Acanthamoeba castellanii has ubiquitous distribution and causes primary acanthamoebic keratitis (AK). AK is a common disease in contact lens wearers and results in permanent visual impairment or blindness. In this study, we observed the cytopathic effect, in vitro cytotoxicity, and secretion pattern of cytokines in human corneal epithelial cells (HCECs) induced by A. castellanii trophozoites and/or cysts. Morphological observation revealed that panked dendritic HCECs co-cultured with amoeba cysts had changed into round shape and gradually died. Such changes were more severe in co-culture with cyst than those of co-cultivation with trophozoites. In vitro cytotoxicity assay revealed the highest cytotoxicity to HCECs in the co-culture system with amoeba cysts. A. castellanii induced the expression of $IL-1{\alpha}$, IL-6, IL-8, and CXCL1 in HCECs. Secreted levels of $IL-1{\alpha}$, IL-6, and IL-8 in HCECs co-cultured with both trophozoites and cysts were increased at an early incubation time (3 and 6 hr). These results suggested that cytopathic changes and pro-inflammatory cytokines release of HCECs in response to A. castellanii, especially amoebic cysts, are an important mechanism for AK development.

Mitochondrial NADP+-dependent isocitrate dehydrogenase 2 (IDH2) produces NADPH, which is known to inhibit mitochondrial oxidative stress. Ureteral obstruction induces kidney inflammation and fibrosis via oxidative stress. Here, we investigated the role and underlying mechanism of IDH2 in unilateral ureteral obstruction (UUO)-induced kidney inflammation using IDH2 gene deleted mice (IDH2-/-). Eight- to 10-week-old female IDH2-/- mice and wild type (IDH2+/+) littermates were subjected to UUO and kidneys were harvested 5 days after UUO. IDH2 was not detected in the kidneys of IDH2-/- mice, while UUO decreased IDH2 in IDH2+/+ mice. UUO increased the expressions of markers of oxidative stress in both IDH2+/+ and IDH2-/- mice, and these changes were greater in IDH2-/- mice compared to IDH2+/+ mice. Bone marrow-derived macrophages of IDH2-/- mice showed a more migrating phenotype with greater ruffle formation and Rac1 distribution than that of IDH2+/+ mice. Correspondently, UUO-induced infiltration of monocytes/macrophages was greater in IDH2-/- mice compared to IDH2+/+ mice. Taken together, these data demonstrate that IDH2 plays a protective role against UUO-induced inflammation through inhibition of oxidative stress and macrophage infiltration.
Objectives This study aims to identify differences in clinical manifestations of COVID-19 between different Sasang constitution. Methods Subjects were recruited from August 29, 2022, to July 11, 2023. COVID-19 clinical symptoms were self-reported via questionnaires. Sasang constitutional diagnosis was performed using the K-PRISM, and Sasang constitutional specialist. Results A total of 66 subjects were recruited for the study, and the Sasang constitutional distribution of the subjects was 19 soyangin, 25 taeeumin, and 22 soeumin. For most of the COVID-19 clinical manifestations, the study found that soyangin experienced symptoms at a higher rate than other constitutions. Among the symptoms observed in the study, sore throat, pantalgia, and cough were severe in all subjects regardless of constitution. Soeumin was more likely to experience abdominal pain, chest pain, and diarrhea, while soyangin was more likely to experience chest pain, nausea/vomiting, diarrhea, and cutaneous symptoms compared to other constitutions. Taeeumin had more sputm, nasal congestion, and skin symptoms, but fewer digestive symptoms. There were differences in the onset and duration of symptoms by constitution. Conclusions This study is an important contribution to our understanding of the differences in response to the COVID-19 virus among different Sasang constitutions. Symptomatic differences between constitutions may have important implications for prevention and treatment strategies for infectious diseases, and personalized treatment and management based on these differences may be needed in the future.

The government revised the regulations electronic financial supervision(hereafter EFS) in response to needs for mitigation of network separation in the IT environment and fintech companies. Some amendments to the EFS, which took effect on 01/01/2023, mitigate network separation only for research and development purposes in cloud environments. If software developed in a cloud development environment is applied to an operating system through a distribution system the existing perimeter-based security model will not satisfaction the network separation conditions. In this Study, we would like to propose a way to maintain the DevOps system in a network separation environment by Using the zero trust security system.
The fibrous layer of mandibular condyle of the neonatal, 1-, 7-, 14-, 27-, 55-days and 1 year old rats were examined in the electron microscope with particular attention to the ultrastructure and diameter of collagen fibrils. In the 1-day rats, most of the cells of the fibrous layer were undifferentiated mesenchymal cells and fibroblasts with rough a little developed rough endoplasmic reticulum(RER) and golgi apparatus(GA). In 7-, 17 and 27-days old rats, most of the fibroblast showed well developed GA and RER with widely distended cisternae containing granular materials. In many of these cells contained intracytoplasmic filaments among the cytoplamic organelle. In 55-day and 1-year old rats, three types of cells were observed, ie, cells containing well developed cytoplasmic organelle presumed to be involved in the collagen fibril synthesis, cells showing well developed lysosomes, golgi apparatus, mitochondria and short cytoplasmic process presumed to be involved in the active resorption of the injured collagen fibrils or cellular debris, cells containing many intracytoplasmic filaments and a little organelle presumed to be cells of inactive state. The average diameters of collagen fibrils were similar in 1- and 7-day old rats as $38.48{\pm}3.81nm$, $38.06{\pm}3.86nm$. That was thickest in 14 days old rats as $50.21{\pm}3.93nm$ among experimental groups. They were gradually thinner in 27-, 55-day rats as $40.05{\pm}2.52nm$, $43.63{\pm}1.20nm$ and thinnest in 1-year old rats as $37.38{\pm}2.17nm$. The distribution pattern of diameters of collagen fibrils were unimordal with peak of 30-60nm in rats from 1-day to 17-day old. With aging from 27-day to 1 year old rats, collagen fibril diameters showed wide distribution pattern and percentage of thin collagen fibrils increased. These results may show the functional adaptation of fibrous layer of mandibular condyle to the increased mechanical forces with aging.
Natural disasters of large scale such as typhoon, heat waves and snow storm have recently been increased because of climate change according to global warming which is most likely caused by greenhouse gas in the atmosphere. Increase of greenhouse gases concentration has caused the augmentation of earth's surface temperature, which raised the frequency of incidences of extreme weather in northern hemisphere. In this paper, we present spatial analysis of future typhoon genesis based on IPCC AR5 RCP 8.5 scenario, which applied latest carbon dioxide concentration trend. For this analysis, we firstly calculated GPI using RCP 8.5 monthly data during 1982~2100. By spatially comparing the monthly averaged GPIs and typhoon genesis locations of 1982~2010, a probability density distribution(PDF) of the typhoon genesis was estimated. Then, we defined 0.05GPI, 0.1GPI and 0.15GPI based on the GPI ranges which are corresponding to probability densities of 0.05, 0.1 and 0.15, respectively. Based on the PDF-related GPIs, spatial distributions of probability on the typhoon genesis were estimated for the periods of 1982~2010, 2011~2040, 2041~2070 and 2071~2100. Also, we analyzed area density using historical genesis points and spatial distributions. As the results, Philippines' east area corresponding to region of latitude $10^{\circ}{\sim}20^{\circ}$ shows high typhoon genesis probability in future. Using this result, we expect to estimate the potential region of typhoon genesis in the future and to develop the genesis model.
Background: Melanoma is the most fatal form of skin cancer due to its rapid metastasis. Recently, several studies reported that selenium can induce apoptosis in melanoma cells. However, the precise mechanism remains to be elucidated. In this study, we investigated the effect of selenium on cell proliferation in murine melanoma and on tumor growth and metastasis in C57BL/6 mice. Methods: Cell proliferation was measured by MTT assay in selenium-treated melanoma cells. Cell cycle distribution was analysized by staining DNA with propidum iodide (PI). mRNA and protein expression related to cell cycle arrest was measured by reverse transcription PCR and western blot. Tumor growth and metastasis was measured by in vivo model. Results: Selenium was suppressed the proliferation of melanoma cells in a dose dependent manner. The growth inhibition of melanoma by selenium was associated with an arrest of cell cycle distribution at G0/G1 stage. The mRNA and protein level of CDK2/CDK4 was suppressed by treatment with selenium in a time-dependent manner. In vivo, tumor growth was not suppressed by selenium; however tumor metastasis was suppressed by selenium in mouse model. Conclusion: These results suggest that selenium might be a potent agent to inhibit proliferative activity of melanoma cells.

This study was conducted to develop a large scale map of species-specific plantation sites based on selected environmental variables such as topography, soil, and climatic factors in Namwon city. Site index equations by tree species were first regressed to 27 environmental variables that could influence the productivity of forest sites using digital forest site maps, digital climate maps, and the 5th National Forest Inventory data. Site index equations by tree species were all evaluated to estimate site productivity using 4-5 environmental variables, and the models' reliability was confirmed based on evaluation statistics. The determination coefficients of site index equations by species ranged from 0.42 to 0.76. With the site index equations, the site conditions appropriate for productive sites by species were considered to assess spatial distribution of productive areas for each species. The final map for optimal plantation in Namwon city was produced based on both site index equations and site conditions appropriate for productive sites by each species using GIS technique. Field survey was conducted to evaluate the suitability of selected species on the map of species-specific plantation sites. Results showed that the plantation map provides relatively reasonable spatial distribution of productive areas for selected species. It was revealed, however, that the sites evaluated as 'not suitable' for any tree species should be revised and complemented with additional information, especially with the site conditions appropriate for productive sites by species of interest. The outcomes of this study are expected to provide information for making customized species-specific plantation maps.

In order to provide important information for the efficient management of the identified farm ecosystem in Jinju Bay, we investigated the spatial and temporal distribution of the phytoplankton community using a UPLC pigment analysis and a CHEMTAX program from the timeframe of February 2013 to January 2014. In addition, we measured the available physical and chemical parameters controlling the distribution of the phytoplankton communities. As a result of this comprehensive pigment analysis, it was noted that the Diatoms were the predominant species with an average of 77.1% as noted located in Jinju Bay. It was discovered that during the summer season, the phytoplankton community composition was changed by a reduction of diatoms and noted increases of the Cryptophytes, Prasinophytes, and Dinoflagellates. Especially, it was noted that the Cryptophytes and Prasinophytes were shown with an average of 18.8% and 17.8% in June, respectively. However, it was revealed that the Cryptophytes and Prasinophytes were not shown by a microscopic observation. The phytoplankton community composition was correlated with the temperature and salinity variations as noticed in the Jinju Bay. Therefore, the water temperature and freshwater inputs in the Jinju Bay were important environmental factors for controlling the phytoplankton community composition and the varying Cryptophytes and the noted amounts of Prasinophytes as well.
This paper proposes quay crane (QC) scheduling algorithms that determine the working sequence of QCs over ship bays in a container vessel in automated container terminals. We propose two scheduling algorithms that examine the distribution of export containers in the stacking yard and determine the sequence of ship bays to balance the workload distribution among the yard blocks. One of the algorithms is a simple heuristic algorithm which dynamically selects the next ship bay based on the entropy of workloads among yard blocks whenever a QC finishes loading containers at a ship bay and the other uses genetic algorithm to search the optimal sequence of ship bays. To evaluate the fitness of each chromosome in the genetic algorithm, we have devised a method that is able to calculate an approximation of loading time of container vessels considering the workloads among yard blocks. Simulation experiments have been carried out to compare the efficiency of the proposed algorithms. The results show that our QC scheduling algorithms are efficient in reducing the turn-around time of container vessels.
