In Vitro Antioxidant Activities and Antimicrobial Activity of Lotus (Leaf, Stem, and Seed Pod) Extracts (연잎, 연 줄기 및 연자방 추출물의 In Vitro 항산화 활성과 항균 활성에 관한 연구)

  • The purpose of this study was to investigate and analyze the total phenolic content (TPC), antioxidant activities (FRAP, ABTS, and DPPH), and antibacterial properties of lotus (Nelumbo nucifera) extracts. Lotus leaves, stems, and seed pods were extracted with deionized water at $95^{\circ}C$, and with 70.5% ethanol at $85^{\circ}C$. The TPC ranged from 8.12 to 215.12 GAE mg/g. The ethanol extract of the seed pod had the highest TPC, and the TPC of the corresponding deionized water extract was 161.45 mg/g. FRAP values ranged from 104.03 to $3,546.39TEAC\;{\mu}mol/g$, ABTS radical cation scavenging activities ranged from 105.11 to $3,956.94TEAC\;{\mu}mol/g$, and DPPH radical scavenging activities ranged from 37.29 to $2,549.46TEAC\;{\mu}mol/g$. $EC_{50}$ values ranged from 0.26 to 9.63 mg/mL, and 0.31 to 21.21 mg/mL for ABTS and DPPH, respectively. The ethanol and deionized water extracts of the seed pod showed higher TPC and stronger antioxidant properties (FRAP, ABTS, and DPPH) than those of characteristic of the leaf extracts. The ethanol and deionized water extracts of the seed pod showed antimicrobial activity against Bacillus subtilis, Staphylococcus aureus, and Pseudomonas aeruginosa with inhibition zones of 9.0 to 14.0 mm, and the ethanol extract of the leaf showed antimicrobial activity against B. subtilis and S. aureus with inhibition zones of 9.0 and 10.0 mm, respectively. Thus, the lotus seed pod could be used to produce novel teas, and could be a potential source of therapeutic ingredients for food and medicine.

Analysis and Enzymatic Production of Structured Lipids Containing DHA Using a Stirred-Batch Type Reactor (회분식 반응기를 이용한 DHA 함유 재구성지질의 효소적 합성 및 이화학적 특성 분석)

  • Structured lipid (SL) was synthesized by enzymatic interesterification with algae oil containing docosahexaenoic acid (DHA) and soybean oil in the stirred-batch type reactor. The reaction was performed for 15hr at $65^{\circ}C$ with 300 rpm catalyzed by sn-1,3 specific Lipozyme RM 1M lipase from Rhizomucor miehei ($11\%$ by weight of total substrates) in the absent organic solvent. SL contained $87.1\;area\%$ triacylglycerol (TAG), $12.1\;area\%$ diacylglycerol (DAG), $0.6\;area\%$ monoacylglycerol (MAG), and $0.2\;area\%$ free fatty acid (FFA). Major fatty acid profiles of SL were DHA $(15.7\;mol\%)$, linoleic $(31.1\;mol\%)$, palmitic $(20.2\;mol\%)$, oleic $(13.5\;mol\%)$ and eicosapentaenoic acid $(EPA,\;6.6 mol\%)$. SL contained the newly synthesized several peaks. Iodine and saponification of SL were 206.7 and 183.8. SL color showed darker and redder than soybean oil, and appeared the most yellowish color among SL, soybean, and algae oil.

DFT Studies on Two Novel Explosives Based on the Guanidine-Fused Bicyclic Structure

  • Density functional theory (DFT) calculations at the B3LYP/6-31G(d,p) theoretical level were performed for two novel explosives (compounds B and C) based on the guanidine-fused bicyclic skeleton $C_4N_6H_8$ (A). The heats of formation (HOFs) were calculated via isodesmic reaction. The detonation properties were evaluated by using the Kamlet-Jacobs equations. The bond dissociation energies (BDEs) for the thermolysis initiation bond were also analyzed to investigate the thermal stability. The results show that the compounds have high positive HOF values (B, 1064.68 $kJ{\cdot}mol^{-1}$; C, 724.02 $kJ{\cdot}mol^{-1}$), high detonation properties (${\rho}$, D and P values of 2.04 $g{\cdot}cm^{-3}$ and 2.21 $g{\cdot}cm^{-3}$, 9.98 $km{\cdot}s^{-1}$ and 10.99 $km{\cdot}s^{-1}$, 46.44 GPa and 59.91 Gpa, respectively) and meet the basic stability requirement. Additionally, feasible synthetic routes of the these high energy density compounds (HEDCs) were also proposed via retrosynthetic analysis.

The Improvement of Thermal Stability and Tensile Toughness by the Photocrosslinking of Poly(phenylene sulfide) containing Acetophenone (아세토페논을 함유한 Poly(phenylene sulfide)의 광가교에 의한 내열성과 인장인성 강화)

  • Poly(phenylene sulfide) films containing acetophenone as a photoinitiator were photocrosslinked under UV irradiation using a continuous UV irradiator. The gel fraction of the irradiated PPS in 1-Chloro naphthalene reached 94.7% with increasing UV energy and the photoinitiator concentration in the film upto $200J/cm^2$ and 12wt% respectively. Solid state $^1C$ NMR analysis suggested that the crosslinking occurred between the phenylene chains in PPS, indicating that the acetophenone may the phenylene hydrogens and subsequently adjacent polymer radicals could be recombined to form the crosslinked structure. The crosslinking improved the thermal behavior of PPS such as loss of $T_g$ and $T_c$, higher melting point and lower melting enthalphy as well as significantly higher peak pyrolysis temperature as much as $63.5^{\circ}C$. Surprisingly the tensile toughness of the most crosslinked PPS increased by 842%, resulting from the substantial enhancements in tensile modulus, strength and strain as much as 76%, 236% and 240% respectively. Also dynamic mechanical measurement indicated that the distance between crosslinks in the crosslinked PPS reached 85.3 g/mol corresponing to a crosslink density of 0.012 mol/g.

Synthesis and Characterization of Structured Lipids from Evening Primrose Seeds Oil and Rice Bran Oil (달맞이꽃 종자유와 미강유로부터 효소적 합성한 재구성 지질의 이화학적 특성 분석)

  • Structured lipids (SLs) were synthesized by enzymatic interesterification with evening primrose oil (EPO) and rice bran oil (RBO) in a batch-type reactor. The interesterification was performed using a water shaker for 24 hr at $55^{\circ}C$. Mixing speed was set at 200 rpm and Lipozyme RM IM (immobilized lipase from Rhizomucor miehei, 10% by weight of total substrates) was used as a biocatalyst. Rice bran oil and evening primrose oil were interesterified with various molar ratios (RBO : EPO, 1:3, 1:4, and 1:5 mol/mol). Reversed-phase high performance liquid chromatography connected with evaporative light-scattering detector was performed to separate the triacylglycerol (TAG) species of SLs. In the fatty acid analysis, $\gamma$-linolenic acid (7.9 mol%), linoleic acid (67.3 mol%) and oleic acid (13.2 mol%) were the most abundant fatty acids in the SLs. During 24 hr reaction, most of the reaction occurred within 3 hr. TAG compositions, tocopherols and phytosterols were also analyzed. In the TAG species analysis, LLL (ECN=42, L=linoleic acid) dramatically decreased when the reaction time increased.

Microstructure, Electrical Properties, and Stability of ZPCCE Based Varistors (ZPCCE계 바리스터의 미세구조와 전기적 성질 및 안정성)

  • The electrical procerties and stability of ZnO-Pr$_{6}$O$_{11}$-CoO-Cr$_2$O$_3$-Er$_2$O$_3$ (ZPCCE) based varistors were investigated in the Er$_2$O$_3$content range of 0.0 to 4.0 mol%. ZPCCE ceramics containing 2.0 mol% Er$_2$O$_3$ exhibited the highest density of 5.74 g/㎤ corresponding to 99.3% of theoretical density. The varistors with 0.5 mol% and 2.0 mol% Er$_2$O$_3$exhibited a relatively satisfying nonlinearity, which the nonlinear exponent is 40.50 and 47.15, respectively and the leakage current is 2.66 $mutextrm{A}$, respectively. Under more severe d.c. stress, such as (0.80 V$_{1mA}$/9$0^{\circ}C$/12h)+(0.85 V$_{1mA}$115$^{\circ}C$/12h)+(0.90 V$_{1mA}$12$0^{\circ}C$/12h)+(0.95 V$_{1mA}$1$25^{\circ}C$12h), they showed a very excellent stability, which the variation rate of the variator voltage is -0.89% and -0.15%, the variation rate of the nonlinear coefficient is -4.67% and -3.56%, and the variation rate of leakage current is -6.02% and -19.56%, respectively. It is surely bellived that ZnO-0.5 mol% Pr$_{6}$O$_{11}$-1.0 mol% CoO-0.5 mol% Cr$_2$O$_3$-x mol% Er$_2$O$_3$(x=0.5, 2.0) based varistors will be greatly contributed to develop the advanced Pr$_{6}$O$_{11}$-based ZnO varistors in future.uture. future.uture.

  • Effects Of organic compound, photosensitizer and $K^+$ ion influx. On the light-induced ATPase of mitochondria in L. edodes purified by linear sucrose density gradient centrifugation were studied. The mitochondrial ATPase activity was investigated by various wavelength illumination at dark state. The mitochondrial ATPase was activated 139% and 128% by 10m mol dithiothreitol and 0.1m mol quinacrine, respectively. This enzyme also was activated 36% by 0.1m mol phenazine methosulfate as photosensitizer. But, 100 mg oligomycin and 1m mol phlorizin inhibited activity of enzyme to 48% and 45%, respectively. Its optimum wavelength was 690 nm on the effect of $K^+$ ion influx, its optimum pH and temperature were found to be 7.2 and $55^{\circ}C$.

  • Two recombinant Escherichia coli strains, GCSC6576 harboring a plasmid pSYL107 containing the Ralstonia eutropha polyhyclroxyalkanoate (PHA) biosynthesis genes and a fadR atoC mutant LS5218 harboring a plasmid pJC4 containing the Alcaligenes latus PHA biosynthesis genes were compared for their ability to synthesize poly(3-hydroxybutyrate-co-3-hydroxyvalerate) [P(3HB-co-3HV)] from whey as the sale carbon source. With the pH-stat fed-batch culture of E. coli LS5218, 、 ,ve obtained a cell concentration, a P(3HB-co-3HV) concentration. a P(3HB-co-3HV) content, and a 3HV fraction of 31.8 g/L, 10.6 g/L, 33.4 wt%. and 6.26 mol%, respectively at 39 h.

  • Water-based magnetic fluids were made of the synthesized ultrafine magnetite with surfactants such as sodium oleate and loeic acid. The water-based magnetic fluid was prepared under the condition that added amounts of sodium oleate and oleic acid dissolved with NH4OH were 2.64$\times$10-2 mol, 1.5~2.0$\times$10-2 mol respectively. The water-based magnetic fluid was stable and should be dispersed well dispersion in the range of pH 9.0 to pH 11.0. As the solid content increased from 0.05 g/cc to 0.4 g/cc, the viscosity of water-based magnetic fluids increased from 2.5 cP to 152 cP and their magnetization at 5 KOe linearly increased from 3.1 emu/g to 26 emu/g. The aggregated powder after drying the water-based magnetic fluid was also successfully re-dispersed in dilute NH4OH solution and in kerosene.
