Glucose 6-Phosphate Dehydrogenase Activity of Bovine Embryos Produced in vitro (소 체외수정란의 Glucose 6-Phosphate Dehydrogenase 활성)

  • 소 체외수정란에 있어서 pentose phosphate pathway (PPP)를 연구하기 위해서, 한개의 체외수정란으로부터 glucose 6-phosphate dehydrogenase (G6PDH)의 활성을 효소증폭방법으로 측정하였다. Glucose 6-phosphate (G6P) 기질을 처리하지 않은 2, 4, 8세포기, 상실배 및 배반포기 수정란에서의 G6PDH 활성치는 각각 $25.5{\pm}3.3$, $27.8{\pm}3.4$, $40.9{\pm}6.2$, $34.9{\pm}3.6$$52.9{\pm}2.5{\times}10^{-8}mol/embryo/h$ 을 나타내었다. 즉, 8 세포기 이후 수정란들은 2 세포기나 4 세포기보다도 높은 효소활성치를 보여주었다 (P<0.01). 그리고 G6P 기질을 첨가한 2,4,8 세포기, 상실배기 및 배반포기 수정란의 G6PDH 활성치는 각각 $32.3{\pm}3.9$, $29.4{\pm}1.8$, $51.9{\pm}4.2$, $42.6{\pm}2.7$$52.9{\pm}2.5{\times}10^{-8}mol/embryo/h$ 로서 기질 무처리구와 마찬가지로 유의성이 인정되었다 (P<0.01). 전반적으로 수정란의 발달단계에 있어서 G6P 첨가한 수정란들에 G6PDH의 효소활성치가 기질을 처리하지 수정란들의 것보다도 높은 경향을 보였다. 한편, 소 체외수정란의 G6PDH 효소활성치와 발생능과의 관계를 알아보기 위하여, 4 세포기 수정란들을 효소활성치의 정량적 수준 (low, middle, high)에 따라 3 군으로 분류한 다음 $38.5^{\circ}C$, 5% $CO_2$에서 5일간 난구세포들과 공동배양을 실시하였다. 그 결과, G6PDH 효소활성치 차이에 따른 수정란들의 체외발달율에는 유의성이 인정되지 않았다. 본 실험의 결과를 종합하여 볼 때, 소 체외수정란에 있어서 PPP 대사는 8세포기 이후부터 활발히 이루어지고 있음을 알 수 있었다.

Antidiabetic Effect of Ethanol Extract on Phelladindron amurense Rupr (황백 에탄올 추출물의 혈당강하작용)

  • This study was performed to investigate the antidiabetic effect of ethanol extract of Phelladrindron Amurense Rupr (P.A) in Streptozotocin(STZ) induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg,b.w. dissolved in citrate buffer. The ethanol extract of P. A was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The content of serum glucose, was significantly decreased in P.A treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased(p<0.05), and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased(p<0.05) in P.A treated group compared to those of STZ-control group, These results indicated that ethanol extract of P.A have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic Effect of Ethanol Extract on Astragali Radix (황기 에탄올 추출물의 항 당뇨 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Astragali Radix(A.R) in Streptozotocin(STZ) induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of A. R was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in A.R treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased, and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in A.R treated group compared to the those of STZ-control group, These results indicated that ethanol extract of A.R would have antidiabetic effect in STZ-induced diabetic rats.

Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Codonopsis lanceolata root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Codonopsis lanceolata root treated group compared to the those of STZ-control group. Also the content of hepatic glycogen and activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased(p<0.05). These results indicated that ethanol extract of Codonopsis lanceolata root would have antidiabetic effect in STZ-induced diabetic rats.

The Effect of Platycodon grandiflorum Root Ethanol Extract on Blood Glucose, Lipid, Activities of Carbohydrate Metabolism Related Enzyme in Streptozotocin-Induced Diabetic Rats (도라지 뿌리 에탄올 추출물이 streptozotocin으로 유발된 흰쥐의 혈당지질, 당대사에 미치는 영향)

Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Platycodon grandiflorum root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Platycodon grandiflorumt root treated group compared to the those of STZ-control group. Also the contents of hepatic glycogen and HDL-cholesterol, the activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased (p<0.05). These results indicated that ethanol extract of Platycodon grandiflorum root would have antidiabetic effect in STZ-induced diabetic rats.

Effect of Meal Pattern on Lipogenesis and Lipogenic Enzyme Activity in Rat Adipose Tissue Fed High Fat Diet (식이급여형태가 고지방식이를 급여한 흰쥐 지방조직의 지방합성 및 지방합성 효소활성에 미치는 영향)

  • This study was undertaken to investigate the effects of meal pattern on lipogenesis and activities of lipogenic enzyme in rats epididymal and mesenteric adipose tissues. A high fat diet was fed either ad libitum or in 1 meal during the last 3 h of the dark cycle for 4 weeks. Lipogenesis was measured as glucose conversion to total lipid and activities of glucose-6-phosphate dehydrogenase (G6PDH), 6-phophogluconate dehydrogenase (6PGDH) and NADP-malate dehydrogenase (ME) were determined by measuring NADPH production. Lipoprotein lipase (LPL) activity and serum lipoprotein concentrations were also measured. Meal-fed (3 h) rats had a decreased food intake, body weight and carcass fat compared with rats fed ad libitum. The serum triglyceride concentration of meal-fed rats tended to be higher than that of the ad libitum rats. However, there were no differences between meal-fed group and ad libitum group in serum concentrations of HDL-cholesterol, LDL-cholesterol and total-cholesterol. Rates of lipogenesis in both epididymal and mesenteric adipose tissues were significantly higher in the meal-fed group than that in the ad libitum group. In addition, meal-fed group showed higher G6PDH, 6PGDH and LPL activities in both epididymal and mesenteric adipose tissues, but exerted no significant effect on ME activity. These results suggest that meal-fed rats compared with ad libitum rats have marked lipogenic capacity, although such elevation probably does not result in increase in carcass fat concentration. Thus, meal-fed diet can be an important determinant of the alterations in adipose lipid metabolism.

The Study of Biochemical Changes Induced by Fish Oil Diet in Rat(I) - Changes in Hepatic Lipogenic Enzyme Activity - (어유(魚油)식이에 의한 흰쥐체내의 생화학적 변화연구(I) -간장조직내 지방산 합성효소의 변화 -)

  • In order to evaluate the effect of fish oil on lipogenesis, activities of glucose-6-phosphate dehydrogenase ( G6PDH ) and malic enzyme (ME) were measured in liver of rats fed mackered oil(MO) or eel oil (EO) for 10 to 14 days, at the various levels of 0 to 10% (w/w ). In addition to two kinds of fish oil, soybean oil (SO), lard (L), and beef tallow (BT) were fed to the different groups of rats. When fish oil was below 10%(w/w ), soybean oil, lard, or beef tallow was mixed with fish oil to maintain constant 10% (w/w) fat level. Three days of feeding MO brought a marked decrease$({\sim}{50}%)$ both in G6PDH and ME activity, the former of which maintained during 13 days of feeding. L group had highest levels of both enzymes. G6PDH activity of MO was lower than SO, but ME activity was not different between MO and SO. G6PDH activity was decreased with increasing content of fish oil (MO, EO), starting at the 2%(w/w) level of fish oil, when L or BT was used as filler oil. But ME activity was significantly reduced when fish oil content was at least 5%(w/w). Difference between the effects shown by two kinds of fish oil and animal species were also found. The present study suggests that fish oil can suppress hepatic lipogenesis by reducing activities of lipogenic enzymes with the same or higher degree than vegetable oil can exert.

Effects of Aminotriazole on Lung Toxicity of Paraquat Intoxicated Mice (Paraquat중독에 의한 폐독성에 미치는 Aminotriazole의 영향)

  • Background: Paraquat, a widely used herbicide, is extremely toxic, causing multiple organ failure in humans. Paraquat especially leads to irreversible progressive pulmonary fibrosis, which is related to oxygen free radicals. However, its biochemical mechanism is not clear. Natural mechanisms that prevent damage from oxygen free radicals include changes in glutathione level, G6PDH, superoxide dismutase(SOD), catalase, and glutathione peroxidase. The authors think catalase is closely related to paraquat toxicity in the lungs Method: The effects of 3-amino-1,2,4-triazole(aminotriazole), a catalase inhibitor, on mice administered with paraquat were investigated. We studied the effects of aminotriazole on the survival of mice administered with paraquat, by comparing life spans between the group to which paraquat had been administered and the group to which a combination of paraquat and aminotriazole had been administered. We measured glutathion level, glucose 6-phosphate dehydrogenase(G6PDH), superoxide dismutase(SOD), catalase, and glutathione peroxidase(GPx) in the lung tissue of 4 groups of mice: the control group, group A(aminotriazole injected), group B(paraquat administered), group C(paraquat and aminotriazole administered). Results: The mortality of mice administered with paraquat which were treated with aminotriazole was significantly increased compared with those of mice not treated with aminotriazole. Glutathione level in group B was decreased by 20%, a significant decrease compared with the control group. However, this level was not changed by the administration of aminotriazole(group C). The activity of G6PDH in all groups was not significantly changed compared with the control group. The activities of SOD, catalase, and glutathione peroxidase(GPx) in the lung tissue were significantly decreased by paraquat administration(group B); catalase showed the largest decrease. Catalase and GPX were significantly decreased by aminotriazole treatment in mice administered with paraquat but change in SOD activity was not significant(group C). Conclusion: Decrease in catalase activity by paraquat suggests that paraquat toxicity in the lungs is closely related to catalase activity. Paraquat toxicity in mice is enhanced by aminotriazole administration, and its result is related to the decrease of catalase activity rather than glutathione level in the lungs. Production of hydroxyl radicals, the most reactive oxygen metabolite, is accelerated due to increased hydrogen peroxide by catalase inhibition and the lung damage probably results from nonspecific tissue injury of hydroxyl radicals.

Effects of Vegetable Sprout Power Mixture on Lipid Metabolism in Rats Fed High Fat Diet (새싹채소 혼합분말이 고지방 식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effects of vegetable sprout powder on serum and adipose tissue lipid metabolism in rats fed high-fat diet for 4 weeks for induction hyperlipidemic model rat. Weight-matched male Sprague-Dawley rats were assigned to five groups according to dietary fat level (10% or 20% of diet wt.) and mixture of vegetable sprout powder levels (5% or 10% 10% or 20% of diet wt.). Vegetable sprout powder was the mixture of same amounts of dried barley, broccoli, rapeseed, alfalfa, radish, mustard, buckwheat and brussels sprouts. Experimental groups were normal fat diet with 5% cellulose (NF-C), high fat diet without fiber (HF-N), high fat diet with 5% cellulose (HF-C), HF-C diet with 5% vegetable sprout powder (HF-CSL), and HF-C diet with 10% vegetable sprout powder (HF-CSH). The body weight of HF-N group increased 16% compared with the NF-C group, while it was decreased by 15% and 22% for HF-CSL group and HF-CSH group, respectively. Fat mass and fat cell size of adipose tissue were lower in HF-CSL group and HF-CSH group compared with HF-C group, and lower in HF-CSH group compared with HF-CSL group. Serum triglyceride, total cholesterol and LDL-cholesterol contents were markedly decreased by vegetable sprout powder containing diet, while the serum HDL-cholesterol and phospholipid contents were higher in vegetable sprout powder containing diet in a dose-dependent manner. Leptin and insulin levels in serum showed a decrease in HF-CSH group. Significantly increased contents of triglyceride, total cholesterol, LDL-cholesterol, leptin and insulin in the serum of HF-N group were returned to normal or even below normal levels by feeding 10% vegetable sprout powder diet. The increased activities of NADP-malate dehydrogenase (ME), glucose-6-phosphate dehydrogenase (G6PDH) and 6-phosphogluconate dehydrogenase (6PGDH) and lipoprotein lipase (LPL) in adiposetissue by HF-N group were decreased to the activity of normal fat group by feeding vegetable sprout powder in a dose-dependant manner. These results indicate that lipid metabolism in rats fed high-fat diet was suppressed by feeding vegetable sprout powder.

Antidiabetic and Antioxidative Effects of Cibotium barometz in Streptozotocin-Induced Diabetic Rats (구척 에탄올 추출물이 Streptozotocin 유발 당뇨 흰쥐에 대한 항당뇨 및 항산화 작용에 미치는 효과)

The ethanol extract of Cibotium barometz was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Cibotium barometz treated group compared to the those of STZ-control group, The content of glutathione(GSH) and activity of gluthathione-s-transferase(GST) were significantly increased (P<0.05) in Cibotium barometz treated group compared to the those of STZ-control group. and activityes of catalase(CAT) and glutathione peroxidae(GSH-Px) were signiicantly decreased (P<0.05) in Cibotium barometz treated group compared to the those of STZ-control group. Also the content of hepatic glycogen and activities of glucose-phosphate dehydrogenase(G-6-PDH)and glucokinase(GK) were significamtly increased(p<0.05), but activity of glucose-6-phosphatase (G-6-Pase) was significamtly decreased (p<0.05) in Cibotium barometz treated group compared to the those of STZ-control group. These results indicated that ethanol extract of Cibotium barometz would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.
