Direct Fermentation of D-Xylose to Ethanol by Candida sp. BT001

  • A yeast strain, BT001, which can directly ferment D-xylose to ethanol was isolated from forest soils, and then identified as Candida sp. Cultural conditions for the optimum ethanol production, along with the effects of aeration on cell growth and ethanol production were investigated. Aeration stimulated the cell growth and the volumetric rate of ethanol production, but decreased the ethanol yield. Optimum temperature and initial pH for the ethanol production were $33{\circ}^C$ and 6.0, respectively. In a shake flask culture, this strain produced 52.3 g ethanol per liter from 12%(w/v) D-xylose after incubation for 96 hours. Ethanol yield was 0.436 g per g D-xylose consumed. This corresponds to 85.8% of theoretical yield. Also, this yeast strain produced ethanol from D-galactose, D-glucose and D-mannose, but not from L-arabinose and L-rhamnose. Among these sugars, D-glucose was the fastest in being converted to ethanol sugars.

Analysis of Agrochemical Residues in Tobacco Using QuEChERS Method by GC-MS/MS

  • This study was performed to apply the more rapid and accurate sample preparation, and the high selectivity and sensitivity of the analyte detection by gas chromatography-triple quadrupole mass spectrometry (GC-MS/MS). QuEChERS (quick, easy, cheap, effective, rugged and safe) method was validated for 49 agrochemicals in the CORESTA Agrochemical Advisory Committee guide and amenable to GC-MS/MS determination. In QuEChERS method, the effects of sorbents (PSA, $C_{18}$ and GCB) and matrix of the analytes in tobacco types (flue-cured, burley and oriental) were investigated. MS/MS acquisition provided high specificity and selectivity for agrochemicals and low limit of quantification. QuEChERS by using PSA alone and the matrix-matched standards gave good recoveries and RSD values in three types of tobaccos. QuEChERS method was no needed to be complex clean-up procedure and would be used as the fast and easy method for agrochemical residue analysis in tobacco.

Production of Ginsenoside Rd from Ginsenoside Rc by ${\alpha}-{\small{L}}$-Arabinofuranosidase from Caldicellulosiruptor saccharolyticus

  • Ginsenoside Rd was produced from ginsenoside Rc using a thermostable recombinant ${\alpha}-{\small{L}}$-arabinofuranosidase from Caldicellulosiruptor saccharolyticus. The optimal reaction conditions for the production of ginsenoside Rd from Rc were pH 5.5, $80^{\circ}C$, 227 U enzyme/ml, and 8.0 g/l ginsenoside Rc in the presence of 30% (v/v) n-hexane. Under these conditions, the enzyme produced 7.0 g/l ginsenoside Rd after 30 min, with a molar yield of 100% and a productivity of 14 g $l^{-1}\;h^{-1}$. The conversion yield and productivity of ginsenoside Rd are the highest reported thus far among enzymatic transformations.

Transmission of Substituent Effects through 5-Membered Heteroaromatic Rings

  • Ab initio calculations are carried out on protonation equilibria of 5-memberedheteroaromatic aldehydes (5MHAs;heteroatom Y = NH, O, PH,and S and substituentZ = NH2, OCH3, SCH3, CH3, H, Cl, CHO, CN,NO2) at the $MP2}6-31G*$ level. Naturalbond orbital (NBO) analyses show that the optimal localized natural Lewis structures of the protonated aldehydes,(P), are ortho (C3) protonated (for Y = O, PH and S) and N-pro-tonated (for Y = NH) forms in contrast to the standard structural Lewis formula for aldehydes, (R). The delo-calizability of ${\pilone}-pairon$ the heteroatom $(n{\pi}(Y))$ is in the order Y = NH > O > S > PH. The transmission efficiency of (Z) substituent effects to the carbonyl moiety run parallel to the delocalizability of $n{\pi}(Y)$ for R,but is dominantly influenced by the cationic charge on $C{\alpha}(C{\alpha}+)$ for P, which is in the reverse order of thede-localizability of $n{\pi}(Y).$ The Hammett ${\rho}values$ for variation of Z in the protonation are determined by the dif-ference in the transmission efficiencies between Pand R stateat simple interpretation of their magnitude is not warranted. However,the magnitude of the gas-phase ${\rho}z+$ values decreases as the level ofcomputation is raised from RHF/3-21G* to RHF/6-31G* and to $MP2}6-31G*$ but increases again at the MP4SDQ/6-31G* level. Further decrease occurs when solvent effect (water) is accounted for by the SCRF method. Comparison of the SCRF ${\rho}z+values$ with those determined in the aqueous acid solution for Y = S and CHCH shows inadequacy of accounting for the solvent effects on the ${\rho}values$ by a continuum model. It is noteworthy that semiempirical calculations, especially theAM1 method, give even lower magnitude of the gas-phase ${\rho}values.

Effect of Cordycepin on the Expression of the Inflammatory Cytokines TNF-alpha, IL-6, and IL-17A in C57BL/6 Mice

  • Culture supernatants of splenocytes from C57BL/6 mice were exposed to 0.3, 1.0, and 3.0 ${\mu}g/ml$ cordycepin plus 3.0 ${\mu}g/ml$ lipopolysaccharide (LPS) to investigate the effects of cordycepin (3'-deoxyadenosine) on the production of inflammatory cytokines. Coadministration of 3.0 ${\mu}g/ml$ cordycepin with LPS in cultured murine spleen cells significantly diminished expression of the inflammatory cytokines tumor necrosis factor-${\alpha}$ and interleukin-6 (IL-6) in a time-dependent manner. Expression of the inflammatory cytokine IL-17A was substantially downregulated in a timeand concentration-dependent manner at all cordycepin concentrations. These findings suggest that cordycepin downregulates the immediate hypersensitivity reaction stimulated by LPS.

Acute Toxicity of Various Heavy Metals on Mysid, Neomysis japonica (곤쟁이, Neomysis japonica에 대한 여러 가지 중금속의 급성독성)

  • In order to estimate acute toxicity of various heavy metals on mysid, Neomysis japonica, the experiments were conducted by renewal bioassay method at $20\pm0.5^{\circ}C$ and $30\pm0.3$ psu salinity. The 96 hr $LC_{50}$ of Neomysis japonica exposed to cadmium, copper, zinc, arsenic, lead and chromium were $90.4\;{\mu}g/L$, $82.3\;{\mu}g/L$, $646.1\;{\mu}g/L$, $1488.5\;{\mu}g/L$, $1758.8\;{\mu}g/L$ and $1609.6\;{\mu}g/L$, respectively, and were ranked in order of toxicity: copper > cadmium > zinc > arsenic > chromium > lead.

Electrical Noise Reduction and Stiffness Increase with Self Force-Balancing Effect in a High-Resolution Capacitive Microaccelerometer using Branched Finger Electrodes with High-Amplitude Sense Voltage (고감지전압 및 가지전극을 이용한 고정도 정전용량형 미소가속도계의 전기적 잡음 감소 및 자율 균형력 발생에 의한 강성 증가)

  • This paper presents a high-resolution capactive microaccelerometer using branched finger electrodes with high-amplitude sense voltage. From the fabricated microacceleromcter, the total noise is obtained as 9 $\mu\textrm{g}$/√Hz at the sense voltage of 16.5V, while the conventional microaccelerometers have shown the noire level of 25~800 $\mu\textrm{g}$/√Hz. We reduce the mechanical noise level of the microaccelerometer by increasing the proof-class based on deep RIE process of an SOI wafer. We reduce the electrical noise level by increasing the amplitude of AC sense voltage. The nonlinearity problem caused by the high-amplitude sense volage has been solved by a new electrode design of branched finger type, resulting in self force-balancing effects for the enhanced linearity and bandwidth. The fabricated microaccelerometer shows the electrical noise of 2.4 $\mu\textrm{g}$/√Hz at the sense voltage of 16.5V, which is an order of magnitude reduction of the electrical noise of 24.3 $\mu\textrm{g}$/√Hz measured at 0.9V. For the sense voltage higher than 2V, the electrical noise of the microaccelerometer is lower than the voltage-independent mechanical noise of 11 $\mu\textrm{g}$/√Hz. Total noise, composed of the electrical noise and the mechanical noire, has been measured as 9 $\mu\textrm{g}$/√Hz at the sense voltage of 16.5V, which is 31% of the total noise of 28.6 $\mu\textrm{g}$/√Hz at the sense voltage 0.9V. The self force-balancing effect in the blanched finger electrodes increases the stiffness of the microaccelerometer from 1.1N/m to 1.61N/m as the sense voltage increases from 0V to 17.8V, thereby generating additional stiffness at the rate of 0.0016$\pm$0.0008 N/m/V$^2$.

  • Marine fungus, Thraustochytrium aureum ATCC 34034, was incubated in artificial sea water media in order to produce docosahexaenoic acid(DHA). Cultures were performed at $24^{\circ}C$ in light for 13 days with orbital shaker at 100rpm. Maximum cell productivity of 1.34g/L and DHA yield of 41.4 mg/L were obtained by using this method, which is almost the twice level of DHA yield obtained for the strains reported previously. This strain did produce much more DHA after sporangium disruption.

Distribution of Polycyclic Aromatic Hydrocarbon at Kongsfjorden in Spitsbergen, Svalbard Islands (북극 스발바드 군도 스피츠베르겐섬 콩스피요르드에서의 다환 방향족 탄화수소화합물의 분포 특성)

  • In order to elucidate the polycyclic aromatic hydrocarbon concentration and its origin in arctic area, four arctic brown algae (Laminaria saccharina, L. digita, Alaria esculenta, Desmarestia aculeata), one marine invertebrate (Echinoidea) and sediments were collected from Kongsfjorden in Spitsbergen from the late July to early August, 2003. In case of macroalgae, the young blade part above growth point and the old stipes and blades beneath growth point were separated and analyzed for polycyclic aromatic hydrocarbons (PAHs) in an attempt to check the mechanism of uptake in macroalgae to accumulate PAH. There was no difference in PAH concentrations between sampling sites (Stations B and C), species, and blades beneath and above growth point. PAH concentrations in all samples collected in this study were relatively higher than those reported in other areas of arctic. Especially, station C, which is known as an unpolluted area, showed 10 times higher PAH concentration (8,765 ng/g) in sediment than station A (694 ng/g) around harbor. In addition high PAH concentration, station C had very higher proportion of methylated PAH to parent PAH in sediment than station A. Source analysis using PAH isomer pair ratios as indicators showed that Kongsfjorden area seemed to be relatively contaminated with PAH derived from direct petroleum input.
