Effects of Dietary n-3 Highly Unsaturated Fatty Acids on Growth and Biochemical Changes in the Korean Rockfish Sebastes schlegeli II. Changes of Blood Chemistry and Properties of Liver Cells (사료의 n-3계 고도불포화지방산 함량에 따른 조피볼락 Sebastes schlegeli의 성장 및 생화학적 변화 II. 혈액성분 변화 및 간세포 성상)

  • To define the effects of various levels $(0\~1.5\%)$ of dietary n-3HUFA on the physiological changes in the Korean rockfish, variations in blood variables and hepatocytes were studied. Biochemical serum analyses, lactate dehydrogenase (LDH) activity of the liver cytosol and ATPase activity of the liver microsomal membrane were also studied. The haematological values (red blood cell, hemoglobin, hematocrit, MCHC, MCV and MCH) were not significantly different in the experimental groups $(P\geq0.05)$. The total protein and glucose levels in the serum were affected by dietary n-3HUFA levels. These levels in groups fed n-3HUFA insufficient diets were significantly lower than those of n-3HUFA sufficient groups (P<0.05). Serum levels of total cholesterol, free cholesterol, glutamic pyruvic transaminase (GPT) and glutamic oxaloacetatic transaminase (GOT) showed significantly higher values in the fish fed n-3HUFA deficient diets (P<0.05). The LDH in the serum was dropped with increasing dietary n-3HUFA levels, but the LDH activity of the liver cytosol was elevated. Histologically, the hepatic cell in the fish fed n-3HUFA free diet was abnormal and showed a necrotic condition. $Ca^{2+}-ATPase$ activities of the liver microsomal membrane were significantly lower in the fish fed n-3HUFA deficient diets than in those fed n-3HUFA sufficient diets (p<0.005). These results suggested that the liver cell membrane was affected by dietary fatty acid compositions and cell membrane of the fish fed n-3HUFA insufficient diets showed abnormalities.

  • In orther to find the characteristics of each constitution, the examinee of Kyung Hee medical center was diagnosed constitution, and resulted body measures and diagnosis. That was considered and the results are as follows 1. The Diagnosis result of Sa-sang Constitutional Medicine is that there are 110 persons of Taeum-In(56. 1%), 58 persons of Soum-In(29.6%), 28 persons of Soyang-In(14. 3%). 2. The distribution of occupation, there are many of Taeum-In who are engaged in business, administeration, and management and many of Soeum-In who are engaged in reserch. 3. QSCC(I) has a tendency that other constitutions diagnose to Taeyang-In, the quastionare 1 has the accuracy of 76. 4% to diagnose Soeum-In. 4. Taeum-In sweats easily but Soeum-In doesn't sweat easily, Taeum-In has a good appetite and likes cold food and digests well, but Soeum-In has a poor appetite and like hot food and digest poorly. 5. The degree of obesity is the highest in Taeum-In. 6. The systoric blood pressure and diastolic blood pressure is high in Taeum-In and the high blood pressure are frequent in Taeum-In. 7. Triglyceride is the highest in Taeum-In and the Hyperlipidemia is the most frequent in Taeum-In, but Total cholesterol has no difference among constitutions. 8. GPT GGT is higher in Taeum-In than Soyang-In, but GOT has no difference among constitutions. 9. The frequency of fatty liver is the highest in Taeum-In.

Effect of Garlic Enzymatic Hydrolysates and Natural Color Resource Composites on Lipid Metabolism in Rat Fed a High Fat Diet (마늘 효소분해물과 천연색소 소재의 복합물이 고지방 식이성 흰쥐의 지질대사에 미치는 영향)

  • The study was performed to investigate the effects of enzyme treated garlic (EG) and its natural resources composites on lipid levels in serum and liver of rats fed a high fat diet. Four different types of EG-composite extracts prepared: EG and EG + grape peel (EGG), EG + Persimmon (EGP) and EG + Catechu (EGC) by mixed 9.5:0.5, 9:1 and 8:2 (w/w) ratios, respectively. DPPH and ABTS radical scavenging activity in vitro, show the highest in EG + Catechu (EGC) composite by mixed 8:2 (w/w). EG and EG-composites extracts (8:2, w/w) were administered orally to SD-male rats at a concentration of 2.5 g/kg/day for 5 weeks. Total lipid and cholesterol contents in serum were significantly lower in EGC group than control group, and triglyceride content was the lowest in EGP group by 54.29 mg/dL. HDL-cholesterol contents were significantly higher in EGP and EGC groups. LDL-cholesterol content was lower in EG group than EG-composite groups, and VLDL cholesterol content was the lowest in EGP group. GOT, GPT and ALP activity was significantly lower in EGP group. Total lipid, cholesterol and triglyceride contents in liver were significantly lower in EGP and EGC group than control group. Antioxidant activity in serum was the highest in EGC groups by 50.86%, in liver was the highest in EGP groups. TBARS content in serum and liver was the lowest in EGP group. In these results, we suggest that EGP composites could have hypolipidemic and anti-obesity effects in rats fed a high fat diet.

  • Pine (pinus densiflora) needles have long been used as a traditional health-promoting medicinal food in Korea. This study was conducted to investigate the effects of pine needle extracts on the hepatic antioxidant system in the damaged liver of carbon tetrachloride ($CCl_4$)-treated rats. Nine-week-old Sprague Dawley rats were divided into four groups: normal group (NOR), $CCl_4$-treated group (CCL), pine needle hot water extract and $CCl_4$-treated group (CCL-P), and Vitamin C and $CCl_4$-treated group (CCL-V). The enzyme activities and antioxidant effects of the pine needle hot water extracts were investigated at the levels of liver homogenates and serum of rats intoxicated with $CCl_4$. Serum GOT and GPT activities by $CCl_4$ treatment increased compared to those of the NOR group. However, they tended to decrease in the hot water extract-administered group. Liver SOD activity in the CCL group was significantly lower than the NOR group (p<0.05). However, they increased in the CCL-P group compared to the CCL group. Further, the CAT and GPx activities of serum treated with $CCl_4$ were higher compared to those of the NOR group but lower in the CCL-P group compared to CCL group. These results suggest that pine needle hot water extract increases antioxidant activities.

The Effect of Extract Powder from Fresh and Black Garlic on Main Components in Serum and Organs of Streptozotocin-Induced Diabetic Rats (생마늘 및 흑마늘 추출분말이 Streptozotocin 유발 당뇨 흰쥐의 혈청 및 장기 내 주요성분에 미치는 영향)

  • In this study, we examined the biological activity and synergistic effects of an extract powder of 1% and 3%, each fresh (FGP) and black garlic (BGP) in streptozotocin-induced diabetic rats. Blood glucose content was significantly lower in FGP and BGP groups than control group. Glycosylated Hb was significantly higher in streptozotocin induced diabetic control group than normal group, but significantly lower in FGP and BGP groups. Total cholesterol content of the FGP and BGP groups were lower than control group, but not shown the significant difference between garlic fed groups. HDL-cholesterol concentrations of the FGP and BGP fed groups were significantly higher than control group, except of 1% BGP group. LDL and VLDL-cholesterol contents were significantly lower in 3% FGP group, and the same tendency atherogenic index and cardiac risk factor. GOT, GPT, and ${\gamma}$-GTP activity of serum were lower in FGP and BGP fed groups than control group. Glycogen contents in liver significantly higher than control group, and has not significantly difference between normal group. TBARS content was no significantly difference in the liver and serum, but in the kidney, 3% FPG and BGP fed groups were significantly lower than other experimental group. DPPH radical scavenging activity of liver has not significant difference among experimental groups, but activity was higher garlic extract powder fed groups in serum and kidney. These results indicate that dietary supplements of fresh and black garlic extract powder was contributed to lower of blood glucose, loss prevention of glycogen in liver and improve of lipid metabolism.

Effect of Medicinal Herb Composites on Antioxidative and Cognition-Enhancing Activities in Rats (생약복합물이 흰쥐의 체내에서 항산화 및 인지개선활성에 미치는 영향)

  • The purpose of this experiment was designed to investigate the effects of medicinal herbs (MH) extracts on dementia induced by trimethyltin chloride (TMT) in rats. Six-week-old male Sprague-Dawley rats were randomly divided into five groups; normal group (group 1), control group (group 2), MH extracts group (250, 500 mg/kg) (group 3, group 4) and positive control group (tacrine group, group 5). In the control group to induce dementia, a 2.5 mg/kg of TMT intraperitoneal injection was used for 14 days (1 per day) in the rats. In the MH extracts group 250 mg/kg and 500 mg/kg of MH extracts were medicated in an oral inoculation for 20 days (1 per day). After 30 minutes, a 2.5 mg/kg of TMT intraperitoneal injection, which causes dementia, was used for 14 days (1 per day). In the positive control group (Tacrine group) 10 mg/kg of Tacrine, the dementia treatment, was medicated in an oral inoculation. After 30 mintues, 1 mg/kg of TMT intraperitoneal injection, which causes dementia, was used for 14 days (1 per day). The present author observed the passive avoidance performance test, and memory ability test (Y maze test), the values of MDA, acetlycholinesterase (AchE) activity in the brain and antioxidant enzyme in serum. MH extracts significantly improved memory of AD model rats in the Y-maze test, and also significantly improved memory of AD model rats in the passive avoidance test. MH extracts significantly reduced AChE activity, and significantly increased the SOD level, but not catalase and MDA. From the results above, MH extracts is thought to be effective in the improvement of antioxidant enzymes and memory ability.

Effects of Agarum cribrosum on the Improvements in Treating Constipation and Plasma lipid Profiles (구멍쇠 미역의 변비 완화 효과 및 혈액지방에 미치는 영향)

  • The present study was conducted to evaluate the effects of Agarum cribrosum on weight change and defecation states in rats with constipation induced by loperamide. Food intake and body weight both decreased in the 5% Agarum cribrosum and loperamide-treated group (SD5) and 10% Agarum cribrosum and loperamide-treated group (SD10), whereas fecal water contents increased by 1.6 and 2.1-fold in the SD5 and SD10 groups, respectively. The concentrations of total-cholesterol, HDL-cholesterol and triglyceride in the sera of the SD5 and SD10 groups were lower than those in the control (C) group. However, the biochemical parameters, GOT (glutamic oxaloacetic transaminase), GPT (glutamic pyruvic transaminase), and glucose levels, were not affected by the level of Agarum cribrosum. In addition, the concentrations of total-cholesterol and triglyceride in the livers of the SD5 and SD10 groups were also lower than those in the control group. The results of the present study demonstrated that Agarum cribrosum might ameliorate constipation symptoms, and lower lipid concentrations in the blood and liver.

Effect of Pig Skin Gelatin Hydrolysates on the Bone Mineral Density of Ovariectomized Rats (돈피 젤라틴 효소분해물이 난소 적출쥐의 골밀도에 미치는 영향)

  • This study was conducted to examine the effects of low molecular weight gelatin hydrolysates (GH, less than 3kDa), extracted from pig skin collagen on the bone metabolism of ovariectomized (OVX) rats. The rats in the experimental groups were randomly segregated into six different treatment groups such as 1) NC, the normal rat fed AIN 93 diet (basal diet) only; 2) OC, the OVX rat fed the basal diet only; 3) GH 0.1, the OVX rat fed the basal diet with 0.1% GH; 4) GH 0.8, the OVX rat fed the basal diet with 0.8% GH; 5) G 0.1, the OVX rat fed the basal diet with 0.1% gelatin; 6) G 0.8, the OVX rat fed the basal diet with 0.8% gelatin. Body weight gain in the GH 0.1, GH 0.8, and G 0.8 was significantly higher than those in the NC and OC. Feed intake of the GH 0.1 and GH 0.8 was higher than that of the NC and OC, while no significant difference was found in feed efficiency ratio (FER). BMD of the GH 0.8 was higher than that of the OC. However, gelatin hydrolysates and gelatin resulted in higher BMC level compare to the OC. Serum HDL-cholesterol of rat fed GH and gelatin was higher than that of OC (p<0.05). LDL-C of the GH 0.1 and the GH 0.8 tended to be less than that of OC. Serum alkaline phosphatase (ALP) of the GH 0.1 was lower than that of the OC. The serum of GH 0.8 showed lower osteocalcin value than the OC (p<0.05). In addition, GOT and GPT levels significantly decreased in all treatment groups. These results indicated that gelatin hydrolysates from pig skin gelatin hydrolysates enhanced BMD and serum biochemical parameters related to bone metabolism. Therefore, the gelatin hydrolysates could be used as a beneficial material to improve bone health.

Growth Performance, Immune Response and Carcass Characteristics of Finishing Pigs by Feeding Stevia and Charcoal (스테비아와 숯이 급여된 비육돈의 사양성적, 면역력 및 도체특성)

  • This study was conducted to determine the effects of stevia (Stevia rebaudiana bertoni) and charcoal supplementation on growth performance, immune response and carcass characteristics of finishing pigs. A total of 420 pigs (LYD) were randomly allocated into 7 treatments with 3 replications. Dietary treatments were 1) T1 (basal diet), 2) T2 (basal diet+0.3% stevia), 3) T3 (basal diet+0.6% stevia), 4) T4 (basal diet+0.3% charcoal), 5) T5 (basal diet+0.6% charcoal), 6) T6 (basal diet+0.3% stevia+0.3% charcoal) and 7) T7 (basal diet+0.6% stevia+0.6% charcoal). During the experimental period, average daily gain (ADG) was higher in T2 and T6 groups than the other treatments (p<0.05). Feed conversion ratio (FCR) was higher in T6 group compared to the others (p<0.05). There were no significant differences in total cholesterol level and glutamic pyruvic transaminase (GPT) activity of blood among treatments. In glutamic oxaloacetic transaminase (GOT) activity, T3, T5, T6 and T7 groups showed lower values (p<0.05) compared to T1. Insulin-like growth factor-1 concentration was higher in T2 and T6 groups than the others (p<0.05), but there were no significant differences in immunoglobulin G, lymphocyte, eosinophil, basophil and atypical lymph levels among treatments. In neutrophil, T6 showed higher level compared to the others (p<0.05). In the carcass characteristics, T6 showed higher level of a carcass grade compared to the other treatments. However, carcass length did not show any significant difference among treatments. As a result, dietary supplementation of 0.3% stevia and 0.3% charcoal showed higher ADG, higher FCR and better immune response resulting in better growth performance and carcass characteristics of finishing pigs.

Basic Studies on Developing Equipment for Waterless Transportation of Live Fish (활어의 무수 수송 장치 개발을 위한 기초적 연구)

  • This study was undertaken to get basic data for the cold-waterless transportation of live fish. The optimal cold temperature of plaice, Paralichthy olivaceus was determined by checking the changes of dissolved oxygen and ammonia in the sea water and the survival times during storage at various temperatures. After determination of optimal temperature for transportation, the changes of serum components and muscle components of live plaice were also carried out during storage at cold($5^{\circ}C$)-waterless conditions and the recovery conditions($10\%$ density at $15^{\circ}C$). At higher storage temperature, decreases in dissolved oxygen and the increases in ammonia in seawater were observed. In addition, the survival time was short at low temperature($0^{\circ}C\;and\;3^{\circ}C$). Almost all of the serum components(hemoglobin, glucose, LDH, GOT and GPT) of live plaice gradually increased during storage in cold-waterless conditions, and then those values decreased to the initial levels after $3{\sim}10hrs$ storage in conditions of recovery. The concentration of ATP in the muscle steadily decreased during storage in cold-waterless conditions. The contents of ADP and IMP seemed to be directly related to the extent of ATP breakdown. ADP and IMP thus showed a gradual increase during storage. The level of lactate in the muscles gradually increased during these storage times, also. On the other hand, the levels of those components in the muscle entirely recovered to their original levels within $3{\sim}6hrs$ storage after they were returned to conditions of recovery. The ratio of ATP to the ATP and its related compounds${ATP/(ATP+ADP+AMP+IMP){\times}100}$ in the muscle showed $45\%$ after 18hrs storage in cold-waterless conditions. Otherwise, ratios returned to their original levels within $3{\sim}6hrs$ of storage in recovery conditions.
