Effects of Tea Fungus/Kombucha Beverage on Growth, Serum Protein Level and Enzyme Activities in Male Rats (발효홍차가 흰쥐의 성장률, 혈청 단백질농도 및 효소활성에 미치는 영향)

  • The current study was undertaken to determine the prolinged effects of fasting-refecding on the lipoprotein lipase (LPL) activity and lipogenesis of adipose tissus in male Sprague-Dawley rals and to investigate the effects of various degrecs of food restriction during refeeding on the LPL activity and lipogenesis. The control group (n=5) was fed ad libitum and killed in the fed state at the beginning of the experiment. All rats except conteol group were fasted for 2 days (n=50). Five rats were killed at the end of fasting and others (n=45) were refed either ad libitum (ad libitum group) or mildly restricted (20% food restricted group), or esverely restricted diet (40% food restricted group). Rats were killed on the day of 7th, 14th, and 21st of refeeding. Lipogenesis was determined by the amount of glucose converted to the total lipid. Body weight and epididymal adipose tissue weight returned to control states by 5 days in ad libitym group and by 14 days in 20% food restricted group. As expected, in 40% food restriction during refeeding weight and epididymal adipose tissue weight did not rcturn to control states until day 21. On day 21 after refeeding, the serum total cholesterol concentration of ad libitum group was significantly (p<0.05) higher than that of control group. The serum HDL-C concentration of 40% food restricted group during refeeding was significantly (p<0.05) higher than that of control group. However, there were no significant dif-ferences in serum HDL-C/total cholesterol (TC) ratio and trigluceride concentration among the groups. Fasting for 2 days decreased lipogenesis and LPL activity (p<0.05). On day 21 after refecding, the lipogencsis of ad libitum group was significantly (p<0.05) lower than that of control group. The lipogenesis of 40% food restricted group during refeeding was significantly (p<0.05) higher than that of conrtol group. Ad libitum group and 20% food restricted group during refecding allowed heparin-releasable (HR) LPL or total extractable (TE) LPL activity to return to control states. 40% food restricted group during refeeding delayed the return of HR-LOL or TE-LPL activity to return to control states until day 21 of refeeding. These results suggest that food restriction during refeeding can partially or completely prevent the overshoot of LPL activity, and this may influence the rate of lipid accumulation in adipose tissue during refeeding.

A Study on Vitamin A Levels in Measles Patients (홍역 환아의 비타민 A치에 관한 연구)

  • Purpose : This study was done to compare the vitamin A levels between hospitalized patients of measles symptoms and of the other infectious diseases common in childhood (bronchiolitis, penumonia, gastroenteritis and etc.). Methods : We have set up a population of 133 hospitalized patients of measles symptoms in the pediatric department of Kangnam Sacred Heart Hospital between Nov. 2000~Apr. 2001 and compared them to a group of 30 hospitalized patients of other infectious diseases between Dec. 2001~Feb. 2002. The whole patients were divided into three groups by diagnosing measles-specific IgM antibody positive as measles group(n=104), negative as measles-susepcted group(n=29) and other infectious disease group. And we compared the vitamin A levels, clinical symptoms, and general hematological and biochemical results. Results : Among the 104 measles patients, the concentrated age groups were 10 12 months(n=24, 23.1%), 13~15 months(n=16, 15.3%) and 16 months~4 years of age(n=24, 23.1%). The mean levels of vitamin A of measles and measles-susepcted groups were $69.6{\pm}28.6{\mu}g/dL$ and $75.2{\pm}34.4{\mu}g/dL$, respectively, and that of other infectious disease group was $28.1{\pm}17.5{\mu}g/dL$. Conclusion : The mean levels of vitamin A of both measles and measles-suspected groups were significantly higher than that of other infectious disease group(P<0.05) and showed no significant correlations with clinical symptoms and hospitalization duration.

Effect of the Plants Mixture and Garlic Composition on Serum Lipid Level of Hypercholesterolemic Rats (식물류 혼합물과 마늘의 복합 조성이 고콜레스테롤혈증 흰쥐의 혈청 지질에 미치는 영향)

  • This study was designed to investigate the effects of 4 kinds of plant water extract mixture and garlic extract (PMC) administration on serum lipid metabolism in hypercholestrolemic rats. The normal group was administered a cholesterol free diet, the control group a 1% cholesterol diet, and each experimental group was given a diet of 1% cholesterol, 1% plant mixture and 0.3, 0.5, 0.7% garlic extract (PMC-I, PMC-II, PMC-III), respectively. Each diet was administered orally to SD-male rats for 4 weeks. Total cholesterol content decreased by about 20% with administration of PMC. Triglyceride content also decreased from 9.3 to 15.0% compared to the control group, and phospholipid was similar to triglyceride. There was no significant difference in HDL-cholesterol content between the control and experimental groups. LDL-cholesterol content of the normal group was 9.4 times lower than the control group and its content was significantly lower in the PMC-II ($68.45{\pm}12.83\;mg/dl$) and PMC-III ($66.35{\pm}5.18\;mg/dl$) groups than the PMC-I group. VLDL-cholesterol content of the PMC-II and III groups were similar to the normal group. Atherogenic index (AI) and cardiac risk factor (CRF) were significantly lower in the PMC group. Blood glucose content was the lowest in the PMC-II ($189.37{\pm}12.02\;mg/dl$) group among all groups tested. Total protein content was $9.56{\pm}0.87{\sim}10.05{\pm}2.69\;mg/dl$ in the PMC-I~III groups and was significantly higher than the normal group. CPT activity did not show a significant difference among the experimental groups, while COT activity was effective only in the PMC-I group. Serum TBARS content in the PMC-III group was lower than in the normal group. Serum antioxidant activity by DPPH radical scavenging was $83.75{\pm}2.32%$ in the PMC-III group, which was significantly higher than the control group.

Effect of Paecilomyes japonica on Lipid Metabolism In Rats Fed High Fat Diet (눈꽃동충하초가 고지방 식이를 섭취한 흰쥐의 지질대사에 미치는 영향)

  • The effects of Paecilomyes japonica on weight gains, food intakes, food efficiency ratios, serum and hepatic lipid concentrations, serum protein levels and serum enzyme activities, were studied in adult male rats. Sprague-Dawley rats,35 weeks old, were given four different types of diets for a succeeding period of five weeks: either a normal diet (5% corn oil), a control diet (high fat; 5% corn oil + 15% lard), a PF diet (control diet + 3% fruiting body of Paecilomyes japonica), or a PM diet (control diet+.3% mycelium of Paecilomyes japonica). The body weight gains, hepatic weights and food efficiency ratios of rats fed the PF or PM diets were significantly lower than those fed the control diet, but were similar to those fed the normal diet. The concentrations of hepatic total lipids, cholesterol and triglyceric, and serum triglyceride, of rats given the PF or PM diets were significantly lower than those given the control diet. Hut the concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol and phospholipid in the serum of rats fed the control, PF or PM diets were significantly higher than those fed the normal diet. In the serum of rats fed the PF diet, the HDL-cholesterol/total cholesterol ratio was significantly higher and the atherogenic index was significantly lower than those fed the control diet, while such effect was not observed in rats fed the PM diet. The alkaline phosphatase activity in the serum of rats fed the control and PM diets was more significantly decreased compared to rats fed the PF and normal diet. No differences were noted in the weights of the pancreas, kidney and heart, the serum concentrations of glucose, hemoglobin and albumin, and the activities of GOT, GPT and ${\gamma}$-GTP, among the rats on all the experimental diets. In conclusion, the rats fed the PF or PM diets maintained normal body and hepatic weights. Despite of the high intake of fats in the PF and PM diets, the concentrations of hepatic total lipids, cholesterol and triglyceride, and serum triglyceride were decreased.

  • Recently, it has been reported that acute and chronic intoxication of Cd was concerned with environmental hygienic studies. In this study the effect of Ca and V-D was given directly into the diet with water containing the 20 ppm of Cd during so days. The results obtained were summarized as follows; 1. Administration of Ca and V-D inhibited the reduction of body weight induced by Cd. The internal organ weights of rats receiving Cd solution without Ca and V-D were generally increased except lung. 2. The values of hematocrit were higher in rats receiving Cd solution supplemented Ca and V-D than those doing Cd solution without Ca and V-D, and the levels of cholesterol were in- creased in experimental rats compared with control group. Total protein concentration in serum of the both group intaking the Ca and Y-D, and without Ca and V-D was not significant. 3. The concentration of Cd and Mg in liver was increased in rats feeding only Cd solution than those doing Ca and V-D group, whereas the concentration of Cu, Zn and Ca was decreased. The concentration of Cd, Cu and Ca in kidney was decreased in only Cd solution group, whereas by supplementing Ca and vitamin D was increased the concentration of Cd, Cu and Zn.

Effects of Minerals Added to Medicinal Plant Extracts on Alcohol-Induced Oxidative Stress and Alcohol Metabolism in Rats (약용식물 추출물이 첨가된 미네랄이 알코올에 의한 산화적 스트레스 및 숙취해소에 미치는 효과)

  • This study investigates the effects of a hangover beverage (MIX) that contains minerals (highly-salty mineral water, HSMW) and several medicinal plant extracts, on antioxidant and alcohol-metabolizing enzymes in alcohol administered Sprague-Dawley rats. HSMW is pumped from below the sedimentary rock layer of Dadaepo, Busan, South Korea, which is 1,050 m below the land surface; it tastes salty, like sea water. In terms of medicinal plant extracts, the total phenolic and flavonoid contents of Rubus coreanus and Cornus officinalis were measured as being significantly higher than those in Curcuma longa. The results suggest that treatment with MIX significantly increases superoxide dismutase (SOD) activity and DPPH radical scavenging activity. In the 10% HSMW-, for MIX- and company product (CP)-treated groups, the concentration of blood alcohol was significantly reduced 1~5 hr after alcohol loading, compared to that in the control group. In hepatic alcohol-metabolizing enzyme activities, alcohol dehydrogenase (ADH) activity was found to be higher in the MIX- and CP-treated groups than in controls, whereas acetaldehyde dehydrogenase (ALDH) activity was significantly higher in the CP-treated groups than other groups. This study concludes, therefore, that MIX (HSMW) minerals, like as Zn, Ca, Mg, Mn, and others stimulate alcohol-metabolizing enzymes, while the antioxidants of plant extracts prevent the damage otherwise incurred by alcohol toxicity. These results suggest that the hangover beverage (MIX) alleviates alcohol hangover symptoms by stimulating activities related to hepatic alcohol-metabolizing enzymes and antioxidant effects.

Effect of buchu (Allium tuberosum) on lipid peroxidation and antioxidative defense system in streptozotocin-induced diabetic rats (부추가 Streptozotocin 유발 당뇨쥐의 지질과산화와 항산화방어체계에 미치는 영향)

  • The pathogenic effort of high glucose, possibly in concert with fatty acids, is mediated to vascular complications of diabetes via increased production of reactive oxygen species(ROS), reactive nitrogen species(RNS), and subsequent oxidative stress. This study was carried out to investigate the suppressive effect of buchu(Allium tuberosum) on oxidative stress in streptozotocin(STZ)-induced diabetes in Sprague Dawley male rats. The effect of buchu supplementation (10%) on lipid peroxidation, and antioxidative defense system in blood and liver was compared among normal rats fed basal diet(normal) and diabetic rats fed basal diet(DM-control) or 10% buchu-supplemented diet(DM-buchu). Diabetes was experimentally induced by the femoral muscle injection of 50 mg STZ per kg of body weight. Animals were sacrificed after 4 wks of experimental diets feeding. The induction of diabetes by STZ elevated the level of lipid peroxidation represented by thiobarbituric acid-reactive substances(TBARS) and conjugated dienes in plasma, LDL, liver, and erythrocytes. 10% buchu-supplemented diet significantly reduced the levels of conjugated dienes in erythrocytes(p<0.05) and lowered TBARS in liver and LDL to the levels of control. Induction of diabetes by STZ elevated Mn-superoxide dismutase(Mn-SOD) activity and lowered activities of glutathionine reductase(GSH-red) and glutathionine peroxidase(GSH-px). Catalase activity was not affected by the induction of diabetes by STZ. However, buchu supplementation to diabetic rats significantly elevated catalase activity(p<0.05) and slightly elevated GSH-px and GSH-red activities in liver. GSH levels of blood and liver were lowered or not changed by induction of diabetes by STZ, respectively, while buchu supplementation to diabetic rats significantly elevated hepatic GSH level (p<0.05). In conclusion, it can be concluded that buchu might be a food source to attenuate oxidative stress in diabetic patients by inhibiting lipid peroxidation, by increasing hepatic GSH level, and by inducing anti-oxidative enzyme systems.

Effects of Pasteurization and Frozen Storage on Changes in Quality Characteristics of 10% Salted Egg Yolk (저온살균 및 냉동저장이 10% 가염난황의 품질특성에 미치는 영향)

  • To obtain the basic data for commercial 10% salted egg yolk for mayonnaise preparation, 3 types of egg yolks [pasteurized egg yolk (Yolk A)-not salted, pasteurized before salting (Yolk B)-salted, and pasteurized after salting (Yolk C)-salted] were prepared, and the changes in quality characteristics of these egg yolks with frozen storage were tested. The results obtained were as follows; Yolk A gelatinized during frozen storage, thus could not used for mayonnaise preparation. The viscosity of the egg yolk increased $3{\sim}5$ times after salting. Viscosity of the salted egg yolk increased with frozen storage time. Viscosity of Yolk B was higher at $-20^{\circ}C$ than $-15^{\circ}C$. Viscosity of Yolk C, however, was higher at $-15^{\circ}C$ than $-20^{\circ}C$. Frozen storage of pasteurized salted egg yolk showed some effects on the emulsification capacity. The effect, however, was smaller than that of unpasteurized salted egg yolk. Microbes of salted egg yolk were decreased with frozen storage, but there was no difference between Yolk B and Yolk C. It was suggested that commercially pasteurized 10% salted egg yolk for mayonnaise preparation can be successfully stored for 12 months at the temperature of $-15{\sim}-20^{\circ}C$.

Observation of Molecular Relaxation Behavior of Glucose Powders with Different Structures and Particle Sizes Using Low Field Nuclear Magnetic Resonance (NMR) (NMR을 이용한 구조 및 입도 차이에 따른 분말 포도당의 molecular relaxation behavior의 관찰)

  • Molecular relaxation behaviors of crystalline glucose anhydrous, crystalline glucose monohydrate, and amorphous glucose with different particle sizes were observed by measuring spin-spin relaxation time constant $(T_2)$ at the temperature range of $-20\;to\;110^{\circ}C$ using temperature-controlled low field nuclear magnetic resonance spectroscopy. No change in $T_2$ values of crystalline glucose anhydrous was observed throughout the temperature range, whereas $T_2$ values of crystalline glucose monohydrate and amorphous glucose increased from around $45\;and\;65^{\circ}C$, respectively. These results indicate that molecular mobility of crystalline glucose anhydrous does not change even at temperature higher than $100^{\circ}C$ and that the stability of powdered glucose could be improved by increasing the particle size of materials.

Pharmacological Studies of Cefoperazone(T-1551) (Cefoperazone(T-1551)의 약리학적 연구)

  • The pharmacological and microbiological studies of Cefoperazone (T-1551, Toyama Chemical Co., Japan) were conducted in vitro and in vivo. The studies included stability and physicochemical characteristics, antimicrobial activity, animal and human pharmacokinetics, animal pharmacodynamics and safety evaluation of Cefoperazone sodium for injection. 1) Stability and physicochemical characteristics. Sodium salt of cefoperazone for injection had a general appearance of white crystalline powder which contained 0.5% water, and of which melting point was $187.2^{\circ}C$. The pH's of 10% and 25% aqueous solutions were 5.03 ana 5.16 at $25^{\circ}C$. The preparations of cefoperazone did not contain any pyrogenic substances and did not liberate histamine in cats. The drug was highly compatible with common infusion solutions including 5% Dextrose solution and no significant potency decrease was observed in 5 hours after mixing. Powdered cefoperazone sodium contained in hermetically sealed and ligt-shielded container was highly stable at $4^circ}C{\sim}37^{\circ}C$ for 12 weeks. When stored at $4^{\circ}C$ the potency was retained almost completely for up to one year. 2) Antimicrobial activity against clinical isolates. Among the 230 clinical isolates included, Salmonella typhi was the most susceptible to cefoperazone, with 100% inhibition at MIC of ${\leq}0.5{\mu}g/ml$. Cefoperazone was also highly active against Streptococcus pyogenes(group A), Kletsiella pneumoniae, Staphylococcus aureus and Shigella flexneri, with 100% inhibition at $16{\mu}g/ml$ or less. More than 80% of Escherichia coli, Enterobacter aerogenes and Salmonella paratyphi was inhibited at ${\leq}16{\mu}/ml$, while Enterobacter cloaceae, Serratia marcescens and Pseudomonas aerogenosa were somewhat less sensitive to cefoperagone, with inhibitions of 60%, 55% and 35% respectively at the same MIC. 3) Animal pharmacokinetics Serum concentration, organ distritution and excretion of cefoperazone in rats were observed after single intramuscular injections at doses of 20 mg/kg and 50 mg/kg. The extent of protein binding to human plasma protein was also measured in vitro br equilibrium dialysis method. The mean Peak serum concentrations of $7.4{\mu}g/ml$ and $16.4{\mu}/ml$ were obtained at 30 min. after administration of cefoperazone at doses of 20 mg/kg and 50 mg/kg respectively. The tissue concentrations of cefoperazone measured at 30 and 60 min. were highest in kidney. And the concentrations of the drug in kidney, liver and small intestine were much higher than in blood. Urinary and fecal excretion over 24 hours after injetcion ranged form 12.5% to 15.0% in urine and from 19.6% to 25.0% in feces, indicating that the gastrointestinal system is more important than renal system for the excretion of cefoperazone. The extent of binding to human plasma protein measured by equilibrium dialysis was $76.3%{\sim}76.9%$, which was somewhat lower than the others utilizing centrifugal ultrafiltration method. 4) Animal pharmacodynamics Central nervous system : Effects of cefoperazone on the spontaneous movement and general behavioral patterns of rats, the pentobarbital sleeping time in mice and the body temperature in rabbits were observed. Single intraperitoneal injections at doses of $500{\sim}2,000mg/kg$ in rats did not affect the spontaneous movement ana the general behavioral patterns of the animal. Doses of $125{\sim}500mg/kg$ of cefoperazone injected intraperitonealy in mice neither increased nor decreased the pentobarbital-induced sleeping time. In rabbits the normal body temperature was maintained following the single intravenous injections of $125{\sim}2,000mg/kg$ dose. Respiratory and circulatory system: Respiration rate, blood pressure, heart rate and ECG of anesthetized rabbits were monitored for 3 hours following single intravenous injections of cefoperazone at doses of $125{\sim}2,000mg/kg$. The respiration rate decreased by $3{\sim}l7%$ at all the doses of cefoperazone administered. Blood pressure did not show any changes but slight decrease from 130/113 to 125/107 by the highest dose(2,000 mg/kg) injected in this experiment. The dosages of 1,000 and 2,000 mg/kg seemed to slightly decrease the heart rate, but it was not significantly different from the normal control. All the doses of cefoperazone injected were not associated with any abnormal changes in ECG findings throughout the monitering period. Autonomic nervous system and smooth muscle: Effects of cefoperazone on the automatic movement of rabbit isolated small intestine, large intestine, stomach and uterus were observed in vitro. The autonomic movement and tonus of intestinal smooth muscle increased at dose of $40{\mu}g/ml$ in small intestine and at 0.4 mg/ml in large intestine. However, in stomach and uterine smooth muscle the autonomic movement was slightly increased by the much higher doses of 5-10 mg/ml. Blood: In vitro osmotic fragility of rabbit RBC suspension was not affected by cefoperazone of $1{\sim}10mg/ml$. Doses of 7.5 and 10 mg/ml were associated with 11.8% and 15.3% prolongation of whole blood coagulation time. Liver and kidney function: When measured at 3 hours after single intravenous injections of cefoperaonze in rabbits, the values of serum GOT, GPT, Bilirubin, TTT, BUN and creatine were not significantly different from the normal control. 5) Safety evaluation Acute toxicity: The acute toxicity of cefoperazone was studied following intraperitoneal and intravenous injections to mice(A strain, 4 week old) and rats(Sprague-Dawler, 6 week old). The LD_(50)'s of intraperitonealy injected cefoperazone were 9.7g/kg in male mice, 9.6g/kg in female mice and over 15g/kg in both male and female rats. And when administered intravenously in rats, LD_(50)'s were 5.1g/kg in male and 5.0g/kg in female. Administrations of the high doses of the drug were associated with slight inhibition of spontaneous movement and convulsion. Atdominal transudate and intestinal hyperemia were observed in animals administered intraperitonealy. In rats receiving high doses of the drug intravenously rhinorrhea and pulmonary congestion and edema were also observed. Renal proximal tubular epithelial degeneration was found in animals dosing in high concentrations of cefoperazone. Subacute toxicity: Rats(Sprague-Dawley, 6 week old) dosing 0.5, 1.0 and 2.0 g/kg/day of cefoperazone intraperitonealy were observed for one month and sacrificed at 24 hours after the last dose. In animals with a high dose, slight inhibition of spontaneous movement was observed during the experimental period. Soft stool or diarrhea appeared at first or second week of the administration in rats receiving 2.0g/kg. Daily food consumption and weekly weight gain were similar to control during the administration. Urinalysis, blood chemistry and hematology after one month administration were not different from control either. Cecal enlargement, which is an expected effect of broad spectrum antibiotic altering the normal intestinal microbial flora, was observed. Intestinal or peritoneal congestion and peritonitis were found. These findings seemed to be attributed to the local irritation following prolonged intraperitoneal injections of hypertonic and acidic cefoperazone solution.
