Effect of Hominis Placenta on cutaneous wound healing in normal and diabetic mice

  • BACKGROUND/OBJECTIVES: The number of diabetic patients has recently shown a rapid increase, and delayed wound healing is a major clinical complication in diabetes. In this study, the wound healing effect of Hominis placenta (HP) treatment was investigated in normal and streptozotocin-induced diabetic mice. MATERIALS/METHODS: Four full thickness wounds were created using a 4 mm biopsy punch on the dorsum. HP was injected subcutaneously at the middle region of the upper and lower wounds. Wounds were digitally photographed and wound size was measured every other day until the 14th day. Wound closure rate was analyzed using CANVAS 7SE software. Wound tissues were collected on days 2, 6, and 14 after wounding for H/E, immunohistochemistry for FGF2, and Masson's trichrome staining for collagen study. RESULTS: Significantly faster wound closure rates were observed in the HP treated group than in normal and diabetes control mice on days 6 and 8. Treatment with HP resulted in reduced localization of inflammatory cells in wounded skin at day 6 in normal mice and at day 14 in diabetic mice (P < 0.01). Expression of fibroblast growth factor (FGF) 2 showed a significant increase in the HP treated group on day 14 in both normal (P < 0.01) and diabetic mice (P < 0.05). In addition, HP treated groups showed a thicker collagen layer than no treatment groups, which was remarkable on the last day, day 14, in both normal and diabetic mice. CONCLUSIONS: Taken together, HP treatment has a beneficial effect on acceleration of cutaneous wound healing via regulation of the entire wound healing process, including inflammation, proliferation, and remodeling.

  • Recently, ITU-T/VCEG and ISO/IEC MPEG have started a new joint standardization activity on video coding, called High Efficiency Video Coding (HEVC). This new standard gives significant improvement in terms of picture quality for high resolution video. The main challenge in this upcoming standard is the time complexity. In this paper we have focused on CU splitting algorithm. We have proposed a novel algorithm which can terminate the CU splitting process early based on the RD cost of the parent and current level and the motion vector value of the current CU. Experimental result shows that our proposed algorithm gives on average more than about 10% decrement in time over ECU [8] with on average 1.78% of BD loss on the original.

Characterization of a Chitinase Gene and Screening of Cold Active Chitinase from Polar Microorganisms (극지유래 저온활성 Chitinase 생산균주의 스크리닝과 Chitinase 유전자 클로닝)

  • Of the 169 strains of microorganisms stored in Polar and Alpine Microbial Collection of Korea Polar Research Institute, 27 strains were selected for their chitinase activity on ZoBell plates supplemented with 0.4% colloidal chitin. Among them, PAMC 21693 strain have shown the highest chitinolytic enzyme activity toward pNP-$(GlcNAc)_1$ at low temperature and the highest growth rate at $4^{\circ}C$. We cloned a full-length chitinase gene of 2,857 bp which contains an open reading frame of 2,169 bp encoding 872-amino acid polypeptide. Recombinant chitinase protein was expressed in E. coli and its molecular weight was confirmed 96 kDa. In this paper, we suggest the potential use of cold-active chitinase from polar microorganisms in the field of biotechnology.

Development of Automatic Nutrient-Solution Mixing System Using a Low-Cost and Precise Liquid Metering Device (액제 정밀계량 장치를 이용한 양액 자동조제 시스템 개발)

  • This study was conducted to develop an automatic nutrient-solution mixing system for small-scale sewers. The nutrient-solution mixing system consisted of a low-cost and precise metering device and data acquisition & control system with a personal computer. and, the metering device was composed of three parts those were supply pumps, metering cylinders and venturi tube. The system controlled electric conductivity(EC) and pH of nutrient-solution based on the time-based feedback control method with the information about temperature, EC, and pH of the nutrient-solution. The performance of the nutrient-solution mixing system was evaluated through the control of EC and pH while compared with those of commercial system. Also an experimental cultivation of tomato was conducted to verify and to improve the developed system. Results of this study were as follows. 1. The correlation coefficient of meteing device between the flow rate and operating time was 0.9999, and the linear reuession equation computed was y=21.759x, where y is the discharge($g$) and x is the operating time(s). 2. Calculated errors for the developed metering device and two commercial pump were $\pm$0.3% $\pm$2.45% and $\pm$1.38 % FS error respectively. 3. An automatic nutrient-solution mixing system based on a low-cost and precise metering device was developed. 4. The full scale errors of the developed system in controlling EC and pH at 23$\pm$1$^{\circ}C$ were $\pm$0.05mS/cm and $\pm$0.2, respectively 5. When using the commercial system, the controlled values of EC and pH of the 500 $\ell$ of water were 1.29 mS/cm and 6.1 pH for the setting points of 1.4 mS/cm and 6.0 pH respectively at 23$pm1^{\circ}C$. 6. The developed nutrient-solution control system showed $\pm$0.05 ms/cm of deviation from the setting EC value over the experimental cultivation period. 7. The deviation from the average values of Ca and Mg mass content in the several nutrient-solution were 0.5% and 1.8% respectively.

Effects of Dietary Nutrient Composition on Compensatory Growth of Grower Olive Flounder Paralichthys olivaceus under Different Feeding Regimes at Suboptimal Temperature

  • The effects of dietary nutrient composition on compensatory growth of grower olive flounder (Paralichthys olivaceus) under different feeding regimes at suboptimal temperature were determined. Four hundred five fish weighing 271.2 g were distributed into 27 300 L flow-through tanks (15 fish per tank). Nine treatments were prepared in triplicate: fish were hand-fed with control (C) diet for 12 weeks (12WF-C); four groups of fish were starved for 1 week and then fed C, high-protein (HP), high-lipid (HL), or combined high-protein and high-lipid (HPL) diets for 11 weeks; these groups are referred to as 11WF-C, 11WF-HP, 11WF-HL, and 11WF-HPL, respectively. Four other groups of fish were starved for 2 weeks and then fed C, HP, HL, and HPL diets for 10 weeks; these groups are referred to as 10WF-C, 10WF-HP, 10WF-HL, and 10WF-HPL, respectively. Weight gain and specific growth rate of fish from 12WF-C group were greater than those of fish from 11WF-C, 11WF-HP, 11WF-HL, 10WF-C, 10WFHP, and 10WF-HL groups, but not different from those of fish from 11WF-HPL and 10WF-HPL groups. Feed-efficiency ratio of fish from 12WF-C, 11WF-HP, 11WF-HPL, 10WF-HL, and 10WF-HPL groups was higher than that of fish in 11WF-C, 11WF-HL, 10WF-C, and 10WF-HP groups. The results of this study demonstrated that grower olive flounder subjected to 1- or 2-week feed deprivation were able to achieve full compensatory growth at suboptimal temperature only when fed HPL diet.

Screening for Resistance to Downy Mildew among Major Commercial Cucumber Varieties (주요 오이 품종의 노균병에 대한 저항성 검정)

  • This study was carried out for the downy mildew resistant test between 2010 and 2012. A set of 22 accessions belonging to 2 wild species and 20 varieties of the genus Cucumis, originating mainly from the Asian Vegetable Research and Development Center (AVRDC) Gene Centre, was evaluated for resistance to Pseudoperonospora cubensis, causal agent of cucumber downy mildew. The youngest fully expanded true leaves were found suitable for in vitro screening. Both leaf discs and full leaves could be kept fresh longer when applying 0.2 ${\mu}g/ml$ of gibberellin acid (GA). The incubation temperature of $20^{\circ}C$ was found to be the most suitable temperature for symptom development comparing with 15 and $25^{\circ}C$. Symptom development was faster when contact diseased leaf discs (2 weeks after inoculation) on to fresh leaf samples comparing with using conidia suspension ($10^5$ spores/ml). The numbers of spots in 'C-19' were lower than other varieties. 'C-19' variety was also showed the highest level of downy mildew resistant at $20^{\circ}C$ chamber in 6 days after inoculating with pathogen and displayed 0.90 (under 10%) of the infected rate. However, other varieties displayed susceptible in the pathogen sprayed plots. 'C-19' was the most resistant variety and no lesion was observed. Based on all data, 'C-19' can be a useful variety for the prevention of downy mildew.

Investigation of Dental Plaque, Calculus and Gingival Inflammation in Beagle Dogs (비글견에서 치태와 치석, 치은 염증의 조사)

  • This study was performed to investigate of dental plaque, calculus and gingival inflammation in Beagle dogs. Forty adults Beagle dogs (28 male and 12 female) were used in this study. The dogs weighed 9.5 kg and were in good oral and systemic health as determined by physical examination, and all dogs had full and normal dentition. The dogs were given a commercial pellet feed during 2 years period. For all examination procedures, the dogs were premedicated with a subcutaneous injection of atropine sulfate (0.04 mg/kg). Anesthesia was induced and maintained by intravenous administration of ketamine (8 mg/kg) and xylazine (2 mg/kg). Dental plaque, calculus and gingival inflammation were assessed by Logan and Boyce clinical plaque index. Calculi covering the maxillary carnassial and first molar teeth were extensive and were accompanied by severe gingival inflammation and pocket formation. Calculi, accompanied by gingival inflammation, were clearly evident on buccal surfaces of other teeth. Calculi didn't showed on the lingual surfaces, but linguogingival inflammation formed in premolar teeth. Although the general pattern was clear, there was considerable variation among dogs in the rate of deposition of calculus and extend of gingival inflammation. This investigation suggest that feeding of the commercial dry food without dental hygiene increase plaque accumulation and may be a contributing factor in calculi formation and periodontal disease.

Effect of Amniotic Fluid Stem Cells and Amniotic Fluid Cells on the Wound Healing Process in a White Rat Model

  • Background Amniotic-fluid-derived stem cells and amniocytes have recently been determined to have wound healing effects, but their mechanism is not yet clearly understood. In this study, the effects of amniotic fluid stem cells and amniocytes on wound healing were investigated through animal experiments. Methods On the back of Sprague-Dawley rats, four circular full-thickness skin wounds 2 cm in diameter were created. The wounds were classified into the following four types: a control group using Tegaderm disc wound dressings and experimental groups using collagen discs, amniotic fluid stem cell discs, and amniocyte discs. The wounds were assessed through macroscopic histological examination and immunohistochemistry over a period of time. Results The amniotic fluid stem cell and amniocyte groups showed higher wound healing rates compared with the control group; histologically, the inflammatory cell invasion disappeared more quickly in these groups, and there was more significant angiogenesis. In particular, these groups had significant promotion of epithelial cell reproduction, collagen fiber formation, and angiogenesis during the initial 10 days of the wound healing process. The potency of transforming growth factor-${\beta}$ and fibronectin in the experimental group was much greater than that in the control group in the early stage of the wound healing process. In later stages, however, no significant difference was observed. Conclusions The amniotic fluid stem cells and amniocytes were confirmed to have accelerated the inflammatory stage to contribute to an enhanced cure rate and shortened wound healing period. Therefore, they hold promise as wound treatment agents.

A fundamental study on the installation methods of automatic identification buoy on coastal gill net (연안자망 부이에 어구자동식별 장치 설치방안에 관한 기초적 연구)

  • As a series of fundamental researches on the development of an automatic identification monitoring system for fishing gear. Firstly, the study on the installation method of automated identification buoy for the coastal improvement net fishing net with many loss problems on the west coast was carried out. Secondly, the study was conducted find out how to install an automatic identification buoy for coastal gill net which has the highest loss rate among the fisheries. GPS for fishing was used six times in the coastal waters around Seogwipo city in Jeju Island to determine the developmental status and underwater behavior to conduct a field survey. Next, a questionnaire was administered in parallel on the type of loss and the quantity and location of fishing gear to be developed and the water transmitter. In the field experiment, the data collection was possible from a minimum of 13 hours, ten minutes to a maximum of 20 hours and ten minutes using GPS, identifying the development status and underwater behavior of the coastal gillnet fishing gear. The result of the survey showed that the loss of coastal net fishing gear was in the following order: net (27.3%), full fishing gear (24.2%), buoys, and anchors (18.2%). The causes were active algae (50.0%), fish catches (33.3%) and natural disasters (12.5%). To solve this problem, the installation method is to attach one and two electronic buoys to top of each end of the fishing gear, and one underwater transmitter at both ends of the float line connected to the anchor. By identifying and managing abnormal conditions such as damage or loss of fishing gear due to external factors such as potent algae and cutting of fishing gear, loss of fishing gear can be reduced. If the lost fishing gear is found, it will be efficiently collected.

Recent Development in Rice Seedling Raising in Japan, with Special Reference to the "Nursling Seedlings"

  • Recently, a nursery mat made from rock wool has realized transplanting of the younger seedlings with the ordinary transplanting machines for Chibyo and Chubyo(3 and 4~5 leaved seedling, respectively). The seedlings defined as the 'Nyubyo' or 'Nursling seedlings' became possible to achieve economic profits from the reduction in both working time and costs. It being widely noticed as a strategy to solve the difficulties in current rice cultivation. The nursling seedlings are 1.4 to 2.5 leaves and height at 4.5 to 7cm, grown 4 to 7 days after seeding. They maintain still up to 50 to 80% of their nutrients in the endosperm, and can grow by using only their own nutrients for a certain period of time after transplanting. Nursling seedlings take 2 days in the nursery chamber at 32$^{\circ}C$ after seeding, and 2 days in the greening house at $25^{\circ}C$. This is only 4 days, all together, to make the nursling seedlings of 1.5 leaves which are ready for transplanting. Watering is only needed once at the sowing time. It only takes 1 or 2 waterings even to raise a seedlings for a period of 7 days. The number of nursery boxes can be reduced because it is possible to sow more densely(220 to 240g per box), thus it only needs seedlings of 15 to 16 boxes per 10 a which leads to a reduction in facilities and space needed. Temperature during the nursery period can be artificially adjusted more precisely which may lead to the prevention of temperature stress. The nursling seedlings can root rapid by because the crown roots from the coleoptile node begin to emerge immediately after transplanting. They show strong resistance to low temperature (12$^{\circ}C$) and deep-planting. There is no danger in the rooting of the seedlings even if half of their height is buried into the soil. Moreover, it can root at a rate of up to 65 to 80% even if the full height of the seedlings is buried. They show also strong resistance to submergence (10~15cm). The nursling seedlings tend to grow by producing tillers from lower nodes. It is therefore, necessary to control to keep the proper numbers of tillers per unit area. They have no fear in the delay of heading and their yield components can be so well balanced that the same level of yield was achieved with the nursling seedlings compared to that with Chibyo. It was further suggested that if the surplus tillers can be avoided by such cultivation practices, the number of grain per panicle can be kept greater and higher yield can be realized. Practical experiments with the nursling seedlings conducted in 1989 and 1990 by farmers in various areas showed exciting results. The nursling seedlings will become widely spread, or at least occupy an important position in Japanese and also in Korean rice cultivation techniques.tivation techniques.
