Volatile Substances and Physicochemical Characteristics of Pyroligneous Liquor (목초액의 휘발성 성분과 이화학적 특성)

  • To develop food additives from pyroligneous liquor, we first examined the physicochemical characteristics of volatile substances. The pH, gravity, acidity, solution tar, and scorched remains in the pyroligneous liquor were 2.50, 1.020, $0.8\%$ $2.31\%$ and $0.008\%$ respectively. The main compounds of pyroligneous liquor were 2,6-dimethoxyphenol (syringol), 2-hydroxy-2-cyclopenten-l-one, 1,2-benzenediol (catechol), phenol, 2 (5H)-furanone, 2-methoxyphenol (guaiacol), 5-hydroxymethyl-2 furancarboxaldehyde were found in organic phase, whereas catechol, 1,4 : 3,6-dianhydro-${\alpha}d$-glucopyranose, cyclopropyl carbinol, 5-hydroxymethyl-2 furancarboxaldehyde in aqueous phase.

Changes in Volatile Compounds of Schizandra chinensis Fruits According to Drying and Extracting Methods (건조 및 추출방법에 따른 오미자 휘발성 성분의 변화)

  • To understand the effects of drying conditions on changes of volatile compounds in fruits of Schizandra chinensis, we analyzed SDE (steam distillation and extraction) extract and Headspace vapor of fresh and dried samples using GC/MS (Gas chromatograph/Mass spectrometer). Contents of essential oils from samples with different drying conditions were 0.58% in fresh ones, 0.60% in freeze dried ones, and 0.30% in hot-air dried ones. In SDE extract, major volatile compounds in fresh samples were terpinen-4-ol(9.01%), ${\gamma}-terpinene(7.02%),\;{\beta}-myrcene(7.55%)$, unidentified sesquiterpenes(28.48%), showing almost the same composition as that in freeze-dried ones, but those in hot-air dried samples at $60^{\circ}C$ were ${\gamma}-terpinene(5.40%),\;{\alpha}-elemene(8.28%)$, unidentified sesquiterpenes(50.38%), indicating the chemical changes during drying procedure. In Headspace vapor, major compounds in fresh samples were ${\beta}-myrcene(22.05%),\;{\gamma}-terpinene(9.47%),\;{\alpha}-pinene(8.91%)$, sabinene(8.48%), which were different from those in SDE extract. In chemical compositions of volatile compounds in dried samples, ${\beta}-myrcene,\;{\alpha}-terpinene$ decreased in the order of freeze-drying > hot-air drying at $60^{\circ}C$ > hot-air drying at $60^{\circ}C$, and ${\alpha}-ylangene,\;{\alpha}-pinene$, camphene increased in the reverse order of the former. We observed the changes of the contents and compositions of essential oils compounds during drying procedure, especially a decrease in monoterpenes and alcohols and an increase in sesquiterpenes with relatively weak volatility.

Residual Polychlorinated Biphenyls(PCBs) in the Sediment of the Kumho River (금호강(琴湖江) 저니토중(底泥土中) Polychlorinated Biphenyls(PCBs)의 잔류(殘留))

  • The residual Polychlorinated biphenyls(PCBs) in the sediment of the Kumho River basin were measured because the Nakdong River was considered to be affected by the environmental quality of Kumho River. PCBs in the sediment were chlorinated and converted into Decachlorinated biphenyl(DCB) which showed the single peak in the GC-ECD chromatogram. The detectable concentrations of PCBs was 0.5ng/g in the sediment. PCBs in the 21 sediment samples was of Kumho River located at 7.5km from conjunction of Nakdong River were not detected from 100 samples. 49% of samples was showed lower than 50ng/g, and 9% did more than 400ng/g. The mean of PCBs concentrations was 131ng/g. The means of COD and organic carbon were 14.5mg/g and 3.41%, respectively. The relationship between PCBs were concentration and the distance from conjunction with Nakdong River was not significant. The concentration of PCBs were varied with the depth of the sediment down to the 250cm depth. The concentrations of PCBs in the depth 0-20cm and 50-90cm were higher than the mean concentration of PCBs. Along the cross of the stream, the concentrations of PCBs were 142ng/g at the center and 126 and 131ng/g at the river sides.

Effect of Drying Condition on the Colors and Flavors Change of Fresh pepper (건조조건에 따른 고추의 색도와 향기 성분의 변화)

  • The moistures, colors and flavors of red pepper were analyzed to observe the changes of the qualities of red pepper with various conditions of drying. The moisture was 11.8%, known as optimal condition for storing red pepper, where dried at $50^{\circ}C$ for 36 and 48 hours. The color of red pepper air-dried at 50% for 48 hours was estimated to be the most execllent in comparison of sun drying and air drying. The optimal absorbances of hexnae extract from red pepper were examined. The ratios between the absorbances at 280nm and at the typical wavelengths of caretenoids(430nm, 450nm, 474nm) were from 2.5-3.4 and these wete also confirmed by the sight of the eye. Hexane fraction showed more peaks of flavors than benzene fraction and both of the two were silylated by with BSTFA to analyze the flavors by GC. CG profiles for the compositions of flavors in red perpper are thought to be useful for extimating the quality of favors in red pepper. Eight flavors including benzene dicarbozylic acid, were identified from red pepper and major components of them were oleic palmitic acids. Drying temperatures and times did not have effects on the changes of specific flavor components but did influenced the compositions of them in red pepper.

Comparison of the Quality Characteristics of Korean Native Black Pork and Modern Genotype Pork During Refrigerated Storage (재래종 흑돼지육과 개량종 돼지육의 냉장저장중 품질비교)

  • This study was carried out to compare the quality characteristics including aroma compounds and pattern of M. Biceps femoris from Korean native black pigs (70kg, 6 months, KNP) and modern genotype pigs (103kg, 6 months, Landrace×Yorkshire, MGP) during vacuum-storage at 2±0.2℃ for 12 days. KNP had high contents of crude protein and ash, but low content of moisture as compared to MGP (p<0.05). KNP showed lower pH and higher cooking loss than MGP (p<0.05) during storage. CIE a* and b* values of KNP significantly higher during storage as compared to those of MGP (p<0.05). TBARS value of KNP significantly increased after 8 days as compared to that of MGP (p<0.05). Shear force value of KNP was higher than that of MGP until 8 days (p<0.05). Volatiles by SPME-GS/MS was slightly different between KNP and MGP, but aroma pattern by electronic nose was not different. Therefore, KNP had redder and darker color, higher cooking loss and harder texture than MGP. The sensory quality of KNP was better than those of MGP (p<0.05), although aroma characteristics by instruments were not different between KNP and MGP.

Accurate Analysis of Trace Phthalates and Method Validation in Cosmetics using Gas Chromatography with Mass Spectrometric Detection (화장품에 함유된 미량의 프탈레이트 함량을 정확히 분석하기 위한 가스크로마토그래피-질량분석 시험법 및 그 시험법의 유효성)

  • An effective, environmentally friendly analytic methods using gas chromatography with mass spectrometric detector (GC-MSD) have been developed for the quantitative analysis of trace phthalate levels in cosmetics such as nail lacquer and hair spray. Since such cosmetics are largely comprised of organic solvents, conventional clean-up methods that have been widely used for phthalate analyses are in adequate. In addition, analysis of trace phthalate levels is notorious for its sensitivity to contamination, which causes high analytical values. A direct sample dilution method using an organic solvent was adopted to the sample preparation process to determine the exact amounts of phthalates and simultaneously avoid the high risk of secondary contamination. The method has many advantages including high accuracy, sensitivity, and simplicity in sample preparation. Dibutyl phthalate (DBP) and di (2-ethylhexyl) phthalate (DEHP) were selected for analysis because they have been frequently detected in cosmetics and consistently reported as endocrine disruptors in humans and animals. Internal standard method using two deuterium substitutes (DBP-$d_4$, DEHP-$d_4$) as the internal standard was also used. The results of 'Method validation' showed the capabilities of this method for the routine analysis of phthalates at the ppm level. The recovery ranges were between 95 % and 106.1 %, and relative standards deviations (RSD) were less than 3.9 % in fortified nail lacquer and hair spray samples at the concentration of $25{\mu}g/g$.

Study on The Thermochemical Degradation Features of Empty Fruit Bunch on The Function of Pyrolysis Temperature (반응온도에 따른 팜 부산물(empty fruit bunch)의 열화학적 분해 특성에 관한 연구)

  • We performed fast pyrolysis of empty fruit bunch (EFB) in the range of temperature from $400{\sim}550^{\circ}C$ and 1.3 s of residence time. The effect of temperature on the yields and physicochemical properties of pyrolytic products were also studied. Elemental and component analysis of EFB showed that the large amount of potassium (ca. 8400 ppm) presents in the feedstock. Thermogravimetric analysis suggested that the potassium in the feedstock catalyzed degradation of cellulose. The yield of bio-oil increased with increasing temperature in the range of temperature from $400{\sim}500^{\circ}C$, while that of gas and biochar decreased and showed monotonous change each with increasing temperature. When the EFB was pyrolyzed at $550^{\circ}C$, the yield of bio-oil and char decreased while that of gas increased. Water content of the bio-oils obtained at different temperatures was 20~30% and their total acid number were less than 100 mg KOH/g oil. Viscosity of the bio-oils was 11 cSt (centistoke), and heating value varied from 15 to 17 MJ/kg. Using GC/MS analysis, 27 chemical compounds which were classified into two groups (cellulose-derived and lignin-derived) were identified. Remarkably the concentration of phenol was approximately 25% based on entire chemical compounds.

Analysis of the Volatile Organic Compounds of Persimmon Flower according to Tree Age and Floral Organ (감나무 수령과 감꽃 기관에 따른 휘발성 향기성분 분석)

  • This study was conducted to investigate volatile organic compounds (VOCs) of persimmon (Diospyros kaki Thunb) flower. VOCs of persimmon flower was collected via SPE (solid phase micro extraction) and determined by GC-MS according to tree age and organs such as flower and calyx. The ratio of early bloom was higher in more than 15 year old tree than other trees showing tree age was related with flowering rate. Major VOCs of persimmon flower was a-pinene, butane, caryophyllene, cubebene, lavandulol, D-limoneneylangene, ylangene, mainly included green, fruit, and floral flavors. The number of VOCs in persimmon flower was 30 compounds in 5-9 years old tree, 24 compounds in 10-14 years old tree, and 32 compounds in more than 15 years old tree. In comparison with VOCs in organs of sweet persimmon 'Fuyu' cultivar, flower has 10 compounds of VOCs and 26.35% of relative peak area, while calyx has 14 compounds and 46.28%, respectively. In astringent persimmon, flower has 6 compounds of VOCs and 17.58% of relative peak area, while calyx has 9 compounds and 50.27%, showing calyx of both cultivars has various volatile compounds. This study will contribute to provide a basic data for the fragrance industry to use the flavor of persimmon flower.

Effects of Extracts of Monascus-fermented Angelica gigas Nakai in High-fat Diet-induced Obesity in Rats (고지방 식이 유도 비만 흰쥐에서 홍국발효 참당귀 추출물의 항비만 효과)

  • This study aimed to identify the effects of extracts of fermented Angelica gigas Nakai (A. gigas) with a Monascus purpureus strain on anti-obesity in a high-fat diet (HFD)-induced obesity rat model. Male Sprague-Dawley rats were randomly divided into seven dietary groups (n=8 per group), as follows: a normal diet group (N) and six HFD groups (C: control, HFD and no treatment; AG: HFD +10% A. gigas extracts; FAG2.5: HFD +2.5% fermented A. gigas extracts; FAG5: HFD +5% fermented A. gigas extracts; FAG10: HFD +10% fermented A. gigas extracts; GC: HFD + Garcinia cambogia extracts). FAG-fed rats exhibited effectively lowered rates of increasing body weight and visceral fat accumulation in the HFD-induced obesity model. The activities of several hepatic marker enzymes, including aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactate dehydrogenase (LDH), and alkaline phosphatase (ALP), were increased with HFD-induced obesity, but levels of these marker enzymes were significantly decreased in FAG-fed rats. The consumption of FAG reduced triglyceride concentrations in serum to normal levels. FAG-fed rats showed effectively increased leptin concentrations in the HFD-induced obesity model. HFD ingestion induced a significant increase in the thiobarbituric acid reactive substances (TBARS) levels, which was decreased in FAG-fed rats. Hematoxylin and eosin staining and Oil Red O staining of the liver showed that the lipid deposits were decreased via FAG feeding. Moreover, hematoxylin and eosin staining of epididymal adipose adipose tissue showed that the adipocyte were decreased by feeding FAG.

Simultaneous Analysis of Antimicrobial Three Straight Chain 1,2-alkanediols in Cosmetics by Gas Chromatography (가스크로마토그래피를 이용한 화장품 중 항균작용의 3종의 직쇄상 1,2-알칸디올류의 동시분석)

  • Preservatives such as paraben, phenoxyethanol, and chlorphene are commonly used in cosmetics thanks to cheap price and good antiseptic effect. Recently, consumers' concerns about their possible toxicity and skin irritation forced them to be replaced with straight 1,2-alkanediols. However, as the alkanediols may also irritate skin, limited amount of them should be used in cosmetics. Three 1,2-alkanediols including 1,2-pentanediol, 1,2-hexanediol, and 1,2-octanediol in cosmetics were analyzed simultaneously by gas chromatogrphy with flame ionization detector. As a result of method validation, the specificity was confirmed by the calibration curves of 1,2-pentanediol, 1,2-hexanediol, and 1,2-octanediol showing good linearity correlation coefficient of above 0.999 over the concentration range of $100{\sim}1,200{\mu}g/g$. The limit of detection (LOD) and quantification (LOQ) of 1,2-pentanediol, 1,2-hexanediol, and 1,2-octanediol were 31, 40, and $19{\mu}g/g$ and 98, 108, and $57{\mu}g/g$, respectively. The precision (Repeatability) of the amount in cosmetics showed less than 2.0%. Relative Standard Deviation (% RSD) and the Accuracy (% recovery) of the amount in cosmetics showed 99.3 ~ 103.3, 99.4 ~ 106.7, 97.5 ~ 107.3% respectively. As a result, simultaneous analysis of antimicrobial three 1,2-alkanediols in cosmetics were possible. This method can be utilized in accurate quantitative analysis of 1,2-alkanediols in cosmetics.
