열처리에 따른 돼지감자 Methanol 추출물의 항산화 및 α-glucosidase 저해 효과 (Antioxidant Activity and α-Glucosidase Inhibitory Effect of Jerusalem Artichoke (Helianthus tuberosus) Methanol Extracts by Heat Treatment Conditions)

  • This study investigated the changes of antioxidant activity and ${\alpha}$-glucosidase inhibitory effect of Jerusalem artichoke (Helianthus tuberosus) 100% methanol extracts by various heat treatment. The contents of total phenolic and flavonoid compounds in methanol extract tended to increased gradually with the rise of temperature to 180$^{\circ}C$. The maximum yield of gallic acid (51.52 ${\pm}$ 2.17mg/g extract weight) and quercetin (13.39 ${\pm}$ 0.03mg/g extract weight) were obtained with extraction temperature of 180$^{\circ}C$ for 120min. In addition, the improving extraction efficiency resulted in the increased biological activities, such as electronic donation ability (EDA, 90.36${\pm}$ 0.57%), reducing power (Abs 1.14) and ${\alpha}$-glucosidase inhibitory effect (92.14 ${\pm}$ 1.14%). Overall, the results of this study indicate that the optimum conditions for the extraction process were an extraction temperature at 180$^{\circ}C$ for 120 min, and will provide the basis for future research on the improving extraction yield of phenolic and flavonoid compounds.

마황(麻黃) 추출물이 항산화에 미치는 영향 (Effects of Ephedras Herba on the activity of antioxidant)

  • Objectives : The purpose of this study was to investigate the antioxidative effects of Ephedrae Herba extract. Methods : Total antioxidant status was examined by total antioxidant capacity(TAC), total antioxidant response(TAR) against potent free radical reactions, total phenolic content, DPPH radical scavenging activities, and the inhibitory effect of the extract on lipid peroxidation in rat liver mitochondria. Results : 1. TAC and TAR of Ephedrae Herba extract at the concentration of 5mg/ml were 3.76 and 1.65 mM Trolox equivalents , respectively. 2. Total phenolic content of Ephedrae Herba extract at the concenuation of 5mg/ml was 3.52 mM gallic acid equivalent. 3. Concentration of Ephedrae Herba extract at which DPPH radical scavenging activity was inhibited by 50% was 0.46mg/ml as compared to 100% by pyrogallol solution as a reference. 4. Ephedrae Herba extract at the concentration of 2mg/ml significantly decreased TBARS concentration by 97%, showing that Ephedrae Herba extract prevented lipid peroxidation at the low concentration. Conclusion : Thus antioxidant effects of Ephedrae Herba extract appear to be due to, at least in part, the prevention from free radicalsinduced oxidation, followed by inhibition of lipid peroxidation.

Mineral, Nutritional, and Phytochemical Profile, Total Phenolic Content, and Radical Scavenging Activity of Philippine Bamboo "Bolo" Gigantochloa levis (Blanco) Merr. Leaves

  • The study is a pioneering effort to determine the mineral, nutritional, and phytochemical composition and phenolic content and to determine the free radical scavenging activity of Gigantochloa levis (Blanco) Merr, a native bamboo species (locally known as "bolo") in the Philippines. Proximate analysis showed that air-dried G. levis leaves contain 15.8% ash, 22.6% crude protein, 1.2% crude fat, 29.3% crude fiber, and 19.7% total sugar. Phytochemical tests indicated the presence of diterpenes, triterpenes, saponins, phenols, tannins, and flavonoids in both the ethanolic and aqueous leaf extracts, while phytosterols were only detected in the ethanolic extract. Folin-Ciocalteu assay determined the total phenolic content in gallic acid equivalents (GAE) to be $85.86{\pm}3.71$ and $32.32{\pm}1.01mg\;GAE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The total phenolic content in quercetin equivalents (QE) was $74.44{\pm}3.11$ and $29.43{\pm}0.85mg\;QE/100g$ dried sample for the ethanolic and aqueous extracts, respectively. The radical scavenging activity of the different solvent fractions containing varying concentrations of the extract was determined using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) assay. The ethyl acetate and 1-butanol fractions were found to have the highest radical scavenging activity. Mineral analysis via Energy Dispersive X-Ray Spectrometry (EDS) of the ash of G. levis leaves showed that Si is the major component, followed by K and Mg. These results point to the potential of G. levis leaves as a source of minerals and bioactive compounds with medicinal value.

찔레꽃 에탄올추출물의 생리활성과 화장품 방부효과 (Biological Activity and Cosmetic Preservative Effects of Rosa multiflora Ethanol Extracts)

  • Background: The Rosa multiflora, a well-known plant belonging to Rosacea, is widely used in orthodox medicine in worldwide. However, its biological activity and cosmetic preservative efficacy have not yet been studied. Thus, this species is yet to be defined as a functional cosmetic material. Accordingly, an investigation of the above mentioned atrributes was performed on a 50% ethanol extract of Rosa multiflora. Methods and Results: The antioxidant activity was assessed through free radical scavenging assays with 2,2-diphenyl-1-picrylhydrazyl (DPPH). Additionally, the contents of total phenols and flavonoids were analyzed. The phenolic compounds were detected using HPLC. The antimicrobial activity against Staphylococcus aureus, Escherichia coli, and Candida albicans was assessed using the disc diffusion assay. The preservative effect (challenge test) on a formulation of soothing gel was performed for 28days. The DPPH radical scavenging ability, denoted by the $SC_{50}$ (half maximal inhibitory concentration for DPPH radical scavenging) value was found to be $131.63{\mu}g/m{\ell}$. The content of total polyphenol and flavonoid content were 202 mg/g and 86.77 mg/g, respectively. In additon, astragalin and gallic acid were identified in the extract. The antimicrobial activity of the extract against S. aureus and E. coli was observed to be 5 - 0.5%, and no significant activity was noted against C. albicans. The ethanol extracts (5% and 10%) met the preservation standards of the Cosmetics, Toiletry, and Fragrance Association (CTFA). Conclusions: Thus the ethanol extract of R. multiflora can be used in cosmetics as a natural preservative and antioxidant.

녹차 카테킨류의 pH에 대한 안정성 연구 (Effect of pH on the Stability of Green tea Catechins)

  • 시중에 유통되고 있는 녹차중의 catechin류에 대하여 열수추출 및 HPLC분석을 통하여 함량을 조사하고 이를 효율적으로 이용하기 위하여 pH조건이 녹차 catechin류의 안정성에 미치는 영향에 대하여 분석하였다. 시판 중인 전남 보성산 녹차의 catechin류 함량은 (+)-catechin>(-)-EGCg>(-)-EGC>(-)-EC>(-)-ECg순으로 추출되었고 Total catechin류의 함량은 103.72mg/g이었다. 녹차에서 분리·정제되어진 catechin류인 (+)-catechin, (-)-EC, (-)-ECg, (-)-EGCg를 pH 3∼11에 대한 안정성을 흡광도측정장치를 이용하여 측정하였다. 그 결과 catechin류의 pH에 대한 영향을 최대흡수파장 및 흡광도 변화로 보면 pH가 높을수록 (+)-catechin과 (-)-EC은 흡광도가 증가되었고 (-)-ECg와 (-)-EGCg는 최대흡수파장이 증가되었다. 또한 7일간 실온에서 저장하면서 기간에 따른 변화를 살펴본 결과에서 산성조건에서는 큰 변화가 없지만 알칼리조건에서는 흡광도가 증가되었다. (+)-catechin과 (-)-EC은 알칼리조건에서 흡수파장 400∼430 nm에서 저장기간이 길수록 흡광도가 증가되었다. 이는 녹차의 갈변과 관계가 있을 것으로 생각되었다. 이상의 결과로, 녹차 Catechin유도체의 pH 및 저장기간에 대한 영향은 ECg, EGCg에 gallic acid의 결합에 의한 것으로 생각되고, C, EC의 불안정은 중합반응으로 생각된다. 녹차 catechin류는 대부분 산성에서 저장기간이 길고 안정한 것으로 나타났으나 생리활성이 강한 (-)-EGCg는 산성에서도 불안정한 것으로 나타났다. 결론적으로 녹차의 기능성 성분을 효율적으로 이용하기 위해서는 pH를 낮게 유지하여야 할 것으로 사료되어진다.

당조고추 70% 에탄올추출물의 항산화 및 항암활성측정 (Anti-Oxidative and Anti-Proliferative Effect of 70% Ethanol Extracts from Green Pepper (Capsicum annuum L. cv. DangZo))

  • 당조고추의 기능성 소재로서의 가능성을 알아보기 위하여 당조고추 70% 에탄올 추출물에 대한 항산화 활성 및 항암활성을 측정하였다. 당조고추 70% 에탄올 추출물의 총 폴리페놀은 30.29 mg GAE/g extract로 나타났다. 당조고추 70% 에탄올 추출물에 대한 DPPH radical 및 hydroxyl radical 소거활성을 $IC_{50%}$ 측정한 각각 2.87, 10.55 mg/mL의 소거활성을 보여 주었으며, ${\alpha}$-glucosidase 및 ${\alpha}$-amylase 저해활성은 10 mg/mL의 농도에서 각각 35.67, 58.41%로 나타났다. 당조고추 70% 에탄올 추출물에 대한 항암활성은 대장암 세포주의 0.5 mg/mL 농도에서 50% 암세포 성장억제를 보였지만, 다른 암세포에서는 억제효과를 나타내지 않았다.

명아주의 일반성분, 항산화활성 분석 및 흰쥐의 혈중 생화학적 분석에 관한 연구 (Studies on the general analysis and antioxidant component analysis of Chenopodium album var. centrorubrum and biochemical analysis of blood of mice administered C. album)

  • The purpose of this study was to develop new food materials by analyzing nutritional components and antioxidant activities of Chenopodium album var. centrorubrum. The highest amount of mineral obtained in C. album was found to be $1.01{\pm}0.07mg$ per 100 g of iron. The total phenolic content in C. album was found to be 3.77~9.57 GAE (gallic acid equivalent) mg/g. The reducing activities of leaves and roots of C. album determined using FRAP (ferric-reducing antioxidant power) were higher in ethanol extracts than water extracts. The ABTS radical scavenging activities of leaves and roots of C. album were $204.29{\pm}4.98{\mu}mol/g$ and $106.96{\pm}2.81{\mu}mol/g$, respectively. The DPPH radical scavenging activity was high upon extraction using ethanol (roots $20.71{\pm}0.04{\mu}mol/g$, leaves $71.08{\pm}0.33{\mu}mol/g$). HDL-cholesterol was significantly higher in the high fat diet groups supplemented with C. album than the control groups (p<0.05). These results suggest that C. album can be used as a natural antioxidant and a functional dietary supplement.

Gallotannins from Nut Shell Extractives of Camellia oleifera

  • Camellia nut shell was collected, dried at room temperature and ground to get fine powder. The powder was extracted three times with 95% EtOH, combined, evaporated, and then freeze dried. The crude powder was dissolved in H2O and then sequentially fractionated with n-hexane, CH2Cl2, EtOAc and n-BuOH. A part of EtOAc fraction was chromatographed on a silica gel and on a Sephadex LH-20 columns using MeOH, aqueous MeOH, EtOAc-n-hexane and EtOH-n-hexane to isolate gallotannins. Three gallotannins, 1,2-di-O-galloyl-β-D-glucopyranoside (2), 1,2,6-tri-O-galloyl-β-D-glucopyranoside (3) and 1,2,3,6-tetra-O-galloyl-β-D-glucopyranoside (4), including gallic acid (1), were isolated and elucidated by NMR and Mass spectroscopies. Although nothing new, these gallotannins were first reported from the nut shell extractives of camellia tree (Camellia oleifera C. Abel). This study was to investigate the chemical constituents, especially hydrolysable tannins, of nut shell extractives of Camellia oleifera and to provide basic information for the future chemical utilization of this species.

Antioxidant potential of a soft cheese (paneer) supplemented with the extracts of date (Phoenix dactylifera L.) cultivars and its whey

  • Objective: The present study was conducted to evaluate the antioxidant potential of paneer, a soft cheese supplemented with various water soluble date extracts during storage. Further, the whey obtained from all the paneer samples was also investigated for its antioxidant potential. Methods: The date cultivars were evaluated for their physico-chemical characteristics and date extracts were assessed for their antioxidant potential. Physico-chemical evaluation, microbiological quality and further antioxidant potential of the prepared paneer were carried out during storage period (0 to 8 days, $5^{\circ}C$). Results: All the date extracts were found to have considerable antioxidant activity due to presence of total phenolics and flavonoids. Owing to the presence of phenolics and flavoinds in date extracts, supplemented paneer showed higher trolox equivalent antioxidant capacity, reducing power and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity than control paneer. Paneer supplemented with Rabi extracts had the highest total phenolics ($190.7{\mu}g$ gallic acid equivalent/g paneer), DPPH radical scavenging activity ($928.1{\mu}mol$ equivalent of Trolx/g paneer) and trolox equivalent antioxidant capacity ($9.2{\mu}mol$ equivalent of Trolx/g paneer). The whey obtained from control paneer showed lower values of total phenolics, total flavonoids, DPPH, trolox equivalent antioxidant capacity and reducing power as compared to the values of whey obtained from paneer supplemented with date extracts. Conclusion: Paneer supplemented with date extracts and its whey may offer potent antioxidant activity.

바이칼레인(baicalein)이 peptidoglycan으로 자극된 RAW 264.7 mouse macrophages의 hydrogen peroxide 생성에 미치는 영향 (Effects of Baicalein on hydrogen peroxide productions in RAW 264.7 mouse macrophages stimulated by peptidoglycan)

  • Objectives : The aim of this study was to investigate the effect of baicalein (BA) on the production of hydrogen peroxide in peptidoglycan-stimulated RAW 264.7 mouse macrophages. Methods : Peptidoglycan-stimulated RAW 264.7 were incubated with baicalein at concentrations of 50 and 100 µM. Incubation time is 30 min, 2 h, 12 h, and 18 h. After incubation, The production of hydrogen peroxide in RAW 264.7 was measured with dihydrorhodamine 123 assay. Berberine and gallic acid were used as the comparative materials. Results : BA at the concentration of 50 and 100 µM did not show cytotoxicity on RAW 264.7 for 24 h incubation. For 30 min, 2 h, 12 h, and 18 h incubation, BA at the concentration of 50 and 100 µM significantly inhibited the production of hydrogen peroxide in RAW 264.7 stimulated by peptidoglycan (p<0.05). In details, production of hydrogen peroxide in peptidoglycan-stimulated RAW 264.7 treated for 30 min with BA at concentrations of 50 and 100 µM was 93.91% and 93.52% of the control group treated with peptidoglycan only, respectively; the production of hydrogen peroxide for 2 h was 93.8% and 92.71%, respectively; production of hydrogen peroxide for 12 h was 94.86% and 95.93%, respectively; production of hydrogen peroxide for 18 h was 95.37% and 96.48%, respectively. Conclusions : BA might have anti-oxidative activity related to its inhibition of hydrogen peroxide production in peptidoglycan-stimulated RAW 264.7 macrophages.
