To investigate potential characteristics of clay minerals (illite, kaolin, zeolite, vermiculite, and bentonite) for food industry application, antioxidative properties of clay minerals, electron-donating ability (EDA), peroxide value (POV), and thiobabituric acid (TBA) were measured, and antimicrobial activity against several food spoilage microorganisms were evaluated by minimum inhibition concentration (MIC) method. Changes in components by cooking clay minerals added to stored rice were measured by GC-MS. DPPH (1,1-diphenyl-2-picrylhydrazyl) analysis results revealed bentonite has strongest EDA at 20.6%. Antioxidant activities measured based on POV were similar to DPPH results. Induction period of linoleic acid with vermiculite was longest among. TBA results revealed zeolite hasstrongest antioxidant ability. Growth inhibition against E. coli and S. aureuswas observed in illite, vermiculite, and zeolite. Aroma components indicated decrease in hex anal, pent anal, non anal, linoleate, stearic acid, and oleic acid when clay minerals were added to stored rice. These results indicate that several clay minerals have antioxidative and antimicrobial abilities and improve flavor profiles in stored rice.

Measuring in vivo rate of bone collagen synthesis has so far been technically difficult and often subject to quite large errors. In the present study, bone collagen synthesis rate was measured using a precursor-product method, based on the exchange of $^2$$H_2O$ into amino acids. Mass isotopomer abundance in hydroxyproline from bone collagen was analyzed by gas chromatography/mass spectrometry. The $^2$$H_2O$ labeling protocol consisted of an initial intraperitoneal injection of 99.9% $^2$$H_2O$, to achieve approximately 2.5% body water enrichment followed by administration of 4% $^2$$H_2O$ in drinking water for 9 weeks. Body $^2$$H_2O$ enrichments were stable at 2.7 ∼ 3.0% over labeling Period. In growing rats, the fractional synthesis rate ( $k_{s}$) of bone collagen was 0.066 $\pm$ 0.049 w $k^{-1}$ . The unique features of stable $^2$$H_2O$ pools and label incorporation allowed the precursor-product approach to be used for measuring bone collagen synthesis rate..

Trichoderma species are a rich source of metabolites, but less known for biomedical potential. This work deals with antibacterial and antioxidant potentials of intracellular non-cytotoxic metabolites, extracted from Trichoderma atroviride (KNUP001). A total of 53 fractions was collected by column chromatography and tested for cytotoxicity by MTT assay. Only one fraction (F41) was found to be non-toxic to Vero cells with $95.4{\pm}0.61%$ of survival. The F41 was then subjected to chemical analysis, antibacterial and antioxidant assays. The F41 at $500{\mu}g.ml^{-1}$ showed the total antioxidant of $48.70{\pm}2.90%$, DPPH radical scavenging activity of $37.25{\pm}2.25$, nitric oxide (NO) radical scavenging activity of $54.55{\pm}1.95$ and $H_2O_2$ radical scavenging activity of $43.75{\pm}3.21$. The F41 at $25{\mu}g.ml^{-1}$ displayed antibacterial activity against E. coli ($14.25{\pm}0.2mm$), P. mirabilis ($10.4{\pm}0.6mm$), S. dysenteriae ($18.6{\pm}03mm$), S. paratyphi A ($14.1{\pm}1.1mm$), E. aerogenes ($5.6{\pm}0.4mm$) and S. marcescens ($14.25{\pm}0.2mm$). GC-MS analysis revealed the dominant presence of oleic acid C 18.1 (63.18%), n-hexadecanoic acid (6.17%), and ethyl oleate (4.93%) and potent molecules such as 8-[(2E)-2-(3-hydroxybenzylidene)hydrazinyl]-1,3,7-trimethyl-3,7-dihydro-1H-purine-2,6-dione, 2-(Dimethylamino)ethyl (1Z)-N-hydroxy-2-(4-morpholinyl)-2-oxoethanimidothioate, Fluorene in the F41, and virtual study revealed that these molecules are likely responsible for the antibacterial activities of F41. Hence, further investigation deserves on purification and characterization of the active metabolites from T. atroviride strain KNUP001 towards developing molecular leads to effective antibacterial drugs, and non-toxic to host cells.
Pumpkin seed oil (PSO) was investigated for its parasite elimination activity and efficacy in treating disorders of the prostate gland and urinary bladder. We confirmed the composition of PSO and identified its ability to improve vessels. Gas chromatography coupled with mass spectrometric (GC-MS) system was used for PSO composition analysis. Cytotoxicity and cell proliferation were confirmed by Cell Counting Kit-8 (CCK-8) assay. Nitric oxide(NO) production was measured with Griess reagent, and intercellular adhesion molecule (ICAM)-1 and vascular cell adhesion molecule (VCAM)-1 mRNA expression levels were measured by reverse transcription polymerase chain reaction (RT-PCR). As a result, PSO revealed the presence of several components such as linoleic acid, oleic acid, palmitic acid and stearic acid. Cytotoxic effects of PSO were not observed, and PSO increased nitric oxide production in human umbilical vein endothelial cells (HUVEC). Additionally, TNF-${\alpha}$-induced cell proliferation and ICAM-1 expression in HUVEC were inhibited by PSO treatment, whereas VCAM-1 expression was not significantly reduced. Taken together, these results show that PSO is worthy of study as a candidate food material for improvement of vascular disease.

Headspace volatile compounds of leaf, stem, and root of Peucedanum japonicum Thunberg (PJT) harvested in March and July were isolated, separated, and identified by using a combined system of solid phase microextraction and gas chromatography/mass spectrometry. Identified numbers of volatile compounds were 72, 75, and 63 in March PJT as well as 78, 73, and 69 in July, respectively. Total compounds identified from PJT consisted of alcohols (1~7), aldehydes (7~15), esters (1~4), hydrocarbons (5~9), ketones (1~2), monoterpenes (13~16), monoterpene alcohols (1~5), monoterpene aldehydes (2~4), monoterpene esters (1~4), monoterpene ketones (1~2), sesquiterpenes (16~24), and miscellaneous compounds (2~3). Major volatile compounds of PJT were monoterpenes at all harvest times and in all parts of PJT. The amounts of ${\beta}$-pinene in leaf and stem harvested in March were highest in monoterpenes, followed by ${\alpha}$-pinene and ${\beta}$-myrcene. However, amounts of volatile compounds of monoterpenes in leaf and stem from July PJT were in the decreasing order of ${\beta}$-pinene, ${\beta}$-phellandrene, and ${\beta}$-myrcene. Of the 39 sesquiterpenes identified in PJT, (E)-caryophyllene in March leaf was the most abundant volatile compound, followed by acoradiene in March leaf, ${\beta}$-elemene, and ${\alpha}$-copaene in July stem.

The present paper was attempted to obtain the basic date concerning a reasonable preparing method and optimum fermentation conditions of Kochujang (Red pepper paste). To establish the standard quality of Kochujang, the chemical compositions and the volatile components of Kochujang was discussed. The native Kochujang collected from 80 households contained 40.51% of moisture, 6.00% of salts, 3.25 % of crude fat, 10.30 % of crude ash, pH 4.79, 9.28ml of tillable acidity. 19.60% of reducing sugar, 179.51 mg/100g of amino nitrogen and 4.43% of total nitrogen. The volatiles of Kochujang were extracted by a steam-distillation under the reduced pressure and determined by gas chromatography-mass spectrometry (GC-MS). Total volatile components identified in the native Kochujang were 46 components, i. e., 30 components in neutral fraction, 8 components in phenolic fraction and 8 components in acidic fraction, respectively. But no components were detected basic fraction.
Objectives: This study was conducted to provide scientific and effective information on phytoncides, which are associated with forest healing, and to activate recreational forests. Methods: The target sites were natural recreation forests, a forest park and an arboretum, and the control sites were three urban parks. The samples were collected at a volume of 6.0 L and a flow rate of 0.1 L/min for one hour using a low volume pump and the solid adsorbent sampling method. The phytoncide compounds adsorbed in the Tenax TA tube were analyzed by a automatic heat desorption unit and GC-MS. Results: By type of recreational forest, the annual concentrations of phytoncide (monoterpene) for the forest park showed the highest concentration with $1.450{\mu}g/m^3$, while those for the arboretum showed the lowest concentration at $0.892{\mu}g/m^3$, and thus the concentration of the forest park was approximately 1.6 times higher than the arboretum. The season showing the highest concentration of phytoncides was summer (June) and the forest park was the highest among the recreational forests. The concentrations of major components for phytoncide showed in descending order: ${\alpha}-pinene$, ${\beta}-pinene$, camphene, 3-carene and limonene. The seasonal concentration of ${\alpha}-pinene$, camphene and ${\beta}-pinene$ by type of recreational forest increased in April, which is characterized by low temperature and humidity, and the seasonal concentration of camphene decreased with higher humidity. The meteorological factors which had the high correlation with the concentration of total terpene were temperature and humidity. $CO_2$ and $O_2$ showed an inverse correlation. Conclusion: The major components of phytoncide were ${\alpha}-pinene$, ${\beta}-pinene$, camphene, 3-carene and limonene in descending order of concentration. Further and systematic study on the chemical nature of individual phytoncides, and on the effect of phytoncides on humans needs to be performed.

This study was designed to analyze the chemical compositions of 'Setoka' branch essential oils (SEBO) and to test their biological activities. 'Setoka' is a Citrus species widely cultivated in Jeju Island. At the present, 'Setoka' branches produced by thinning process were mostly discarded as a waste. Therefore, utilization of this branch waste has received much attention. 'Setoka' branch essential oils (SBEO) were prepared by treatment of its ethanol extracts with jojoba oil. SBEO were chemically analyzed using gas chromatograph-mass spectrometry (GC-MS), and following components were identified; ethyl linoleate (64.14%), ethyl palmitate (16.50%), neophytadiene (11.06%) and beta-citronellol (5.09%). The anti-inflammatory activity in the SBEO was examined using RAW 264.7 murine macrophage cells stimulated with LPS. As a result, the SBEO inhibited nitric oxide (NO) productions with a dose-dependent manner. In addition, SBEO showed good anti-microbial activities against drug-susceptible and -resistant skin pathogens such as Staphylococcus aureus and Propionibacterium acnes, which are acne-causing bacteria. Based on these results, we suggest that SBEO has the possibility for use as an anti-inflammatory and anti-microbial agent in cosmetic applications.

Gibberellin(GA) 3-$\beta$ hydroxylation is very important for the shoot elogation in the higher plants, since only 3$\beta$-hydryoxylated GAs promote shoot elogation in several plants. Fluctuation of 3$\beta$-hydryoxylase activity was examined during seed maturation using two cultivars of , P. vulgaris, Kentucky Wonder (normal) and Masterpiece (dwarf). Very immature seeds of both cultivars contain high level of 3$\beta$-hydroxylase activity (per mg protein). Both cultivars showed maximum of enzyme activity (per seed) in the middle of their maturation process. Gibberellin 3$\beta$-hydroxylase catalyzing the hydroxylation of GA20 to GA1 was purified 313-fold from very early immature seeds of P. vulgaris. Crude soluble enzyme extracts were purified by 15% methanol precipitation, hydrophobic interaction chromatogrphy, DEAE ion exchange column chromatography and gel filtration HPLC. The 3$\beta$-hydroxylase activity was unstable and lost much of its activity duting the purification. The molecular weight of purified enzyme was extimated to be 42, 000 by gel filtration HPLC and SDS-PAGE. The enzyme exhibited maximum activity at pH 7.7. The Km values for [2.3-3H] GA20 and [2.3-3H]GA9 were 0.29 $\mu$M and 0.33 $\mu$M, respectively. The enzyme requires 2-oxoglutarate as a cosubstrate; the Km value for 2-oxoglutarate was 250 $\mu$M using 3H GA20 as a substrate. Fe2+ and ascorbate significantly activated the enzyme at all purification steps, while catalase and BSA activated the purified enzyme only. The enzyme was inhibited by divalent cations Mn2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+ and Hg2+. Effects of several GAs and GA anaogues on the putrified 3$\beta$-hydroxylase were examined using [3H]GA9 and GA20 as a substrates. Among them, GA5, GA9, GA15, GA20 and GA44 inhibited the enzyme activity. [13C, 3H] GA20 was converted by the partially purified enzyme preparation to [13C, 3H]GA1, GA5 and GA6, which were identified by GC-MS, GA9 was converted only GA4, GA15 and GA44 were converted to GA37 and GA38, respectively. GA5 was epoxidized to GA6 by the preparation. This suggests that 3$\beta$-hydroxylation of GA20 and epoxidation of GA5 are catalyzed by the same enzyme in P, vulgaris.
Sarcodon aspratus (Berk.) S. Ito is a wild mushroom commonly consumed in Korea due to its beneficial effects on health. However, only limited information on the volatile and nonvolatile constituents of S. aspratus is available. In the present study, the total concentration of mushroom soluble sugars, including glucose, trehalose, sucrose, and xylose, was found to be 202.5 mg/kg. The total contents of free and essential amino acids were 2,592.1 mg/kg and 1,249.5 mg/kg, respectively; arginine, lysine, methionine, and valine were the major amino acids present. The contents of total 5'-nucleotides and flavor 5'-nucleotides in S. aspratus were 2,510.7 mg/kg and 773.4 mg/kg, respectively. The volatile components of S. aspratus were collected by simultaneous distillation-extraction (SDE) and analyzed by gas chromatography-massspectrometry (GC-MS). A total of 27 volatile compounds were isolated and identified. The most abundant was l-octen-3-ol, which accounted for more than 68% of total volatiles; other important compounds were 2-octen-1-ol, 1-octen-3-one, and 2-octenol. Our results provide preliminary data for the development of S. aspratus as a food material.
