건강기능식품 기능성원료로서 창녕양파추출액의 지표성분 Quercetin 분석법 (Analytical Method Validation of Quercetin in Changnyeong Onion Extract as a Functional Ingredient for Functional Health Food)

  • 개별인정형 건강기능식품 기능성 원료로 개발한 창녕양파추출액의 표준화를 위해 지표성분으로 quercetin을 설정하였으며, HPLC를 이용하여 지표성분 quercetin의 분석법을 확립하며 그에 따른 유효성 검정을 실시하고자 하였다. 유효성 검정 결과, 본 시험법에서 표준용액의 피크유지시간과 창녕양파추출액의 피크유지시간이 일치하였으며 동일한 spectrum을 나타내어 특이성을 확인하였다. 검량선의 상관계수($R^2$)는 0.9986로 높은 직선성을 보여 분석에 적합함을 알 수 있었으며, 검출한계는 0.2 mg/L, 정량한계는 0.5 mg/L 로 설정되었다. Quercetin의 회수율은 0.05 mg/mL에서는 82.36~95.26%, 0.075 mg/mL는 82.70~98.24%, 0.1 mg/mL은 87.91~95.11%의 범위의 회수율을 보였으며, intra-day에서의 정밀도(RSD)는 0.10~3.28%, inter-day에서는 0.96~5.79%의 정밀도를 나타내어 창녕양파추출액의 지표성분인 quercetin의 분석법은 적합한 시험법임이 검증되었다.

노니 지표성분 6종과 발효노니의 면역활성 증진 효과 (Enhancement of Immune Activities of Fermented Morinda citrifolia L. (Noni) and Six Marker Compounds)

  • 본 연구에서는 발효노니를 건강기능식품 소재로 활용 시 기초자료로 제공하고자 발효노니 추출물 및 지표성분의 면역활성 증진 효과를 평가하였다. RAW 264.7 대식세포에서 발효노니 추출물 및 지표성분 6종을 처리하여 XTT 세포독성평가, Nitric Oxide 생성 측정, Cyokine 생성 측정, immune marker genes 발현분석 수행하였다. 뿐만 아니라 양성대조군으로 LPS와 기능성 원료로 사용되고 있는 발효홍삼 추출물을 사용하였다. 그 결과 모든 처리 농도 및 처리군에서 세포독성이 관찰되지 않았으며, 지표성분 6종 중 SCP 및 ASE에서 NO 생성이 증가됨을 확인하였다. 뿐만 아니라 ASE 처리군에서는 IL-6 및 IL-1β의 생성이 증가되었으며, iNOS 및 TNF-α의 immune marker genes 발현이 증가됨을 확인하였다. 발효노니 추출물 효능 평가에서는 발효시 NO 생성 및 IL-6, IL-1β의 생성, COX2의 발현이 증가되는 것으로 나타났다. 이러한 연구 결과는 발효노니 추출물 및 지표성분의 선천면역 활성증가를 타나내며 노니 및 발효노니 표준화 연구에서 지표성분으로 사용 가능성을 제시한다. 따라서 발효노니는 면역증진 활성을 갖는 제품 개발에 있어서 유용한 기능성 식품소재로써 사용될 수 있으며, 우수한 효능을 나타내는 지표성분은 유용성분으로 이용이 가능할 것으로 사료된다.

증숙처리에 따른 재래생강 및 중국종자생강의 항산화 및 기능성물질 비교 (Comparison of Antioxidant and Functional Compounds in Korean Conventional and Chinese Seed Ginger (Zingiber officinale Roscoe) Following Steam Treatment)

  • 본 연구에서는 재래생강과 중국종자생강 간의 증숙 시간별 총 페놀화합물, 총 플라보노이드, ABTS 및 DPPH 라디칼 소거능, gingerol, shogaol 함량을 비교 분석하였다. 총 플라보노이드와 총 페놀 화합물, ABTS radical 소거능의 경우 대체적으로 증숙 6시간에서 가장 높은 함량을 나타내다가 그 이상으로 증숙할수록 오히려 감소하는 경향을 보였으며, 생강 중에서는 재래생강인 WO-2가 가장 높게 나타났다. DPPH 라디칼 소거능은 증숙 시간별로 유의미한 차이가 없었다. 생강의 gingerol 함량은 대조군에서 WO-2가 1,351 mg/kg으로 가장 높은 함량을 나타냈다. 증숙 시간에 비례하여 gingerol의 함량은 감소하고, shogaol의 함량은 증가하였는데 증숙 8시간에서 WO-2가 생강 중에서도 가장 높았으며, 또한 종자별로도 재래생강이 중국 종자생강보다 shogaol 함량이 높은 것으로 나타났다. 본 연구결과 재래생강이 중국종자생강보다 기능성 함량이 높음을 확인하였으며 증숙 처리된 재래생강을 활용하여 다양한 식품을 개발할 수 있을 것으로 판단된다.

쥐눈이콩 첨가량을 달리한 초콜릿 제조 조건의 최적화 (Optimization of the Addition of Jinuni Beans to Chocolate Using the Response Surface Methodology)

  • The purpose of this study was to determine the optimal mixing conditions of different amounts of Jinuni beans and fresh cream for the preparation of Jinuni bean chocolate. An experiment was designed using the central composite design of the response surface, which required 10 experimental treatments including 2 replicates for the Jinuni bean and fresh cream. The compositional and functional properties of the treatments were measured and these values were applied to the mathematical model. A canonical form and perturbation plot showed the influence of each ingredient on the mixture products. The color value L (Lightness) decreased with an increase in the amount of added Jinuni bean but the color value a (redness) and color value b (yellowness) did not change. In addition, the hardness value increased with the addition of the Jinuni bean. The results of the sensory evaluation showed very significant differences in the values of the texture (p<0.01), taste (p<0.05), and overall quality (p<0.01). As a result, the optimum formulations as determined by the numerical and graphical methods were 27.98 g of Jinuni bean and 67.0 g of fresh cream.

Antioxidant activity of silkworm powder treated with protease

  • The antioxidant activity of silkworm powder treated by proteolytic enzyme was investigated. Total protein content of silkworm power was assayed using BCA, Bradford assays and SDS-polyacrylamide gel electrophoresis (PAGE) with alkaline protease treatment conditions including temperature and pH. The optimum condition of alkaline protease treatment for silkworm powder was found to be $60^{\circ}C$ and pH 7. The alkaline protease treatment resulted in increased contents of free amino acids, total polyphenol and total flavonoid compared to control group. The silkworm hydrolysates showed excellent antioxidant activities in various in vitro models such as 2,2 diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2 - azino-bis(3-ethylbenzthiazoline-6)-sulfonic acid (ABTS) radical scavenging activity. These results provide useful information for using silkworm powder as an ingredient in functional foods and for exploiting alkaline protease treatment to improve the extractability and bioactivity of a raw material.

Study on Physiologically Active Compounds and Antioxidant Activity of Korean Yam (Dioscorea batatas DECNE.)

  • The bioactive compound and antioxidant property of Korean yam (Dioscorea batatas DECNE.) were studied using in vitro methods. Yam available in Korea was analyzed for lycopene, chlorophyll a, b, tannin, phytic acid and total saponin contents. 70% Methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v) were used to extract yam. Then the antioxidant activity evaluated through ferrous ion chelating activity, ${\beta}$-carotene bleaching method, lipid peroxidation inhibition and nitric oxide (NO) radical scavenging activity. 70% Methanol extract showed the highest ferrous ion chelating activity and NO radical scavenging activity. And CM extract was the most effective in inhibition of linoleic acid peroxidation evaluated by ${\beta}$-carotene bleaching assay and lipid peroxidation inhibition assay. Based on the results obtained, yam is a potential active ingredient that could be applied in antioxidation as well as bio-health functional food to take a good part in prevention of human diseases and aging.

  • Background: Traditionally, East-Asians (Korea, Japan, and China) utilize seaweeds as a food source and ingredient in traditional medicine. Korea is one of the biggest seaweed producer and consumer in the global trade. Especially, side dishes made from seaweeds are very popular in the traditional Korean cuisines. Seaweeds are popular as fresh vegetable salads and soup or eaten as snacks. Main body: Seaweeds are rich in essential nutrients, minerals, and vitamins as well as a promising source of novel bioactive compounds. The compounds (polysaccharides, polyphenols, and sterols) present in the edible Korean seaweeds possess important bioactive properties such as antioxidant, anti-inflammation, anticancer, anti-diabetic, and anticoagulant properties. Thus, the long-term consumption of seaweed has a potential to reduce the risk of cancer, diabetes, obesity, and inflammation-related complications. However, seaweed consumption is limited to the small population around the globe. Thus, it is important to increase the awareness of the health benefits of seaweeds consumption among the general population. Short conclusion: In the present study, we discussed some popular green and red edible Korean seaweeds and their health-promoting properties. This study might be useful to increase the public awareness of the consumption of seaweed as a food source.

유도비만 흰쥐에서 발효 서목태가 미치는 생리활성 (Bioactivities of Fermented Rhynchosia nulubilis in Dietary Obese Rats)

  • This study was to examine the lipid metabolism of fermented Rhynchosia nulubilis (FRN) in obese rats. The abnormal content of blood lipids often results in metabolic diseases, such as obesity and hyperlipidemia. Seven weeks female Sprague-Dawley rats were divided into four groups and fed high fat diets for 44 days. Also FRN was administered orally for 44 days at 7.5 ml/kg of body weight of rats. The effects of the lipid metabolism were evaluated by total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL), low density lipoprotein (LDL), glutamic pyruvate transaminase (GPT) and glutamic oxalacetic transaminase (GOT) levels in sera. The levels of TC, TG, LDL and GPT in FRN-treated groups were lower than those in obese groups. While HDL levels were significantly increased. These results demonstrated that FRN had improving effects of lipid metabolism in the obese rats, suggesting that FRN would be used as an ingredient of the useful functional products.

A novel L-ascorbic acid and peptide conjugate with increased stability and collagen biosynthesis

  • L-ascorbic acid (Vitamin C) and peptide are both useful compounds for collagen biosynthesis in cosmeceuticals (cosmetic and pharmaceutical fields). The instability of these compounds, however, limit their application in these industries. In this report, we describe the development of a novel compound, Stabilized Ascorbyl Pentapeptide (SAP), which physically is much more stable than L-ascorbic acid in water. The inhibitory effects of this SAP compound on tyrosinase and melanin synthesis is comparable to that of L-ascorbic acid. Importantly, the SAP compound displays no cytotoxicity at a high concentration (5 mM). The ability of SAP to promote collagen biosynthesis is greater than that of L-ascorbic acid or the KTTKS peptide alone. Considering the in vitro stability and functional effects, our data strongly suggest that the SAP compound is a good candidate not only as a cosmetic ingredient, but also as a wound healing agent.

반응표면분석법을 이용한 천년초 분말 첨가 머핀 제조 조건의 최적화 (Optimization of Muffin with Opuntia humifusa Powder using Response Surface Methodology)

  • The objective of this study was to determine the optimum mixing ratio of Opuntia humifusa powder and sugar in the preparation of muffin. The optimal sensory composite recipe was determined by following the central composites for muffin with different levels of Opuntia humifusa powder (A) and sugar (B). The analysis was performed using the response surface methodology, and the sensory evaluation was performed with the data. Ten experimental recipes, including two with reference points in the composition, were selected. The compositional and functional properties were measured, and the physical and sensory values were applied to the mathematical models. Perturbation plots showed the effects of each ingredient on the final product. Measurements showed significant values in lightness (p<0.05), sweetness (p<0.05), hardness (p<0.05), gumminess (p<0.05) and cohesiveness (p<0.01). The sensory measurements showed significant values in color (p<0.05), flavor (p<0.05), appearance (p<0.05), softness (p<0.05) and overall quality (p<0.05). Based on the numerical and graphical methods, the optimal formulation was determined to be 6.10 g of Opuntia humifusa powder and 82.61 g of sugar.
