A study on the denitrification and microbial community characteristics by the change of C/N ratio of molasses and nitrate nitrogen (당밀과 질산성 질소의 C/N ratio 변화에 따른 탈질 및 미생물 군집 특성에 관한 연구)

  • To compare the denitrification efficiency, this study used molasses and methanol were used as external carbon sources. Specific experimental conditions were classified according to C/N ratio conditions. The batch test showed that the denitrification efficiency increased as C/N ratios of molasses and methanol rose. The most suitable C/N ratio of molasses turned out 4:1 considering the concentration of the residue chemical oxygen demand (COD) and the denitrification efficiency, which was 91.4%. Specific denitrification rate (SDNR) drawn as a kinetic factor demonstrated that molasses and methanol showed similar SDNR values as C/N ratios of molasses and methanol increased. Under the condition of C/N ratio 4:1, 0.0292 g $NO_3{^-}-N$ removal/g mixed liquor volatile suspended solid (MLVSS)/day (molasses), 0.0299 g $NO_3{^-}-N$ removal/g MLVSS/day (methanol) were found. Sludge adapted to molasses showed that Bacterium Pseudomonas sp. and Bergeylla sp. dominated through an analysis of microbial community. In addition, some bacteria were high convergences than the variety of microbial community. Accordingly, it was assumed that molasses focus on growing microorganisms specialized in denitrification and applied as a replaceable external carbon source that can enhance denitrification performance.

The Effect of Nutritional Balance between Carbon and other Nutrient Sources on the Growth of Sporobolomyces holsaticus (탄소원과 다른 영양원간의 영양균형이 Sporobolomyces holsaticus의 균체생육도에 미치는 영향)

  • Direct production of biomass from starch using amylolytic yeast, Sporobolomyces holsaticus FRI Y-5 was studied with varying the ratios between carbon and other nutrient sources in the medium. It was investigated under condition of constant C/P and C/S ratio to influence the initial concentration of starch $(S_o)$ and C/N ratio on its growth which is described as the specific growth rate $({\mu})$, cell yield (Y), the maximum concentration of cell $(X_m)$, and productivity (P). They were very dependent on both $S_o$ and C/N ratio. The form of the relationship between and ${\mu}$ and $S_o$ was observed to be similar to saturation kinetics at C/N = 100 but presented substrate inhibition at other C/N ratios. As $S_o$ was changed from 22.5 to 90 g/l, Y was observed to vary with C/N ratios but seemed to decrease as a wholes. $X_m$ was linearly related to $S_o$ at more than C/N = 50 but at less than C/N = 10 substrate inhibition was presented. P increased suddenly to $S_o$ = 45 g/l and then changed decreasingly at less than C/N = 50, but at more than C/N = 100 it changed increasingly. The effect of C/P ratio and C/S ratio on the yeast growth was also investigated at constant $S_o$ and C/N ratio. ${\mu}$ was dependent on C/P and C/S ratios, but Y, independent on them. But $X_m$ was reliant upon C/P ratio but not upon C/S ratio.

  • Pseudomonas oleovorans was cultivated to produce medium chain length polyhydroxyalkanoates (MCL-PHA) fram octanoic acid and ammonium nitrate as carbon and nitrogen source, respectively, by a pH-stat fed-batch culture technique. The octanoate concentration of the culture broth was maintained below 4 g/L by feeding the mixture of octanoic acid and ammonium nitrate when the culture pH rose above high limit. The effect of the ratio of octanoic acid to ammonium nitrate (C/N ratio) in the feed on the PHA production was examined. The final cell concentrations of 62.5, 54.7, and 9.5 g/L, PHA contents of 62.9, 75.1, and 67.6% of dry cell weight, and productivities of 1.03, 0.632, and 0.161 g/L/h were obtained when the C/N ratio in the feed were 10, 20, and 100 g octanoic acid/g ammonium nitrate, respectively.

Effect of C/N ratio on polyhydroxyalkanoates (PHA) accumulation by Cupriavidus necator and its implication on the use of rice straw hydrolysates

  • The effects of carbon-to-nitrogen (C/N) ratio in simulated rice straw hydrolysates using glucose and ammonium chloride on polyhydroxyalkanoates (PHA) accumulation by Cupriavidus necator was investigated. In general, PHA accumulation rate was higher under higher degrees of N-deficient conditions (e.g., C/N ratio of 360:1) than lower degrees of N-deficient conditions (e.g., C/N ratio of 3.6:1 and 36:1). Also, the most PHA accumulation was observed during the first 12 h after the PHA accumulation initiation. This study showed that the similar PHA accumulation could be achieved by using different accumulation periods depending on C/N ratios. N source presence was important for new cell production, supported by approximately ten times greater PHA accumulation under the N-deficient condition ($NH_4Cl$ 0.01 g/L) than the N-free (without $NH_4Cl$) condition after 96 h. C/N ratio of the rice straw hydrolysate was approximately 160:1, based on the glucose content, and this accumulated $0.36{\pm}0.0033g/L$ PHA with PHA content of $21{\pm}3.1%$ after 12 h. Since external C or N source addition for C/N ratio adjustment increases production cost, an appropriate accumulation period may be used for PHA accumulation from organic wastes, based on the PHA accumulation patterns observed at various C/N ratios and C and N concentrations.

Denitrifications of Swine Wastewater with Various Temperature and Initial CM Ratio in Anoxic Reactor (무산소조에서 온도 및 초기 C/N비에 따른 축산폐수의 탈질특성)

  • The biological denitrification batch tests were conducted to optimize the operating conditions with various temperature and initial SCO $D_{Cr}$ /N $O_3$-N ratio. and the denitrification rates were analyzed various SCO $D_{Cr}$ /N $o_3$-N ratio of influent with swine wastes fermented and temperature. The finishing time of denitrification was within 15 hours, 12 hours, and 6 hours as the temperature of denitrification applied were 15$\pm$1$^{\circ}C$, 25$\pm$1$^{\circ}C$, and 31$\pm$1$^{\circ}C$, respectively. From the batch tests, denitrification rate was operated with over 3 of SCO $D_{Cr}$ /N $O_3$-N ratio. Denitrification rate was increased as the temperature of denitrification, increased such as 2.40-3.90 mg N $O_3$-N/gMLVSSㆍhr, 6.10-7.60 mgN $O_3$-N/gMLVSSㆍhr, and 14.40-15.88 mgN $O_3$-N/gMLVSSㆍhr, respectively. The denitrification rate was increased as the ratio of initial SCO $D_{Cr}$ N $O_3$-N increased. However, it was found that the suitable ratio of SCO $D_{Cr}$ /N $O_3$-N for denitrification should be considered because the ratio of mg SCO $D_{Cr}$ , consumed per mg N $O_3$-N removed varied depend on the influent SCO $D_{Cr}$ /TKN ratios.

Influence of COD/Nitrate-N Ratio on Denitrification and Methanogenesis in Anaerobic Sludge (COD/Nitrate-N비가 혐기성 슬러지에서 탈질과 메탄화에 미치는 영향)

  • Simultaneous denitrification and methanogenesis at various influent COD/Nitrate-N(below C/N) ratio was investigated with two different anaerobic sludges of which one was inoculated with glucose only and the other was inoculated with glucose and nitrate. Three different C/N ratios of 30, 20, and 10 were applied with each anaerobic sludge in serum bottles agitated in a rotary shaker to monitor the time related changes of amount of gas production and gas composition. Glucose was used as a single carbon source, which was adjusted to 1,000 mg/L as COD and potassium nitrate was used as a nitrogen source to adjust the each C/N ratio. COD removal by methanogenesis was dominant at higher C/N ratios, while COD removal by denitrification was dominant at lower C/N ratios. With the sludges cultured at C/N ratios below 10, SMA and SDNR rising different carbon substrates were investigated. The most efficient carbon substrate was acetate and the highest SMA and SDNR were 0.76 g COD/g VSS day with pure anaerobic sludge and 1.38 g ${NO_3}^--N/g$ VSS day with sludge cultured at C/N ratio of 5, respectively.

Effect of Ratios Carbon Source to Nitrogen Source on the Yields of PHB Fermentation Variables (탄소원과 질소원의 비가 PHB 발효특성치 수율에 미치는 영향)

  • The effects of ratios of initial concentration of carbon source to the initial concentration of nitrogen source in the fermentation media on both the yields of PHB fermentation variables and the accumulation of poly-${\beta}$-hydroxybutyric acid(PHB) were investigated. The fermentation media were composed of the combination of varing glucose concentrations, 10, 20, 25, 30, 40, $50g/\ell$ and the NH4Cl concentrations 0.33, 0.4, 0.5, 1.5, 3, $5g/\ell$. The yield of biomass on glucos, Yx/s, decreased very slowly according to the increase of the ratio of C to N. And the yield became constant at 0.35(g biomass/g glucose) with the ratio higher than 70. The yield of residual biomass, Yx/s, also decreased with the ratio of C to N and finally showed a constant value of 0.065(g residual biomass/g glucose) when the ratio was higher than 65. In accordance with the augmentation of the ratio, the yield of PHB, YPHB/S, however, increased and showed the maximum value of 0.35 (g PHB/g glucose) between 40 and 60 of the ratio. The maximum yield of PHB to the change of biomass, YPHB/S, was 0.87(g PHB/g biomass), and the yie1d YPHB/RX, was 4.2(g PHB/g residual biomass). The maximum accumulation percent of PHB to the final biomass was 81% when the ratio was higher than 67.

A Study on Generalized Fibonacci Sequence (피보나치 수열의 일반화에 관한 고찰)

  • In this paper we investigate several properties and characteristics of the generalized Fibonacci sequence $\{g_n\}$={a, b, a+b, a+2b, 2a+3b, 3a+5b,...}. This concept is a generalization of the famous Fibonacci sequence. In particular we find the identities of sums and the nth term $g_n$ in detail. Also we find the generalizations of the Catalan's identity and A. Tagiuri's identity about the Fibonacci sequence, and investigate the relation between $g_n$ and Pascal's triangle, and how fast $g_n$ increases. Furthermore, we show that $g_n$ and $g_{n+1}$ are relatively prime if a b are relatively prime, and that the sequence $\{\frac{g_{n+1}}{g_n}\}$ of the ratios of consecutive terms converges to the golden ratio $\frac{1+\sqrt5}2$.

Effects on the Stability of Aerobic Granular Sludge (AGS) at Different Carbon/Nitrogen Ratio (C/N비 변화가 호기성 그래뉼 슬러지의 안정성에 미치는 영향)

  • In this study, the effect on the stability of Aerobic Granular Sludge (AGS) with different Carbon/Nitrogen (C/N) ratios was investigated. The C/N ratios were controlled to 10.0, 7.5, 5.0, and 2.5 using the sequencing batch reactor, and the results showed that the removal efficiency of organic matter and total nitrogen decreased simultaneously with the decrease of C/N ratio. The removal efficiency of organic matter and total nitrogen at C/N ratio of 2.5 was 70.7% and 52.3% respectively. In addition, the AGS/mixed liquor suspended solids (MLSS) ratio showed a tendency to decrease from 85.7% to 73.7%, while the sludge volume index showed a tendency to increase from 82 mL/g to 102 mL/g as the C/N ratio decreased. At the same time, the apparent deviation of polysaccharide (PS) content in extracellular polymeric substances was observed, and polysaccharides/protein (PS/PN) ratio decreased from 0.62 to 0.31 as the C/N ratio decreased. Optical microscope observations showed that the reduction in C/N ratio caused the growth of filamentous bacteria and significantly affected the stability of AGS.

Relationships of Midrib Ratio to Some Agronomic Characters in Burley Tobacco(N.tabacum L.) (버어리종 잎담배의 주맥비율과 형질 상호간의 관련성)

  • This study was conducted to find out the relationship among agronomic characteristics including midrib ratio in Burley tobacco(N.tabacum L.) Four cultrivars, Burley 21, KB108, KB111 and KB301, were grown at Suwon Experimental Station in 2001. Several agronomic characteristics of these cultivars were measured for the analysis of correlations among agronomic characteristics. Midrib ratio of KB301 was somewhat higher than those of the other cultivars, but there is no significant differences among cultivars at $5\%$ probability level. Among four stalk positions, midrib ratio of cutters was the highest. Midrib ratio was correlated positively with length of leaf at $1\%$ level and negatively with weight of lamina at $5\%$ level, respectively.
