Enhancement of centelloside production from cultured plants of Centella asiatica by combination of thidiazuron and methyl jasmonate

  • In order to produce centellosides from whole plant cultures of Centella asiatica (L.) Urban, we evaluated the synergistic effects of thidiazuron (TDZ) and methyl jasmonate (MJ) on whole plant growth and centelloside production. After 4 weeks of treatment with 0.025 mg/L of TDZ coupled with 0.1 mM MJ, the production of made-cassoside and asiaticoside from whole plant cultures was estimated to be 2.40- and 2.44-fold, respectively, above that of MJ elicitation alone. When whole plants were treated with a growth regulator and an elicitor, the growth of whole plants, as compared to the controls, did not differ. Additionally, total phytosyterol content in the leaves of whole plants co-treated with MJ and TDZ was 1.08-fold greater than those of MJ alone. These results demonstrate that combined treatments not only stimulate the accumulation of centellosides in the leaves but also inhibit the reduction of phytosterol levels caused by MJ elicitation.

  • Previous phytochemical studies of Vigna angularis (Ohwi) Ohwi & Ohashi (Leguminosae) have shown the presence of saponins and flavonoids. From the seed of V. angularis, genistein-7-O-${\beta}$-D-glucopyranoside (genistin) was isolated. Lifespan-extending effect of genistin was elucidated using Caenorhabditis elegans model system. Genistin showed potent lifespan extension of worms under normal culture condition. This compound also exhibited the protective effects against thermal and oxidative stress conditions. In the case of heat stress, genistin-treated worms exhibited enhanced survival rate, compared to control worms. In addition, genistin-fed worms lived longer than control worms under oxidative stress induced by paraquat. To verify the possible mechanism of genistin-mediated increased lifespan and stress resistance of worms, we investigated whether genistin might alter superoxide dismutase (SOD), catalase activities and intracellular ROS levels. Our results showed that genistin was able to elevate SOD and catalase activities of worms and reduce intracellular ROS accumulation in a dose-dependent manner.

Development of Multiplex RT-PCR for Simultaneous Detection of Garlic Viruses and the Incidence of Garlic Viral Disease in Garlic Genetic Resources

  • Garlic generally becomes coinfected with several types of viruses belonging to the Potyvirus, Carlavirus, and Allexivirus genera. These viruses produce characteristically similar symptoms, they cannot be easily identified by electron microscopy (EM) or immunological detection methods, and they are currently widespread around the world, thereby affecting crop yields and crop quality adversely. For the early and reliable detection of garlic viruses, virus-specific sets of primers, including species-specific and genus-specific primers were designed. To effectively detect the twelve different types of garlic viruses, primer mixtures were tested and divided into two independent sets for multiplex polymerase chain reaction (PCR). The multiplex PCR assays were able to detect specific targets up to the similar dilution series with monoplex reverse transcription (RT)-PCR. Seventy-two field samples collected by the Gyeongbuk Agricultural Technology Administration were analyzed by multiplex RT-PCR. All seventy two samples were infected with at least one virus, and the coinfection rate was 78%. We conclude that the simultaneous detection system developed in this study can effectively detect and differentiate mixed viral infections in garlic.

Risk Acceptance and Expectations of Laryngeal Allotransplantation

  • Background Laryngeal allotransplantation (LA) is a technique involving transplantation of a deceased donor's larynx into a recipient, and it may be substituted for conventional laryngeal reconstruction. There are widely different views on LA, as the recipient is administered continuous, potentially life-threatening, immunosuppressive therapy for a functional or aesthetic result, which is not directly related to life extension. The purpose of this study was to analyze the difference in risk acceptance and expectations of LA between four population groups. Methods A survey was performed to examine patients' risk acceptance and expectations of LA. The survey included 287 subjects in total (general public, n=100; kidney transplant recipients, n=53; post-laryngectomy patients, n=34; doctors, n=100), using a Korean translated version of the louisville instrument for transplantation (LIFT) questionnaire. Results All four groups responded differently at various levels of their perception in risk acceptance and expectations. The kidney transplant recipients reported the highest risk acceptance and expectations, and the doctor group the lowest. Conclusions This study examined the disparate perception between specific population groups of the risks and benefits of using LA for the promotion of the quality of life. By addressing the information gaps about LA in the different populations that have been highlighted from this survey, we suggest that LA can become a more viable alternative to classical surgery with resultant improved quality of life for patients.

녹두 식물체 부위별 Flavonoids 함량 변화 (Variation of Flavonoids Contents in Plant Parts of Mungbean)

  • 녹두의 식물체 부위, 수확시기, 생육단계, 파종기 등에 따른 주요 flavonoids의 함량 차이를 구명하고자 본 연구를 수행하였다. 1. Vitexin과 isovitexin은 녹두 식물체 부위 중에서 종실에만 분포하고 잎, 엽병, 줄기, 뿌리에서는 검출되지 않았다. 2. 녹두 종피에만 vitexin은 $52.1\;mg{\cdot}g^{-1}$, isovitexin은 $51.7\;mg{\cdot}g^{-1}$ 분포하고 자엽에서는 검출되지 않았다. 3. 녹두 조 적 만기 수확에 따른 vitexin과 isovitexin 함량은 차이가 없었다. 4. 녹두 잎에 다량 함유된 기능성 물질을 분리 동정한 결과 rutin이었다. 5. Rutin함량은 녹두 잎, 엽병, 줄기 순으로 많고, 종실, 뿌리, 협에서는 검출되지 않았다. 6. 녹두 잎, 엽병, 줄기에 함유된 rutin은 3, 5, 7엽기 순으로 유묘기일수록 많았다.

지질분해효소 저해물질 함유 도둑놈의 갈고리 추출물의 영양특성 및 생리기능성 (Nutritional Characteristics and Physiological Functionality of Lipase Inhibitor-Containing Desmodium oxyphyllum DC. Extracts)

  • 천연물로부터 lipase 저해물질을 개발하여 기능성 식품 산업에 응용하고자 lipase 저해 활성이 우수하여 전보에서 최종 선발된 도둑놈의 갈고리(Desmodium oxyphyllum DC.) 에탄올 추출물의 영양성분, 온도, pH 안정성과 생리기능성 등을 측정하였다. Lipase 저해물질을 함유한 도둑놈의 갈고리 에탄올 추출물의 지질과 단백질 함량은 각각 26.5%와 5.7%이었고 유리아미노산으로 asparagine과 tryptophan 함량이 각각 4,860 mg/건조중량 100 g, 1,209 mg/건조중량 100 g으로 물 추출물보다 높았다. 또한, 도둑놈 갈고리의 에탄올 추출물은 pH 3.0~9.0, $80^{\circ}C$ 이하에서 비교적 안정하였고, 이에 대한 생리 기능성을 측정한 결과, lipase 저해 활성은 74.2%, 항 치매성 Acetylcholinesterase 저해 활성은 68.4%로 비교적 높은 활성을 나타내었다.

Comparison of open reduction and internal fixation with total elbow arthroplasty for intra-articular distal humeral fractures in older age: a retrospective study

  • Background: Intra-articular distal humeral fractures can be surgically challenging. It remains under discussion whether open reduction and internal fixation (ORIF) or total elbow arthroplasty (TEA) is more beneficial for treatment of the elderly. This study aimed to compare the clinical and functional outcomes of ORIF and TEA for managing intra-articular distal humerus fractures in patients aged 65 years or older. Methods: Patients who underwent ORIF (n=28) or TEA (n=43) for in intra-articular distal humerus fracture between May 2008 and December 2018 were reviewed. Range of motion, Mayo Elbow Performance Score (MEPS), Disabilities of the Arm, Shoulder, and Hand (DASH) score, radiologic outcomes, and surgical complications were evaluated at the final follow-up visit. Results: The ORIF and TEA groups showed a mean arc of flexion-extension of 97°±21° and 101°±12°, respectively. The mean MEPS and DASH scores were 94±15 and 27±12 points, respectively, in the ORIF group and 81±27 and 47±28 points in the TEA group. This difference was statistically significant. The incidence of total complications was similar between the groups. Conclusions: In patients older than 65 years with intra-articular distal humerus fracture, ORIF had better outcomes than TEA.

RHadoop 기반 보건의료 빅데이터 분석의 성능 평가 (Performance Evaluation of Medical Big Data Analysis based on RHadoop)

  • 빅데이터 시대에 각광받고 있는 데이터 분석 도구인 R은 강력한 통계 분석 기능과 데이터 가시화 기능을 제공함으로 인해 그 사용자를 급속히 넓혀 가고 있다. 오픈소스 기반으로서의 다양한 기능 확장성이 R의 강점인데 반해 규모 확장성이 미흡함으로 인해 대용량 데이터 처리에서의 성능 제약이 발생한다. 이를 보완하기 위한 확장 패키지 중 하나인 RHadoop은 R로 작성된 코드에 대해 하둡 플랫폼 기반 병렬 분산 처리를 지원하므로 데이터 분석 성능을 높일 수 있다. 본 논문에서는 인터넷을 통해 공개되는 실제 보건의료 빅데이터를 이용한 데이터 분석에서 RHadoop을 활용할 때 얻을 수 있는 성능 개선을 평가함으로써 RHadoop의 유효성을 검증한다. 본 연구를 통해 R과 RHadoop에서 국민건강보험 진료내역정보를 각각 분석한 결과 8개의 데이터 노드로 구성된 RHadoop 클러스터가 R과 비교하여 최대 8배 이상 성능을 개선시킬 수 있음을 입증하였다.

  • 폐질환은 대부분 흡연과 대기 오염으로 발병하며 한 번 손상된 폐는 기능 회복이 어려워 생명 연장의 위험 질환이며 현대인의 6대 질병 중 하나로 사회적 관심이 증대되고 있다. 따라서 평소 흡연을 하지 말며 폐 기능 강화를 위한 행위를 행해야 호흡기 질환으로 고통 받는 경우의 수를 줄일 수 있을 것으로 여겨진다. 본 논문에서는 귀의 혈 자리가 인체 장기와 연계되어 있다는 이혈요법을 적용하여 폐와 상응하는 귀의 혈 자리를 자극했을 때 실제 폐 기능 강화에 도움이 되는지를 음성 분석 기술에 의해 규명하는 방법을 제안하고자 한다. 이를 위해 우선 인체가 폐에 미치는 영향을 분석하기 위한 선행 연구로 흡연 전/후의 음성 변화를 고찰하고자 한다. 이를 기반으로 폐와 상응하는 이혈점을 자극하기 전과 후의 음성 특징 차이를 분석하여 폐의 이혈점 자극이 실제 폐 기능 향상에 도움이 되는지를 수치적, 정량적으로 규명해 내고자 한다.

Immediate Effect of Spinal Mobilization on Lower Limb Strength in Healthy Individuals: A Pilot Study

  • Background: Spinal Mobilization is one of the manual therapy technique that clinicians have used to treat pain, however, there is still a lack of research on changes in strength in healthy people. Objectives: To investigate the effect of posterior-anterior lumbar mobilization on lower limb strength in healthy individuals. Design: Two-group pretest-posttest design. Methods: In this study, 23 healthy subjects aged 20 years were assigned to 12 lumbar mobilization group (LMG) and 12 sham group (SG) to perform intervention and measurement through pre- and post-design. Intervention was performed in LMG with grade III~IV on L3-5 of the lumbar spine, and lumbar mobilization was performed for each segment. After intervention, knee flexion and extension strength were measured. To measure the main effect on muscle strength, a comparative analysis was conducted using paired t-test and independent t-test. Results: In LMG, knee flexor and extensor strength were increased significantly at 60°/s (P<.05). In addition, the extensors of LMG and SG were significantly different only at 60°/s, and the flexors were significantly different between groups at both 60°/s and 180°/s (P<.05). Conclusion: In healthy individuals, lumbar mobilization results in improvement of strength of knee flexor and extensor, and additional experiments on the effect of mobilization on the lumbar spine on functional changes in the lower limbs will be needed.
