Thermal Effects on Bioaccumulation and Antioxidant Enzyme Response in the Liver and Gills of Tilapia Oreochromis niloticus Exposed to Arsenic

  • The effects of waterborne arsenic (As) exposure on bioaccumulation and antioxidant defenses were examined in the liver and gills of tilapia, Oreochromis niloticus, under thermal stress. Tilapia were exposed to different As concentrations (0, 200, and $400{\mu}g\;L^{-1}$) at three water temperatures (20, 25, and $30^{\circ}C$) for 20 days. After As exposure, higher levels of As accumulation were observed in the gills compared with the liver in elevated water temperatures. In terms of the antioxidant response, glutathione (GSH) levels and the activities of glutathione reductase (GR), glutathione peroxidase (GPx), and glutathione S-transferase (GST) significantly decreased in the liver and gills of tilapia exposed to As for 20 days, regardless of the As concentration (200 and $400{\mu}g\;L^{-1}$) or water temperature. These enzyme activities exhibited greater fluctuations in the liver and gills of tilapia after As exposure in water warmer than $20^{\circ}C$. The present findings suggest that the simultaneous stress of temperature change and As exposure can accelerate As accumulation and alter the antioxidant enzymes activities of tilapia.

Oyster mushroom extract protects antioxidant defence system in cisplatin induced nephrotoxicity in mice

  • Cisplatin is a highly effective and extensively used anticancer drug. Higher doses of cisplatin manifest acute nephrotoxicity and this is one of the limiting factors of this drug in cancer chemotherapy. The effect of the oyster mushroom extract to ameliorate cisplatin ( cis platinum (II) diammine dichloride) induced nephrotoxicity and restoration of antioxidant defence system in mice was investigated. The investigations showed that prior administration of methanolic extract of Pleurotus florida at a dose of 500 and 1000mg/Kg body weight significantly reduced elevated serum creatinine and urea levels and increased superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) activities in the kidney, consequent to cisplatin treatment, in a dose dependent manner. The extract restored the decreased reduced glutathione (GSH) activity and increased malondialdehyde (MDA) level due to cisplatin administration. The results thus indicated that oyster mushroom extract rendered significant protection against cisplatin induced nephrotoxicity and depletion of antioxidant defence system in a dose dependent manner. Since oyster mushrooms are excellently edible and non-toxic, the finding reported here is of significant use in cancer chemotherapy.

Antioxidant Activities of Isoflavones from the Rhizomes of Belamcanda chinensis on Carbon Tetrachloride-Induced Hepatic Injury in Rats

  • The present study was carried out to clarify whether tectorigenin and tectoridin isolated from the rhizomes of Belamcanda chinensis (Iridaceae) inhibit hepatic damage induced by carbon tetrachloride ($CCl_4$)-intoxication in rats by the experimental methods in vitro and in vivo. Tectorigenin and tectoridin exhibited a significant decrease in serum transaminase activities elevated by hepatic damage induced by $CCl_4$-intoxication in rats, as well as in a lipid peroxidation causing a significant decrease in malondialdehyde (MDA) production by thiobarbituric acid (TBA)-reactant assay. Both compounds also showed strong increase in the antioxidant enzymes such as hepatic cytosolic superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-px) activities in $CCl_4$-intoxicated rats. These results suggested that tectorigenin and tectoridin isolated from the rhizomes of B. chinensis possess not only the antioxidative, but also the hepatoprotective activities in $CCl_4$ -intoxicated rats.

Dietary Nigella sativa and Peganum harmala Oils Reverses Hyperglycaemia, Hepatotoxicity, and Metabolism in Rats

  • This study aims to evaluate the therapeutic action of administration of Nigella sativa (NS) and Peganum harmala (PH) oils in diabetes and hepatic toxicity. Results show that treatment of diabetic rats with NS oil or PH oil ameliorate hyperglycaemia induced stress oxidative and hepatic dysfunction in diabetic rats. Administration of NS or PH oil to diabetic rats caused an anti-diabetic and antioxidant activities by the decrease in plasmatic glucose level and increase in hepatic superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) activities, reduced glutathione (GSH) and glycogen contents compared to untreated diabetic rats. Besides, NS and PH oils protect the hepatic function observed by decrease of triglyceride (TG), total cholesterol (TCh), and increase of high density lipoprotein-cholesterol (HDL-Ch) levels in serum and hepatic tissues. Moreover, a diminution in the bilirubin, transaminase glutanic pyruvic (TGP), and transaminase pyruvic oxaloacetic (TPO) contents in serum and the thiobarbituric acid-reactive substances levels (TBARs) in hepatic tissues are also detected.

Antioxidant and Anti-Inflammatory Effects of NCW Peptide from Clam Worm (Marphysa sanguinea)

  • Clam worms (Marphysa sanguinea) are a rich source of bioactive components such as the antibacterial peptide, perinerin. In the present study, we explored the physiological activities of a novel NCWPFQGVPLGFQAPP peptide (NCW peptide), which was purified from clam worm extract through high-performance liquid chromatography. Tandem mass spectrometry (MS/MS) revealed that NCW was a new peptide with a molecular weight of 1757.86 kDa. Moreover, NCW peptide exhibited significant antioxidant effects, causing a 50% inhibition of DPPH radical at a concentration of 20 μM without showing any cytotoxicity. These were associated with a reduction in the activity of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) in LPS-stimulated RAW264. 7 cells. Furthermore, NCW peptide exhibited anti-inflammatory effects in LPS-stimulated RAW264.7 macrophages via inhibition of the abnormal production of pro-inflammatory cytokines including nitric oxide (NO), inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2). These anti-inflammatory effects of NCW peptide were associated with the inhibition of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α). Our results therefore suggest that this novel NCW peptide with antioxidant and anti-inflammatory effects could be a good therapeutic agent against inflammation-related diseases.

Effects of Swimming Training on Hepatic Antioxidant Enzyme and Serum Lipid in Cholestrol-Dietary Rats (유영운동이 콜레스테롤 식이 흰쥐에 있어서 간 조직의 항산화물질 및 혈액 성분에 미치는 영향)

  • The purpose of this study was to investigate effects of swimming training and cholesterol diet on the activation of hepatic antioxidant enzymes and serum lipid in Sprague Dawley rats(24 weeks of age). They were divided into five groups which were made up of normal-diet detraining group(C), 2% cholesterol-diet detraining group(CC), 2% cholesterol-diet swimming training groups which were classified according to their training time(CSA: 12min, CSB:8min, CSC:4min). They were given normal diet for the first 6weeks and then, separated normal-dietary and 2% cholesterol-dietary for 14 weeks. During these periods, 10 weeks’ swimming training was performed after 4 weeks later. And then we analyzed blood and liver by decapitating those rats. Swimming training showed a tendency to increase the activation of GSH-peroxidase, Nonprotein-SH and malondialdehyde, and decrease total- cholesterol, LSL-C/HDL-C and VLDL significantly. Whereas, cholesterol diet which has no training showed decrease the activation of hepatic antioxodant enzymes, and increase total-cholesterol and LDL-C/HDL-C absolutely. These results suggest that swimming training should stimulate the activation of hepatic antioxidant enzymes and decrease total-cholestrol even if they had cholesterol diet.

Effects of Ovariectomy and Calcium Intake on Lipid Profile and Antioxidant Capacities in Rats (난소절제와 칼슘 섭취수준이 흰쥐의 지질패턴과 항산화능에 미치는 영향)

  • The purpose of this research was to investigate the effect of the calcium intake on lipid profile and antioxidant capacities in ovariectomized (OVX) rats. Rats were divided into 3 groups and fed diet with different levels of calcium (low 0.1%, adequate 0.5%, high 1.5%) for 4 weeks. The half of rats in each group was ovariectomized and the others were sham-operated. And rats were fed same diets for 8 weeks after operation. Feed intake and weight gain were significantly higher in OVX group than those in sham-operated. Serum HDL-cholesterol was the highest in high-calcium group of OVX. Hepatic triglyceride of low-calcium group in sham-operated was the highest, while that of highcalcium group in OVX was the highest. Hepatic activities of glutathione-S-transferase (GST), glutathione peroxidase (GSH-Px), and catalase were significantly decreased by increasement of calcium intake. Hepatic TBARS level was the lowest in high-calcium group of OVX. And hepatic level of TBARS induced by AAPH was significantly decreased by increasement of calcium intake. These results may indicate that the high calcium intake have the potential role to improve lipid profiles and antioxidant capacities in OVX rats.

Effect of Dietary Selenium Binding Yeast Peptide on Growth Performance, Tissue Se, Serum Glutathione Peroxidase Activity and Meat Quality in Finishing Pigs (비육돈에 있어서 Selenium Binding Yeast Peptide의 첨가가 생산성, 조직내 Se함량, 혈청내 GSH-Px의 활성 및 돈육의 품질에 미치는 영향)

  • This study was conducted to evaluate the effects of selenium binding yeast peptide supplementation on growth performance, tissue Se, serum glutathione peroxidase activity and meat quality in finishing pigs. A total of eighty (Duroc${\times}$Yorkshir${\times}$Landrace) pigs (82.88$\pm$1.23 kg average initial body weight) were used in a 35-day assay. Dietary treatments included 1) CON (basal diet), 2) SY1 (CON diet+0.05% selenium binding yeast peptide), 3) SY2 (CON diet+0.l% selenium binding yeast peptide) and 4) SY3 (CON diet+0.2% selenium binding yeast peptide). Overall period, average daily gain of pigs fed selenium binding yeast peptide diet was higher than that of pigs fed CON diet, however, there was not significant difference (p>0.05). L* (lightness) value of M. longissimus dorsi was higher in SY2 than CON and SY3 (p<0.05). a* (redness) value of M. longissimus dorsi was lower in CON than other treatments (p<0.05). Selenium content in serum was increased as adding selenium binding yeast peptide compared to pigs fed CON diet. However, there was not significantly different among the treatments (p>0.05). Selenium content of M. longissimus dorsi was higher in SY2 (0.021 $\mu$g/g) and SY3 (0.031 $\mu$g/g) than CON diet (0.008 $\mu$g/g) (p<0.05). Selenium content of kidney was increased in SY2 I and SY3 compared to pigs fed CON and SY1 (p<0.05). Selenium content of liver was higher in SY1 than CON (p<0.05). In conclusion, it is suggested that selenium content could be accumulated in M. longissimus dorsi, kidney and liver by selenium binding yeast peptide supplementation, and meat color of M. longissimus dorsi could be affected by selenium binding yeast peptide supplementation.

In Vitro Hepatoprotective Effects of Fermented Curcuma longa L. by Aspergillus oryzae against Alcohol-Induced Oxidative Stress (알코올성 산화적 손상에 대한 발효울금의 간세포 보호 효과)

  • Protective effects of fermented Curcuma longa L. (CL) against alcoholic liver damage were investigated in HepG2/2E1 cells. Fermented CL was extracted by cold water (FCC), hot water, 80% ethanol, and methanol. Of the four extracts, the strongest hepatoprotective effect against ethanol-induced oxidative stress was observed in FCC. Pretreatment with FCC also reduced intracellular reactive oxygen species formation compared to ethanol-alone treated cells. FCC also enhanced catalase, glutathione-S-transferase, glutathione reductase, glutathione peroxidase, and superoxide dismutase, and non-enzymatic antioxidative activities such as glutathione compared to alcohol-treated HepG2/2E1 cells. Our findings suggest that FCC might be considered as a useful agent in the prevention of liver damage induced by oxidative stress by increasing the antioxidant defense mechanism.

Changes in Lipid Peroxidation Level and Antioxidant Enzyme Activities of Rats Supplemented with Dietary Cholesterol and/or Taurine (콜레스테롤 및 타우린 첨가식이가 흰쥐 혈장과 간의 지질과산화물 농도와 항산화효소 활성에 미치는 영향)

  • Effects of dietary cholesterol and/or taurine supplementation on plasma and hepatic lipid peroxidation status and antioxidant enzyme activities were evaluated in rats fed one of the following semisynthetic diets for 5 weeks: control diet (CD, cholesterol-free and taurine-free diet); high cholesterol diet (HCD, CD+1.5% cholesterol): high taurine diet (HTD, CD+1.5% taurine): high cholesterol and high taurine diet (HCHTD, HCD + 1.5% taurine). Plasma malondialdehyde (MDA) level was not influenced by dietary cholesterol or taurine supplementation, while hepatic MDA level was 70% higher in rats fed HCD compared to the value for CD rats (p<0.05). Our observation that taurine supplementation significantly decreased the hepatic MDA level of rats fed HCD, but failed to decrease lipid peroxidation of rats fed CD indicates that the protective effect of taurine in the liver against lipid peroxidation is manifested only under the hypercholesterolemic environment. Plasma and hepatic glutathione peroxidase (GSH-Px) activities were not affected by dietary supplementation of cholesterol or taurine. However, hepatic superoxide dismutase (SOD) activity was significantly reduced by dietary taurine supplementation (p <0.05), and thus significantly lower in rats fed HTD compared to the value for CD (p<0.05). Plasma total cholesterol concentration was positively correlated with hepatic cholesterol concentration as expected (r=0.712, p<0.001). Plasma (r=0.399, p<0.05) and hepatic cholesterol levels (r=0.429, p<0.05) showed a significantly positive correlation with hepatic MDA concentration, respectively. Plasma taurine concentration was negatively correlated with hepatic SOD activity (r=-0.481, p<0.01), and tended to be negatively correlated with hepatic GSH-Px activity without showing statistical significance (r=-0.188, p<0.05). These results indicate an antioxidative effect of taurine in rats with elevated level of lipid Peroxidation due to high intake of dietary cholesterol. Future application of taurine as a safe candidate for a hypolipidemic agent without adversely affecting body's antioxidant defense system is speculated.
