Soil properties in Panax ginseng nursury by parent rock (모암별 인삼묘포지의 토양특성에 관한 연구)

  • A research has been done for growing characteristics of Korean ginseng in Geumsan of Chungnam Province. It had been made to determine the transitional element concentrations of the rocks, divided by biotitic granite(GR) and phyllite(PH). The physical and chemical properties of their weathering soils and ginseng nursery soils were analyzed. The texture in the GR weathering and ginseng nursery soils were sandy clay, and the texture of the PH weathering and ginseng nursery soils were heavy or silty clay. The bulk densities of the GR and PH weathering soils were $1.21{\sim}1.32g/cm^3$ and $1.26{\sim}1.38g/cm^3$, respectively. Also, the bulk densities of the GR and PH ginseng nursery soils were $1.02{\sim}1.10g/cm^3$, respectively. The pH (4.80) of the GR weathering soil were lower than the pH of the PH(5.34) weathering soil. The pH in the 2 year and 4 year-ginseng nursery soil of the GR were 4.39 and 4.40. In addition, those of the PH were 5.24 and 5.34, respectively. The difference in pH of the two nursery soils could be from the pH difference between the two parent materials. The organic matter contents of the GR weathering soils(0.24%) were higher than those of the PH(1.02%) weathering soils. The organic matter of the 2 and 4 year-ginseng GR nursery soils were 0.87% and 1.52%, and of the PH nursery soils were 2.06% and 2.96%, respectively. The total nitrogen contents of the GR weathering soils were 259.43ppm and of the PH weathering soils were 657.22ppm. Those of 2 and 4 year-ginseng GR nursery soils were 588.04ppm and 657.22ppm and those of the PH nursery soils were 1037.72ppm and 1227.96ppm, respectively. The nitrate and ammonium contents of the GR weathering soils were the extremely small, and those of the PH weathering soils were 6.7ppm and 9.94ppm. Those of 2 year-ginseng GR nursery soils(223.09ppm and 26.96ppm) were higher than those of PH(19.46ppm and 8.23ppm) nursery soils. And those of 2 year-ginseng PH nursery soils(14.22ppm and 16.84ppm) were lower than those of PH(306.93ppm, 34.21ppm) nursery soils. The difference was due to fertilizer types and more deposits of nitrate after oxidation of ammonium. The phosphate contents of the GR and PH weathering soils were 14.41ppm and 38.60ppm. Those of GR 2 and 4 year-ginseng nursery soils were 46.89ppm and 102.44ppm and those of the PH nursery soils were 147.04ppm and 38.60ppm. The cation exchange capacities of the GR weathering soils were 12.34me/100g and those of the PH weathering soils were 15.40me/100g. Those of 2 and 4 year-ginseng GR nursery soils were 15.80me/100g and 7.70me/100g and those of PH nursery soils were 12.14me/100g and 12.83me/100g. All of exchangeable cation($K^+$, $Ca^{2+}$, $Mg^{2+}$, $Na^+$) contents in the nursery soils were higher than those in the weathering soils. The $SO_4{^2-}$ contents of the weathering soils in both of the GR(5.98ppm) and PH(9.94ppm) were higher than those of the GR and PH ginseng nursery soils. The $Cl^-$) contents of the GR and PH weathering soils were a very small and those of the nursery soils(2-yr GR: 39.06ppm, 4-yr GR: 273.43ppm, 2-yr PH: 66.41ppm, 4-yr PH: 406.24ppm) were high because of fertilizer inputs.

A Study on the Effect of Herbal-acupuncture with Gentianae macrophyllase Radix Solution at Joksamni(ST36) on Collagen-induced Arthritis DBA/1J mice (족삼리(足三里) 진구약침이 생쥐의 Collagen-induced Arthritis에 미치는 영향(影饗))

  • Objectives & Methods : The purpose of this study is to observe the effects of Gentianae macrophyllase Radix herbal-acupuncture solution(GR-HAS) at Joksamni(ST36) on Collagen-induced arthritis(CIA) in DBA/1J mice. The author performed several experimental items to analyze arthritis evaluation, change of weight, spleen size and adhesion rate, change of cytokine level, IgG, IgM and anti-collagen II , change of immunocyte count, histological change of CIA mouse joint related with RA. Results : 1. In the GR-HA group, the arthritis index, the incidence of arthritis and joint edema were significantly decreased. 2. In GR-HA group, there were no weight loss and similarly maintained as normal group. Spleen size, adhesion rate and the edema and transformation of knee joint were low and similarly maintained as normal group. 3. In the GR-HA, cytokine level, IgG, IgM and anti-collagen II were significantly decreased. 4. In the GR-HA, the change of immunocyte count were similarly maintained as normal group. 5. In the histological changes of the CIA mouse joint, the cartilage destruction and synovial cell proliferation were decreased in the GR-HA(Hematoxylin and eosin stain). The collagen fiber expressions in the GR-HA were similar with that of the normal group(Masson's Trichrome stain) Conclusions : These results suggest that GR-HA at ST36 has an effect to control immune reaction and suppress inflammation, synovial cell proliferation and cartilage destruction in rheumatoid arthritis.

Changes of Growth and Antioxidative Enzyme(SOD, APX, GR) Activities of Spinach Beet(Beta vulgaris var. cicla) Under Saline Condition (염 환경하에서 근대(Beta vulgaris var. cicla)의 생장과 항산화효소(SOD, APX, GR)의 활성변화)

  • Antioxidative enzymes (superoxide dismutase; SOD, ascorbate peroxidase; APX, glutathione reductase; GR) play major roles in scavenging mechanism of reactive oxygen species which were involved in various stress conditions including salt. In order to investigate the relation between their growth responses (dry weight) and the changes of antioxidative enzymes activity, salt-tolerant spinach beet having 15cm of shoot length were treated with various salt levels (0, 50, 200, 1000 mM NaCl) for 24 hours. Spinach beet exhibited an increase in the activity of antioxidative enzymes by salt, the maximal activity at 200 mM NaCl and the lowest activity at 50 mM NaCl in 2 hrs. after treatments. As a result of PAGE, it has been confirmed that spinach beet contained 3 isoforms (Fe-SOD, CuZn-SOD and Mn-SOD) of SOD and main isoform was CuZn- SOD form. In case of APX, isoforms of the low molecular weight(No. 7, 8) were showed strong expression especially at 200 and 400 mM NaCl treatment. Meanwhile, GR did not show specific pattern of isoforms among the salt treatments. Especially, in case of 50 mM treatment, plant showed the lowest activity of SOD with the best growth, a low enzyme activity was induced by inactivation of the Mn-SOD. Therefore, we suggested that the decrease of SOD activity at a low salt level (50 mM NaCl) or the increase of enzyme activity at a high salt level (200 mM NaCl) may be related to expression of the Mn-SOD isoform. These antioxidative enzymes showed the increase of activity in a short time by salt addition. So, it is considered that spinach beet copes effectively with a stressful condition such as salt by operating effective antioxidative defense mechanism rapidly under high salt level.

Study on Antioxidant and Staminal Activities of Kejihongsamatang (계지홍삼탕의 항산화 및 지구력 효과에 대한 연구)

  • KJHST (Kejihongsarntang) is a modified oriental prescription that consists of five herbs such as Ginseng Radix rubra Koreana, Atractylodis Rhizoma, Zingiberis Rhiaoma, Cinnamomi Ramulus and Glycyrrhizae Radix. For the evaluation of antioxidant and staminal activities of KJHST (Kejihongsarntang), the study was done in comparison of Ginseng Radix rubra (GR). For the antioxidant study, KJHST inhibited hemolysis of erythrocyte and decolored the DPPH(2,2-diphenyl-1-picrylhydrazyl) free radical in a dose depenrlent manner more effectively than GR alone in vitro. KJHST and GR significantly suppressed the time course (1 hr∼6 hr)-level of MDA (malondialdehyde) following AAPH (2,2'-azo-bis-(2-amidino-propane) dihydrochloride) treatment in vivo as compared with control data with no statistical difference. From the evaluation of stamina by swimming test GR and KJHST significantly increased the swimming time in a time and dose dependent manner as compared with control data, while GR was more effective than KJHST in 2 weeks after treatment, though KJHST was more effective than GR at low dose (25 m/kg) 4 weeks after treatment. From the results it can be concluded GR and KJHST had antioxidant and staminal activities.

Soil buffer capacities from the differrent host rocks by the treatment of artificial acid precipitation

  • To investigate the weathering soil buffering capacities of the artificial acidic precipitation, the weathering soils and their leachate solutions were sampled from the host rocks(granite;GR, rhyolite;RH, gabbro;GA, basalt;BA, two serpentinite;SE1, SE2 and limestone;LI) and analyzed for pH and chemical properties. 1n the soil pH of the GR and RH ,the acidic rocks, were 5.02 and 5.95, respectively. And the GA and BA, basic rocks, were 6.52 and 7.57. The SE1 and SE2 were 8.90 and 8.89. While the LI was 7.84. These results means the typical soil pH properties by host rocks. After the artificial acidic precipitation input 5OOml, the average changes of soil leachate solutions treated by pH levels(pH 5.0, 4.0 and 3.0), were pH 5.73, 5.00 and 4.40. in GR soil, and pH 6.19, 5.99 and 5.57 in RH. GA were pH 6.31, 6.04 and 5.86, BA were pH 7.05, 6.85 and 6.56 and SE1 were pH 8.31, 8.26 and 7.71. SE2 were pH 8.29, 8.24 and 7.96. LI were pH 7.55, 7.46 and 6.79. The soil leachate pHs from volcanic rocks were higher than those from the plutonic rocks and GR soils showed greater response than other soils. With increasing 100ml input-solution, the soil leachate pHs were mainly decreased. Cation concentrations, CEC, EC and total nitrogen concentrations of RH and BA soils, the volcanic rocks, were higher than those of GR and GA soil, the plutonic rocks. On the contrary, Al concentrations of the GR and GA soils were higher than those of RH and BA soils, partly because of high quartz content in GR and Al content in the biotite and plagioclase in GA.

  • The present study investigated the expression of glucocorticoid receptor (GR) mRNA as a stress response during salinity changes (35, 10, and 0 psu) and water temperature changes (from $20^{\circ}C$ to $30^{\circ}C$, $1^{\circ}C$/day) in black porgy. We cloned the full-length GR cDNA from the kidney and examined its expression in the gill, kidney, and intestine by quantitative real-time PCR (QPCR) during salinity and water temperature changes. During salinity changes, the levels of GR mRNA in the gill, kidney, and intestine were highest at 0 psu, and the levels of plasma cortisol and glucose were elevated, but triiodothyronine ($T_3$) decreased. Also, during water temperature changes, the levels of GR mRNA in the gill, kidney, and intestine increased at $30^{\circ}C$. Plasma parameters also increased with an increase in water temperature. Therefore, this upregulation of GR mRNA was a good indicator of stress, such as those resulting from changes in salinity and water temperature.

  • Mercury (Hg) poses a threat to marine ecosystem due to continuous inflow from various industries and bioaccumulation to higher trophic level via food web. Mercury can adversely affect growth, development, reproduction and metabolism to aquatic organisms. In the present study, acute toxicity and oxidative stress markers (total glutathione content, and activities of GST, GR and GPx) were investigated in brackish water flea Disphanosoma celebensis exposed to HgCl2 for 24 h. As results, Hg showed negative effect in survival of D. celebensis. 24 h-LC50 value was determined as 0.589 mg/l (95% C.I. 0.521~0.655 mg/l). After exposure to Hg (0.08 and 0.4 mg/l) for 24 h, total glutathione content was significantly decreased, whereas GST, GPx and GR activities were enhanced. These findings indicate that Hg induced oxidative stress in D. celebensis, and oxidative stress markers may be involved in cellular defense against Hg - mediated toxicity. This study provides a better understanding of molecular mode of action of Hg toxicity in this specie and potent of molecular markers for heavy metal monitoring in marine ecosystem.

  • For the evaluation of protective effect on hepatic damage by Korean red ginseng mixed formula, we used GR(Korean red ginseng), GRF-A(Korean red ginseng-mixed formula) as a materials. The study was performed on protective effect against hepatic damage induced by $CCl_4$. In vitro assay with 1.1 mM galactosamine, protection(%) was 44%(GR), 58%(GRF-A) at 50ug/ml, while maxium protection(%) was 5%(GR) and 24% (GRF-A) against acute hepatotoxicity by $CCl_4$. GRF-A significantly protected ethanol induced-liver damage by lowering ALT and ALP and fatty degenertion in liver tissue.

Outcomes of Preoperative Chemoradiotherapy and Combined Chemotherapy with Radiotherapy Without Surgery for Locally Advanced Rectal Cancer

  • Purpose: To evaluate the treatment outcomes of patients with locally advanced rectal cancer treated with preoperative concurrent chemoradiotherapy (CCRT) or combined chemotherapy together with radiotherapy (CMT-RT) without surgery. Materials and Methods: A total of 84 patients with locally advanced rectal adenocarcinoma (stage II or III) between January $1^{st}$, 2003 and December $31^{st}$, 2013 were enrolled, 48 treated with preoperative CCRT (Gr.I) and 36 with combined chemotherapy and radiotherapy (CMT-RT) without surgery (Gr.II). The chemotherapeutic agents used concurrent with radiotherapy were either 5-fluorouracil short infusion plus leucovorin and/or capecitabine or 5-fluorouracil infusion alone. All patients received pelvic irradiation. Results: There were 5 patients (10.4%) with a complete pathological response. The 3 year-overall survival rates were 83.2% in Gr.I and 24.8 % in Gr.II (p<0.01). The respective 5 year-overall survival rates were 70.3% and 0% (p<0.01). The 5 year-overall survival rates in Gr.I for patients who received surgery within 56 days after complete CCRT as compared to more than 56 days were 69.5% and 65.1% (p=0.91). Preoperative CCRT used for 12 of 30 patients in Gr.I (40%) with lower rectal cancer demonstrated that in preoperative CCRT a sphincter sparing procedure can be performed. Conclusions: The results of treatment with preoperative CCRT for locally advanced rectal cancer showed comparable rates of overall survival and sphincter sparing procedures as compared to previous studies.

Effect of Sinhyo-Taklee-San on the Proliferation of Fibroblast Cell(Balb/c 3T3) (섬유아세포(纖維芽細胞)(Balb/c 3T3)의 증식(增殖)에 미치는 신효탁리산(神效托裡散)의 영향(影響))

  • The studies were conducted to investigate the effect of Sinhyo-Taklee-San(STS), which is composed of Astragali Radix(AR), Lonicerae Flos(LF), Angelicae gigantis Radix(AGR) and Glycyrrhizae Radix(GR), on the proliferation of fibroblast cell(Balb/c 3T3). STS, GR and glycyrrhizin increased the proliferation of 3T3 cells. The 10% serum obtained from STS, AR, LF, AGR and GR treated mice also increased the proliferation of 3T3 cells markedly. GR, glycyrrhizin and glycyrrhetinic acid inhibited protein synthesis, but did not affect on DNA synthesis.
