Oxidative Stress in Ovariectomy Menopause and Role of Chondroitin Sulfate

  • Oxidative stress due to reactive oxygen species (ROS) can cause oxidative damage to cells. Cells have a number of defense mechanisms to protect themselves from the toxicity of ROS. Mitochondria are especially important in the oxidative stress as ROS have been found to be constantly generated as an endogen threat. Mitochondrial defense depends mainly on super-oxide dismutase (SOD) and glutathione peroxidase (GPx), whereas microsomal defense depends on catalase (CAT), which is an enzyme abundant in microsomes. SOD removes superoxide anions by converting them to $H_2O$$_2$, which can be rapidly converted to water by CAT and GPx. Also, GPx converts hydroperoxide (ROOH) into oxidized-glutathione (GSSG). Ovariectomized (OVX) rats are used as an oxidative stress model. An ovariectomy increased the levels of MDA, one of the end-products in the lipid peroxidative process, and decreased levels of the antioxidative enzymes; SOD, CAT and GPx. However, Chondroitin sulfate (CS) decreased the levels of MDA, but increased the levels of SOD, CAT and GPx in a dose-depen-dent manner. Moreover, inflammation and cirrhosis of liver tissue in CS- treated rats were sig-nificantly decreased. These results suggest that CS might be a potential candidate as an anti oxidative reagent.

어성초 추출물의 ICR생쥐와 L1210 세포에 대한 항암작용 및 SOD, GPx 효소활성변화 (Anticancer Effect of Houttuynia cordata Extract on Cancered ICR Mouse and L1210 Cells With Changes of SOD and GPx Activities)

  • The present investigation was undertaken to examine the anticancer activity of the methanol extract from Houttuynia cordata on ICR mouse with induced abdominal cancer and L1210 cancer cells. When the methanol extract of Houttuynia cordata (10∼200 $\mu\textrm{g}$/$m\ell$) was administered orally to ICR mouse with abdominal cancer, 47.8% of the best life prolonging effect was obtained. In case of cytotoxicity study (inhibition of cell proliferation) of Houttuynia cordata extract against L1210 cells, $IC_{50}$/ was found to be 62.8 $\mu\textrm{g}$/$m\ell$. In contrast to such considerable toxicity against cancer cell line, the toxicity demonstrated by the identical extract against normal lymphocytes was very meagre as shown to be < 5% compared with 86.5% in case of L1210 cells at the same condition. To get an insight into the reaction mechanism undelying the anticancer activity, $O_2$ion quantity and antioxidant enzyme activities such as superoxide dismiutase (SOD) and glutathione peroxidase (GPx) of L1210 cells in the presence of Houttuynia cordata extract were measured. The increased values of SOD and GPx enzyme activities in addition to the augmented generation of $O_2$ ion in L1210 cells implied that the reactive oxygen species induding $O_2$ion which were presumably induced by Houttuynia cordata extract might have participated in the process of L1210 cells cytotoxicity.

  • 우리나라의 주요 해산양식어종인 돌돔, Oplegnathus fasciatus을 대상으로 30, 60, 120 및 240 mg/kg의 아연을 40일 동안 경구 투여에 따른 아가미와 간의 항산화효소 활성의 변화를 검토하였다. 돌돔 간의 superoxide dismutase(SOD) 활성은 30~240 mg/kg 아연농도, glutathione peroxidase (GPx) 활성은 30~120 mg/kg의 아연농도에서 유의적으로 증가하였다. 돌돔 아가미의 SOD 및 Glutathione(GSH) 활성은 120 및 240 mg/kg 아연농도, GPx활성은 60~240 mg/kg의 아연농도에서 유의한 증가가 관찰되었다.

  • 본 연구에서는 호랑나비의 용기 동안 혈림프, 지방체, 표피, 큐티클 및 중장에서 항산화효소의 활성을 조사하였다. 혈림프와 지방체에서 항산화효소 활성의 변화가 두드러지게 나타났다. SOD, CAT 및 GST의 활성은 높은 활성을 보인 반면, GPX와 GR은 상대적으로 활성이 매우 낮으므로 곤충의 변태기 동안 항산화과정에서 이들 효소의 역할은 매우 미미할 것으로 생각된다. 더불어 CAT의 활성은 대부분의 조직에서 용화 직후에 높은 활성을 보이며 상대적인 활성도 매우 높게 나타나고 있어 과산화수소의 분해에는 GPX보다는 CAT가 주로 관여할 것으로 생각된다. 또한 GPX와 GR의 활성보다는 GST의 활성이 전 조직에서 비교적 높게 나타나는 것으로 보아 lipid peroxidation을 통한 항산화 과정에도 GPX보다는 주로 GST가 관여할 것으로 생각된다.

황해쑥 추출정제물 SD-994의 L1210암세포에 대한 세포독성과 항산화효소의 유발 (Cytotoxicity of SD-994 from Artemisia argyi against L1210 Cells with Concomitant Induction of Antioxidant Enzymes)

  • SD-994 was prepared from methanol extract of Artemisia argyi by stepwise purification of solvent partioning and silica gel chromatography. In the course of this purification, fractions obtained at each step were investigated for their cytotoxicities against L1210 cells. Fractions A~G prepared from chloroform fraction showed considerable cytotoxicities raging 40~90% against L1210 cells. Subfractions I~IX obtained from fraction A exhibited various cytotoxicities and subfraction I (SD-994) was found to be the most effective compound. $IC_{50}$ values of SD-994 were measured to be $0.5{\;}{\mu\textrm{g}}/ml and less than $0.05{\;}{\mu\textrm{g}}/ml against L1210 cells and normal lymphocytes, respectively: When SD-994 was added to L1210 cell as cytotoxic agent, significantly increased amount of superoxide ($O_2^-$) and dramatically augmented activities of superoxide dismutase (SOD), specially MnSOD and glutathione peroxidase (GPx) were observed according to the concentration and incubation time. Whereas, in case of normal lymphocytes under the same condition, cytotoxicities were not apparent and the generation of superoxide ($O_2^-$) or the activity changes of SOD and GPx were insignificant. These results together indicate that the cytotoxic action of SD-994 against L1210 cell may be achieved via necrosis and/or apoptosis induced by reaction oxygen species which could not probably be completely abolished even by drastically increased antioxidant enzymes, SOD and GPx activities.

미강 함유 Tocotrienol의 항산화 효과 (Antioxidant Effects of Tocotrienol in Rice Bran)

  • 미강으로부터 정제된 tocotrienol은 DPPH를 기질로 확인한 결과 매우 뛰어난 항산화력을 가지는 것으로 판명되었다. 또한 정상세포와 암세포를 배양하면서 tocotrienol을 처리하고 세포내의 항산화에 가장 큰 역할을 하는 superoxide dismutase와 glutathione peroxidase 활성을 측정한 결과 두 효소 모두 tocotrienol에 의하여 활성이 증가되는 것을 볼 수 있었으며, 전체적으로 암세포에서 GPX가 SOD보다 더 민감하게 증가함을 알 수 있었다.

쥐의 대두 단백질 섭취가 국소 뇌허혈/재관류 후 뇌경색 크기와 항산화효소 활성도에 미치는 영향 (Effect of Dietary Soybean Protein on Cerebral Infarction Size and Antioxidant Enzyme Activities in Rat Focal Brain Ischemia Model)

  • Purpose: The purpose of this study was to investigate the cerebral infarction size, antioxidant enzyme activities and lipid peroxidation changes after 6 weeks of dietary soybean protein intake in a rat focal brain ischemia model. Method: Weaning Sprague-Dawley rats were fed with either modified AIN-93G diet containing casein 20% (control), 20% soybean protein isolate-based diet (S20), or 40% of soybean protein isolate-based diet (S40) for 6 weeks. The animals were subject to right middle cerebral artery occlusion for 2 hr. After 24 hr of recirculation, the rats were sacrificed. Antioxidant enzymes activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx) and thiobarbituric acid reactive substance (TBARS) level in the right brain were also measured. Result: There were no significant differences in the right cortical infarction volume, TBARS level, SOD and CAT activities among the three groups whereas the GPx activities of the S20 group were significantly higher than those of the control group (p=.02). Conclusion: Our results suggest that 20% of soybean protein may have a modulating effect on GPx and possibly have some protective effect against oxidative stress although it may enough to decrease cerebral infarction volume in rat focal brain ischemia model.

Correlations of Physical Fitness Factors, Antioxidant Enzymes, Lipid Peroxidation, Lipid Profiles, Lactate Levels and Cardiovascular Variables in an Exercising Group and Controls

  • Purpose: This study was designed to investigate correlations between physical fitness, antioxidant enzymes (SOD, CAT, GPX), lipid peroxidation levels (MDA), lipid profiles, lactate levels and cardiovascular variables in an exercising group and a control group. Methods: Twelve healthy young males (Exercise group: 6, Controls: 6). All subjects took physical fitness tests and blood samples were collected while subjects were resting. Results: In the exercise group, there were several significant correlations: between back strength and SOD enzyme levels (r=0.82, p=0.04), back strength and MDA (r=0.94, p=0.00), agility and GPX (r=0.81, p=0.04), and balance and GPX (r=0.81, p=0.04). In the control group, there were significant correlations between: dominant grip strength and MDA (r=-0.84, p=0.03), and agility and GPX (r= -0.82, p=0.04). In the exercise group, there were no significant correlations between physical fitness factors, TC, TG, HDL-C and lactate levels. In the control group, there were significant correlations between: back strength and TG (r=0.88, p=0.01), and agility and HDL-C (r= -0.84, p=0.03). In the exercise group, there were significant correlations between: non-dominant grip strength and SBP (r=0.94, p=0.00), dominant grip strength and SBP (r=0.85, p=0.03), and power and SBP (r=0.82, p=0.04). In controls, there were significant correlations between: dominant grip strength and DBP (r=-0.85, p=0.03), muscular endurance and ST level (r=-0.93, p=0.00), and muscular endurance and HR (r=-0.88, p=0.01). Conclusion: That cardiovascular patients and controls who participated in regular exercise maintained their antioxidant capacity suggests that long-term physical activity can counteract the negative dysfunction that characterizes sedentary lifestyle, probably by maintaining plasma antioxidant defenses and thereby preventing oxidative stress.

고 pH 돈육 내의 항산화 기작 (Factors Affecting Lipid Oxidation Development in High pH Ground Pork)

  • 돈육의 pH가 5.0에서 7.0으로 증가할수록 저장중 산패치는 유의차 있게 감소하였다. 본 실험 결과 고 pH 돈육의 낮은 유리철 함량과 높은 GPx 활성은 저장중 돈육의 산패 억제에 관여하는 중요한 요인들인 것으로 밝혀졌다. 그러나 pH 조절에 의한 산패 억제에 관여할 수 있는 그 밖의 다른 가능한 인자들을 찾는 연구 또한 중요하다고 하겠다. 나아가서 유리철 ion의 방출 억제나 불활성화 방안, GPx의 또 다른 활성화 방안 및 천연항산화제로서의 활용 가능성등이 계속 연구, 검토되어야 할 것이다. 본 연구의 결과는 돈육의 저장성 증진은 물론, 규명된 항산화 기작들을 다른 작품 system에 응용하는 데에도 이용될 수 있을 것으로 본다.

고체상 추출과 친화 크로마토 그라피-유도결합 플라즈마 질량분석법을 이용한 한국인 혈청에서의 셀레노 단백질 정량 (Quantification of seleno proteins in Korean blood serum using solid phase extraction and affinity chromatography-inductively coupled plasma/mass spectrometry)

  • 본 연구에서는 음이온 고체상 추출법 (AE SPE; anion exchange solid phase extraction)을 사용하여 간섭요인을 제거한 후, 친화 크로마토그래피 AF HPLC; affinity high performance liquid chromatography)와 유도결합 플라스마 질량분석법 (ICP/MS; inductively coupled plasma/mass spectrometry)을 사용하여 한국인의 혈청에서의 셀레노 단백질을 분리하고 정량하였다. 먼저 동위원소 희석법으로 셀레늄 총량을 분석한 결과, 건강한 한국 사람의 혈청내 평균농도는 $94.3{\pm}2.3ngg^{-1}$ 이었다. AE SPE와 AF 컬럼을 online으로 연결하여 셀레노단백질인 glutathione peroxidase (GPx), selenoprotein P (SelP), selenoalbumin (SeAlb)을 분리하고, 후 컬럼 동위원소 희석법 (PC ID; post column isotope dilution)으로 정량하였다. 혈청 인증표준물인 BCR-637을 분석한 결과 전체 셀레노 단백질의 합은 $85.4{\pm}3.4ngg^{-1}$으로 문헌값인 $81{\pm}7ngg^{-1}$과 일치하는 결과를 얻을 수 있었다. 20 명의 건강한 한국인의 혈청에서 얻은 셀레노 단백질 GPx, SelP 및 SeAlb 의 농도는 각각 $12.1{\pm}1.4ngg^{-1}$, $57.2{\pm}2.0ngg^{-1}$, 그리고 $20.0{\pm}1.9ngg^{-1}$ 이었으며 이들의 합인 $89.3ngg^{-1}$은 셀레늄의 총량값인 $94.3ngg^{-1}$과 거의 같은 값으로 혈청에서의 셀레늄은 주로 셀레노 단백질로 구성되어 있음을 알았다. 이 중 GPx의 농도는 간섭을 제거하기 전인 $25.0ngg^{-1}$에 비해 50% 이상 크게 감소하였는데 이로서 간섭은 주로 GPx에 포함되어 있음을 확인할 수 있었다. AE SPE는 간섭요인인 Cl과 Br을 제거 하는데 매우 효과적임을 보여주었다.
