyl acetate extract from fresh R. coreanus $M_{IQ}$. possesses remarkable radical scavenging activity. The content of total phenolics of the ethanol extract from dried R. coreanus $M_{IQ}$. ($IC_{50}$=155.87${\pm}$8.8${\mu}$g/mL) is the highest among all of the samples. The stronger radical-scavenging activity ($IC_{50}$=77.72${\pm}$2.1${\mu}$g/mL) of ethyl acetate extract from dried R. coreanus $M_{IQ}$. should be related to its phenolic content as measured by gallic acid test. Conclusion: These results suggest that dried R. coreanus $M_{IQ}$. may act as a potential antioxidant agent.
The hepatoprotective effects of bergenin and its derivative, acetylbergenin, were evaluated against D-galactosamine-induced liver damage in rats. Bergenin is a C-glucoside of 4-O-methyl gallic acid that has been isolated from the cortex of Mallotus japonicus (Euphorbiaceae). Acetylbergenin was synthesized from acetylation of bergenin to increase lipophilic and physiological activities. Bergenin (50, 100 and 200 mg/kg) and acetylbergenin (25, 50 and 100 mg/kg) were administered orally once daily for successive 5 days after the injection of galactosamine (400 mg/kg, i.p.), respectively. The substantially elevated serum enzyme activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and ${\gamma}-glutamyltransferase$ due to galactosamine treatment were dose-dependently restored towards normalization by post-treatment with bergenin and acetylbergenin. Bergenin and acetylbergenin also significantly prevented the elevation of hepatic malondialdehyde formation and depletion of reduced glutathione content induced by galactosamine in a dose-dependent manner. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored towards normalization. These results suggest that effects of bergenin and acetylbergenin may be related to complex mechanisms that involve prevention of lipid peroxidation and preservation of hepatic glutathione. The results of this study clearly indicate that bergenin and acetylbergenin have potent hepatotherapeutic action against galactosamine-induced hepatotoxicity in rats, and lipophilic acetylbergenin is more active in the antihepatotoxic effects against galactosamine than much less lipophilic bergenin.
As part of ongoing study focused on the discovery of natural antioxidants from Korean plants by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, ethanol extract of the aerial parts of Curcuma longa (Zingiberaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the ethanol extract led to the isolation of two phenolic compounds, 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucopyranoside (1) and gallic acid (2), as antioxidant compounds. Their structures were elucidated by spectroscopic studies. Compounds 1 and 2 were isolated for the first time from this plant. These compounds showed the significant antioxidative effects during the DPPH free radical scavenging test, and the riboflavin- and xanthine-originated superoxide quenching activity tests.
The EtOAc and $H_2O$ soluble fractions of Katsura tree (Cercidiphyllum japonicum Sieb. Et Zucc) bark extracts were chromatographed on a Sephadex LH-20 column with various aqueous MeOH. Gallic acid (1), methyl galate (2), kurigalin (3), 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose (4) and 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucose (5) were isolated from EtOAc fraction. Isocorilagin (6) and methyl galate (2) were separated from $H_2O$ fraction. The structure determination was done by $^1H$ and $^{13}C$ NMR. Of these isolated compounds, methyl galate (2), kurigalin (3) and isocorilagin (6) were isolated, for the first time, from the bark extracts of Cercidiphyllum japonicum.

The substantially elevated serum enzymatic activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and ${\gamma}$-glutamyltransferase induced by $CCl_4$ were restored towards normalization by posttreatment with bergenin and acetylbergenin. Bergenin and acetylbergenin also significantly prevented the elevation of hepatic malondialdehyde formation and depletion of glutathione content induced by $CCl_4$ in a dose dependent fashion. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored towards normalization. These results suggest that therapeutic effects of bergenin and acetylbergenin may be related complex mechanisms that involve prevention of lipid peroxidation and preservation of hepatic GSH. The results of this study clearly indicate that bergenin and acetylbergenin have potent hepatothrapeutic action against $CCl_4$-induced hepatotoxicity in rats. In addition, acetylbergenin 50 mgHg showed almost the same levels of hepatoprotective activity as those of bergenin 100 mgAg, indicating the fact that lipophilic acetylbergenin is more effective in the hepatoprotective action against $CCl_4$ than bergenin.
This study has been performed to identify the phenolic contents and antioxidant properties of methanol and ethanol extracts of young Camellia leaves in Jeju. Total polyphenol and flavonoid contents were 74.9 and 46.1 mg gallic acid equivalent/100 g, and 30.6 and 21.4 mg RE/100 g in the methanol and ethanol extracts, respectively. Based on $IC_{50}$ values, both extracts were also found to possess potent scavenging capacities of 1,1-diphenyl-2-picrylhydrazyl, nitric oxide and superoxide anion radicals, and ferrous ion chelating and reducing power activities. These results showed that young Camellia leaves is a good antioxidant source and holds promise as a natural ingredient in functional food and pharmaceutical supplement.

Antioxidant activities of the extract of red seaweed, Polysiphonia morrowii, were evaluated using several in vitro assay systems. Activity-guided fractionation revealed that the 90% MeOH fraction of the P. morrowii extract exhibited the highest antioxidant activity, and that this fraction had a high total phenolic content ($135.7{\pm}5.0\;mg$ gallic acid/g extract). Therefore, the antioxidant activities of the 90% MeOH fraction against 2,2-diphenyl-1-picrylhydrazyl (DPPH), hydroxyl radical, reducing power, ferrous chelating, and hydrogen peroxide were investigated. The results revealed that the antioxidant activities of the 90% MeOH fraction were similar and/or superior to that of commercial antioxidants such as butylated hydroxyanisole (BHA) and butylated hydroxytoluene (BHT). In addition, the ability of the 90% MeOH fraction to inhibit oxidative damage to DNA was assessed by measuring the conversion of the supercoiled pBR322 plasmid DNA to the open circular form. The 90% MeOH fraction was found to significantly protect this hydroxyl radical-induced DNA damage in a dose-dependent manner. Taken together, these findings suggest that the 90% MeOH fraction of P. morrowii extract and/or its constituents has the potential for use as a new bioresource of antioxidants.
To evaluate the antioxidant and potential of Humulus japonicus, total polyphenol and flavonoid content, radical scavenging activities, and antitumor activities were measured. The total polyphenol and flavonoid contents of the methanol extracts from Humulus japonicus were $30.13{\pm}1.13$ and $13.61{\pm}0.49$ mg gallic acid equivalent/g extract, respectively. DPPH radical and hydroxyl radical scavenging activities of methanol extracts of Humulus japonicus were 60% and 35%, respectively. The Humulus japonicus higher activities of anticancer activities on liver cancer cell lines compared to other cancer cell lines.
Antioxidative and free radical scavenging properties of different stem extracts of Euphorbia trigona were evaluated and correlated with its total phenolic content. Aqueous, acetone and methanolic extracts of shade dried stem were obtained and were concentrated in vacuo. The antioxidant and free radical scavenging activities of stem extracts was determined by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) scavenging assay, reducing power assay, deoxyribose degradation assay and $Fe^{2+}$ chelating assay. Total phenolic contents (TPC) were evaluated using Folin-Ciocalteu reagent. The results confirmed that the plant is a rich source of polyphenolic compounds which are invariably higher compared to other herbs. All extracts showed TPC in the range of 146.6 - 168.6 mg/g gallic acid equivalents at $300{\mu}g/ml$ of extract. Among the three extracts ME showed highest scavenging activity as evidenced by maximum scavenging of DPPH (83.2%), $OH{\bullet}$ radicals (94.81%), $Fe^{2+}$ chelating activity (88.59%) and a high reducing power 0.623 at $300{\mu}g/ml$. Our results demonstrate that Euphorbia trigona, an unexplored xerophytic plant could be potential source of natural antioxidants and phytotherapeutic agents. The plant possess invariably high amount of polyphenolic compounds with a broad spectrum of antioxidant properties and could be further used for food, feed and pharmaceutical applications.
