Initial Control of Microorganism in Kimchi by the Modified Preparation Method of Seasoning Mixture and the Pretreatment of Electrolyzed Acid-Water (전해산화수 전처리 및 양념류 제조방법에 따른 김치의 초기 미생물 제어)

  • To lessen the initial level of microorganism in kimchi, the preparation method of seasoning mixture was modified and electrolyzed acid-water was substitute for washing water and brine water. Changes in the microbial counts of the red pepper powder-garlic mixture prepared with red pepper powder, garlic and 4.8% (w/w) water (tap water and electrolyzed acid-water) of manufactured kimchi showed that coliform count and E. coli count were reduced to 93% and 98%, of the initial level in the tap water added red pepper powder-garlic mixture and 97% and 99% in the electrolzed acid-water added mixture after 6 hours of mix. After 24 hours of mix, no E. coli was recovered in borth mixtures. Microbial levels were revealed as $2.4{\times}10^5\;CFU/g$ for total count, $2.3{\times}10^5\;CFU/g$ for Lactobacillus count. $1.0{\times}10^2{\;}CFU/g$ of coliform count and $10^1\;CFU/g$ of E. coli count in the seasoning mixture prepared by the general method, and $4.2{\times}10^5\;CFU/g$ of total count, $4.0{\times}10^5CFU/g$ of Lactobacillus count, $1.0{\times}10^3\;CFU/g$ of coliorm count and $4.0{\times}10^2\;CFU/g$ of E. coli count in the mixture propared by the modified method. During fermentation at $10^{\circ}C$, the levels of total and Lactobacillus counts in kimchi prepared by the modified method were significantly lower than those of kimchi were recovered in kimchi prepared by the modified method, whereas there were at the level of $10^1\;CFU/g$ in kimchi prepared by the general method. The pH and acidity of kimchi prepared by the modified method were 4.66 and 0.54%, respectively, whereas those in kimchi prepared b the general method were 4.51 and 0.70%, respectively. But after 14 days of fermentation significant differences were not observed in the changes of microbial, pH and acidity for both kimchi.

Development of Diffusion - Precipitation Method to Determine AVS Concentrations in Freshwater Sediments

  • A diffusion - precipitation method was developed to determine acid volatile sulfide (AVS) concentrations in freshwater sediments. This method uses silver nitrate as a sulfide trap solution and the concentration of trapped sulfide is determined gravimetrically. The proposed diffusion - precipitation method is more rapid and less expensive than previously developed purge- and - trap methods. Spiked sodium sulfide recoveries using this method $(97\~120\%)$ were similar with a previously developed diffusion - absorption method $(93.8\~115\%)$ and about $20\%$ greater than a previously developed purge-and-trap method $(74.6\~105\%)$. Detection limit of this method $(0.1\;{\mu}mole\;S\;g^{-l})$ was comparable with that of diffusion-absorption method $(0.06\;{\mu}mole\;S\;g^{-l})$ and purge-and-trap method $(0.05\~0.5\;{\mu}mole\;S\;g^{-l})$.

A Survey on the Amount used of Toothpaste According to the Size of Head of Toothbrush and Squeezing Method (칫솔두부크기와 짜는 방법에 따른 일회 세치제 사용량 조사)

  • The purpose of this study was to prevent excessive use of fluorine toothpaste. To comparatively evaluate the amount used of the fluorine toothpaste in children from 3 to 6 years old, the toothpaste amount was compared according to toothbrush size and toothpaste squeezing method targeting 84 children in 5 child care centers where were located in Hongseong-gun, Chungnam. The toothpaste amount was compared with a method of squeezing to pea-size and a method of smear on toothbrush, targeting 141 students for the Dept. of Dental Hygiene, who are actually carrying out oral health education to targeting children. The statistical analysis was used SPSS 14.0 K for Windows program(Copyright(c) SPSS Inc.). The following results were obtained. 1. The amount of fluorine toothpaste squeezed on toothbrush for children was 0.29 g. The amount of fluorine toothpaste squeezed on toothbrush for adults was 0.34 g. It was surveyed to use the less amount of toothpaste when squeezing to smaller toothbrush (p<0.05). 2. The amount that children aged 3-6 squeezed as the method of squeezing commonly at ordinary times was 0.31g. The amount that was squeezing to pea-size was 0.21 g. The amount that was squeezed by using smear method was measured to be 0.26g. Less amount of fluorine toothpaste was used with the method of squeezing to pea-size and smear method rather than a method of squeezing commonly at ordinary times(p<0.05). 3. As a result of surveying the students of the department of dental hygiene, the amount of having squeezed to pea- size was 0.23 g. It was surveyed to be 0.15 g when having used the smear method. Thus, it was surveyed to use the less amount of toothpaste when using the smear method(p<0.05). 4. Using smaller toothbrush, the amount used of fluorine toothpaste can be reduced. Both smear method and the method of squeezing to pea-size are available for minimizing careless fluorine intake by less used amount of fluorine toothpaste.

A Comparative Evaluation of Sampling Methods for Airbone Mercury Concentration (공기중 수은 농도의 측정방법에 대한 비교평가연구)

  • This study was performed to evaluate methods for determination of airbone mercury in industrial environments. Three methods, such as Hopcalite method, passive monitoring method and Jerome method were evaluated at two (2) fluorescent lamp manufacturing Plants in Korea during a period from May 24 to May 31, 1994 and the result are as follows: 1. The average were concentrations of airbone mercury were $26{\mu}g/m^3$ by Hopcalite method, $25{\mu}g/m^3$ by passive monitoring method, and $38{\mu}g/m^3$ by the Jerome method, respectively, which were below the permissible exposure limit of $50{\mu}g/m^3$ established by both the Korean Ministry of Labor and ACGIH. However, 12 out of 49 cases(24.4%) in plant A and 2 out of 31 cases(6.5%) in plant B were in excess of the standard. 2. The relationship between the results by Hopcalite method and the passive monitoring method was significant(r=0.892). 3. The variation among three results by eath of three methods were different by process. The highest variation was determined at quality control process("process b") which invoved in large variation of concentrations. 4. When short term high concentrations were produced, the Hopcalite method was more efficient then the passive method.

Optimum Determination Method of Protein-Nitrogen Improve Mohr Method in Leaf Tobacco (잎담배 단백태 질소 분석법(Mohr법) 개선)

  • This study was carried out in order to improve the Mohr method for protein-nitrogen concentration determining. Existing Mohr's method takes seven hours to analyze for one sample. In order to make up for these things tried to utilize filter bag(F75, ANKOM) for sample treatment. As a result of this implementation, have saved sample loss amount and analyzing time for a quarter comparing with Mohr's method. In addition, it have gotten high efficiency through five samples hydrolyze per one trial by using ANKOM hydrolysis system with 0.5% acetic acid. Besides, had a good reproducibility for analysis results in relation to nitrogen concentration determining with Dumas methodology. Thus, new Mohr's method takes one day to analyze for 40 sample. It is more efficiency about 6 times compare with existing Mohr's method. And, this modified Mohr's method was verified that is substitutable for the existing Mohr's method in statistical analysis.

Evaluation of Parameter Estimation Method for Design Rainfall Estimation (설계강우량 산정을 위한 매개변수 추정방법 평가)

  • Determining design rainfall is the first step to plan an agricultural drainage facility. The objective of this study is to evaluate whether the current method for parameter estimation is reasonable for computing the design rainfall. The current Gumbel-Kendall (G-K) method was compared with two other methods which are Gumbel-Chow (G-C) method and Probability weighted moment (PWM). Hourly rainfall data were acquired from the 60 ASOS (Automated Synoptic Observing System) stations across the nation. For the goodness-of-fit test, this study used chi-squared (𝛘2) and Kolmogorov-Smirnov (K-S) test. When using G-K method, 𝛘2 statistics of 18 stations exceeded the critical value (𝑥2a=0.05,df=4=9.4877) and 10, 3 stations for G-C method, PWM method respectively. For K-S test, none of the stations exceeded the critical value (Da=0.05n=0.19838). However, G-K method showed the worst performances in both tests compared to other methods. Subsequently, this study computed design rainfall of 48-hour duration in 60 ASOS stations. G-K method showed 5.6 and 6.4% higher average design rainfall and 15.2 and 24.6% higher variance compared to G-C and PWM methods. In short, G-K showed the worst performance in goodness-of-fit tests and showed higher design rainfall with the least robustness. Likewise, considering the basic assumptions of the design rainfall estimation, G-K is not an appropriate method for the practical use. This study can be referenced and helpful when revising the agricultural drainage standards.

A Study on the Validity of Filter Paper Method in Estimation of the Amount of Daily Salt Intake (여과지법에 의한 식염섭취량추정의 타당성에 관한 조사연구)

  • For the purpose of the determination of the most proper sampling time on using spot urine which can represent the general status of electrolytes in 24 hour urine and for test of the validity of filter paper method which was developed recently in estimation of the amount of daily salt intake, the author investigated this study by different sampling time and various measuring methods in 21 healthy men and 12 women. The summarized results were as follows; 1) The mean excretion amount of urinary electrolytes were Na 3.93 g/l, K 1.47 g/l, and creatinine 1.08 g/l in male, and Na 3.83 g/l, K 1.86 g/l, and creatinine 0.99 g/l in female. 2) In using spot urine for estimation of the amount of daily salt intake, morning urine was the most approximate to that of 24 hour urine in both sexes. 3) There was validity in estimation of daily salt intake by filter paper method using spot urine instead of 24 hour urine. 4) The estimated amounts of daily salt intake in male was higher than the values in female as 16.04-16.22 g and 13.35-13.82 g respectively.

HPLC Method for Simultaneous Quantitative Detection of Quercetin and Curcuminoids in Traditional Chinese Medicines

  • Objectives: Quercetin and curcuminoids are important bioactive compounds found in many herbs. Previously reported high performance liquid chromatography ultraviolet (HPLC-UV) methods for the detection of quercetin and curcuminoids have several disadvantages, including unsatisfactory separation times and lack of validation according the standard guidelines of the International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use. Methods: A rapid, specific, reversed phase, HPLC-UV method with an isocratic elution of acetonitrile and 2% v/v acetic acid (40% : 60% v/v) (pH 2.6) at a flow rate of 1.3 mL/minutes, a column temperature of $35^{\circ}C$, and ultraviolet (UV) detection at 370 nm was developed. The method was validated and applied to the quantification of different types of market available Chinese medicine extracts, pills and tablets. Results: The method allowed simultaneous determination of quercetin, bisdemethoxycurcumin, demethoxycurcumin and curcumin in the concentration ranges of $0.00488-200{\mu}g/mL$, $0.625-320{\mu}g/mL$, $0.07813-320{\mu}g/mL$ and $0.03906-320{\mu}g/mL$, respectively. The limits of detection and quantification, respectively, were 0.00488 and $0.03906{\mu}g/mL$ for quercetin, 0.62500 and $2.50000{\mu}g/mL$ for bisdemethoxycurcumin, 0.07813 and $0.31250{\mu}g/mL$ for demethoxycurcumin, and 0.03906 and $0.07813{\mu}g/mL$ for curcumin. The percent relative intra day standard deviation (% RSD) values were $0.432-0.806{\mu}g/mL$, $0.576-0.723{\mu}g/mL$, $0.635-0.752{\mu}g/mL$ and $0.655-0.732{\mu}g/mL$ for quercetin, bisdemethoxycurcumin, demethoxycurcumin and curcumin, respectively, and those for intra day precision were $0.323-0.968{\mu}g/mL$, $0.805-0.854{\mu}g/mL$, $0.078-0.844{\mu}g/mL$ and $0.275-0.829{\mu}g/mL$, respectively. The intra day accuracies were 99.589%-100.821%, 98.588%-101.084%, 9.289%-100.88%, and 98.292%-101.022% for quercetin, bisdemethoxycurcumin, demethoxycurcumin and curcumin, respectively, and the inter day accuracy were 99.665%-103.06%, 97.669%-103.513%, 99.569%-103.617%, and 97.929%-103.606%, respectively. Conclusion: The method was found to be simple, accurate and precise and is recommended for routine quality control analysis of commercial Chinese medicine products containing the flour flavonoids as their principle components in the extracts.

  • In order to observe the K contents, Korean common foods, especially processed foods were analyzed by Atomic Absorption Spectrophotometer. The results obtained were summarized as followed; (I) The K contents of instant noodle(ramyon) was 105-180 mg/100g and K ontents of their soup powder was 500-1000 mg/100g. (2) The K contents of potato products and bean products were less than those of original foods. So it was recongnized that the K contents might reduced during the food process. (3) The K contents of meats was 130-475 mg/I00g and those of meats products was relatively low, 80-260 mg/100g. (4) The K contents of carbonate beverage was very low but that of instant coffed was very high, 2400-2600 mg/100g. (5) There was no Statistical difference between the results of wet ash method and dry ash method in the K contents of all food groups.

The Extraction Method for the G-Sensitivity Scale-Factor Error of a MEMS Vibratory Gyroscope Using the Inertial Sensor Model (관성센서 오차 모델을 이용한 진동형 MEMS 자이로스코프 G-민감도 환산계수 오차 추출 기법)

  • In this paper, we present a new approach to extract the g-sensitivity scale-factor error for a MEMS gyroscope. MEMS gyroscopes, based on the use of both angular momentum and the Coriolis effect, have a g-sensitivity error due to mass unbalance. Generally, the g-sensitivity error is not considered in general use of gyroscopes, but it deserves our attention if we are to develop for tactical class performance and reliability. The g-sensitivity error during vehicle flight increases navigation error; so it must be analyzed and compensated for the use of MEMS IMU for high dynamics vehicle systems. Therefore, we analyzed how to extract the g-sensitivity scale-factor error from the inertial sensor error model. Furthermore we propose a new method to extract the g-sensitivity error using flight motion simulator. We verified our proposed method with experimental results.
