Effects of Riboflavin-photosensitized Oxidation on the Formation of Volatile Compounds in Oleic Acid Model Systems (리보플라빈 광산화에 의한 올레산 모델시스템에서 휘발성 물질 발생연구)

  • Formation mechanisms of light-induced volatile compounds were studied using riboflavin-photosensitized oleic acid model systems. Volatile compounds in model systems with 4000 ppm riboflavin at $35^{\circ}C$ under light or in the dark for 39 hr were isolated and identified by a combination of solid phase microextraction (SPME), gas chromatography (GC), and mass spectrometry (MS). Total volatiles in oleic acid with riboflavin under light for 13, 26, and 39 hr increased by 90, 190, and 270%, respectively, compared to those in oleic acid without riboflavin under light. Total volatiles in samples without riboflavin under light or samples with riboflavin in the dark were not significant in tested conditions (p>10.05). Riboflavin did not act as a photosensitizer when it was dispersed in oleic acid. Heptane, octane, heptanal, octanal, nonanal, and 2-nonenal were significantly increased in riboflavin-photosensitized samples compared to those in samples without riboflavin (p<0,05). Light-induced volatile compounds including heptanal and 2-nonenal from oleic acid could be explained using singlet oxygen oxidation, the formation mechanism of which were not understood using triplet oxygen oxidation. These results will help to understand volatile formation in oleic acid containing foods stored under light.

  • The chemical composition and content of essential oils extracted by ASTM standard D 889-93 method from the leaves of thirty introduced varieties in Perilla frutescens Britton var. japonica $H_{ARA}$ was investigated and compared. The oil was analysed by gas chromatography. More than forty compounds were found and thirteen compounds were identified. The major three constituents representing about $56{\sim}88%$ of total oil were shown caryophyllene and 2 unknown oils in 25 varieties. The chemical composition and content had much differences in each of varieties. This result seemed to be corresponding to introduce that varieties had diverse essential oils.

  • Volatile organic compounds (VOCs) are an important public health problem throughout the world. Many important questions remain to be addressed in assessing exposure to these compounds. Because they are ubiquitous and highly volatile, special techniques must be applied in the analytical determination of VOCs. Personal exposure measurements are needed to evaluate the relationship between microenvironmental concentrations and actual exposures. It is also important to investigate exposure frequency, duration, and intensity, as well as personal exposure characteristics. In addition to air monitoring, biological monitoring may contribute significantly to risk assessment by allowing estimation of absorbed doses, rather than just the external exposure concentrations, which are evaluated by environmental and personal monitoring. This study was conducted to establish the analytic procedure of VOCs in air, blood, urine and exhaled breath and to evaluate the relationships among these environmental media. The subjects of this study were selected because they are occupationally exposed to high levels of VOCs. Environmental, personal, blood, urine and exhalation samples were collected. Purge & trap, thermal desorber, gas chromatography and mass selective detector were used to analyze the collected samples. Analytical procedures were validated with the“break through test”, 'quot;recovery test for storage and transportation”,“method detection limit test”and“inter-laboratory QA/QC study”. Assessment of halogenated compounds indicted that they were significantly correlated to each other (p value < 0.01). In a similar manner, aromatic compounds were also correlated, except in urine sample. Linear regression was used to evaluate the relationships between personal exposures and environmental concentrations. These relationships for aromatic and halogenated are as follows: Halogen $s_{personal}$ = 3.875+0.068Halogen $s_{environmet}$, ($R^2$= .930) Aromatic $s_{personal}$ = 34217.757-31.266Aromatic $s_{environmet}$, ($R^2$= .821) Multiple regression was used to evaluate the relationship between exposures and various exposure deter-minants including, gender, duration of employment, and smoking history. The results of the regression model-ins for halogens in blood and aromatics in urine are as follows: Halogen $s_{blood}$ = 8.181+0.246Halogen $s_{personal}$+3.975Gender ($R^2$= .925), Aromatic $s_{urine}$ = 249.565+0.135Aromatic $s_{personal}$ -5.651 D.S ($R^2$ = .735), In conclusion, we have established analytic procedures for VOC measurement in biological and environmental samples and have presented data demonstrating relationships between VOCs levels in biological media and environmental samples. Application of Multiresidue Analysis Method of Unregistered Pesticides in Korea for Imported Food (수입식품 중 국내 미등록 농약의 다성분 잔류분석법 적용)

  • BACKGROUND: Recently in Korea, the import of agricultural products is rising due to the increasing amount of trade. Unregistered pesticides, allidochlor, propachlor, propham, cycloate, diallate and pebulate are widely used as pesticides for rice cultivation in foreign countries, while they are not registered in Korea. Therefore, the residue amount of imported agri-foods should be verified using the proper official analytical method for each of them that has not registered in Korea. METHODS AND RESULTS: This work was conducted to apply the official method of Korea Food & Drug Administration (KFDA) for determining multi class pesticide multiresidues in agricultural commodities. Brown rice and orange which have different characteristics as a matrix were selected as representative samples for residue analysis. The recoveries of cycloate, diallate and pebulate by GC/MS in fortified brown rice and orange with levels of 0.04~0.4 mg/kg were ranged from 82.8% to 110.3%. The quantification limits of three pesticides in brown rice and orange were 0.04 mg/kg. CONCLUSION: As a result, this method can surely be used as an official method for routine analysis of unregistered pesticides in Korea for imported agri-food.

Characteristics of pollutant emission from wallpapers - Around TVOC and HCHO - (벽지에서 발생되는 오염물질 방출특성 - TVOC와 HCHO를 중심으로 -)

  • This study is to investigate the characteristics of emission concentration according to wallpaper sort and emission time using small chamber method. The target compounds included 45 VOCs and formaldehyde, which were respectively determined by adsorption sampling and thermal desorption coupled with GC/MS method, and by sampling in DNPH cartridge and HPLC method. The emission factor of TVOC and HCHO was detected to $1.1mg/m^2{\cdot}h$ and $0.01mg/m^2{\cdot}h$ respectively, and the wallpapers of 25 satisfied emission standard. TVOC emission factor appeared in order of the concentration of PVC, natural, and Non-PVC wallpaper, while HCHO was detected very low concentration without relation to wallpaper sort. The paraffin hydrocarbons appeared to be the most contributable class of hydrocarbons in terms of their concentrations, followed by aromatics, and olefins, halogenated hydrocarbons was not detected. PVC wallpapers plentifully emitted TVOC above other wallpapers, and toluene was showed higher concentration of 10 times than natural wallpaper. In addition to, emission factor according to elapse was gradually decreased.

Mechanism of Action of Prochloraz and Triadimefon on Valsa ceratosperma (Prochloraz 와 Triadimefon의 사과나무 부란병균(腐爛病菌)에 대(對)한 작용기작(作用機作))

  • In order to elucidate the action mechanism of prochloraz and triadimefon, the mycelia of the Valsa ceratosperma were treated with the compounds in vitro. Analysis by GLC of the sterol extract from Valsa ceratosperma mycelia revealed one major peak and two minor peaks. Their relative retention time(RRT) relative to chloresterol were 1.29, 1.48 and 1.82, The compounds with RRT 1.29 and RRT 1.82 were identified as ergosterol and 24-methylenedihydrolanosterol by GC/MS, respectively. Five ppm of prochloraz and triadimefon applied to mycelia caused decrease in ergosterol content, whereas increase in 24-methylenedihydrolanoserol content in mycelia. The longer treatment time and the higher concentrations of the chemicals resulted in the greater decrease in ergosterol and the greater increase in 24-methylenedihydrolanosterol. Based on the analysis, it is considered that the two chemicals inhibit the ergosterol biosynthesis in Valsa ceratosperma by blocking C-14 demethylation as found previously in other fungi arid yeasts.

Screening of Edible Mushrooms for the Production of Lovastatin and its HMG-CoA Reductase Inhibitory Activity (Lovastatin을 생산하는 식용버섯 선발과 HMG-CoA reductase 저해 효과)

  • This research was performed to determine the production of lovastatin and its HMG-CoA reductase activity produced by fruit bodies and mycelial liquid cultures of domestic edible mushrooms (8 fungal strains). By deter-mining TLC analysis for the confirmation of the presence of lovastatin, all the extracts from fruit bodies and mycelial liquid culture showed same Rf value (0.46), whick was identical to that of the standard lovastatin. In order to extract lovastatin from fruit body, the mixture of water/acetonitrile/methanol was chosen as the most effective solvent. Extracts from fruit body and mycelial liquid culture of pleurotus ostreatus produced the high-est lovastatin 0.98 mg/g based on dry biomass, and 21.90 mg/L, respectively. In the inhibition rate of 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) reductase, the highest was obtained in P. ostreatus as 67.8% among fruit bodies, and the rates of mycelial liquid culture extracts from P. ostreatus and Laetiporus sulphureus were 37.2% and 29.1%, respectively. Unusually L. sulphureus showed high inhibition rate with low content of lovastatin due to the contribution of campesterol and gamma-sitosterol with hypocholesterolemic activity as metabolites.

  • 새로운 지능기반사회(ubiquitous society) 패러다임이란 전제 아래 미국을 비롯해 일본, 유럽 등 세계 각국에서는 모바일, 브로드밴드, 극소형 컴퓨터, IPv6의 기술이 창출해 내는 컴퓨팅 혁명의 실체를 유비궈터스 IT로 파악하고 각 국의 걸부, 기업, 연구소들이 주도권을 잡기 위해 많은 노력을 기울이고 있다. 현재 유비쿼터스 IT를 활용한 u-비즈니스를 활성화하고 관련 시장을 선점하기 위한 전 세계적인 경쟁은 매우 치열한 양상을 보이고 있다. 주요 국가들은 서로 지향하는 영역은 다르지만 궁극적으로 지능기반사회 구현이라는 대명제 아래 독자적인 산업 구축을 위해 노력하고 있다. 이러한 이유는 기본적으로 지능기반사회로의 이행과정에서 발생하는 경제적 파급효과, 예컨대 디지털 가전, 이동통신기기, 전자의료기기, 디스플레이, 2차 전지, 보안 등 관련 산업에 미치는 영향 및 새로운 시장 창출의 기회가 있기 때문이다. 기존의 많은 연구 및 문헌에서는 아직도 유비쿼터스 IT에 대한 개념적인 논의에 치중되어 있고 시장 중심적 또는 수요자 중심적 사고의 논의가 부족하다. 성공적으로 유비쿼터스 산업을 육성하기 위해서는 유비쿼터스 IT자체기술 개발사업(tangible service) 뿐만 아니라 비가시적(intangible) 서비스, 그리고 컨버전스(convergence) 서비스에 대한 상용화 모델과 성공적 사업 전개 방법론에 대한 깊은 연구가 수행되어야 한다. 본 연구에서는 먼저 유비쿼터스 IT의 기술동향과 시장 동향 및 산업 규모에 대한 문헌연구를 통해 u-비즈니스의 가능성을 고찰해보고, u-비즈니스 추진 전략을 탐색적으로 제시하고자 한다.되었다. SPME-GC/MS에 의한 휘발성분 분석 결과, 기능성 유지에서 홍국쌀 에탄올 추출물로부터 기인되는 특유의 휘발성분은 관찰되지 않았으나, RT=12.1 min인 hexanal의 함량이 control에 비하여 유의적인 감소 경향을 나타내는 것이 확인되었다(p<0.05).히지 못하여, 이의 기전을 밝히기 위한 추후 연구가 필요하다./TEX>로서 p<0.01 수준에서 현저히 증가하였다. 혈청의 인지질과 콜레스테롤 에스테르의 지방산 조성에서 vitamin C를 혼합 투여한 3,5,7군에서 PUFA 함량이 증가한 반면, SFA 함량은 감소하여 P/S 비율, n-3P/n-6P 비율은 증가하는 경향이었으며 이는 간장의 인지질, 콜레스테롤 에스테르, 총 지질의 지방산조성에서도 같은 경향을 볼 수 있었다.X>$(C_{18:2})$와 n-3계 linolenic acid$(C_{18:3})$가 대부분을 차지하였다. 야생 돌복숭아 과육 중의 지방산 조성은 포화지방산이 16.74%, 단불포화지방산 17.51% 및 다불포화지방산이 65.73%의 함유 비율을 보였는데, 이 중 다불포화지방산인 n-6계 linoleic acid$(C_{18:2})$와 n-3계 linolenic acid$(C_{18:3})$가 지질 구성 총 지방산의 대부분을 차지하는 함유 비율을 나타내었다.했다. 하강하는 약 4일간의 기상변화가 자발성 기흉 발생에 영향을 미친다고 추론할 수 있었다. 향후 본 연구에서 추론된 기상변화와 기흉 발생과의 인과관계를 확인하고 좀 더 구체화하기 위한 연구가 필요할 것이다.게 이루어질 수 있을 것

Isolation and Identification of Low Molecular Phenolic Antioxidants from Ethylacetate Layer of Korean Black Raspberry (Rubus coreanus Miquel) Wine (복분자(Rubus coreanus Miquel) 와인의 에틸아세테이트층으로부터 저분자 페놀성 항산화 화합물의 단리·동정)

  • Five antioxidative active substances were isolated from the EtOAc layer (20 g/56.2 g) of Korean black raspberry (Rubus coreanus Miquel) wine (11 L, black raspberry 15.7 kg fresh wt. eq.) by various column chromatography and high performance liquid chromatography (HPLC). Proton nuclear magnetic resonance ($^1H$-NMR) spectroscopy and gas chromatography Electro Ionization-Mass Spectrometry (GC-EI-MS) identified these as 4-hydroxybenzoic acid (1, 0.1 mg), 3,4-dihydroxybenzoic acid (2, 0.3 mg), 4-(2-hydroxyethyl)-phenol (3, 0.6 mg; tyrosol), pyrocatechol (4, 0.3 mg), 3,4,5-trihydroxybenzoic acid ethyl ester (5, 0.6 mg; ethyl gallate). The presence of 1 and 2 in Korean black raspberry has previously been reported. However, the presence of 3-5 in Korean black raspberry, and the identification of 1-5 from the Korean black raspberry wine have never before been reported.
