Phenotypic and genetic characteristics of Vibrio ichthyoenteri isolated from the olive flounder, Paralichthys olivaceus of culturing size (미성어 양식 넙치, Paralichthys olivaceus에서 분리한 Vibrio icthyoenteri의 표현형 및 유전형적 특성)

  • From 2002 to 2004, various vibrios were isolated from the olive flounder, Paralichthys olivaceus of culturing size with disease signs. During this survey, it was known that the high proportion of Vibrio ichthyoenteri was occupied among the isolated vibrios. Generally, V. ichthyoenteri is well known as the pathogen of bacterial enteritis of olive flounder larvae. The aim of the present study was the compare the characteristics of two groups of V. ichthyoenteri, culturing sized olive flounder, and larvae of olive flounder showing the intestinal necrosis. The research was focused on the physiology, biochemistry, genetics in the two bacterial groups. The physiological and biochemical characteristics of the tested strains were very similar. The intergenic spacer (IGS) region between the 16S and 23S rRNA genes of 21 isolated strains and 3 reference strains, V. ichthyoenteri, were investigated by PCR fragment length typing and DNA sequencing. After the isolated strains were identified as V. ichthyoenteri, not only phenotypic characteristics of the isolated and reference strains but also homology of 16S-23S IGS of all isolated strains and reference strains as 99.1~100%. The V. ichthyoenteri showed 4 specific 16S-23S patterns and contained no-tRNA, tRNAGlu(TTC) , tRNAIle(GAT) tRNAAla(TGC) type .

Transcriptional Analysis Responding to Propanol Stress in Escherichia coli (대장균에서 프로판올 스트레스에 관한 전사분석)

  • We compared the transcriptome in response to propanol stress in wild-type and propanol-resistant mutant Escherichia coli using the DNA microarray technique. The correlation value of RNA expression between the propanol-treated wild type and the untreated-one was about 0.949, and 50 genes were differentially expressed by more than twofold in both samples. The correlation value of RNA expression between the propanol-treated mutant and the untreated one was about 0.951, and 71 genes in two samples showed differential expression patterns. However, the values between the wild type and mutant, regardless of propanol addition, were 0.974-0.992 and only 1-2 genes were differentially expressed in the two strains. The representative characteristics among differentially expressed genes in W3110 or P19 treated with propanol compared to untreated samples were up-regulation of hest shock response genes and down-regulation of genes relating to ribosome biosynthesis. In addition, many genes were regulated by transcription regulation factors such as ArcA, CRP, FNR, H-NS, GatR, or PurR and overexpressed by sigma factor RpoH. We confirmed that RpoH mediated an important host defense function in propanol stress in E. coli W3110 and P19 by comparison of cell growth rate among the wild type, rpoH disruptant mutant, and rpoH-complemented strain.

Analysis of Thin Aggregative Fimbriae Genes csgA, csgB of S. typhimurium and S. enteritidis Strains (S. typhimurium과 S. enteritidis 균주의 Thin Aggregative Fimbriae 유전자 csgA, csgB 분석)

  • The purpose of this study was investigated to observe the relatively low amino acid mutations in six different strains of ATCC or KCTC Salmonella strains (four species and two isolates). The mutations in the strains were to be detected and compared with the genes csgA and csgB corresponding to the Thin aggregative fimbriae. Amino acid mutations in the strains were observed from Ser20→ Gly(AGT→GGT) in the csgA gene, Asp25→Ala(GAT→GCT) and Lys66→Thr(AAA→ACA) in the csgB gene. Among the six strains, the two most common amino acid variations were observed in S. typhimurium - TH strains. On the other hand, no mutation of nucleotide sequence was observed in the strains of S. typhimurium ATCC 13311 and S. typhimurium KCTC 1925. In conclusion the genes csgA and csgB in the strains may be useful for the evaluation and detection of amino acid mutation.

  • The cloned glyceraldehyde-3-phosphate dehydrogenase (GAP) gene of Saccharomyces cereviszae (Holland et al., 1983) has been characterized. Based on the communication, we have also cloned 2.1 kb CAP DNA fragment and modified this fragment as a portable promoter. Two yeast expression vectors, one is YCp type vector being maintained at low copy number (1 or 2) and the other is YEp type vector at high copy number, have been constructed with the GAP promoter and the PH05' gene as a reporter. Our plasrnids were introduc,ed into S. cerevisiae HY-1, which has been improved. The $Trp^+$ transformants expressed APase activity efficiently and showed high level of PH05' transcripts.

  • Congenital pulmonary airway malformation (CPAM), previously known as congenital cystic adenomatoid malformation, is a rare developmental lung abnormality associated with rhabdomyosarcoma, pleuropulmonary blastoma, and mucinous adenocarcinoma of the lung. We report an unusual case of a 10-day-old male newborn with a left lower lobe pulmonary cyst who underwent lobectomy, which revealed type II CPAM complicated by multifocal mucinous adenocarcinoma. KRAS sequencing revealed a somatic mutation in Codon12 ($GGT{\rightarrow}GAT$), suggesting the development of a mucinous adenocarcinoma in the background of mucinous metaplasia. Mucinous adenocarcinoma is the most common lung tumor associated with CPAM, but it generally occurs in older children and adults. Further, all cases in the literature are of type I CPAM. This case in a neonate indicates that malignant transformation can occur very early in type II CPAM.

A Survey on the Notion and Intake of Kimchi among College Women (여대생의 김치에 대한 의식과 섭취실태 조사)

  • A survey of the notion and intake on Kimchi among college women in Pusan was conducted to get some basic information on kimchi. Two hundred sixty seven students in Pusan participated in this survey. Seventy five percent of students answered that they like kimchi. They preferred well-fermented kimchi with anchovy extracts, refreshing taste and crispy texture the saltly and sweet. Chinese cabbage kimchi (87.6%) was found to be the most favored kimchi and Kakdugi(seasoned pickles of cubed radish), Nabak kimchi and Chonggak kimchi (ponytail kimchi) were followed in the order. The most favorite food made from kimchi was stir fried kimchi with rice. They disliked traditional special kimchi, such as Puchu kimchi (leek kimchi), Pa kimchi (green onion kimchi), Kkennip kimchi (perilla leaf kimchi), Godulbaegi kimch (Korea wild lettuce kimchi) and Gat kimchi (Leaf mustard kimchi). About 93 grams of kimchi was consumed daily and this amount was a little. Seventy percent of students did not have any experiences preparing kimchi. Experiences of kimchi preparation were given by mother through kimchii-making event for the winter(71.7%), cooking practice in middle or high school (14.1%) and college(10.9%) and general cooking education (33%). They preferred to buy kimchi at the Agricultural Cooperative Association (48.5%) or a large kimch factory (32.75). College students believe that kimchi is a healthy food and are willing to learn how to make kimchi.

  • The purpose of this study is to focus on the townscape of Daegu based on the urban characteristics of the landscape cognition and images captured by citizens. The analysis was performed by the data obtained from questionnaires and interviews. This study methods were deals the cognition characteristics, landmark landscape, visual preference landscape, image and satisfaction. The results are summarized as follows: 1. The orders of cognition landscapes were estimated Apple> Weather> Texture> Mt.Palgong> Daegu Tower> Pretty Girl> Mt. Apsan> Dalsung Park> Conservative> Dongsung Road, etc. That is constructed Nonphysical elements(62.0%) and Physical elements(38.0%) 2. The orders of representative landscape(Landmark) in city were estimated Mt.Palgong> Daegu Tower> Gat Rock> Mt.Apsan> Dalsung Park, etc. As a whole middle and old people(over 30gen) preferenced as a history landscape or natural landscape, but youth people(10-20gen) preferenced as visible and interesting artifical places. 3. While the positive attitudes for the image of city were traditional(3.30), intimacy(3.58), and rest(2.90), the negative attitudes were unnewly(2.34), closing(2.37) and narrow(2.40). Also total satisfaction for that was estimated 5.51. 4. Psychological factors, related to the satisfaction of the image of city were composed of four factors, individuality character, pleasure character, amenity character, formation character. And the presumption formula of satisfaction was: Satisfaction = 5.477 + 0.752(Individuality) + 0.470(Pleasure) + 0.413(Amenity) + 0.241(Formation).

  • Many countries in the world have regulated cross-border supply in basic telecommunications services based on GATS exception rule for telecommunications because of anti-competition behavior control, domestic consumer protection, tax levy, national security, etc. Korea partially regulates cross-border supply in telecommunications service through the WTO and the domestic law and regulation. However, WTO and FTA negotiations, cross-border supply regulation are strongly demanded to deregulation. Therefore, it is required to review the effectiveness of cross-border supply regulation to prepare the counter plan for that regulation. Thus, the major countries of the telecommunication services regulation regarding the cross-border supply are reviewed. In addition, it is retrieved some implications for Korean regulation policy by reviewing the actual regulation of many countries cases.

  • Effective border security is crucial in managing and mitigating firearm-related threats. While prior research has focused on firearm detection, it lacks contextual analysis. This paper advances firearm-related incident assessment by integrating pose estimation to improve gun violence detection. Our novel approach extracts body and firearm pose graphs and employs Graph Attention Networks(GAT) for graph analysis to accurately identify gun violence incidents. By recognizing associated actions, our system provides greater situational awareness beyond mere firearm detection. Utilizing Graph-LSTM, we capture spatial and temporal information. As a result, our proposed algorithm is lighter and more accurate than the CNN-LSTM model used as a baseline, achieving test F1-scores of 82.04 % on our collected data.

Growth Inhibition of Escherichia coli during Heterologous Expression of Bacillus subtilis Glutamyl-tRNA Synthetase that Catalyzes the Formation of Mischarged Glutamyl-$tRNA_{l}$$^{Gln}$

  • It is known that Bacillus subtilis glutamyl-tRNA synthetase (GluRS) mischarges E. coli $tRNA_{1}$$^{Gln}$ with glutamate in vitro. It has also been established that the expression of B. subtilis GluRS in Escherichia coli results in the death of the host cell. To ascertain whether E. coli growth inhibition caused by B. subtilis GluRS synthesis is a consequence of Glu-$tRNA_{1}$$^{Gln}$ formation, we constructed an in vivo test system, in which B. subtilis GluRS gene expression is controlled by IPTG. Such a system permits the investigation of factors affecting E. coli growth. Expression of E. coli glutaminyl-tRNA synthetase (GlnRS) also amelio-rated growth inhibition, presumably by competitively preventing $tRNA_{1}$$^{Gln}$ misacylation. However, when amounts of up to 10 mM L-glutamine, the cognate amino acid for acylation of $tRNA_{1}$$^{Gln}$, were added to the growth medium, cell growth was unaffected. Overexpression of the B. subtilis gatCAB gene encoding Glu-$tRNA^{Gln}$ amidotransferase (Glu-AdT) rescued cells from toxic effects caused by the formation of the mis-charging GluRS. This result indicates that B. subtilis Glu-AdT recognizes the mischarged E. coli Glu-$tRNA_{1}$$^{Gln}$, and converts it to the cognate Gln-$tRNA_{1}$$^{Gln}$ species. B. subtilis GluRS-dependent Glu-$tRNA_{1}$$^{Gln}$ formation may cause growth inhibition in the transformed E. coli strain, possibly due to abnormal protein synthesis.
