Expression of Human Cytochrome b5 in Zebrafish (Zebrafish에서 human cytochrome b5의 발현)

  • In this study, we sought to develop an effective cloning system by which human cytochrome $b_5$ (cyt $b_5$) is introduced and expressed in zebrafish. First, the 414 bp human cyt $b_5$ gene was amplified from RNA extracts of HeLa cells using RT-PCR, and the amplicon was subsequently sequenced to confirm that it was intact. Next, cyt $b_5$ was cloned into the pEGFP-N3 vector, which also encodes a fluorescent gene. One-cell stage zebrafish embryos were microinjected with the recombinant vector containing the cyt $b_5$ gene. Fluorescence microscopy confirmed high expression of the fluorescent gene in the injected fry compared to the non-fluorescent control fry. Finally, we extracted RNA from the injected fry and performed RT-PCR to determine whether the human cyt $b_5$ gene is expressed in the transgenic zebrafish. Sequencing analysis further confirmed that the cloned human cyt $b_5$ gene was intact. The transgenic zebrafish model produced in this study will be a useful tool to study therapeutic approaches to cure various diseases related to the deficiency of functional human cyt $b_5$ as well as tools for cloning useful genes in fish.

Evaluation of Physical Properties and Biocompatibility of HA-Dex Fusion Hydrogel Patch for Atopic Healing Ability (HA-Dex 융복합 하이드로겔 패치의 아토피 치유 능력에 대한 물리적 특성 및 생체 적합성 평가)

  • Recently, since atopic dermatitis is sensitive to skin irritation, it has been suggested that the development of a patch that can effectively exhibit adhesion and absorption to a specific local area while minimizing skin irritation, and capable of appropriate drug release should be given priority. In this study, we tried to develop a hydrogel patch that minimizes skin irritation, adheres effectively to a specific area, and promotes absorption. The atopic patch was formulated into a super-absorbent hydrogel sheet using a freeze drying method. Cell viability assay was carried out using keratinocytes (HaCaT cell) and fibroblasts (L929 cells). In order to investigate the physical properties, FT-IR, FE-SEM, porosity analysis and swelling behavior were investigated. As a result, the newly prepared HA-Dex hydrogel patch was verified by biocompatibility and physical evaluation. In addition, the manufactured hydrogel patch has sufficient moisture absorption capacity and can relieve itching of atopic skin, and is expected to be applied to various drug delivery products for the treatment of atopic dermatitis in the future.

  • Remarkable scientific and technological achievements are mainly shown in the 'super-convergence' or 'convergence of convergence' among cross- disciplinary fields, and advanced countries are promoting the 'high-risk, high-return research' ecosystem. Google LLC is carrying out numerous new challenges in terms of a non-failure perspective. Innovative research by the US Defense Advanced Research Projects Agency (DARPA) has produced such breakthroughs as the Internet, GPS, semiconductors, the computer mouse, autonomous vehicles, and drones. China is pioneering a 'Moon Village' and planning the world's largest nuclear fusion energy and ultra-large particle accelerator project. Japan has also launched 'the moonshot technology development research system' to promote disruptive innovation. In Korea, the government is preparing a new research program to tackle the global scientific challenges. Therefore, it is necessary to determine the reasonable geoscientific challenges to be addressed and to conduct a preliminary study on these topics. For this purpose, it is necessary to conduct long-term creative research projects centered on young researchers, select outstanding principal investigators, extract innovative topics without prior research or reference, simplify research proposal procedures, innovate the selection solely based on key ideas, and evaluate results by collective intelligence in the form of conferences.

Analysis of MODIS LAI and NDVI Patterns of Broad-leaved Trees by the Timesat Program on the Korean Peninsula (Timesat 프로그램에 의한 한반도 활엽수의 지역별 MODIS LAI 및 NDVI 패턴 분석)

  • This paper analyzed MODIS data from 2006 to 2013 to determine relationship between meteorological changes and vegetation index. The experimental area was divided into the northern, central and southern regions according to the regional characteristics, and the smoothed MODIS LAI and NDVI were obtained using Timesat. In the case of precipitation, MODIS NDVI had correlation coefficients of 0.66, 0.44 and 0.35 in the northern, central and southern regions and the correlation was the highest in the northern region. In the case of temperature, MODIS LAI had correlation coefficients of 0.66, 0.64 and 0.68, and MODIS NDVI had 0.89, 0.89 and 0.80. The correlation of MODIS NDVI was higher and showed similar positive correlation regardless of region. In addition, The accuracy between Timesat plant seasonal start and actual plant seasonal start in MODIS NDVI was higher than MODIS LAI. The average error in MODIS LAI was 19 days in the central region and 20 days in the southern region. And the average error in MODIS NDVI was 6 days in the central region and 8 days in the southern region.

  • This research was conducted to identify plant regulatory regions by gene tagging method. A promoterless GUS coding sequence was introduced to Populus nigra ${\times}$ P. maximowiczii via Agrobacterium strains(LBA4404/EHA101), and putative transgenic poplars were selected by culturing on medium containing G418($60mg/{\ell}$) and by GUS assay. Among them one positive plant was to amplify the native sequences flanking to the introduced GUS gene in plant genome by inverse PCR method and from this 730 by DNA product was obtained. After subcloning and sequencing, it has 88% homology to the Eucalyptus gunnii CAD(cinnamyl alcohol dehydrogenase) gene. The GUS gene fused with the putative promoter reinserted into poplar leaves by particle bombardment method to test the funtional promoter activity. Upon staining with X-gluc, many blue spots appeared on the leaf segments bombarded by the chimeric gene 2-3 days, thus the isolated DNA fragment contain some possible coding region as well as a putative regulatory sequences of poplar CAD gene.

Relative Navigation Study Using Multiple PSD Sensor and Beacon Module Based on Kalman Filter (복수 PSD와 비콘을 이용한 칼만필터 기반 상대항법에 대한 연구)

  • This paper proposes Kalman Filter-based relative navigation algorithms for proximity tasks such as rendezvous/docking/cluster-operation of spacecraft using PSD Sensors and Infrared Beacon Modules. Numerical simulations are performed for comparative analysis of the performance of each relative-navigation technique. Based on the operation principle and optical modeling of the PSD Sensor and the Infrared Beacon Module used in the relative navigation algorithm, a measurement model for the Kalman filter is constructed. The Extended Kalman Filter(EKF) and the Unscented Kalman Filter(UKF) are used as probabilistic relative navigation based on measurement fusion to utilize kinematics and dynamics information on translational and rotation motions of satellites. Relative position and relative attitude estimation performance of two filters is compared. Especially, through the simulation of various scenarios, performance changes are also investigated depending on the number of PSD Sensors and IR Beacons in target and chaser satellites.

A novel human KRAB-related zinc finger gene ZNF425 inhibits mitogen-activated protein kinase signaling pathway

  • Zinc finger (ZNF) proteins play a critical role in cell growth, proliferation, apoptosis, and intracellular signal transduction. In this paper, we cloned and characterized a novel human KRAB-related zinc finger gene, ZNF425, which encodes a protein of 752 amino acids. ZNF425 is strongly expressed in the three month old human embryos and then is almost undetectable in six month old embryos and in adult tissues. An EGFP-ZNF425 fusion protein can be found in both the nucleus and the cytoplasm. ZNF425 appears to act as a transcription repressor. Over-expression of ZNF425 inhibits the transcriptional activities of SRE, AP-1, and SRF. Deletion analysis indicates that the C2H2 domain is the main region responsible for the repression. Our results suggest that the ZNF425 gene is a new transcriptional inhibitor that functions in the MAPK signaling pathway.

Expression of Recombinant Human Epidermal Growth Factor as a Active Form through Codon Optimization with E. coli and Co-expression of Chaperone (코돈 최적화 및 샤페론 공발현을 통한 활성 형태의 재조합 인간 상피세포성장인자의 발현)

  • Epidermal growth factor (EGF) is a hormone protein that affects cell growth and proliferation, and has various medical applications. In the present study, the gene of human EGF was codon-optimized with E. coli and the expression vector was constructed by cloning into pRSET. In order to obtain the recombinant human EGF in an active form rather than an inclusion body, chaperone co-expression was attempted along with codon optimization, for the first time. The expressed human EGF was isolated in the pure form by performing Ion Exchange Chromatography in two consecutive runs. ELISA analysis showed that the activity of purified EGF was greater than 99%, which is similar to commercially available EGF. Cell proliferation test confirmed that the recombinant human EGF has the ability to promote cell proliferation of human skin fibroblasts. The human EGF expression system of this study gives a significant amount of protein, and does not require the renaturation step and the additional chromatographic system to remove a fusion contaminant, thereby providing a very useful alternative to conventional expression systems for the preparation of recombinant human EGF.

Laboratory Validation of Bridge Finite Model Updating Approach By Static Load Input/Deflection Output Measurements (정적하중입력/변위출력관계를 이용한 단경간 교량의 유한요소모델개선기법: 실내실험검증)

  • This paper presents a laboratory validation of a new approach for Finite Element Model Updating(FEMU) on short-span bridges by combining ambient vibration measurements with static load input-deflection output measurements. The conventional FEMU approach based on modal parameters requires the assumption on the system mass matrix for the eigen-value analysis. The proposed approach doesn't require the assumption and even provides a way to update the mass matrix. The proposed approach consists of two steps: 1) updating the stiffness matrix using the static input-deflection output measurements, and 2) updating the mass matrix using a few lower natural frequencies. For a validation of the proposed approach, Young's modulus of the laboratory model was updated by the proposed approach and compared with the value obtained from strain-stress tests in a Universal Testing Machine. Result of the conventional FEMU was also compared with the result of the proposed approach. It was found that proposed approach successfully estimated the Young's modulus and the mass density reasonably while the conventional FEMU showed a large error when used with higher-modes. In addition, the FE modeling error was discussed.

A Study on the Progression and Prevalence of Myopia according to Age for the Last Five Years : from 2008 to 2012 (최근 5년간 연령에 따른 근시 유병률 진행에 관한 연구 : 2008년에서 2012년 중심으로)

  • Purpose: In this study, we analyzed the progression and prevalence of myopia according to age for the last five years. Methods: We have done a comparative analysis of the progression and prevalence of myopia with the Korean National Health and Nutrition Examination Survey document from 2008 to 2012. Results: According to classification of myopia by age group for the last five years, the prevalence of low myopia was 25.5% for 5-11ages group, 25.1% for 12-18ages, 27.3% for 19-29ages, 30.7% for 30-39ages, 29.6% for 40-49ages, 19.2% for 50-59ages, 11.8% for 60-69ages, and 20.2% for over 70ages respectively. The prevalence of moderate myopia was 21.7% for 5-11ages group, 43.6% for 12-18ages, 36.2% for 19-29ages, 30.0% for 30-39ages, 20.4% for 40-49ages, 9.9% for 50-59ages, 5.2% for 60-69ages, and 7.6% for over 70ages respectively. The prevalence of high myopia was 2.1% for 5-11ages group, 11.7% for 12-18ages, 11.5% for 19-29ages, 6.9% for 30-39ages, 5.6% for 40-49ages, 1.9% for 50-59ages, 1.5% for 60-69ages, and 1.0% for over 70ages respectively. Conclusions: We must recognize an importance to the increase of the progression and prevalence of myopia, so it is necessary to provide a social interest in prevention of deteriorating vision and eye health welfare.
