Effects of the Fermented Black Garlic Extract on Lipid Metabolism and Hepatoprotection in Mice (발효흑마늘 추출물이 흰쥐의 지질대사 및 간기능 개선에 미치는 영향)

  • This study was conducted to evaluate the functionality of fermented black garlic extracts under various conditions. Black garlic powder was prepared by aging for 0~72 hours at 80℃ depending on relative humidity (RH). It showed the highest antioxidant effects among the samples; the total antioxidant activity of black garlic powders at RH 75%, 84%, and 90% for 72 hours was increased 31.9 times, 28.2 times, and 22.6 times compared with that of the fresh garlic powder, respectively. Also, the alliin content was gradually decreased. S-ally-L-cysteine and S-ethyl-cysteine levels were increased; the highest values were 495.9 ㎍/g and 1,769.7 ㎍/g after aging for 72 hours at RH 75%. Aspartate transaminase (AST) and alanine transaminase (ALT) levels were increased following high fat diet feeding, but the rise was obviously reduced by administration of black garlic extract. The total cholesterol, LDL/VLDL-cholesterol, and triglyceride contents in serum were significantly lower in methionine and choline deficient (MCD) diet treatment groups than in the positive control group. The concentration was increased following the intake of black garlic and fermented black garlic extracts. Therefore, black garlic extracts could be an ideal material as a dietary supplement in healthy functional foods to improve the effects on fatty liver.

Effects of Yeast Fermentation on the Production of Soy-oilgosaccharides from Bean Cooking Water (대두침출액으로부터의 대두올리고당 생산을 위한 효모발효 효과)

  • Bean cooking water was used as a raw material for the production of soy-oligosaccharides. To maximize the yield of the physiologically functional oligosaccharides such as raffinose and stachyose, a fermentation process was introduced to reduce sucrose content. Yeast strains utilizing sucrose, but scarcely affecting the raffinose and stachyose for the growth were initially selected to reduce the sucrose content in the bean cooking water. The selected strains were Saccharomyces cerevisiae ATCC 9763, S. cerevisiae KCTC 7039 and Hansenula anomala KFRI 626. Bean cooking water with 2% inoculation volume of these yeasts was fermented until 48 hrs, respectively. The results of sugar analysis according to the fermentation time showed, bean cooking water treated with S. cerevisiae KCTC 7039 and H. anomala KFRI 626 used almost sucrose without decreasing stachyose for the growth.

Quality Characteristics of Inner Beauty Food (Mosidae Tofu) by the Addition of Adenophora remotiflora Powder (모시대 분말을 첨가한 미용식품(모시대 두부)의 품질 특성)

  • In this study, physio-chemical, mechanical, and sensory characteristics of Tofu containing 0, 0.5, 1.0, and 2.0% Adenophora remotiflora powder were examined. In addition, we examined the potential of utilizing Adenophora remotiflora powder as a functional food material by estimating total phenol contents, electron-donating abilities, and hydroxyl radical scavenging activities of the hot water and ethanol extracts of Adenophora remotiflora powder in the range from 0~2.0%. The total phenol content of the ethanol extracts of Adenophora remotiflora powder was $487.93{\mu}g/mL$ while the that of the water extract of Adenophora remotiflora powder was $403.70{\mu}/mL$. The electron-donating abilities of the ethanol and water extracts of Adenophora remotiflora powder were 75.37 and 86.10%, respectively. The hydroxyl radical scavenging activities of the ethanol and water extracts of Adenophora remotiflora powder were 65.50 and 66.22%, respectively. We also evaluated the quality characteristics of Tofu containing Adenophora remotiflora powder. In the case of color values, as the level of Adenophora remotiflora powder increased, the values of L (lightness) and a (redness) decreased, whereas that of b (yellowness) increased. In the case of mechanical properties, as the level of Adenophora remotiflora powder increased, hardness, gumminess, and chewiness values increased (p<0.05), whereas springiness and cohesiveness values decreased (p<0.05). In the case of sensory evaluation, MPT1.0 scored significantly higher in color, flavor, tenderness, texture, and overall quality. To sum up, Tofu containing 1% Adenophora remotiflora powder showed the highest overall preference.

Quality Characteristics of Pound Cake Added to $\beta$-Glucan During Storage ($\beta$-Glucan 첨가 파운드 케이크의 저장 중 품질 특성)

  • The aim of this study was to evaluate the quality characteristics of pound cake made of flour substituted with $\beta$-glucan (3, 6 and $9\%$), which is a functional fuod material produced from Agrobacterium Spp. R259 KCTC 10197BP. A rapid decrease of moisture content during storage at $20^{\circ}C$, $70\%$ relative humidity, was observed in the control while the moisture content in 3 and $6\%$ $\beta$-glucan added pound cake was not decreased until the 12th day of storage. During storage, the hardness of the control cake was increased whereas that of $\beta$-glucan $6\%$ and $9\%$ added pound cake was not changed until the 6th day of storage. The addition of $\beta$-glucan was therefore conflmed to delay the staling of cake. During storage, the Hunter color L ud b values of the crust and crumb colors of the $\beta$-glucan added pound cake were not significantly different from those of the control, although the a value did increase with longer storage. The sensory results showed that the scores of over-all acceptability of the 3 and $6\%$ $\beta$-glucan added pound cake at the 6th day of storage days were higher than that of the control. These study results confirmed that the addition of $\beta$-glucan to pound cake maintained the moisture content and delayed hardness during storage.

The antioxidant activity of steamed ginger and its protective effects on obesity induced by high-fat diet in C57BL/6J mice

  • BACKGROUND/OBJECTIVES: Ginger, a root vegetable, is known to have antioxidant and antiobesity effects. Preparation, such as by steaming, can affect the chemical composition of prepared root vegetables or herbs and can change their functional activities. In the present study, we investigated the protective effects of steamed ginger against oxidative stress and steatosis in C57BL/6J mice fed a high-fat diet. MATERIAL/METHODS: The levels of polyphenols and flavonoids in two different extracts of steamed ginger, i.e., water extract (SGW) and ethanolic extract (SGE); as well, their antioxidant activities were examined. Forty male C57BL/6J mice were fed a normal diet (ND, n = 10), high-fat diet (HFD, 60% fat, w/w, n = 10), HFD supplemented with 200 mg/kg of SGE or garcinia (GAR) by weight (SGED or GARD, respectively, n = 10) for 12 weeks. Serum chemistry was examined, and the expressions of genes involved in lipid metabolism were determined in the liver. Histological analysis was performed to identify lipid accumulations in epididymal fat pads and liver. RESULTS: The SGE had higher contents of polyphenols and flavonoids and higher DPPH and $ABTS^+$ free radical scavenging activities compared to those of SGW. Treatment with SGE or GAR significantly decreased the HFD-induced weight gain. Both SGE and GAR significantly reduced the high serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein levels induced by HFD. Compared to ND, HFD significantly increased hepatic TC and TG levels. SGE or GAR supplementation significantly decreased the increase of hepatic lipids by HFD. Interestingly, SGE had a more significant effect in reducing hepatic TC and TG levels than GAR. Furthermore, hepatic genes involved in lipogenesis and lipolysis were altered in both the SGED and GARD groups. CONCLUSIONS: The present study indicates that steamed ginger supplementation can decrease plasma TC and TG and can inhibit liver steatosis by regulating the expressions of hepatic genes.

Anti-Inflammatory Effects of Abalone (Haliotis discus hannai) Viscera via Inhibition of ROS Production in LPS-Stimulated RAW 264.7 Cells

  • Haliotis discus hannai called abalone, is the valuable marine mollusks and the by-products of abalone processing are viscera. Brownish abalone male viscera (AMV), which have not been reported as having anti-inflammatory effects, was extracted with acetone and fractionated by different six acetone/hexane ratios (0, 10, 20, 30, 40, and 100%) using a silica column via in vitro ABTS and DPPH radical and nitric oxide (NO) production assay-guided fractionation. Among the fractions, the acetone/hexane ratio 40%, A40 exhibited the most potent radical scavenging activities and inhibition of lipopolysaccharide (LPS)-induced NO production without cytotoxicity. A40 inhibited LPS-induced intracellular reactive oxygen species (ROS) production in a dose-dependent manner. Western blot analysis revealed that A40 down-regulated the activation of NF-κB, MAPK (ERK 1/2, p-38, and JNK), and inflammatory enzymes, inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2. Moreover, this fraction inhibited the generation of pro-inflammatory cytokines such as interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α. These results suggested that AMV containing A40 with anti-inflammatory and anti-oxidantive effects, is the effective therapeutic and functional material for treating inflammatory disorders.

Antioxidant and Physiological Activities of Different Solvent Extracts of Cnidium japonicum (갯사상자(Cnidium japonicum) 추출물의 항산화성 및 생리활성)

  • Cnidium japonicum a biennial plant belonging to the family Umbelliferae, is a halophyte that grows in high-salinity areas of coastal sand dunes and sandy shores. This study was conducted to investigate the constituents, antioxidant activities, and physiological activities of C. japonicum. Mineral analyses revealed that potassium, sodium, calcium, and magnesium were the most prevalent minerals in C. japonicum. We used 80% ethanol, 80% methanol, and distilled water as solvents to prepare extracts from C. japonicum tissues, and the obtained extraction yields ranged between approximately 26% and 32%. Among the three extracts, the ethanol and methanol extracts had higher total polyphenol and flavonoid levels than the water extracts did. The antioxidant activities of methanol extracts were the highest among the various solvent extracts of C. japonicum as was the elastase/collagenase inhibitory activity. In contrast, the ethanol extract exhibited the highest tyrosinase inhibitory activity. Furthermore, the methanol extract possessed over 80% BACE1 (β-secretase) inhibitory activity at a final concentration of 20 ㎍/mL. Therefore, these results indicate that methanol and ethanol extracts of C. japonicum may be useful as antioxidant and functional substances in food and pharmaceutical material.

Analytical Validation of Rosmarinic Acid in Water Extract of Perilla frutescens Britton var. acuta Kudo as Functional Health Ingredient (건강기능식품 기능성 원료로써 장흥 차조기 열수 추출물의 지표성분인 로즈마린산 분석법 검증)

  • This study attempted to establish an HPLC analysis method for determination of marker compounds as a part of material standardization for the development of health functional food materials from Perilla frutescens Britton var. acuta Kudo. The quantitative determination method of rosmarinic acid as a marker compound of P. frutescens Britton var. acuta Kudo extract (PFE) was optimized by HPLC analysis using a C18 column ($4.6{\times}150mm$, $5{\mu}m$) with 0.1% acetic acid as the elution gradient and methanol as the mobile phase at a flow rate of 1 mL/min and detection wavelength of 280 nm. The HPLC/UV method was applied successfully to quantification of the marker compound in PFE after validation of the method with linearity, accuracy, and precision. The method showed high linearity in the calibration curve at a coefficient of correlation ($R^2$) of 0.9995, and the limit of detection and limit of quantitation were $0.36{\mu}g/mL$ and $1.2{\mu}g/mL$, respectively. Relative standard deviation (RSD) values of data from intra- and inter-day precision were less than 3.21% and 1.43%, respectively. Recovery rate test at rosmarinic acid concentrations of 12.5, 25 and $50{\mu}g/mL$ scored between 97.04~98.98% with RSD values from 0.25~1.97%. These results indicate that the established HPLC method is very useful for the determination of marker compound in PFE to develop a health functional material.

Antioxidant efficacy of Jeju crop extracts using Jeju lava seawater as a solvent (제주 용암해수를 용매로 한 제주 작물 추출물의 항산화 효능)

  • Jeju lava seawater's abundant minerals are known to exert antioxidant effects that remove the free radicals responsible for aging. Therefore, lava seawater reportedly possesses high commercial value as a functional food material. This study compared and analyzed the antioxidant activities of extracts from crops produced in Jeju (carrots, blueberries, and mandarins) using distilled water and lava seawater as solvents. Lava seawater extracts exhibited higher total polyphenol and flavonoid contents of blueberries and mandarins than distilled water extracts. Furthermore, the antioxidant enzymatic and 2, 2-diphenyl-1-picrylhydrazyl radical-scavenging activities of these crops were higher in lava seawater extracts than in distilled water extracts. In particular, using Vero cells, the ROS-scavenging efficacies of blueberries and mandarins were found to be higher in lava seawater extracts. Meanwhile, the antioxidant activities of carrots were higher in lava seawater extracts, despite no difference in total polyphenol and flavonoid contents. These results suggest that lava seawater exhibits favorable potential as a solvent in the functional food industry, and lava seawater-based Jeju crop extracts are potentially useful as functional food ingredients.

  • In an attempt to estimate the life projection of LED packages, IESNA published a paper regarding an LED package measurement test method in 2008, and a life projection technical document in 2011, to be used for LED life estimation. IESNA's publications regarding LED package measurement methods were functional, but they were not internationally standardized before 2017. In order to develop a standardized method, the International Standard chose to use the LM-80 as a measurement method for LED life projection in their publication in 2017. Many projection methods have been discussed by the IEC Technical Committee 34 working group, including the method using an exponential function, which reflects lumen degradation characteristics well. This study is designed to explore alternative LED package life estimation methods using an exponential function with statistical analysis, other than the one suggested by the International Standard.
