Microtubule acetylation has been proposed as a marker of highly heterogeneous and aggressive triple-negative breast cancer (TNBC). The novel microtubule acetylation inhibitors GM-90257 and GM-90631 (GM compounds) cause TNBC cancer cell death but the underlying mechanisms are currently unknown. In this study, we demonstrated that GM compounds function as anti-TNBC agents through activation of the JNK/AP-1 pathway. RNA-seq and biochemical analyses of GM compound-treated cells revealed that c-Jun N-terminal kinase (JNK) and members of its downstream signaling pathway are potential targets for GM compounds. Mechanistically, JNK activation by GM compounds induced an increase in c-Jun phosphorylation and c-Fos protein levels, thereby activating the activator protein-1 (AP-1) transcription factor. Notably, direct suppression of JNK with a pharmacological inhibitor alleviated Bcl2 reduction and cell death caused by GM compounds. TNBC cell death and mitotic arrest were induced by GM compounds through AP-1 activation in vitro. These results were reproduced in vivo, validating the significance of microtubule acetylation/JNK/AP-1 axis activation in the anti-cancer activity of GM compounds. Moreover, GM compounds significantly attenuated tumor growth, metastasis, and cancer-related death in mice, demonstrating strong potential as therapeutic agents for TNBC.
Enzyme-linked immune sorbent assay (ELISA) was applied to quantify soy protein in fresh soybean curd (bean curd) produced by combination of genetically modified (GM) and genetically not modified (non-GM) soybeans. Antibodies against 113 and 24 kDa proteins, which appeared only in non-GM bean curd (specific band), and in both non-GM and GM bean curds (non-specific band) based on SDS-PAGE results, were prepared by immunization to rabbit. Through ELISA using either antibody, GM bean curd protein content was determined at dilution rates of $10^{-1}-10^{-6}$. Standard curve showing relationship between ELISA optical density and non-GM protein content was produced using antibody against 113 kDa protein at protein dilution between $10^{-7}\;to\;10^{-6}$, highly antigen content-dependent dilution. Bean curd prepared by random combinations of GM and non-GM soybeans were analyzed by ELISA, and standard curve was produced. Results reveal non-GM protein content of bean curd could be quantified with higher than 93% accuracy.
Two feeding experiments were conducted to investigate the effects of dietary genetically modified (GM) soya and com on growth performance, feed utilization and body composition of juvenile olive flounder, Paralichthys olivaceus and rockfish, Sebastes schlegelii. For each fish species, four isonitrogenous (50% crude protein) and isocaloric (4.1 kcal/g) diets (designated as nGM soya, GM soya, nGM com and GM com) were formulated to contain 20% non-GM (nGM) and GM soya and com. Thirty olive flounder (initial body weight, 15.4${\pm}$0.4 g) and fifty rockfish (initial body weight, 3.1${\pm}$0.02 g) were distributed in each 400 L tank (200 L water) in a flow through system. Each experimental diet was fed to triplicate groups of fish to visual satiation, twice a day (9:00 hand 17:00 h) for 6 weeks. Growth performance was measured every three weeks. No effects of GM feedstuffs on survival were observed. Dietary inclusion of GM feedstuffs did not affect growth performance and feed utilization of fishes, except for rockfish fed GM com. Rockfish fed the GM com diet showed higher weight gain, daily feed intake and daily protein intake than did fish fed the nGM com diet, but no significant differences were observed in final body weight between the dietary treatments. Condition factor, hepatosomatic index, visceral somatic index and body composition were not altered by the inclusion of GM feedstuffs. These results indicate that dietary inclusion of GM soya and com could have no effects on growth performance and feed utilization of juvenile flounder and rockfish. Lower weight gain and feed intake in flounder and rockfish fed the diets containing 20% soya were likely due to anti-nutritional factors, rather than transgenic factors in the feedstuffs. Dietary inclusion of GM soya and com at the level tested did not alter the body composition of fishes. Further studies to investigate the effects of GM feedstuffs on health conditions and the development of fishes, as well as those of residue of transgenic fragments in ambient environments and in animals are necessary for safe use of the ingredients in aquaculture.
To develop human risk assessment protocols, we explored housekeeping gene and cytokine expression in mouse spleen cells using Rt-PCR. We normalized housekeeping gene expression by RT-PCR; gene expression was highly uniform in potato leafs and mouse spleen cells. We measured the expression of frequently used housekeeping genes, such as those encoding APRT, $\beta$-tubulin, Actin, Hsp 20.2, Cyclophilin, 18S RNA, Efla, Tbp, GAPDH, $\beta$-actin, Tuba2, Hprt, Cyclophlin A, Tfrc, and RPL13A in mice fed GM or non-GM potatoes. Housekeeping gene expression did not show any significant differences between GM and non-GM potato-fed mice. The murine model of potato-fed mice did not express IL-4 and IL-13 at a significant levels.
Recently, the occurrence of a ship capsizing was analyzed as the main cause of the lack of stability or loss because of the improper management of the center of gravity, the movement of cargo or heavy weight when excessive steering occurs or when navigating during bad weather. Thus, to prevent a ship from capsizing, it is necessary to secure stability to enable the ship's return to its upright position, even if a dangerous heel occurs. The GM is a crucial evaluation factor regarding stability, which the navigation officer uses to preserve stability. In this study, based on the stability data collected from the operating of ships for five years, The GM by ship's type according to the operating status was analyzed specifically such as a ship's length, breadth, and gross tonnage. The feature of the GM distribution according to a ship's length was confirmed, and after performing the correlation analysis between the breadth and the GM, the ratio of the GM to breadth was calculated, and the result was compared with the previous ratio. Additionally, a simple approximation formula and minimum GM for the estimation of the GM by ship type were proposed by the regression analysis of the GM using the gross tonnage (GT)/breadth (B) to reflect the trend of larger ships being built. The results of this study are expected to be used as data for the review of securing a stable GM on ships.

Background: The functionality of myofibrillar proteins is a major factor influencing the quality attributes of muscle foods. Nonetheless, the relationships between muscle type and oxidative changes in chevon during ageing are meagrely elucidated. Postmortem changes in antioxidant status and physicochemical properties of glycolytic gluteus medius (GM) and oxidative infraspinatus (IS) muscles in goats were compared. Methods: Twenty Boer bucks (9-10 months old, body weight of $36.9{\pm}0.725kg$) were slaughtered and the carcasses were subjected to chill storage ($4{\pm}0.5^{\circ}C$). Analyses were conducted on GM and IS muscles sampled on 0, 1, 4 and 7 d postmortem. Results: Chill storage did not affect the antioxidant enzyme activities in both muscles. The IS had greater (P < 0.05) superoxide dismutase and catalase activities than GM. Carotenoid and tocopherol contents did not differ between muscles but decreased (P < 0.05) over storage. The IS had higher (P < 0.05) glycogen and ultimate pH and lower (P < 0.05) shear force and cooking loss than GM. The carbonyl content, % metmyoglobin, drip loss and TBARS increased (P <0.05) while free thiol, metmyoglobin reducing activity (MRA), shear force and myoglobin decreased (P < 0.05) over storage. Muscle type had no effect (P > 0.05) on free thiol, MRA and TBARS. The GM had lower (P < 0.05) redness on d 0 and 1 than IS while the IS had greater carbonyl, % metmyoglobin and drip loss than GM on d 7. The reflective density of slow myosin heavy chain (MHC) was higher (P < 0.05) while the density of fast MHC and actin was lower (P < 0.05) in IS than GM. Regardless of muscle type, the density of MHC decreased (P < 0.05) while that of actin was stable over storage. Nonetheless, the degradation of fast and slow MHC was greater (P < 0.05) in IS than GM. Muscle type had no effect (P > 0.05) on consumer preference for flavour, juiciness and overall acceptability. However, IS had higher (P < 0.05) tenderness score than GM on d 1 and 4 postmortem. Intramuscular fat was higher (P< 0.05) in IS compared with GM. Fatty acid composition did not differ between the muscles. However, GM had lower (P < 0.05) n-6/n-3 ratio than IS. The n-3 and n-6 PUFA declined (P < 0.05) while the SFA increased (P < 0.05) over storage. Conclusion: The changes in myofibrillar proteins and physicochemical properties of goat meat during postmortem chill storage are muscle-dependent.
This study identifies the structural determinants of the social acceptance of genetically modified (GM) foods across European countries. Toward this end, we suggest an integrated theoretical model to explain the social acceptance of GM foods by including both perception factors (perceived benefit, perceived risk, feelings, trust, and knowledge) and value factors (ethical concerns, science optimism, religiosity, and ideology). This model is then tested by analyzing survey data collected from 18,634 Europeans in 32 countries. The results indicate that first, not only perception factors but also value factors significantly contribute to explaining the acceptance of GM foods. Second, perceived benefits, perceived risk, feelings, and ethical concerns tend to be the four biggest determinants for acceptance. Third, this two-factor model could be generalized even with variation across countries. Finally, ethical concerns and scientific optimism play a moderating role between predictors and outcomes in the acceptance of GM foods.
It was determined whether the sandwich ELISA using specific anti-CP4 EPSPS polyclonal and monoclonal antibodies, developed in the previous study, could be applied to detect GM soybean or not. The soybeans (47 imported and 20 domestic soybeans) were analyzed by a sandwich ELISA. The results of imported soybeans were divided into two groups which were high contents $(39.1{\pm}13.5\;{\mu}g/g,\;n=33)$ and low contents of CP4 EPSPS $(2.6{\pm}1.2\;{\mu}g/g,\;n=14)$. The ratio of GM in imported soybeans was about 70.2%. One the other hand, the contents of CP4 EPSPS in domestic soybeans was very low $(0.9{\pm}0.5\;{\mu}g/g,\;n=20)$ which determined to be non-GM soybeans. In case of soybean sprouts, the contents of CP4 EPSPS in soybean sprouts were different between GM and non-GM soybean sprout. The CP4 EPSPS in cotyledon of GM soybeans sprout was higher than that in root hair. The contents of CP4 EPSPS in soybeans sprout of domestic soybeans were very low. Thus, it was possible to determine that the soybeans sprout was made of GM or non-GM soybeans. Also, PCR experiment showed that the sandwich ELISA was accurate to distinguish the soybeans to be GM or non-GM. These results showed the sandwich ELISA could determine the soybeans were GM or non-GM, rapidly and simply.
A new algorithm for planning a collision-free path based on algebraic curve is developed and the concept of collision-free Path Space (PS) is introduced. This paper presents a Geometry Mapping (GM) based on two straight curves in which the intermediate connection point is organized in elliptic locus ($\delta$, $\theta$). The GM produces two-dimensional PS that is used to create the shortest collision-free path. The elliptic locus of intermediate connection point has a special property in that the total distance between the focus points through a point on ellipse is the same regardless of the location of the intermediate connection point on the ellipse. Since the radial distance, a, represents the total length of the path, the collision-free path can be found as the GM proceeds from $\delta$=0 (the direct path) to $\delta$=$\delta$$\_$max/(the longest path) resulting in the minimum time search. The GM of elliptic workspace (EWS) requires calculation of interference in circumferential direction only. The procedure for GM includes categorization of obstacles to .educe necessary calculation. A GM based on rectangular workspace (RWS) using Cartesian coordinate is also considered to show yet another possible GM. The transformations of PS among Circular Workspace Geometry Mapping (CWS GM) , Elliptic Workspace Geometry Mapping (EWS GM) , and Rectangular Workspace Geometry Mapping (RWS GM), are also considered. The simulations for the EWS GM on various computer systems are carried out to measure performance of algorithm and the results are presented.
