A total of twenty-nine species and one species variety belonging to 12 genera was isolated from 30 samples of molasses on 1% glucose(10 genera, 22 species and 1 variety) and 50% sucrose(7, 21 and 1) Czapek's agar at $25^{\circ}C$ media. Aspergillus, Mucor, Mycosphaerella and Penicillium were the most common genera on the two types of media. From the above genera, the most prevalent species were: Aspergillus flavus, A. fumigatus, A. niger, Mycosphaerella tassiana, Penicillium chrysogenum, P. oxalicum and P. purpurogenum. Also, some species were only isolated on 50% sucrose such as Eurotium amstelodami, E. chevalieri, E. repens, Humicola fuscoatra, Penicillium aurantiogriseum and P. puberulum. About 65 fungal isolates isolated from 50% sucrose agar were tested for their ability to produce invertase enzyme in liquid medium and 93.8% of the isolates could produce this enzyme. From the positive isolates, 32 showed high invertase activity, 21 had moderate activity and the remaining 8 isolates were of weak activity. Sixty isolates of Aspergillus, Emericella, Eurotium, Mycosphaerella and Penicillium from the preceding study were screened for the presence of their respective mycotoxins. Larva of brine shrimp(Artema sauna L.) were used for toxicity test of the fungal crude extracts. Three isolates out of 60 tested were toxic. Using thin-layer chromatographic technique, 5 different known mycotoxin were detected aflatoxins : B1, B2, G1, G2 and citrinin.
Twelve fungal isolates out of 123 isolates obtained from cysts and soils of potato cyst nematode (PCN)-infested fields in Central Java, Indonesia had parasitic abilities of over 50% on PCN eggs or females (cysts) in vitro pathogenicity tests. Cultural and morphological characters and DNA sequences of ribosomal genes in ITS region revealed that they were four isolates of Gliocladium (Trichoderma) virens, three isolates of Fusarium oxysporum, one of F. lateritium, one of Penicillium tritinum and two of Taralomyces spp. A hundred percent infections occurred in eggs or cysts by three fungal isolates G. virens, F. oxysporum and P. oxalicum, suggesting that these fungi may have a good potential for the PCN biocontrol. Especially, G. virens isolates, which occurred most frequently in the PCN-infested potato fields and are known to be highly adaptable to varying habitats, may be developed as reliable agents for controlling PCN with both egg- and cyst-parasitic capabilities and with high ecological adaptabilities.
Background: Endophytic fungi play an important role in balancing the ecosystem and boosting host growth. In the present study, we investigated the endophytic fungal diversity of healthy Panax notoginseng and evaluated its potential antimicrobial activity against five major phytopathogens causing root-rot of P. notoginseng. Methods: A culture-dependent technique, combining morphological and molecular methods, was used to analyze endophytic fungal diversity. A double-layer agar technique was used to challenge the phytopathogens of P. notoginseng. Results: A total of 89 fungi were obtained from the roots, stems, leaves, and seeds of P. notoginseng, and 41 isolates representing different morphotypes were selected for taxonomic characterization. The fungal isolates belonged to Ascomycota (96.6%) and Zygomycota (3.4%). All isolates were classified to 23 genera and an unknown taxon belonging to Sordariomycetes. The number of isolates obtained from different tissues ranged from 12 to 42 for leaves and roots, respectively. The selected endophytic fungal isolates were challenged by the root-rot pathogens Alternaria panax, Fusarium oxysporum, Fusarium solani, Phoma herbarum, and Mycocentrospora acerina. Twenty-six of the 41 isolates (63.4%) exhibited activity against at least one of the pathogens tested. Conclusion: Our results suggested that P. notoginseng harbors diversified endophytic fungi that would provide a basis for the identification of new bioactive compounds, and for effective biocontrol of notoginseng root rot.

Decay fungi can decompose plant debris to recycle carbon in the ecosystem. Still, they can also be fungal pathogens, which can damage living trees and/or wood material and cause a large amount of timber loss. We isolated and identified basidiomycetous fungi from the decayed bark of Mongolian oak wrapped with sticky roll traps. The degrading enzyme activities were then tested for all fungal isolates. The decay ability of selected isolates was assessed based on the weight loss of wood discs after inoculating with culture suspension of decay fungi under the different humidity levels. A total of 46 basidiomycetous fungal isolates belonged to 12 species, and 10 genera were obtained from Jong Myo (16 isolates), Chang Kyung palace (7 isolates), Cheong Gye (10 isolates), and Gun Po (13 isolates). Gymnopus luxurians was the most dominant fungus in the present study, and this species distributed in all survey sites with 9 isolates in Jong Myo, followed by 3 isolates in Chang Kyung palace, while Cheong Gye and Gun Po had only 1 isolate each. Among 46 isolates, 44 isolates secreted at least one enzyme, while 25 isolates produced both cellulase and phenol oxidase enzymes, and 2 isolates produced neither. The assessment of decay ability by artificial inoculation indicated that the weight loss of wood discs was significantly influenced by humidity conditions when inoculated with bark decay fungi. The percent weight losses by G. luxurians inoculation in RH of 90-100% and RH of 65-75% were 4.61% and 2.45%, respectively. The weight loss caused by Abortiporus biennis were 6.67% and 0.46% in RH of 90-100% and RH of 45-55%, respectively. The humidity reduction approach should be applied for further studies to control the growth and spread of bark decay fungi on the trunks wrapped with sticky roll traps.
Very few studies have addressed the phylogenetic diversity of fungi from Northeast India under the Eastern Himalayan range. In the present study, an attempt has been made to study the phylogenetic diversity of culturable soil fungi along the altitudinal gradients of eastern Himalayas. Soil samples from 24 m above sea level to 2,000 m above sea level altitudes of North-East India were collected to investigate soil micro-fungal community structure and diversity. Molecular characterization of the isolates was done by PCR amplification of 18S rDNA using universal primers. Phylogenetic analysis using BLAST revealed variation in the distribution and richness of different fungal biodiversity over a wide range of altitudes. A total of 107 isolates were characterized belonging to the phyla Ascomycota and Zygomycota, corresponding to seven orders (Eurotiales, Hypocreales, Calosphaeriales, Capnodiales, Pleosporales, Mucorales, and Mortierellales) and Incertae sedis. The characterized isolates were analysed for richness, evenness and diversity indices. Fungal diversity had significant correlation with soil physico-chemical parameters and the altitude. Eurotiales and Hypocreales were most diverse and abundant group of fungi along the entire altitudinal stretch. Species of Penicillium (D=1.44) and Aspergillus (D=1.288) were found to have highest diversity index followed by Talaromyces (D=1.26) and Fusarium (D=1.26). Fungal distribution showed negative correlation with altitude and soil moisture content. Soil temperature, pH, humidity and ambient temperature showed positive correlation with fungal distribution.
Genetic variation among the base isolates and monosporidial strains derived from these isolates of Tilletia indica- the causal agent of Karnal bunt (KB) in wheat, was analyzed by morphological, growth behaviors and RAPD-ISSR based molecular polymorphism. Genetic make up of fungal cultures vary among each other. The magnitude of variation in KBPN group is less (narrow genetic base) when compared to the other groups KB3, KB9 and JK (broad genetic base) reflecting that variability is a genetically governed process. The generation of new variation with different growth characteristics is not a generalized feature and is totally dependant on the original genetic make-up of the base isolate generating new monosporidial strains. Thus, it can be concluded that monosporidial strains derived from mono-teliosporic isolate, consists of genetically heterogeneous population. The morphological and genetic variability further suggests that the variation in T. indica strains is predominantly derived through the genetic rearrangements through para sexual means.

The Korean sawmills have recently recognized the importance of prevention of fungal discoloration due to increased losses in revenue. Before establishing integrated control strategies of fungal discoloration, more complete knowledge about causal organisms is needed. As a first step, we initiated a through survey of fungi colonizing commercially important softwood(Pinus dens flora, Pinus koraiensis, and Pinus radiata) logs and lumber in Korea. In this paper we report results obtained from Japanese red pine(Pinus densiflora) log study. In summer 2000, fungi were isolated from Japanese red pine logs in storage, and identified based on their cultural and morphological characteristics. A total of 595 fungi were isolated, representing 21 genera and 30 species. Mold fungi, mostly Trichoderma species, were the most frequently isolating fungi, representing more than half of all isolates. Dematiaceous fungi represented approximately one fifth of the isolates, and Rhinocladiella atorvirens was the most abundant in all samples. Opiostoma species represented 7% of all isolates from cores planted on malt extract agar(MEA) and the incidence of these species doubled with the addition of streptomycin and cycloheximide to MEA. The results indicate that Japanese red pine sapwood is susceptible to colonization by a variety of fungal species. As a result, control strategies that concentrate on one fungus may have limited success because of interference from competing flora.

Three new records of Ascomycota species (Chaetomium acropullum, Phialemonium globosum, Phialemonium atrogriseum) from field soils in Korea are presented in this study. These newly discovered fungal isolates were isolated from field soils from various places across Gyeongnam, Korea in 2016. All the isolates were identified and described based on morphological characteristics, and rDNA internal transcribed spacer and ${\beta}$-tubulin gene sequence data. Morphological features of these fungal species were studied on different agar media: potato dextrose agar, oatmeal agar, malt extract agar, Czapek yeast extract agar, and yeast extract sucrose agar. Full description and illustrations of their morphological characters are provided. These fungal species have not officially been previously reported in Korea.
Boxwood is a representative ornamental shrub that is widely used in landscaping horticulture. After pruning, damaged leaves or stems of boxwoods are unavoidably vulnerable to infection by various plant pathogens. Several boxwood diseases caused by fungi, such as Volutella blight and Macrophoma leaf spot, have been reported worldwide including Republic of Korea. In this study, we isolated and identified fungal pathogens of boxwood diseases that occurred in Korea and characterized their morphological and taxonomic characteristics. Boxwood samples showing blight symptoms were collected in Seoul, Republic of Korea, and the putative fungal pathogens Pseudonectria buxi, P. foliicola, and Neofusicoccum buxi were successfully identified. Investigation of the morphological features of the field isolates, including mycelial growth and conidial morphology, and phylogenetic analysis of multiple DNA barcode loci revealed that there were some morphological and genetic variations among isolates, but all of the analyzed isolates were closely related to the corresponding reference strains. We also found that P. foliicola strains were more virulent than P. buxi, and the N. buxi strains isolated in this study were weak pathogens or saprophytes. The results of our study will contribute to the development of control strategies for boxwood diseases caused by fungi and accelerate research on the complex ecology of boxwood diseases.
The endophytic fungi were isolated from red pine trees (Pinus densiflora) that were distributed in three sites of Chungbuk, Korea. Twenty fungal isolates were isolated from 16 trees and divided into 8 groups by morphological characters. The fungal isolates were identified using the sequences of ITS region of rDNA; Lophodermium complex, Sydowia polyspora, Hymenula sp., Sistotrema brinkmannii, Septoria pini-thunbergii, Earliella sp. Lophodermium spp. were the most frequently found fungal species the across sites and firstly detected from Pinus species in Korea by molecular work.
