Black pine barks from the southern region of Korea were extracted using pressurized hot water and the water soluble extracts were then separated in a stepwise fashion using a variety of solvents, column chromatography (CC), thin layer chromatography (TLC), and high pressure liquid chromatography (HPLC). The antioxidant activities of each fraction and the active compounds were determined based on the radical scavenging activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH), reductive potential of ferric ion, and total phenol contents. A DPPH test showed that the half maximal effective concentration ($EC_{50}$ value : $6.59{\pm}0.31\;{\mu}g/mL$) of the ethyl acetate fraction (ca. 0.67%) was almost the same as that of the control compounds and inversely proportional to the value of the total phenol contents. The cell viability of the water extracts was confirmed by methyl thiazol-2-yl-2,5-diphenyl tetrazolium bromide (MTT) with enzyme linked immune sorbent assay (ELISA). Catechin, epicatechin, quercetin and ferulic acid were isolated from the ethyl acetate fraction as active compounds and identified by nuclear magnetic resonance. The antioxidant activity as value of DPPH of each of the separated compounds was lower than the ethyl acetate fraction, and ferulic acid was the lowest among these compounds.
Even though some standard developmental bioassay protocols for environmental assessment using sea urchins have already been described, there have not been many attempts to apply and modify these protocols with Korean species. Therefore, there is a strong need to establish standard bioassay protocols using sea urchins commonly found in Korea. Prior to developing a new protocol, it is essential to know the optimal conditions for the bioassay procedures. We investigated the optimal conditions (temperature, salinity, and embryo density) of the sea urchin Strongylocentrotus intermedius. The ideal temperature for developmental bioassay of S. intermedius was determined to be $15^{\circ}C$ and the time required for the embryo to become pluteus larva was 72 hr. The optimal range of salinity for the embryo toxicity using S. intermedius was between 30 to 32 psu, which is similar to the range found in the natural habitats of adult populations. The optimum density of embryos at the beginning of bioassays was 100 embryos/mL. When the assays were carried out at higher densities, the proportion of normally developed larvae decreased significantly.
This study investigated the productivity and growth of copepods, Sinocalanus tenellus 2 strains, Paracyclopina nana 3 strains, Apocyclops royi 2 strains, Tachidius triangularis 1 strain and Tigriopus sp. 1 strain at different temperatures $(24,\;28\;and\;32^{\circ}C)$ under 15 psu and various salinities (10, 20, 30 psu) at $28^{\circ}C$ for an individual culture. And the mass culture was carried out to evaluate the production of these copepods in the 500 mL vessel. For an individual culture of copepod, survival period of a brood female tended to increase with the decrease of temperature, but was not significantly influenced by salinity. With an increase of temperature and a decrease of salinity, maturation period of nauplii in all species tended to decrease. Total nauglii production from a broodo female at $24^{\circ}C$ was higoher than that from a brood .female at either 28 or $32^{\circ}C$ for S. tenellus and at $32^{\circ}C$ than either 24 or $32^{\circ}C$ for A. royi Haenam stram, but was not influenced by temperature for another copepods. And total nauplii production from a brood female at 10 and 20 psu tended to be higher than that from a brood female at 30 psu, expect for Tigriopus sp. and P. nana Hwajinpo strain. Total nauplii production from a brood female was the highest in Tigriopus sp. (350 nauplii) at $28^{\circ}C$ under 20 psu, followed by A. royi Haenam strain (187 nauplii) at $32^{\circ}C$ under 15 psu and P. nana Hwajinpo strain (152 nauplii) at $24^{\circ}C$ under 10 psu. In the mass culture of each copepod, P. nana was the most productive (26.6 mg dry weight) and this value was two and four times higher than productivity of Tigriopus sp. and A. royi, respectively.
This study was carried out to investigate the effect of trans-cinnamaldehyde and high pressure treatment on milk. Cinnamon oil milk was manufactured by high speed homogenization (3,000 rpm) and high pressure homogenization (500 and 2,000 bar) processing UHT milk and trans-cinnamaldehyde of various concentrations (0 to 0.1% (w/v)). Cinnamon oil milk was inoculated with Escherichia coli (6.4 Log CFU/mL) and kept at $7^{\circ}C$ for 10 d to observe the antibacterial effect. The cinnamon oil milk containing 0.05% (w/v) trans-cinnamaldehyde initially began to show an antibacterial effect and Escherichia coli completely died in cinnamon oil milk added 0.1% (w/v) trans-cinnamaldehyde on the 6th day of storage. The result of the TBA value showed that the addition of 0.1% (w/v) trans-cinnamaldehyde was also effective to protect lipid oxidation. In the physical properties of cinnamon oil milk, particle sizes were enlarged in all samples during storage periods and the total color difference of cinnamon oil milk was slightly increased as level of high pressure. The surface tension of cinnamon oil milk treated 2,000 bar was remarkably higher than other samples. It seems that trans-cinnamaldehyde showed antibacterial activity and antioxidation effect at 0.05 and 0.1% (w/v) of concentration. Remarkably, high pressure treatment did not influence its microbial property but slightly affected the physical properties of cinnamon oil milk.
By using a complete rate constant($k_e$) which treats a solvent (water) as a reactant, and a conventional rate constant($k_c$), which ignores the solvent in describing the rate, the parameters ${\Delta}V^{\neq}_s,\;{\Delta}H^{\neq}_s\;and\;{\Delta}S^{\neq}_s$ were introduced. These quantities represent the volume change, the enthalpy change, and the entropy change accompanying the electrostriction which occurs when solvent molecules condense on the activated complex. The authors measured the rates of the solvolysis of benzoyl chloride in water-acetone mixtures at $15^{\circ}$ to $30^{\circ}C$ and 1 bar to 2500 bars. Applying the authors' theory to the experimental results, the parameters, ${\Delta}V^{\neq}_s,\;{\Delta}H^{\neq}_s\;and\;{\Delta}S^{\neq}_s$ were evaluated, and it was found that they are all negative, indicating that water dipoles condense on the activated complex. They also proposed the following equations: ${\Delta}H^{\neq}_c\;=\;{\Delta}H^{\neq}_e\;+\;{\Delta}H^{\neq}_s\;and\; {\Delta}S^{\neq}_c\;=\;{\Delta}S^{\neq}_e\;+{\Delta}S^{\neq}_s\;,\;where\;{\Delta}H^{\neq}_c\;and\;{\Delta}H^{\neq}_c\;and\;{\Delta}S^{\neq}_s $are the activation enthalpy change and the activation entropy change for the conventional reaction rate, respectively, and ${\Delta}H^{\neq}_e$ and ${\Delta}S^{\neq}_e$ are the corresponding quantities for the complete reaction rate. The authors proposed that for the $SN_1$ type, all the quantities, ${\Delta}V^{\neq}_s,\;{\Delta}S^{\neq}_s\;,{\Delta}H^{\neq}_s\;and\;{\Delta}S^{\neq}_s$ are comparatively large, and for the $SN_2$ type, these quantities are smaller than for the $SN_1$ type, and occasionally the case ${\Delta}S^{\neq}_e$ < 0 occurs. Using these criteria, the authors concluded that at high temperature, high pressure and for a high water content solvent, the SN_1$ type mechanism predominates whereas in the reversed case the $SN_2$M type predominates.
We investigated the optimum salinity and temperature conditions for the mass culture of small rotifer, Proales similis. In the salinity experiment ranging from 1, 5, 10, 15, 20, 25 and 33‰, growth tended to decrease with salinity increase. Most agreeable salinity for rotifer growth was 1‰ in which maximum density and specific growth rate (SGR) were obtained. In the temperature experiments ranging from 15, 20, 25, 30 and $35^{\circ}C$, continuous growth of rotifer populations was found up to $35^{\circ}C$. The highest maximum density (2,060 inds./mL) of rotifer was observed at $25^{\circ}C$ in given temperature regime. Also, the SGR of females showed increasing tendencies with the increase of temperature. These results suggest that the optimum salinity and temperature for mass culture of P. similis may be 1‰ and $25^{\circ}C$, respectively.

Developing drugs targeting respiratory pathogen is essential to control respiratory diseases. Many experiments have been performed under in vivo situation. However, in vivo experiments have economical and ethical issues. The objective of this study was to determine the possibility of developing an ex vivo lung culture system with possible application for respiratory infection studies. After isolating lungs from naïve pigs, agarose-inflated lung tissues were prepared and sliced manually. These sliced lung tissues were then subsequently placed on 24-well plates. Eight different combinations of media were used to determine the optimum ex vivo lung culture condition. In addition, lung tissues were infected with porcine reproductive and respiratory syndrome (PRRS) virus at a titer of $1{\times}10^4\;TCID_{50}/mL$. Virus growth was confirmed by titration in MARC-145 cells at 2, 4, 6 days post infection (dpi). We found that ex vivo lung culture in physiological environment was not media specific based on histopathology and cytotoxicity. However, under virus-infected condition, thickened alveolar walls in the lung tissues and stable virus titers at 2, 4, 6 dpi were shown in F12K medium suggesting that it was useful for tissue maintenance and virus infection using PRRS virus infected lung tissues. The present study shows the possibility of using porcine ex vivo lung model for respiratory infection studies.
The objective of this study is to evaluate the antioxidant components, elastase inhibition activities, and melanin synthesis rates of Korean traditional Actinidia (Actinidia arguta) fruits and leaves depending on the ethanol extraction concentrations. The total polyphenol content was the highest in the 50% ethanol extract of both fruits and leaves, with values of 634.1 mg GAE/100 g and 3,985.2 mg GAE/100 g, respectively. The total flavonoid content was the highest in the fruit 90% extract and leaf 50% extract at 191.9 mg/100 g and 2655.6 mg/100 g, respectively. The vitamin C content was the highest in the 50% extract of leaves at 2990.3 mg/100 g. Elastase inhibition was the highest at 56.9% in the leaf 50% extract at a concentration of 1,000 ㎍/mL. Melanin synthesis inhibition showed the highest melanin synthesis inhibitory effect among the extracts, as the leaf 50% extract showed an inhibitory rate of 65% or more. Therefore, the antioxidant components, elastase inhibition activities, and melanin synthesis inhibitory rate were better in leaves than in fruits. The leaf 50% extract was particularly the best among the extracts. Korean traditional Actinidia leaves can be considered as potential sources for new functional materials.
This study developed the procedure and method for the accuracy assessment of unmanned boat survey data, based on the reservoir water depth data of Misan Reservoir, measured by the manned and unmanned boats in 2009 by Korea Rural Community Corporation. In the first step, this study devised the method to extract the contour map of NGIS data in AutoCAD to generate easily the reservoir boundary map used to set the survey range of reservoir water depth and to test the survey accuracy. The surveyed data coordinate systems of the manned and the unmanned boat were also unified by using ArcGIS for the standards of accuracy assessment. In the accuracy assessment, the spatial correlation coefficient of the grid maps of the two measurement results was 0.95, showing high pattern similarity, although the average error was high at 78cm. To analyze in more detail assessment, this study generated randomly the 3,250m transverse profile route (PR), and then extracted grid values of water depth on the PR. In the results of analysis to the extracted depth data on PR, the error average difference of the unmanned boat measurements was 73.18cm and the standard deviation of the error was 55cm compared to the manned boat. This study set these values as the standard for the correction value by average shift and noise removal of the unmanned boat measurement data. By correcting the unmanned boat measurements with these values, this study has high accuracy results, the reservoir water depth and surface area curve with R2 = 0.97 and the water depth and storage volume curve with R2 = 0.999.
