Changes in the growth and biochemical composition of Chaetoceros calcitrans cultures using light-emitting diodes (LED (Light-Emitting Diode)를 이용한 미세조류 (Chaetoceros calcitrans)의 성장 및 생화학적 조성 변화)

  • The marine microalgae Chaetoceros calcitrans was cultured under a fluorescent lamp (CON) and light-emitting diodes (LEDs) of various wavelengths (blue, LB; red, LR; green, LG; white, LW); changes in growth, fucoxanthin, chlorophyll-a, amino acid and fatty acid profiles were investigated. LR-exposed cultures exhibited the highest specific growth rate (SGR) (0.34), whereas LG-exposed cultures showed the lowest SGR (0.26). After cultivation for 10 days, the maximum dry cell weight (g/L) of LR-exposed cultures was significantly higher than that of those exposed to other light conditions (LR${\geq_-}$CON>LB${\geq_-}$LW${\geq_-}$LG). Eicosapentaenoic acid (EPA) levels were significantly higher in CON-exposed cultures compared to those exposed to LW (P<0.05), with no marked difference compared to those exposed to LB, LR and LG (P>0.05). The fucoxanthin content was highest in LB-exposed cultures ($6.3{\mu}g/mL$), whereas LW showed the lowest ($3.6{\mu}g/mL$; P<0.05). Chlorophyll-a content was highest in cultures exposed to LB compared to other light sources. These results suggest consistent differences in growth and biochemical composition after exposure to light of different wavelengths.

Effects of Feeding Nattokinase as Natural Feed Additives on Milk Production and Blood Metabolites in Lactating Dairy Cows (천연 사료첨가제 Nattokinase 공급에 따른 젖소의 산유능력 및 혈액성상에 미치는 영향)

  • This experiment was conducted to determine the effect of nattokinase (NK) additives on milk production and composition, and blood metabolites in dairy cows. The two kinds of nattokinase with high fibrinolytic activity were produced by two strains of bacteria, Bacillus amyloliquefacines (NK1) and Bacillus subtilis (NK2). Total fifteen Holstein cows (average $1.83{\pm}0.37$ parity; average milk yield $23.2{\pm}3.2$ kg/d) were randomly assigned to three treatments (5 animals per treatment). Cows were fed TMR supplemented with 0g, 100g and 100g for control, NK1 and NK2 treatment, respectively for 4 weeks. Milk yield was significantly higher (p<0.05) for NK1 (22.89 kg/d) than for control (21.07 kg/d) and NK2 (21.36 kg/d). Somatic cell counts in NK treatments were significantly lower than that in control group (58,000 vs. 21,000 and 35,000 cells/ml, control vs. NK1 and NK2). Serum ALT levels in all treatment were similar to the range of 32.00~35.83 IU/L, but AST levels in NK1 (85.67 IU/L) was significantly decreased compared with those in control and NK2 (121.67 and 117.67 IU/L respectively). Serum T-CHO levels in NK1 (145.33 mg/dl) was significantly decreased (p<0.05) compared with that in control (179.00 mg/dl) and NK2 (176.17 mg/dl). This finding showed that NK1 additives could possibly have a positive effect in lactation performance of mid-lactation dairy cows by increasing milk yield, reducing somatic cell count, improving liver function and decreasing cholesterol in blood.

Valuable Organic Liquid Fertilizer Manufacturing through $TAO^{TM}$ Process for Swine Manure Treatment

  • $TAO^{TM}$ System is an auto-heated thermophilic aerated digestion process using a proprietary microbe called as a Phototropic Bacteria (PTB). High metabolic activity results in heat generation, which enables to produce a pathogen-free and digested liquid fertilizer at short retention times. TAO$^{TM}$ system has been developed to reduce a manure volume and convert into the liquid fertilizer using swine manure since 1992. About 100 units have been installed and operated in Korean swine farms so far. TAO$^{TM}$ system consists of a reactor vessel and ejector-type aeration pumps and foam removers. The swine slurry manure enters into vessel with PTB and is mixed and aerated. The process is operated at detention times from 2 to 4 days and temperature of 55 to $65^{\circ}C$. Foams are occurred and broken down by foam removers to evaporate water contents. Generally, at least 30% of water content is evaporated, 99% of volatile fatty acids caused an odor are removed and pathogen destruction is excellent with fecal coliform, rotavirus and salmonella below detection limits. The effluent from TAO$^{TM}$ system, called as the "TAO EFFLUX", is screened and has superb properties as a fertilizer. Normally N-P-K contents of screened TAO Efflux are 4.7 g/L, 0.375 g/L and 2.8 g/L respectively. The fertilizer effect of TAO EFFLUX compared to chemical fertilizer has been demonstrated and studied with various crops such as rice, potato, cabbage, pumpkin, green pepper, parsley, cucumber and apple. Generally it has better fertilizer effects and excellent soil fertility improvement effects. Moreover, the TAO EFFLUX is concentrated through membrane technology without fouling problems for a cost saving of long distance transportation and a commercialization (crop nutrient commodity) to a gardening market, for example.

Mass Production of Resting Egg of Korean Rotifer, Brachionus plicatilis (L and S-type) (한국산 Rotifer, Brachionus plicatilis (L과 S-type)의 내구란 대량생산)

  • Mass production of resting egg of the rotifer, Brachionus plicatilis was carried out in 1 $m^3$ tank for L-type rotifer and in 1 $m^3$ and 4 $m^3$ tank for S-type rotifer. L-type rotifer was fed on concentrated Chlorella + baker's yeast and only baker's yeast for 15 days. S-type rotifer was fed on concentrated Chlorella in 1 $m^3$ tank for 7 days and frozen Chlorella + baker's yeast in 4 $m^3$ for $6\~8$ days. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for L-type rotifer fed on Chlorella + baker's yeast were $54.5{\times}10^6$ eggs, $30.5{\times}10^6$ eggs and $100{\times}10^3$ eggs, respectively. These were higher than those of rotifer fed on baker's yeast only. Total number of resting egg and number of resting egg from $10^8$ rotifers and 1 g dry weight diet for S-type rotifer fed on concentrated Chlorella in 1 $m^3$ tank were $50\~104{\times}10^6$ eggs, $47\~82{\times}10^6$ eggs and $136\~260{\times}10^3$ eggs, respectively. In 4 $m^3$ tank with frozen Chlorella + baker's yeast, these were $149\~567{\times}10^6$ eggs, $36\~123{\times}10^6$ eggs and $131\~338{\times}10^3$ eggs, respectively. This result suggests that S-type rotifer are better than L-type rotifer for the mass production of resting egg of the Korean rotifer.

  • The occurrence of falcarinol(n-heptadeca-1, 9-dien-4,6-diyn-3-ol)in the umbelliferous plants such as Ladebouriella seseloides WOLFF., Cnidium officinale MAKINO, Foeniculum vulgare GAERTNER, Torilis japonica(HOUTT.) DC. Bupleurum falcatum L.,Angelica gigas NAKAI, Oenanthe javanica (BL.) DC. and Daucus carota var. sativa DC. was investigated using gas chromatography/mass spectrometry(GC/MS). Falcarinol was found in the root of Ledebouriella seseloides, the stem and the root of Oenanthe javanica, and the root of Daucus carota var. sativa in which its contents were $1,055\;{\mu}g/g$, $289\;{\mu}g/g$, $179\;{\mu}g/g$, and $212\;{\mu}g/g$, respectively, while it was not found in the other plants.

Phytochemicals and Antioxidant Activity of Codonopsis lanceolata Leaves (더덕 잎의 파이토케미컬(phytochemicals)과 항산화 활성)

  • Phytochemicals in Codonopsis lanceolata leaves were saponins, triterpenes, tannins, and flavonoids, not alkaloids. The levels of total polyphenols and flavonoids in Codonopsis lanceolata leaves were measured to evaluate the antioxidant activity. C. lanceolata leaves showed strong 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and potent reducing power. In addition, C. lanceolata leaf extracts inhibited production of nitric oxide (NO) in lipopolysaccharide (LPS)-activated RAW 264.7 cells. To examine active phytochemical for antioxidant activity, aglycone fraction of C. lanceolata leaves was analyzed by high performance liquid chromatography (HPLC). Luteolin was identified as a main component of aglycone fraction and showed potent antioxidant activity as determined by a DPPH radical scavenging assay and reducing power test. These results suggest that C. lanceolata leaves are a good source of antioxidants.

Four compounds were isolated from the extracted and two of these compounds were identified as ergosterol and stigmasterol by T.L.C., G.L.C. and chemical tests.

Chemiluminescence System with Air Pump as a Sensor for Determination of Metal Levels in Rain

  • A lab-made chemiluminescence system with air pump was developed for monitoring of some metal levels in rain. The air pump enabled injection of 17.7 $\mu$g samples into a glass cell filled with luminol-$H_2O_2$ reagent of typically 300 $\mu$L for chemiluminescence measurement. The monitored trend of total metal ions in the rain collected in our campus was compared with analytical results of each metal ion from GFAAS. The system was also demonstrated to determine $Cr^{6+}$ by reduction to $Cr^{3+}$ using $SnCl_2$. The limit of detection for $Cr^{6+}$ obtained by 4 measurements was 85.0 pg $mL^{-1}$ with a relative standard deviation of 3.4%. Although this system doesn’t have selectivity due to the characteristics of chemiluminescence, application of it to environmental monitoring as a sensor for some transition metal ions was demonstrated.

Production of Ascorbic acid-2-Glucoside from Ascorbic acid with Rice ${\alpha}-Glucosidase$ (벼의 ${\alpha}-Glucosidase$에 의한 Ascorbic acid로부터 Ascorbic acid-2-Glucoside의 생산)

  • For the enzymatic production of $2-O-{\alpha}-D-glucopyranosyl-L-ascorbic$ acid (AA-2G) from ascorbic acid, rice seed was used as the source of ${\alpha}-glucosidase$ having transglucosylation activity. Among six rice varieties, cultivated in Korea, ${\alpha}-glucosidase$ activity of Oryza savita L. cv. Ilpumbyeo was the highest with 125.03 unit/ml and it had maximum specific activity with 8.52 unit/mg protein when rice seeds were grown for 3 days after germination. For the production of AA-2G using crude extract of O. savita L. cv. Ilpumbyeo, maltose was most effective glucose donor. The optimum concentration of maltose and ascorbic acid were 125 mM and 175 mM, respectively. The optimum concentration of ${\alpha}-glucosidase$ was 100 unit. The most effective buffer was 100 mM sodium citrate. The optimum pH and temperature were 5.0 and $60^{\circ}C$, respectively. Under the optimum condition, $108.43\;{\mu}M/unit$ of AA-2G was produced from ascorbic acid after 35 minutes of reaction, which corresponds to 6.2% of conversion ratio based on the amount of ascorbic acid used.
