건강기능식품 중 잔류용매 분석법 개발 및 모니터링 (Analytical Method Development and Monitoring of Residual Solvents in Dietary Supplements)

  • Residual solvents in foods are defined as organic volatile chemicals used or produced in manufacturing of extracts or additives, or functional foods. The solvents are not completely eliminated by practical manufacturing techniques and they also may become contaminated by solvents from packing, transportation or storage in warehouses. Because residual solvents have no nutritional value but may be hazardous to human health, there is a need to remove them from the final products or reduce their amounts to below acceptable levels. The purpose of this study was to develop and evaluate an analytical method for the screening of residual solvents in health functional foods. Furthermore, the aim of this study was to constitute a reasonable management system based on the current state of the market and case studies of foreign countries. Eleven volatile solvents such as MeOH, EtOH, trichloroethylene and hexane were separated depending on their column properties, temp. and time using Gas Chromatography (GC). After determining the GC conditions, a sample preparation method using HSS (Head Space Sampling) was developed. From the results, a method for analyzing residual solvents in health functional foods was developed considering matrix effect and interference from the sample obtained from the solution of solvents-free health functional foods spiked with 11 standards solutions. Validation test using the developed GC/HSS/MS (Mass Spectrometry) method was followed by tests for precision, accuracy, recovery, linearity and adequate sensitivity. Finally, examination of 104 samples grouped in suits was performed by the developed HSS/GC/MS for screening the solvents. The 11 solvents were isolated from health functional foods based on vapor pressure difference, and followed by separation within 15 minutes in a single run. The limt of detection (LOD), limit of quantification (LOQ), recovery and coefficient of variation (C.V.) of these compounds determined by the HSS/GC/MS were found to be 0.1 pg/mL, 0.1-125 pg/g, 51.0-104.6%, and less than 15%, respectively. Using the developed HSS/GC/MS method, residual solvent from 16 out of 104 health functional products were detected as a EtOH. This method therefore seems t o be a valuable extension ofanalytical method for the identification of residual solvents in health functional food.

작잠에 관한 연구 (Studies on tussah silkworm, Antheraea pernyi)

  • I. Breeding of tussah silkworm(preliminary report). The preliminary examination for bleeding has been carried out since 1963 in tussah silkworms. 1) The strain(l-MG-B)of the heaviest silk quantity was the green silkworm and brown cocoon in univoltine, and the strains(2-G-B, 2-MG-B) of the heaviest silk quantity were also the green silkwom and brown cocoon in bivoltine in both spring and fall in 1965. 2) It looks like the voltinism, the body color and the cocoon color have reached to pure line up to 1965. II. Best place for the winter of tussah pupa. This work was aimed to find out good ways for the winter of tussah pupa. 1) The hatch of bivoltine was better than that of univoltine. 2) The cocoons covered with the leaves were good in the emergence of moth. 3) The cocoons which were kept at natural temperature till the first emergence of moths would show bad in both hatch and emergence. 4) If some of the pupae kept under natural condition were controled at proper temperature for a few days, hatch and laying eggs were best. 5) The best places for the winter were the egg storage and the rearing room. III. Relation between incubation temperature and voltinism. 1) When the tussah pupa are kept at natural temperature during winter, the moths do not come out of the pupa. 2) There is no difference between about 18$^{\circ}C$ and about 25$^{\circ}C$ during incubation in hatching ratio. 3) The tussah silkworms of univoltine in mortality are stronger than that of bivoltine. 4) There is not any relation between voltinism and high or low temperature for pupa and eggs. IV. Induced mutation by gamma-ray and neutron in tussah silkworm. This work was carried out in order to induce the mutation by treating the pupa or the eggs of tussah silkworm with gamma my and neutron. The results obtained are as follows. 1. Though the whole pupa treated with neutron become moths, the moths have no ability to copulate each other. The only moths emerged from pupa treated with neutron, 4000${\gamma}$ are able to lay all un-fertilized eggs, some of which have a hole on the surface and nothing of contents. 2. The non-diapause eggs are treated with neutron in spring, but the hatching ratio is 50∼60 percent, but the whole eggs treated with gamma ray are never hatched. 3. The sensitivity of the pupa to neutron is weaker than that of the eggs. 4. The hatching ratio is in direct proportion to the gamma ray dose. 5. Author find out a new mutant which is excellent in the cocoon quality, so he will do the progeny test next hear.

벼와 잡초에서 온도조건(溫度條件)에 따른 Bensulfuron의 흡수(吸收), 이행(移行) 및 대사(代謝) (Absorption, Translocation and Metabolism of Bensulfuron in Rice and Weeds at Different Temperatures)

  • 온도조건에 따른 벼의 품종별 bensulfuron의 약해기구(藥害機構) 및 잡초에 대한 살초성(殺草性)을 구명하고자 $^{14}C$-bensulfuron을 사용하여 흡수(吸收), 이행(移行) 및 대사실험(代謝實險)을 수행하였다. 1. $^{14}C$-bensulfuron 흡수실험에서 품종간 벼의 흡수량(吸收量)은 삼강벼보다 상풍벼에서 많았으며 전반적으로 25/$20^{\circ}C$ 보다는 30/$25^{\circ}C$에서 흡수량이 많았다. 잡초의 흡수량은 올미, 너도방동사니, 피 순으로 많아 올미와 너도방동사니에서 살초력이 높은 것으로 생각되고 온도간에 $^{14}C$-bensulfuron의 흡수량은 차이가 적었다. 2. 이행율(移行率)은 벼의 경우 품종간에 큰 차이를 보이지 않았으나 시간이 경과함에 따라 상풍벼에서 높았으며 25/$20^{\circ}C$ 보다 30/$25^{\circ}C$에서 더 높은 경향이었다. 잡초는 너도방동사니, 올미, 피 순으로 이행율이 높았으며 3초종 모두 30/$25^{\circ}C$에서 이행율이 높았다. 따라서 피에서는 이행량이 낮았기 때문에 내성을 보였고, 올미와 너도방동사니는 이행량이 높아 감수성을 나타낸 것으로 생각된다. 3. $^{14}C$-bensulfuron의 대사실험에서 식물체내에 남아있는 친화합물(親化合物)인 bensulfuron은 감수성인 상풍벼가 삼강벼보다 높았으며 잡초에서는 25/$20^{\circ}C$에서 내성을 보인 피가 가장 적었으며 올미와 너도방동사니는 높았으며 30/$25^{\circ}C$에서는 큰 차이를 보이지 않았다. 4. 따라서 벼 품종간의 선택성 차이는 식물체가 가지고 있는 불활성 대사능력의 차이에 의한 것이라 생각되며 잡초에서의 살초성 작용기작은 bensulfuron 흡수량과 이행율이 관계된다고 생각되며 온도의 상승은 흡수 및 이행량을 증대시킴으로써 약해(藥害)와 살초력(殺草力)을 증가시켰다고 사료된다.

흰쥐에서 항염증제 후보물질 KAL-1120의 HPLC 분석 및 약물동태 (HPLC Analysis and Pharmacokinetics of KAL-1120, a Novel Anti-inflammation Agent, in Rats)

  • A rapid and sensitive reversed-phase high performance liquid chromatography (HPLC) method was developed for the determination of N-(-4-Chlorophenyl)-6-hydroxy-7-methoxy-2-chromanecarboxamide (KAL-1120), a novel anti-inflammation agent, in the rat plasma. The method was applied to analyze the compound in the biological fluids such as bile, urine and tissue homogenates. After liquid-liquid extraction, the compound was analyzed on an HPLC system with ultraviolet detection at 275 nm. HPLC was carried out using reversed-phase isocratic elution with a $C_{18}$ column, a mobile phase of a mixture of acetonitril (40 v/v%) at a flow rate of 1.0 mL/min. The chromatograms showed good resolution and sensitivity and no interference of plasma. The calibration curve for the drug in plasma was linear over the concentration range of 0.05-50 ${\mu}g$/mL. The intra- and inter-day assay accuracies of this method ranged from 0.06% to 9.33% of normal values and the precision did not exceed 6.28% of relative standard deviation. The plasma concentration of KAL-1120 decreased to below the quantifiable limit at 1.5 hr after the i.v. bolus administration of 2-10 mg/kg to rats ($t_{1/2,({\alpha})}$ and $t_{1/2,({\beta})$ of 2.15 and 26.7 min at a dose of 2 mg/kg, 3.91 and 33.0 min at a dose of 10 mg/kg, respectively). The steady-state volume of distribution ($V_{dss}$) and the total body clearance ($CL_t$) were not significantly altered in rats given doses from 2 to 10 mg/kg. Of the various tissues tested, KAL-1120 was mainly distributed in the lung and heart after i.v. bolus administration. KAL-1120 was detected in the bile by 30 min after its i.v. bolus administration. However, the concentration in the urine after i.v. bolus administration became too low to measure, suggesting that KAL-1120 is mostly excreted in the bile. In conclusion, this analytical method was suitable for the preclinical pharmacokinetic studies of KAL-1120 in rats.

비열 플라즈마 처리를 이용한 polystyrene, 소시지 케이싱, 그리고 훈제연어에서의 식중독균 저해 (Inhibition of Foodborne Pathogens on Polystyrene, Sausage Casings, and Smoked Salmon Using Nonthermal Plasma Treatments)

  • PS 필름에 접종된 S. Typhimurium, E. coli O157:H7, L. monocytogenes를 저해시킬 때 헬륨을 사용한 플라즈마 처리의 경우에는 S. Typhimurium이, 헬륨과 산소를 동시에 사용한 플라즈마 처리의 경우에는 L. monocytogenes가 가장 많이 저해되었다. E. coli O157:H7과 L. monocytogenes는 헬륨과 산소를 함께 사용하여 플라즈마 처리하였을 때 헬륨만 사용하였을 때보다 더 큰 저해를 보였다. 헬륨과 산소를 함께 사용한 비열 플라즈마 처리는 L. monocytogenes가 소시지 케이싱이나 훈제연어에 감염되었을 때보다 PS 필름에 감염되었을 때 L. monocytogenes를 더 효과적으로 저해시켜 감염된 재료에 따라 다른 저해 효과를 보여 주었다. 비열 플라즈마 처리에 사용된 가스의 종류와 미생물이 감염된 재료는 그 처리의 항균 효과에 대한 평가에서 중요하게 고려되어야 할 것이다. 헬륨과 산소를 사용한 비열 플라즈마 처리는 공정 후 포장재에 감염된 L. monocytogenes를 효과적으로 저해할 수 있을 것으로 기대된다.

Chemosensitization of Fusarium graminearum to Chemical Fungicides Using Cyclic Lipopeptides Produced by Bacillus amyloliquefaciens Strain JCK-12

  • Fusarium head blight (FHB) caused by infection with Fusarium graminearum leads to enormous losses to crop growers, and may contaminate grains with a number of Fusarium mycotoxins that pose serious risks to human and animal health. Antagonistic bacteria that are used to prevent FHB offer attractive alternatives or supplements to synthetic fungicides for controlling FHB without the negative effects of chemical management. Out of 500 bacterial strains isolated from soil, Bacillus amyloliquefaciens JCK-12 showed strong antifungal activity and was considered a potential source for control strategies to reduce FHB. B. amyloliquefaciens JCK-12 produces several cyclic lipopeptides (CLPs) including iturin A, fengycin, and surfactin. Iturin A inhibits spore germination of F. graminearum. Fengycin or surfactin alone did not display any inhibitory activity against spore germination at concentrations less than 30 ug/ml, but a mixture of iturin A, fengycin, and surfactin showed a remarkable synergistic inhibitory effect on F. graminearum spore germination. The fermentation broth and formulation of B. amyloliquefaciens JCK-12 strain reduced the disease incidence of FHB in wheat. Furthermore, co-application of B. amyloliquefaciens JCK-12 and chemical fungicides resulted in synergistic in vitro antifungal effects and significant disease control efficacy against FHB under greenhouse and field conditions, suggesting that B. amyloliquefaciens JCK-12 has a strong chemosensitizing effect. The synergistic antifungal effect of B. amyloliquefaciens JCK-12 and chemical fungicides in combination may result from the cell wall damage and altered cell membrane permeability in the phytopathogenic fungi caused by the CLP mixtures and subsequent increased sensitivity of F. graminearum to fungicides. In addition, B. amyloliquefaciens JCK-12 showed the potential to reduce trichothecenes mycotoxin production. The results of this study indicate that B. amyloliquefaciens JCK-12 could be used as an available biocontrol agent or as a chemosensitizer to chemical fungicides for controlling FHB disease and as a strategy for preventing the contamination of harvested crops with mycotoxins.

Evidence for the Association of Ce11u1ar Iron Loss in Nitric Oxide-induced Apoptosis of HL-60 Cells: Involvement of p38 Kinase, c-Jun N-terminal Kinase, Cytochrome C Release, and Caspases Pathways

  • Nitric oxide has high affinity for iron, and thus it can cause intracellular iron loss. We tested the idea that intracellular iron can be the primary target of NO toxicity by comparing the signaling mechanisms involved in cell death caused by iron depletion and that caused by NO. Treatment of HL-60 cells with a NO donor, S-nitroso-N-acetyl-DL-penicillamine (SNAP), decreased the intracellular iron level rapidly as that observed with the iron chelator deferoxamine (DFO). Iron chelators such as DFO and mimosine could induce death of human leukemic HL-60 cells by a mechanism requiring activation of p38 kinase, c-Jun N-terminal kinase, caspase-3 and caspase-8. DFO and SNAP also caused release of cytochrome c from mitochondria. Inhibition of p38 kinase by a selective inhibitor, SB203580, abolished the NO and DFO-induced cell death, release of cytochrome c, and activation of caspase-3 and caspase-8, thus indicating that p38 kinase lies upstream in the cell death processes. In a parallel situation, the cells that are sensitive to NO showed similar sensitivity to DFO. Moreover, simultaneous addition of ferric citrate, an iron-containing compound, inhibited the SNAP and DFO-induced activation of caspases and also blocked the NO-mediated cell cycle arrest at $G_1$ phase. Collectively, our data implicate that the NO-induced cell death of tumor cells including HL-60 cells is mediated by depletion of iron and further suggest that activation of p38 kinase lies upstream of cytochrome c release and caspase activation involved in this apoptotic process.

즉석식품 제조를 위한 육가공제품의 감마선 조사에 따른 미생물 및 유전독성학적 안전성 평가 (Microbiological and Mutagenical Safety Evaluation of Gamma Irradiated Ready-to-Eat Foods of Animal Origin)

  • 감마선 조사를 통한 즉석식품 제조용 육가공제품의 미생물학적 및 유전 독성학적 안전성 평가를 실시하였다. 미생물 안전성은 총 4종류의 병원성 미생물(Salmonella typhimurium, Escherichia coli, Staphylococcus aureus 및 Listeria ivanovii)을 이용하여 inoculation test를 실시하였으며 유전 독성학적 평가는 ames test를 통해 조사하였다. 4 종류의 병원성 미생물을 볶은 쇠고기 및 햄에 접종했을 경우 초기 균수는 7 log 정도로 나타났으나 1 kGy로 감마선 조사했을 경우 S. typhimurium은 4 log cycle 정도, E. coli 및 S. aureus는 3 log 및 L. ivanovii는 2 log정도 증식이 억제되었다. 볶은 쇠고기 및 햄에 접종된 4 종류의 병원성 미생물은 3 kGy의 선량으로 감마선 조사했을 경우 검출되지 않았다. 볶은 쇠고기 및 햄에 접종된 4종류의 병원성 미생물의 방사선 조사에 대한 감수성을 측정한 결과 접종된 병원성 미생물의 D/sub 10/ value는 볶은 쇠고기에 접종된 L. ivanovii가 0.48로 가장 감수성이 낮은 것으로 나타났다. 볶은 쇠고기 및 햄을 비조사구와 조사구(10 kGy)로 나누어 S. typhimurium TA98 및 TA100 두 균주를 이용하여 유전 독성학적 안전성을 평가한 결과 대사활성을 시킨 경우와 대사활성을 시키지 않은 경우 모두 추출물의 농도에 따른 복귀돌연변이 집락수의 증가는 확인되지 않아, 즉석 식품제조를 위한 볶은 쇠고기 및 햄의 감마선 조사는 유전독성학적으로 안전한 것으로 나타났다.

ENSO 패턴에 대한 MM5 강수 모의 결과의 유역단위 성능 평가: 플로리다 템파 지역을 중심으로 (Combining Bias-correction on Regional Climate Simulations and ENSO Signal for Water Management: Case Study for Tampa Bay, Florida, U.S.)

  • 수자원의 수요 증가와 ENSO (El Ni$\tilde{n}$o/La Ni$\tilde{n}$a Southern Oscillation) 등의 기후변화 현상으로 인한 수자원 공급의 불안정 요소가 제기됨에 따라, 수자원 관리 계획 수립 시 장/단기강우 모의의 중요성이 강조되고 있다. 본 연구에서는 미국 플로리다 템파 지역의 두 개 유역을 대상으로 1986년부터 2008년까지의 MM5 지역기후모델을 이용한 강우모의 결과를 시험지역의 33개 관측자료와 CDF-mapping 기법을 이용하여 통계적으로 보정하였으며 그 결과를 바탕으로 ENSO 패턴에 따른 모델의 성능을 평가하였다. 보정된 MM5일 강우 모의결과는 대체적으로 각 관측소의 월 평균 강우량 (ME: 1.0mm)을 잘 모의하는 것으로 나타났다. 블락-크리깅 기법을 이용하여 추정된 유역 평균 일/월 강우량 또한 관측치를 잘 재현하였다(일 강우 ME: 0.8mm, 월 강우 ME: 7.1mm). 한편, ONI (Oceanic Ni$\tilde{n}$o index)를 이용하여 구분한 ENSO 패턴에 따른 강우 모의치를 분석한 결과, 월별 엘리뇨/라니냐 해에 대한 유역 단위의 강우량 모의 성능이 상이한 것으로 나타났다. 이 원인으로 한정된 모수화 적용 및 모델 경계자료 오차 등을 제시하고 이에 대한 보정 방법개선 등의 추가 연구의 필요성을 지적하였다. 본 연구는 ENSO 패턴을 고려한 월별 기후모델 결과를 활용함에 있어 유의점을 제시하였기에, 우기와 건기에 대한 수자원 관리를 위한 적용 등에 유용하게 활용될 것으로 기대된다.

센서용 거대자기저항 스핀밸브소자의 열처리 효과 (Post-annealing Effect of Giant Magnetoresistance-Spin Valve Device for Sensor)

  • 셀(cell) 단위의 생체분자의 자성특성 검출을 위한 거대자기저항-스핀밸브(giant magnetoresistance-spin valves; GMR-SV) 바이오센서로서 미세 패턴된 모양에 따라 길이 방향과 폭 방향 용이축에 의존하는 자기저항 특성을 연구하였다. 바이오센서(biosensor)로서 사용할 스핀밸브 다층구조는 glass/NiO/NiFe/CoFe/Cu/CoFe/NiFe 이었다. 자성 다층박막의 일축이방성을 만들기 위해 증착시와 소자 패턴닝 후 진공 열처리를 $200^{\circ}C$에서 300 Oe 정도 외부자기장을 인가하였다. 형상자기이방성 효과를 고려하여 광 리소그래피 과정으로 얻은 미세 활성영역 패턴 사이즈는 $2{\times}5{\mu}m^2$로 정하였다. 2단자법으로 길이방향의 센싱전류와 폭 방향의 고정층의 용이축 방향 각도에 의존하는 자장민감도의 변화는 바이오센서 소자로서 활용에 중요한 요인임을 확인하였다.
