5-Hydroxytryptamine 6 Receptor (5-HT6R)-Mediated Morphological Changes via RhoA-Dependent Pathways

  • The $5-HT_6R$ has been considered as an attractive therapeutic target in the brain due to its exclusive expression in the brain. However, the mechanistic linkage between $5-HT_6Rs$ and brain functions remains poorly understood. Here, we examined the effects of $5-HT_6R$-mediated cell morphological changes using immunocytochemistry, Western blot, and live-cell imaging assays. Our results showed that the activation of $5-HT_6Rs$ caused morphological changes and increased cell surface area in HEK293 cells expressing $5-HT_6Rs$. Treatment with 5-HT specifically increased RhoA-GTP activity without affecting other Rho family proteins, such as Rac1 and Cdc42. Furthermore, live-cell imaging in hippocampal neurons revealed that activation of $5-HT_6Rs$ using a selective agonist, ST1936, increased the density and size of dendritic protrusions along with the activation of RhoA-GTP activity and that both effects were blocked by pretreatment with a selective $5-HT_6R$ antagonist, SB258585. Taken together, our results show that $5-HT_6R$ plays an important role in the regulation of cell morphology via a RhoA-dependent pathway in mammalian cell lines and primary neurons.

산화 아연에서의 질소 용해도에 대한 알루미늄의 효과 : 밀도 범함수 이론 (Effect of Aluminum on Nitrogen Solubility in Zinc Oxide: Density Functional Theory)

  • Zinc oxide as an optoelectronic device material was studied to utilize its wide band gap of 3.37 eV and high exciton biding energy of 60 meV. Using anti-site nitrogen to generate p-type zinc oxide has shown a deep acceptor level and low solubility. To increase the nitrogen solubility in zinc oxide, group 13 elements (aluminum, gallium, and indium) was co-added to nitrogen. The effect of aluminum on nitrogen solubility in a $3{\times}3{\times}2$ zinc oxide super cell containing 72 atoms was investigated using density functional theory with hybrid functionals of Heyd, Scuseria, and Ernzerhof (HSE). Aluminum and nitrogen were substituted for zinc and oxygen sites in the super cell, respectively. The band gap of the undoped super cell was calculated to be 3.36 eV from the density of states, and was in good agreement with the experimentally obtained value. Formation energies of a nitrogen molecule and nitric oxide in the zinc oxide super cell in zinc-rich conditions were lower than those in oxygen-rich conditions. When the number of nitrogen molecules near the aluminum increased from one to four in the super cell, their formation energies decreased to approach the valence band maximum to some degree. However, the acceptor level of nitrogen in zinc oxide with the co-incorporation of aluminum was still deep.

  • Radiation technology (RT) has long been applied in various fields for increasing the safety and shelf-life of foods by controlling pathogen-induced poisoning. RT was introduced for the first time in Korea in the 1950s to eliminate harmful microorganisms in food materials. In the 1980s, RT had been scientifically proven to be effective for the sterilization of food and public health products. In recent years, irradiation with gamma rays has also been used for improving physiological properties through the structural modification of natural molecules, which has been proposed to be applicable to various industries. In particular, radiation transformation technology (RTT), which involves the development of new functional compounds through the molecular conversion of natural biomaterials, is becoming a new high-value technology as a fusion technique of RT and biotechnology. The present reports have suggested that RTT can be an effective tool for the development of new functional compounds and improvement of the physiological activity of biomolecules.

리튬이차전지 양극 분말 소재 위 탄소나노튜브의 직접 성장 거동 고찰 (Investigation of direct growth behavior of carbon nanotubes on cathode powder materials in lithium-ion batteries)

  • This study reports a direct growth of carbon nanotubes (CNTs) on the surface of LiCoO2 (LCO) powders to apply as highly efficient cathode materials in lithium-ion batteries (LIB). The CNT synthesis was performed using a thermal chemical vapor deposition apparatus with temperatures from 575 to 625 ℃. Ferritin molecules as growth catalyst of CNTs were mixed in deionized (DI) water with various concentrations from 0.05 to 1.0 mg/mL. Then, the LCO powders was dissolved in the ferritin solution at a ratio of 1g/mL. To obtain catalytic iron nanoparticles on the LCO surface, the LCO-ferritin suspension was dropped in silicon dioxide substrates and calcined under air at 550℃. Subsequently, the direct growth of CNTs on LCO powders was performed using a mixture of acetylene (10 sccm) and hydrogen (100 sccm) for 10 min. The growth behavior was characterized by scanning and transmission electron microscopy, Raman scattering spectroscopy, X-ray diffraction, and thermogravimetric analysis. The optimized condition yielding high structural quality and amount of CNTs was 600 ℃ and 0.5 mg/mL. The obtained materials will be developed as cathode materials in LIB.

Identification of Amino-Acids Residues for Key Role in Dextransucrase Activity of Leuconostoc mesenteroides B-742CB

  • Dextransucrase (DSRB742) from Leuconostoc mesenteroides NRRL B-742CB is a glucosyltransferase that catalyzes the synthesis of dextran using sucrose, or the synthesis of oligosaccharides when acceptor molecules, like maltose, are present. The DSRB742 gene (dsrB742) was cloned and the properties were characterized. In order to identify critical amino acid residues, the DSRB742 amino acid sequence was aligned with glucosyltransferase sequences, and three amino acid residues reported as sucrose binding amino acids in Streptococcus glucosyltransferases were selected for site-directed mutagenesis experiments. Asp-533, Asp-536, and His-643 were independently replaced with Ala or Asn. D533A and D536A dextransucrases showed reduced dextran synthesis activities, 2.3% and 40.8% of DSRB742 dextransucrase, respectively, and D533N, D536N, H643A, end H643N dextransucrases showed complete suppression of dextran synthesis activities altogether. Additionally, D536N dextransucrase showed complete suppression of oligosaccharide synthesis activities. However, modifications at Asp-533 or at His-643 retained acceptor reaction activities in the range of 8.4% to 21.3% of DSRB742 acceptor reaction activity. Thus at least two carboxyl groups of Asp-533 and Asp-536, and His-643 as a proton donor, are essential for the catalysis process.

Dynamic Transcriptome, DNA Methylome, and DNA Hydroxymethylome Networks During T-Cell Lineage Commitment

  • The stepwise development of T cells from a multipotent precursor is guided by diverse mechanisms, including interactions among lineage-specific transcription factors (TFs) and epigenetic changes, such as DNA methylation and hydroxymethylation, which play crucial roles in mammalian development and lineage commitment. To elucidate the transcriptional networks and epigenetic mechanisms underlying T-cell lineage commitment, we investigated genome-wide changes in gene expression, DNA methylation and hydroxymethylation among populations representing five successive stages of T-cell development (DN3, DN4, DP, $CD4^+$, and $CD8^+$) by performing RNA-seq, MBD-seq and hMeDIP-seq, respectively. The most significant changes in the transcriptomes and epigenomes occurred during the DN4 to DP transition. During the DP stage, many genes involved in chromatin modification were up-regulated and exhibited dramatic changes in DNA hydroxymethylation. We also observed 436 alternative splicing events, and approximately 57% (252) of these events occurred during the DP stage. Many stage-specific, differentially methylated regions were observed near the stage-specific, differentially expressed genes. The dynamic changes in DNA methylation and hydroxymethylation were associated with the recruitment of stage-specific TFs. We elucidated interactive networks comprising TFs, chromatin modifiers, and DNA methylation and hope that this study provides a framework for the understanding of the molecular networks underlying T-cell lineage commitment.

Enhanced γ-aminobutyric acid and sialic acid in fermented deer antler velvet and immune promoting effects

  • Deer antler velvet is widely used in traditional medicine for its anti-aging, antioxidant, and immunity-enhancing effects. However, few studies have reported on the discovery of probiotic strains for deer antler fermentation to increase functional ingredient absorption. This study evaluated the ability of probiotic lactic acid bacteria to enhance the concentrations of bioactive molecules (e.g., sialic acid and gamma-aminobutyric acid [GABA]) in extracts of deer antler velvet. Seventeen strains of Lactobacillus spp. that were isolated from kimchi and infant feces, including L. sakei, L. rhamnosus, L. brevis, and L. plantarum, and those that improved the life span of Caenorhabditis elegans were selected for evaluation. Of the 17 strains, 2 (L. rhamnosus LFR20-004 and L. sakei LFR20-007) were selected based on data showing that these strains increased both the sialic acid and GABA contents of deer antler extract after fermentation for 2 d and significantly improved the life span of C. elegans. Co-fermentation with both strains further increased the concentrations of sialic acid, GABA, and metabolites such as short-chain fatty acids and amino acids. We evaluated the biological effects of the fermented antler velvet (FAV) on the antibacterial immune response in C. elegans by assessing worm survival after pathogen infection. The survival of the C. elegans conditioned with FAV for 24h was significantly higher compared with that of the control worm group fed only normal feed (non-pathogenic E. coli OP50) exposed to E. coli O157:H7, Salmonella typhi, and Listeria monocytogenes. To evaluate the protective effects of FAV on immune response, cyclophosphamide (Cy), an immune-suppressing agent was treated to in vitro and in vivo. We found that FAV significantly restored viability of mice splenocytes and immune promoting-related cytokines (interleukin [IL]-6, IL-10, inducible nitric oxide synthase [iNOS], interferon [IFN]-γ, and tumor necrosis factor [TNF]-α) were activated compared to non-fermented deer antlers. This finding indicated the protective effect of FAV against Cy-induced cell death and immunosuppressed mice. Taken together, our study suggests that immune-promoting antler velvet can be produced through fermentation using L. rhamnosus LFR20-004 and L. sakei LFR20-007.

종양괴사인자에 의하여 유도된 혈관내피세포의 Cell Adhesion Molecules 발현을 억제시키는 플라보노이드 선별 (Selection of Flavonoids Inhibiting Expression of Cell Adhesion Molecules Induced by Tumor Necrosis Factor- a in Human Vascular Endothelial Cells)

  • 염증성 cytokines의 분비 또는 혈관손상으로 인한 백혈구의 adhesion과 transmigration을 통하여 죽상경화과정이 시발되는데, 본 연구에서는 이러한 죽상경화의 초기과정에서 플라보노이드가 억제작용을 발휘하는 지를 규명하고자 하였다. 본 연구에서는 화학적인 구조가 서로 다른 플라보노이드를 사용하여 화학적인 구조와 항동맥경화작용과의 상관성을 착인하였다. TNF-$\alpha$는 혈관내피세포를 활성화시켜 THP-1 단핵구의 adhesion을 유의적으로 증가시켰다. 여러형태의 플라보노이드를 전처리하고 TNF-$\alpha$를 가하여 혈관내피세포를 활성화 시 켰을 때, flavonols인 quercetin과 flavones의 luteolin과 apigenin은 THP-1 단핵구의 adhesion억제효과를 보여주었다. 그러나, catechins과 flavanones의 플라보노이드는 이러한 억제효과를 전혀 보여주지 못하였다. 이러한 adhesion 억제작용을 가지는 플라보노이드는 CAMs 단백질의 발현도 차단시킨다는 것을 확인할 수 있었다. Quercetin, luteolin과 apigenin은 TNF-$\alpha$에 의하여 증가된 VCAM-1, ICAM-1 및 E-selectin의 단백질 발현을 일률적으로 감소 또는 차단시켰다. 그 대신, 단핵구의 adhesion을 차단시키지 못한 (-)epigallo-catechin gallate와 (+)catechin은 TNF-$\alpha$에 의한 이러한 CAMs의 발현을 전혀 억제시키지 못하였다. 또한 quercetin, luteolin과 apigenin의 CAMs단백질 발현 억제작용은 유전자 전사단계에서 mRNA의 down-regulation으로 인하여 나타난다는 사실을 알 수 있었다. 결론적으로 quercetin, luteolin, apigenin과 같은 플라보노이드는 TNF-$\alpha$와 같은 염증성 cytokines에 의한 단핵구의 adhesion을 혈관내피세포의 CAMs 단백질 발현을 억제하므로서 차단시킨다는 것이 확인되었다. 여기서 모든 플라보노이드가 이러한 활성을 다 지니고 있지 않아서 화학적인 구조와 초기 항동맥경화작용에는 서로 연관성이 있다는 것이 제시되었다. 또한, 선별된 플라보노이드의 초기 항동맥경화작용은 활성산소를 소거하는 플라보노이드의 항산화능과는 무관한 것 같다고 할 수 있다.

  • Therapeutic antibodies represent one of the fastest growing areas of the pharmaceutical industry. There are currently 19 monoclonal antibodies in the market that have been approved by the FDA and over 150 in clinical developments. Driven by innovation and technological developments, therapeutic antibodies are the second largest biopharmaceutical product category after vaccines. Antibodies have been engineered by a variety of methods to suit a particular therapeutic use. This review describes the structural and functional characteristics of antibody and the antibody engineering for the generation and optimization of therapeutic antibodies.

아조벤젠기를 갖는 장쇄지방산LB막의 광이성화 특성 (Properties of Photoisomerization of Organic-Ultrathin Films)

  • The absorption spectra of synthesis of azobenzene containing long chain fatty acids were investigated by UV spectrophotometer in chloroform solution. Also, the pressure-area isotherms of the molecules on the water-air interface were obtained and the LB films were fabricated onto a quartz slides and quartz crystals by the conventional Langmuir-Blodggett(LB) method. The UV absorption spectra of Langmuir-Blodggett(LB) films on quartz slides have been measured. From these measurements, following conclusions were obtained. Azobenzene containing long chain fatty acids show a photoisomerization by irradiation of UV light and visible light alternatively. At the pressure-area isotherms, the value of surface pressure increment were decreased when the number of $C_n$ increased. Also the LB films show a photoisomerization characteristics. So the LB film of azobenzene containing long chain fatty acid has possibility to being applied to functional molecular device such as photomemory and light switching.
