Isolation of Aspergillus niger K-25 Prroducing Acid-stable ${\alpha}-amylase$ (내산성(耐酸性) 아밀라제를 생산하는 Aspergillus niger 균주의 분리)

  • One strain of Aspergillus niger K-25 producing an acid-stable ${\alpha}-amylase$ was isolated from the soil. The optimum culture conditions were investigated. The production of the acid-stable ${\alpha}-amylase$ was enhanced when the strain was incubated in a medium containing soluble starch 3.5%, peptone 2%, $KH_2PO_4$ 0.5%, $MaSO_4{\cdot}7H_2O$ 0.25% and $FeCI_3$ 1.0% at pH 3 for 7 days. However, higher activity of acid-stable ${\alpha}-amylase$ was demonstrated on wheat bran culture. Amylase production was doubled when A. niger K-25 was incubated on the wheat bran supplemented with fumaric acid buffer (pH 3).

Characterization and Release Behavior of Polymersomes of PEG-Poly(fumaric-sebacic acids)-PEG Triblock Copolymer in Aqueous Solution (PEG-Poly(fumaric-sebacic acids)-PEG 삼중 블록 공중합체로 수용액에서 만들어진 폴리머솜의 분석과 방출특성)

  • Polymersomes made of biodegradable triblock copolymers based on poly(fumaric acid-co-sebacoyl chloride)/PEG (PEG-co-P(FA/SC)-co-PEG) were prepared and studied in aqueous solutions. TEM confirmed the formation of vesicles in aqueous media. Aggregation behavior of the copolymers was studied by fluorescence spectroscopy of 8-anilino-1-naphthalenesulfonic acid, and the critical aggregation concentration (c.a.c.) of the copolymer was found to be ${\sim}26.2{\mu}M$ indicating desirable stability of the vesicles. Dynamic light scattering revealed that the size of the vesicles was distributed within the range of 170-270 nm. Turbidity measurements confirmed the relative short-term stability of the polymersomes. Carboxyfluorescein, a hydrophilic compound, was simply encapsulated in the vesicles during polymersome preparation. The release of encapsulant from the polymersomes at 25 and $37^{\circ}C$ lasted about 3 weeks, and the rate of release followed a first-order kinetics. The release is speculated to be primarily carried out through diffusion. These results confirm that these polymersomes are promising as controlled-release carriers of various drugs.

Chemical Components of Gastrodia elata Blume Powder (천마 분말의 화학적 성분)

  • The chemical components in freeze dried Korean Gastrodia elata blume powder were analyzed. The proximate composition was composed of moisture(2.12%), crude protein (4.41%), crude fat(1.20%), crude ash(2.25%), crude fiber(4.60%) and N-free extract(85.15%). Free sugar was composed of glucose(1,314mg%), sucrose(1,081mg%) and fructose(869mg%). Total organic acid content was 2,095mg% and composed of succinic acid(1,238mg%), citric acid, malic acid, oxalic acid and fumaric acid in the order. Among the 11 fatty acids detected, linoleic acid was the most predominant one and unsaturated fatty acid was 80.55 area percent. 18 and 39 amino acids were detected from total and free amino acid, respectively. Arginine, asparagine, proline and sarcosine were the major components of free amino acid. The content of crude saponin and total phenol were 3,768mg% and 821mg%, respectively. Ascorbic acid(6.4mg%), vitamin B1(1.2mg%), vitamin B2(1.7mg%) and niacinamide(0.2m%) were detected as soluble vitamins of the powder. The content of vitamin A and vitamin E was 1.22mg% and 0.32%, respectively.

Organic Acids Content of the Selected Korean Apple Cultivars (우리나라 사과 일부 품종의 유기산 함량)

  • Total and individual organic acid contents of Malus domestica Borkh, cultivars, Tsugaru, Fuji Jonathan (Hong-Ok), and New Jonagold(Sin-Heung) apples, were investigated. Average titratable acidities measured by titration method and total organic acids content determined by HPLC were 241.64-444.52 and 364.23-680.80mg%, respectively. Average total organic acid contents were Jonathan 630.80mg%, New Jonagold 471.04mg%, Fuji 403.80mg%, and Tsugaru 364.23mg%, Contents of DL-malic, citric, fumaric, and quinic acid were 351.98 (Tsugaru)-579.88mg% (Jonathan), 2.14 (Fuji)-12.95mg% (Jonathan), 0.012 (Fuji)-0.060mg% (Jonathan), and 8.91 (Tsugaru)-14.20mg% (Fuji), respectively. Succinic acid was detected only from Jonathan (27.53mg%) and New Jonagold (5.20mg%), while maleic acid was not detected from all cultivars. Ratio of L-malic acid and DL-malic acid contents were 0.98-1.02 in all apple cultivars.

Development of HPLC-UV method for detection and quantification of seven organic acids in animal feed (사료 중 유기산 7종 동시분석법 개발 및 유통 사료의 모니터링)

  • 본 연구에서는 사료 첨가제로 이용되고 있는 유기산 7종(formic acid, malic acid, lactic acid, acetic acid, citric acid, fumaric acid, propionic acid)의 동시분석법 개발을 위한 연구를 실시하였다. 7종의 화합물은 표준물질의 Retention time과 UV spectra를 통해 구별하였고, 분석법 검증은 직선성, 민감성, 선택성, 정확성, 정밀성을 통하여 검증하였다. 그 결과로 LOD와 LOQ의 범위가 각각 43~26,755 μg/kg, 12-8,026 μg/kg으로 설정하였고, 평균 회수율이 79.3~95.2%로 우수하게 보였으며, intra-day, inter-day에 대한 전반적인 상대 표준 편차(%RSD)는 3.2% 미만으로 나타났다. 이와 같이 검증된 자료를 통해 유기산의 동시분석에 대한 직선성, 민감성, 선택성, 정확성 및 정밀성을 확인하였고, 높은 수준을 나타냄을 알 수 있었다. 이를 바탕으로 유기산이 검출되는 단미사료 46 가지를 분석에 적용하여 진행하였고, 정량과 동시분석 검출을 위한 방법은 RP-HPLC/UV 검출기를 이용하여 성공적으로 개발되었다. 따라서 본 연구결과를 바탕으로 하여 사료 중의 유기산의 분석이 신속하고 정확해졌을 뿐 아니라, 다른 종류의 사료 또한 이를 적용하여 효율적으로 이용할 수 있을 것으로 판단된다.

Organic Acid in Korean Soy-Sauces (한국(韓國) 간장중(中)의 유기산(有機酸)에 대(對)하여)

  • Korean soy-sauces were orepared by the ordinary and impreved method and its analyses on the organic acid. The results obtained is as following: 1. In analysing general components of prepared soy-sauce, total acid, volatile acid and non-volatile acid were found more in improved soy-sauce than in ordinary soy-sauce. 2. Volatile organic acid were analysed by gas-chromatography method. As a result, the followings was attained: a) In the ordinary soy-sauce, formic acid, acetic acid, propionic acid, and butyric acid were detected. Butyric acid was in the highest amount and then propionic acid, acetic acid and formic acid are followed in the order. b) In the improved soy-sauce, formic acid, acetic acid, propionic acid and butyric acid were detected. Acetic acid was in the highest amount and then propionic acid, butyric acid and formic acid are followed in the nrder. 3. Non-volatile organic acid were analysed by paper partition chromatography method. As a result, the followings were attained: a) Lactic, glutaric, fumaric, malonic, malic, glycolic, oxalic, tartaric, and succinic acid and two unknown spots were detected in ordinary soysauce. Lactic acid was in the highest amount and then succinic, glycolic, oxalic, tartaric, glutaric, malic, fumaric and malonic acid are followed in the order. b) Lactic, glutaric, malonic, malic, glycolic, tartaric, succinic and tgalacturonic acid and two unknown spots were detected in the improved soy-sauce. Lactic acid was in the highest amount and then succinic, glycolic, malic, glutaric, tartaric, galacturonic and malonic acid are followed in the order. 4. ${\alpha}-keto$ acid were analysed by paper partition chromatography. As a result, the followings were attained: Pyruvic acid and ${\alpha}-keto$ glutaric acid and an unknown spot were detected in the ordinary and improved soy-sauce. Pyruvic acid was in the highest amount and then ${\alpha}-keto$ glutaric acid are ollowed in the order. 5. Stale flavor in the ordinary soy-sauces seems to be partly affected by butyric acid and propionic acid. 6. Substances influencing taste, such as lactic acid and succinic acid, were found more in improved soy-sauce than ordinary soy-sauce.

Isolation, Identification, and Characterization of a Bacteriocin-Producing Enterococcus sp. from Kimchi and Its Application to Kimchi Fermentation

  • A bacteriocin-producing lactic acid bacterium, which strongly inhibited the Lactobacillus plantarum recognized as an important acid spoilage microorganism in kimchi fermentation, was isolated from kimchi. From morphological, physiological, sugar fermentation, biochemical tests, and l6S rDNA sequencing results, the isolate was identified as an Enterococcus sp. and designated as Enterococcus sp. K25. The bacteriocin produced by Enterococcus sp. K25 inhibited several Gram-positive bacteria, including Lb. plantarum, whereas it did not inhibit Gram-negative bacteria and yeasts. Optimal temperature and pH for the bacteriocin production were $25^\circ{C}$ and 5.5, respectively. Enterococcus sp. K25 was applied to kimchi manufacturing alone and together with other preservatives (i.e., chitosan and fumaric acid). In addition, growth of lactic acid bacteria, pH, and titratable acidity (TA) were measured during aging at $5^\circ{C}$ and $10^\circ{C}$. Inoculation of Enterococcus sp. K25 together with fumaric acid showed the most synergistic effect on extension of kimchi shelf-life. Compared to control (no addition), the treatment prolonged the kimchi shelf-life up to 6 days, whereupon the eight-point TA value recognized as the edible limit was reached.

  • A survey of the free amino acids and organic acids in the shoot of Phyllostachys edulis was made by means of amino acid autoanalyzer and gas chromatograph. The results of the survey are summerized as follows. 1. Eighteen amino acids found in bamboo shoot were lysine, histidine, arginine, tryptophan, aspartic acid, glutamic acid, threonine, serine, proline, glycine, alanine, valine, leucine, isoleucine, methionine, tyrosine and phenylalanine, and an unknown was found. Serine showed the highest amount and more than about 44% of total free amino acids. 2. Oxalic acid was the major organic acid, and formic acid, acetic acid, maleic acid, succinic acid, fumaric acid, citric acid, tartaric acid and sorbic acid were determined, and two unknown were found.

Immediate Drug Release Enhancement of Nateglinide Using Fumaric Acid (푸마르산을 이용한 나테글리니드 함유 속방출형 제형의 약물방출 개선에 관한 연구)

  • The purpose of this study was to improve release rate and bioavailability of nateglinide formulation. Polymorphism selection and particle size control were performed to enhance formulation dissolution rate, and a pH modifier was included in the formulation to overcome pH-dependent solubility of nateglinide. The enhanced dissolution rate was characterized by using a dissolution test. The results showed that H-type raw material had a higher dissolution rate than that of B-type raw material. There was 6.2% difference in dissolution between the two materials at 60 min. With regard to particle size, raw material with a $1.13{\mu}m$ particle size showed a 20% faster release rate than that of raw material with a $2.28{\mu}m$ particle size. Furthermore, fumaric acid was included in formulation as a pH modifier. That addition produced a greater than 50% improvement in dissolution rate. In conclusion, dissolution rate of nateglinide can be enhanced by optimizing its polymorphism and particle size; moreover, a synergistic effect on the enhancement of dissolution rate is obtained by including fumaric acid, a pH modifier, in the formulation.

Characteristics and Stability of the Color of the Cranberry Solution (크랜베리 수용액 색상의 특성 및 안정성)

  • Effects of pH, temperature, light, sugars, organic acids, metal ions, ascorbic acid, thiamine nicotinic acid and pyridoxine on the stability of the color of the cranberry solution were investigated. The pH had marked influences on the color of the cranberry solution: i.e., the color of the cranberry solution was more intense at low pH. It showed characteristic bathochromic shift as the pH of the solution increased. The half-lives of olor value were 34 days at 37$^{\circ}C$, 91 mins at 9$0^{\circ}C$ and 29 mins at 12$0^{\circ}C$. Light gave an adverse effect to the stability of the color. The color degradation can be minimized by shielding the light from the cranberry solution. Among the sugars tested, fructose was the most deleterious followed by sucrose, galactose, maltose and glucose. Fumaric acid was found to be the most effective in stabilizing the color followed by citric acid, malic acid, acetic acid, while tartaric acid was found to be deleterious. Among the metal ions tested N $a^{+}$ and $Mg^{2+}$ were found to be effective in stabilizing the color, while M $n^{2+}$ was found to be the most deleterious followed by F $e^{2+}$, $K^{+}$ and $Ca^{2+}$. Ascorbic acid was found to be deleterious considerably followed by thiamine, while nicotinic acid and pyridoxine were found to be effective in stabilizing the color feebly.or feebly.
