Studies on the processing of rapid fermented anchovy prepared with low salt contents by adapted microorganism. -2. Thermodynamic characteristics of microbial extracellular protease isolated from fermented fish paste- (미생물을 이용한 저식염 멸치젓의 속성발효에 관한 연구 -2. 젓갈에서 분리한 단백질분해효소의 열역학적 특성-)

  • This study was undertaken to determine thermodynamic characteristics of B. subtilis p-4 and B. licheniformis p-5 proteases isolated from fermented anchovy paste. $K_m$ values of two proteases for casein as a substrate were 0.38mM in p-4 protease and 0.18mM in p-5 protease, respectively. Denaturation constants($K_D$) of p-4 and p-5 proteases were $12.2{\times}10^{-5}/sec\;and\;19.0{\times}10^{-5}/sec\;at\;40^{\circ}C,\;and\;35.7{\times}10^{-5}/sec\;and\;46.3{\times}10^{-5}/sec\;at\;50^{\circ}C$, respectively. Activation energies($E_a$) of p-4 and p-S pmteases were 19.6 Kcal/mole and 15.2kcal/mole, respectively. Free energy of activation(${\Delta}G^*$), activation enthalpy(${\Delta}H^*$) and activation entropy(${\Delta}S^*$) at $40^{\circ}C$ were 23.21Kcal/mole, 18.98Kcal/mole and -13.50 eu, respectively for p-4 protease and 22.93Kcal/mo1e, 14.58Kcal/mole and -26.68 eu, respectively for p-5 protease. The major amino acids in p-4 protease(151 residues of amino acid) were Gly, Glu, Pro, Asp, Ser, Ala, Lys and Leu, while those in p-5 protease(247 residues of amino acid) were Gly, Glu, Asp, Ala and Leu. It may be concluded that heat denaturation of two proteases showed liner regression curve and p-5 protease was more sensitive to heat than p-4 protease.

Changes Over Time in the Community Structure and Spatial Distribution of Forest Vegetation on Mt. Yeompo, Ulsan City, South Korea (염포산 산림식생의 군락 구조 및 공간 분포의 경시적 변화)

  • In 2000 and 2018, phytosociological surveys were carried out in the forest vegetation of Mt. Yeompo, a representative isolated urban forest in Ulsan city. The trends of change in forest structure, composition, and spatial distribution were compared between years. Total percent coverage per 100 squaremeters of forest vegetation was similar, but natural vegetation showed a 9% increase. The importance of constituent species changed slightly. Specifically, Lindera erythrocarpa and Styrax japonicus showed very high growth rates of 835% and 269%, respectively. Species richness (S) and diversity (H') decreased by about 22% and 8%, respectively. Both S and H' showed slightly higher rates of decrease in artificial compared with natural vegetation. The constituent species life form spectrums were the same in 2000 and 2018 as 'MM-R5-D4-e'. The similarity (Jaccard coefficient) in the species composition of the forest vegetation was almost homogeneous at approximately 75%. The number of indicator species decreased from 16 species in 2000 to 7 species in 2018. This decrease was mostly due to a decline in herbaceous plants, such as Hemicryptophytes, Geophytes, and Therophytes, which are sensitive to disturbances. The spatial distribution of forest vegetation did not change significantly. The number of forest landscape elements (patches) increased by approximately 25% from 537 in 2000 to 721 in 2018, while the average size decreased by about 20% from 1.28 ha in 2000 to 1.03 ha in 2018.

Changes in the pathogenicity of Naegleria fowleri by serial brain passage in mice (자유생활아메바 Naegleria fowleri의 계대감염에 의한 병원성의 변화에 관한 연구)

  • The pathogenicity of free-living amoeba, Waegleria fcwleri, is influenced according to the strain, cultural condition and host (Culbertson et at., 1968; Carter, 1970; Wong et at., 1975), Phillips (1973) demonstrated that Entamoeba histolytica became avirulent after more than 2 year maintenance in axonic culture in vitro. This study was carried out to compare the difference in pathogenicity between two strains of N. fowleri, one of a prolonged maintenance in arsenic medium and the other one obtained by serial brain passage in mice. The 0 strain was that N. fowleri had cultivated axenically more than 7 years in CGVS medium. The 2-1 strain was obtained from the brain of mouse inoculated intranasally with a strain, which was from the mouse brain infected with 0 strain, and cultured for 15 weeks until the beginning of this experiment. White male mice weighing 18-22 g were used. Mice were anesthetized by an intraperitoneal injection of about 1 mg secobarbital, and inoculated intranasally with $10{\times}$10^4 live N. fowleri trophoBoites in a $5{\;}{\mu}l$ cell suspension. Sluggish behaviour, nervousness, rotation and leg paralysis were developed earlier and more frequently in the 2-1 experimental group than the control 0 group. Pathological changes such as inflammatory and necrotic lesion were observed in the olfactory and anterior portion of brain, and these changes were more extensive in the 2-1 group. The edematous and inflammatory changes in lung were demonstrated in mice died after 13th day post-inoculation. The experimental mice of 2-1 group began to die suddenly from 7th day post-inoculation, and the survival time in 2-1 group mice was shorter than 0 group mice. The typical primary amoebic meningoencephalitis was developed in the mice inoculated intranasally with N. fowleri. The prolonged maintenance of N. fowleri amoebae in axonic CGVS medium was observed to have lost their original pathogenicity for mice, but their pathogenicity was restored by serial brain passage in mice.

The COP9 Signalosome Network in Eukaryotic Microorganisms (진핵 미생물에서의 COP9 signalosome의 역할)

  • COP9 signalosome (CSN), which is originally identified as the regulator of the photomorphogenic development in plant, is highly conserved protein complex in diverse eukaryotic organisms. Most eukaryotic CSN complex is composed of 8 subunits, which is structurally and functionally similar to the lid subunit of 26S proteasome and eIF3 translation initiation complex. CSN play important functions in the regulation of cell cycle and checkpoint response by controlling Cullin-Ring E3 ubiquitin ligases (CRL) activities. CSN exhibits an isopeptidase activity which cleaves the neddylated moiety of cullin components. In fission yeast, S-phase cell cycle progression was delayed and the sensitivity to g-ray or UV was increased in CSN1 and CSN2 deletion mutants, indicating that yeast CSN is also involved in the checkpoint regulation. CSN in fungal system more closely resembles that of the higher organisms in the structure and assembly of their components. Functionally, CSN is associated with the regulation of conidiation rhythms in Neurospora crassa and the sexual development in Aspsergillus nidulans. Recent studies also revealed that CSN functions as an essential cell cycle regulator, playing key roles in the regulation of DNA replication and DNA damage response in Aspergillus. Overall, CSN of microorganisms, such as fission yeast and fungi, share functionally common aspects with higher organisms, implying that they can be useful tools to study the role of CSN in the CRL-mediated diverse cellular activities.

Effects of Leucine on in Vivo Protein Synthesis in Skeletal Muscles of Fed and Food-Deprived Rats (Leucine이 정상 또는 굶게 된 쥐의 골격근육의 단백질 생합성에 미치는 영향)

  • In vivo effects of leucine on skeletal muscle protein synthesis in fed and I-day food deprived young rats were examined. Animals assigned to leucine group were given a single i.p. injection of 80 or 160flmoles of leucine while control group animals were saline sham injected. The rate of protein synthesis was measured by the amount of $^{14}\textrm{C} incorporated into muscle protein after a single injection of $^{14}\textrm{C}-tyrosine, IO$\mu$ Ci/l00g B.W. Examined muscles were two different types of hind limb muscles. the oxidative solues and the glycolytic EDL and plantaris. Administered leucine elevated the concentration of free leucine in soleus muscles by 4-6.8 times the normal level. A massive dose of leucine, 160 flmoles, stimulated protein synthesis in the EDL and plantaris by 24 %, 29 % respectively of straved rats. The soleus of I-day food deprived rats and both types of muscles in fed rats did not respond to the injected leucine. The synthesis rate of the EDL and plantaris was supressed to one-half of the normal while the soleus that was not stimulated by leucine maintained a relatively normal rate, 78 %, of protein synthesis after I-day of food deprivation. Thus, in vivo stimulatory effect of leucine appears to be not a general phenomenon but to be related to the degree of catabolic condition developed by stress such as food deprivation. Although anabolic effects of leucine observed in this study was limited, any applicability of this special property of leucine to human subjects for the purpose of protein sparing in skeletal muscles remains to be examined.

  • 극세섬유는, 1970년대 말부터 일본의 합섬업체들이 천연소재인 Silk를 모방하기 위해 개발을 시작하였으며, 개발된 섬유의 굵기가 0.5d(1denier=1g/9,000m)이하인 것을 말한다. 이후 극세사에 대한 끊임없는 노력으로, 1980년대에는 천연스웨이드 풍의 인조피혁의 제조가 가능한 0.1d이하급 초극세사가 N/P(Nylon/Polyester) 복합방사된 형태로 개발되었으며, 일본, 및 한국 등에서 이들 소재로 제조된 직편물 제품이 첨단 고부가소재로서 호황을 누려왔다. 이러한 초극세사는 다양한 형태로 발전되어왔는데, 해도형(Islands-in-a-Sea Type), N/P(Nylon/PET), P/N(PET/Nylon) 등이 대표적인 형태이며, 가공공정 중에서 분할이 되면서 그 특성을 발현한다고 하여 분할사, 형태에 따라서 N/P 분할사라 하기도 한다. 최근들어 이러한 N/P 분할사는 기존의 의류용 용도뿐만 아니라, Wiping Cloth, 극세사 타울, 항진드기용 침장 등 다양한 비의류용 소재로도 확대 전개되고 있으며, 이렇게 다양화 되어가고 있는 용도에 따른 공정별 최적 가공 방법에 대한 연구가 진행 중이다. 사가공 공정에 있어서는 텍스쳐링 방법이 적용되기도 하는데, 가장 보편화된 텍스쳐링 방법으로는 DTY(Draw Texturing Yarn), ATY(Air Texturing Yarn) 등의 형태가 있으며, 이러한 텍스쳐링 방법은 물성에 민감한 N/P 분할사의 강도, 신도, 분할도에 영향을 미치게 된다. 본 연구에서는 기존의 텍스쳐링 방법이 아닌 Fancy사 형태로 사가공을 하였으며, 직물로 제직하고, 분할 가공하여 직물로써의 물성까지 분석하였다. Fancy사는 색이나 형의 변화로 디자인효과를 준 실을 말하는데, 심사, 부사, 압사로 이뤄지며 의장사, 장식사 등으로 불려지기도 한다. 주요 공정을 보면, 크릴형성용 부사를 500-600T/M으로 가연한 연사를 얻는 제1공정과, 이 가연된 부사와 심사를 합사하는 제2공정 및 합사된 크릴의 뒤틀림을 방지하기 위하여 압사로 크릴을 고정하여 주는 제3공정으로 이루어진다. 사용된 N/P 분할사는 NP30/36dty, NP50/36dty를 사용하였으며, 부사의 오버피드 및 피드되는 사에 따라 각각 8종, 7종의 Fancy사를 제조하여 섬도(Denier), 강도(Tenacity), 신도(Elongation)를 측정하였다. 또한, 이들 사들로 제직 및 분할가공을 하여 인장강도, 인장신도, 인열강도, 마모강도, 공기투과도 등의 물성과 중량, 두께를 측정하였으며, 온도, NaOH 농도, 시간 등의 분할 가공조건에 따른 직물의 인열강도 변화도 측정하였다. 이렇게 공정별 조건에 따른 물성의 변화분석을 통해 추후 N/P의 제품화 전개에 도움이 되고자 하였다.

Rapid Nutrient Diagnosis of Tomato by Test Strips and a Chlorophyll Meter (Test strip과 chlorophyll meter를 이용한 토마토의 신속한 영양진단)

  • This study was performed to develop a more rapid and simple nutrient diagnosis method of plants than the conventional leaf analysis method. tomato (Lycopersicon esculentum Mill. cv. super momotaro) was planted in the mixed media produced by mixing perlite and rock wool at 1 . 1 (v/v) ratio. The Yamazakki nutrient solutions for cucumber and tomato were supplied to the media using the micro-drip irrigation system. Experimental plots produced consisted of no fertilization, deficient fertilization, adequate fertilization and surplus fertilization for N, P and K, respectively. Each experimental plot was replicated four times. Specific color different sensor values (SCDSV) measured by the chlorophyll meter were closely related to total-N concentrations in the leaves measured by the conventional method. Nitrate, $PO_4$ and K concentrations in petiole sap measured by test strips #(Reflectoquant^{\circledR},\;Merck,\;Germany)$ showed a significant relationship with total-N, p and K concentrations in leaves. The linear regression equations between $NO_3,\;PO_4$ and K concentrations in petiole sap and total-N, p and K concentrations in the leaves were prepared. The optimum levels of $NO_3,\;PO_4$ and K in petiole sap were obtained by plugging the optimum concentrations of total-N, p and K in the leaves by other researchers into the equations. In conclusion, the SCDSV by the chlorophyll meter and concentrations of NO3, p and K in petiole sap measured by the test strips would be suitable for rapid estimation of plant nutrient status.

Effect of Immobilization Method in the Biosorption and Desorption of Lead by Algae, Chlorella pyrenoidosa (Chlorella pyrenoidosa에 의한 납 흡.탈착시 고정화 방법의 영향)

  • In this studies, the adsorption test using Chlorella pyrenoidosa was conducted to examine the effect of Pb adsorption according to various immobilized methods such as Ca-alginate, K-carrageenan, and Polyacrylamide. From the results, the duration to need to reach adsorption equilibrium was delayed according to the immobilization. And, the higher adsorption capacity of immobilized Chlorella pyrenoidosa was represented in the higher concentration of Pb, the smaller amount of immobilizing agent, and the higher pH of solution. The maximum adsorption capacity of Pb was shown in the adsorption test using Chlorella pyrenoidosa immobilized with Ca-alginate even though it was sensitive pH. The adsorption results properly represented with Freundlich isotherm equations. And, pseudo second-order chemisorption kinetic rate equation was applicable to all the biosorption data over the entire time range. The FT-IR analysis showed that the mechanism involved in biosorption of Pb by Chlorella pyrenoidosa was mainly attributed to Pb binding of carbo-acid and amide group. Adsorbed Pb on immobilized Chlorella prenoidosa was easily desorbed in the higher concentration of desorbents(NTA, HCl, EDTA, $H_2SO_4,\;Na_2CO_3$). Among the several desorbents, NTA showed the maximum desoption capacities of Pb from Chlorella pyrenoidosa immobilized with Ca-alginate and K-carrageenan and EDTA was the most effective in Chlorella pyrenoidosa immobilized with polyacrylamide. The desoprtion efficiency in the optimum condition was 90.0, 83.0, and 80.0%, respectively.

A Dendroclimatic Analysis on Abies koreana in Cheonwang-bong Area of Mt. Chiri, Korea (지리산 천왕봉지역 구상나무의 연륜기후학적 해석)

  • The relationships between climate (monthly precipitation and temperatures) and tree-ring growth of Korean fir (Abies koreana) growing at subalpine (1,600∼l,700m) zone on the south slope, Joong-Bong and the north slope, Changgun-Bong in the Cheonwang-bong area in Chiri mountains in the southern Korea were analyzed. Two cores from each of 10∼12 trees were extracted. The relationship between tree-ring(standardized) chronologies and climate was analyzed by the response-function method. Climate variables could explain 45.9∼53.8% of total variance in the chronologies. The precipitation response function of Korean fir were similar at both sites in overall ; positive for March∼May and August, and negative for February and July. The south-slope fir of Joong-Bong possessed higher negative temperature response function for February and August than north-slope one. The positive response function for April temperature was significant for both sites. In contrary to other subalpine species (e.g., Pinus koraiensis and Taxus cuspidata) in South Korea, whose growths are positively correlated with temperature in most seasons, the growths of Korean fir trees in Mt. Chiri appeared to be mainly limited by the moisture regime of spring prior to the cambial growth and early growing season.

Antimicrobial Activity and Cell Cytotoxicity of Korean Plum-yem Extract (개비자나무(Cephalotaxus koreana) 추출물의 항균활성 및 세포독성)

  • Antimicrobial activity and cell cytotoxicity of Korean Plum-yem (Cephalotaxus koreana) extract were evaluated for the development of a functional biomaterial. And the chemical compositions of Korean Plum-yem extract (KPE) were analyzed. KPE showed high antimicrobial activities against various microorganisms including gram-positive bacteria, gram-negative bacteria, yeasts and molds. KPE showed cytotoxic activity dose-dependently against blood cancer cell line K562. Additionally, the KPE showed no significant adverse effect in hemolysis and no mutagenicity. It was suggested that KPE could be used as a functional biomaterial for functional food and cosmetics
