개비자나무(Cephalotaxus koreana) 추출물의 항균활성 및 세포독성 (Antimicrobial Activity and Cell Cytotoxicity of Korean Plum-yem Extract)

  • 본 연구에서는 개비자나무(Cephalotaxus koreana)를 기능성 생물소재로 개발하기 위해 개비자나무 추출물(Korean Plum-yem extract, KPE)의 항균활성, 암세포 증식 억제활성 및 독성을 확인하였고, 일반성분을 조사하였다. KPE의 농도에 따른 항균활성은 그램 양성균, 그램 음성균, 효모 및 사상균에 대하여 성장 저해현상을 나타내었으며, 특히 그램 음성균인 Pseudomonas aeruginosae가 KPE에 가장 민감하게 생육 저해현상을 보였고, 마찬가지로 그램 음성균인 Salmonella thyphimurim, Escherichia coli 순이었다. 그램 양성균이 그램 음성균에 비해 KPE에 의한 생육저해효과가 작은 것으로 나타났다. 혈액암 세포주를 이용한 KPE의 암세포 증식 억제활성을 분석한 결과 80 ${\mu}g/mL$의 농도에서 현저한 암세포 저해활성을 보였다. 또한, KPE를 섭취하였을 때 유발되는 용혈율을 측정하여 KPE의 적혈구 막지질 붕괴의 유무를 조사한 결과, KPE 중 hemolysis를 일으키는 물질은 KPE의 유효성분인 HHT 외에 다른 물질이 있는 것으로 판단되었으며, Ame's test에 의한 돌연변이원성을 갖지 않음을 확인하였다. 따라서 개비자나무 추출물은 항균과 암세포에 대한 세포독성 등 기능성을 갖는 천연물 소재로서 이를 응용한 기능성 식품 개발이 가능한 것으로 판단된다.

  • The aim of this study was to better understand the factors of the mentoring function in the foodservice industry that improve the organizational commitment of subordinates, and to asses the roles that trust towards supervisors play in the relationship between the mentoring and organizational commitment. A survey was of subordinates who were conducting the mentoring in foodservice companies (hotels, family restaurants) located in Seoul and Gyeonggi area was conducted from May 1 to May 30, 2008. The questionnaires were distributed to 400 employees in 50 restaurants and, 341 of them were received and a total of 320 were finally analyzed. The results were as follows. First, the effect of mentoring functions on the organizational commitment of subordinates indicated that the career developing function and psychosocial function had a significant positive effect on the emotional commitment and normative commitment (p<.01), and the career developing function, psychosocial function and role modeling function had a significant positive effect on the enduring commitment (p<.01). Second, the mediating roles of supervisory trust in the relationship between mentoring functions and organizational commitment of subordinates indicated that supervisory trust mediated the relationship between the career developing function of the mentoring functions, and enduring commitment and normative commitment in addition, it met the mediating requirements in terms of the relationship between the role modeling function, and the emotional commitment, enduring commitment and normative commitment. Consequently, the high career developing function, psychosocial function and role modeling function in mentoring raised the level of organizational commitment;thus, the mentoring system should used as on of the mentoring functions in the foodservice industry and mentoring functions should be also established.

Momordica charantia Protects against Cytokine-induced Apoptosis in Pancreatic $\beta$-Cells

  • The unripe fruit of Momordica charantia (MC) has been shown to possess antidiabetic activity. However, the mechanism of its antidiabetic action has not been fully understood. In this study, the effects of the aqueous ethanolic extract of MC (AEE-MC) were evaluated on the apoptosis in pancreatic $\beta$-cells treated with a combination of the cytokines, interleukin (IL)-$1{\beta}$, tumor necrosis factor (TNF)-$\alpha$, and interferon (IFN)-$\gamma$. In MIN6N8 cells, the inhibitory effect of AEE-MC was significantly observed at 2 to 50 ${\mu}g/mL$: a 26.2 to 55.6% decrease of cytoplasmic DNA fragments quantified by an immunoassay. The molecular mechanisms, by which AEE-MC inhibited $\beta$-cell apoptosis, appeared to involve the inhibition on the expression of p21, Bax, and Bad, the up-regulation of Bcl-2 and Bcl-$X_L$, and the inhibition on the cleavage of caspase-9, -7, and -3 and poly (ADP-ribose) polymerase. This study suggests that MC may inhibit cytokine-induced apoptosis in $\beta$-cells and, thus, may contribute via this action to the antidiabetic influence in diabetes.

Function of Cell-Bound and Released Exopolysaccharides Produced by Lactobacillus rhamnosus ATCC 9595

  • The physiological characteristics and function of the exopolysaccharide (EPS) produced by Lactobacillus rhamnosus ATCC 9595 were determined. The total quantity of EPS was rapidly increased to 496$\pm$20 mg/l during the exponential phase, and then maintained steadily during the stationary phase. During the exponential phase (18 h), the total EPS consisted of 61% cell-bound EPS (cb-EPS) and 39% released EPS (r-EPS), whereas the relative proportion of EPS during the stationary phase (48 h) was convered to 23% cb-EPS and 77% r-EPS. On gel permeation chromatography, cb-EPS was fractionated as a single peak of 8.6$\times10^6$ Da, whereas r-EPS was fractionated as two peaks with average molecular weights of 4.3$\times$10$^4$ and 8.6$\times10^6$ Da. Interestingly, both EPS species exhibited anticancer properties and cholera toxin-binding activities. Our results suggest that the EPS generated by L. rhamnosus ATCC 9595 might be suitable for use as a functional food or food supplement.

전라남도 일부 지역 노인들의 신체적·정신적 기능 원활 정도에 따른 식생활 패턴의 차이 (Dietary Patterns among the Elderly in Jeollanam-do Area based on Their Physical and Mental Function State)

  • This study investigated dietary patterns among the elderly over 75 years old living in Jeollanam-do area in May 2012. Although structured interviews were conducted with 236 consenting subjects, only 194 who completed the ADL, IADL, and K-MMSE tests were used for statistical data analysis. Using ADL, IADL, K-MMSE scores, cluster analysis was first performed and resulted in two groups: IFG (Insufficiently Functioning Group) and SFG (Sufficiently Functioning Group). Chi-square tests for nominal scales, Mann-Whitney tests for ordinal scales, and ANOVAs and t-tests for interval and ratio scales were conducted to compare two groups. More than 70% of IFG were illiterates compared to 28.1% of SFG. 'Excessive eating', 'appetite', 'digestion', and 'balanced diet' did not differ between groups. SFG more frequently had snacks and ate out and were more likely to take health supplements than IFG. Among the 100 major food items, consumption frequencies of several foods differed between groups. Study implications and limitations were discussed.

Effects of Raw, Cooked, and Germinated Small Black Soybean Powders on Dietary Fiber Content and Gastrointestinal Functions

  • The effects of raw and processed small black soybean powders on dietary fiber content and gastrointestinal function in rats were investigated. The crude oil, protein, and ash contents of raw small black soybean powder were not significantly different from those of processed small black soybean powders. The germination process increased soluble and insoluble dietary fiber contents significantly, as compared to raw small black soybean powder. The germinated small black soybean powder diet led to a significantly different food intake than the basal diet in both normal and loperamide-induced constipated rats. The body weight gains of the experimental groups, however, were not significantly different from that of the basal diet groups for both the normal and loperamide-induced rats. The gastrointestinal transit times and fecal weights for normal and loperamide-induced rats consuming the processed small black soybean powder diet were significantly different from those on a basal diet. These results suggest that the processes of cooking and germinating the small black soybean might contribute to acceleration of fecal excretion in both experimental normal and constipation model rats.

A Rapid and Effective Colony PCR Procedure for Screening Transformants in Several Common Mushrooms

  • In the post-genomic era, gene function analysis has attracted much attention. Transformation is often needed to investigate gene function. In this study, an easy, rapid, reliable, and cost-effective colony polymerase chain reaction (PCR) method for screening mushroom transformants was developed: picking up a suitable amount of transformant's tissue ($1-10{\mu}g$) to $20{\mu}l$ 0.25% Lywallzyme solution, and vortexing for 10 s followed by incubation at $34^{\circ}C$ for 15 min. Finally, $2{\mu}l$ of the suspension was used as templates to perform PCR and single target bands were successfully amplified from respective transformants of Tremella fuciformis, Pleurotus ostreatus, and Pleurotus tuber-regium. This procedure could be widely employed for screening transformants in mushroom transformation experiments.

건강기능식품의 섭취 시 주의사항 표시제도 개선에 대한 연구 (A Study on Improvement of Labeling on the Caution for Intake for Health Functional Food)

  • The objective of this study was to analyze labeling improvements and evaluate willingness to pay for health functional foods with a focus on the caution for intake. For this study, we conducted a survey on health functional food intake behavior, confirmation and improvements of cautions for intake. We assessed the willingness to pay for improvement of the caution for intake. Consumers anticipate improved immune function, and fatigue improvement after consumption of health functional foods. They mainly checked the function components related to efficacy and effectiveness, ingredients and their contents, ingestion amount and method, expiry date and best mode of storage, product name, and cautions associated with ingestion of health functional foods. They has been difficulties in obtaining sufficient caution information for intake from the current labeling method. Therefore, it is necessary to improve the labeling of caution for intake. The analysis indicated that about 5.14% of the respondents were willing to pay more if new labeling was introduces. However, there is still controversy over their safety, which is damaging to the consumers. Therefore, by providing consumers with accurate and detailed information on cautions for intake, it can contribute to securing safety and improving the quality of health functional foods.
