Genetic Diversity of Ascaris in China Assessed Using Simple Sequence Repeat Markers

  • The giant roundworm Ascaris infects pigs and people worldwide and causes serious diseases. The taxonomic relationship between Ascaris suum and Ascaris lumbricoides is still unclear. The purpose of the present study was to investigate the genetic diversity and population genetic structure of 258 Ascaris specimens from humans and pigs from 6 sympatric regions in Ascaris-endemic regions of China using existing simple sequence repeat data. The microsatellite markers showed a high level of allelic richness and genetic diversity in the samples. Each of the populations demonstrated excess homozygosity (Ho0). According to a genetic differentiation index (Fst=0.0593), there was a high-level of gene flow in the Ascaris populations. A hierarchical analysis on molecular variance revealed remarkably high levels of variation within the populations. Moreover, a population structure analysis indicated that Ascaris populations fell into 3 main genetic clusters, interpreted as A. suum, A. lumbricoides, and a hybrid of the species. We speculated that humans can be infected with A. lumbricoides, A. suum, and the hybrid, but pigs were mainly infected with A. suum. This study provided new information on the genetic diversity and population structure of Ascaris from human and pigs in China, which can be used for designing Ascaris control strategies. It can also be beneficial to understand the introgression of host affiliation.

Oxygen Toxicity: Behavioral Assessment by Swim Test in Mouse Exposed to Hypoxia and Hyperoxia (수영 시험 모델에서 산소 농도에 따른 운동성 변화와 독성 평가)

  • This study was aimed to evaluate the effect of oxygen on motor activity and toxicity in male mice. The modified Porsolt forced swim test (FST) was used and the distance and time of movement by mice were analyzed in 15。C water bath for 20 minutes using the automatic Ethovision videotracking system. Analyses were carried out before and after 20 minutes of exposure to 10%-70% concentration of normobaric oxygen. The effects of inspired oxygen tension on the distance and time of movement showed the similar trends, but changes in distance were more prominent. Both the distance and time of movement increased after exposure to 30% and 40% oxygen concentration. The distance and time of movement also increased upon exposure to 50% and 60% oxygen. In contrast, increases En movement and time under exposure to 21% oxygen concentration were suppressed when exposed to over 50% oxygen concentration. With exposure to 10% oxygen, there was a significant decrease in the distance of movement and a slight suppression of movement time. During the swim test, 12.5%, 37.5%, and 87.5% of the mice drowned after exposure to 10%, 60%, and 70% oxygen concentration, respectively. These results suggest that motor activity can be enhanced by inspired oxygen up to 40% concentration. When hypoxic and hyperoxic oxygen exposure over 50%, motor activity is reduced and toxicity may be induced.

Anti-Depressant Like Effect of Methyl Gallate Isolated from Acer barbinerve in Mice

  • In the present study, the anti-depressant like effect of methyl gallate (MG) isolated from the stem bark of Acer barbinerve was examined in ICR mice. Body weight (BDW) and blood glucose (BDG) levels significantly decreased in the repeated restraint stress (RRS) group (2 h/day for 14 days) compared to the no stress (NS) group. To examine the effect of MG on RS-induced BDW loss and hypoglycemia, MG (10 mg/kg) and the anti-depressant fluoxetine (10 mg/kg) were administered daily for 14 days. Orally administered MG and fluoxetine significantly attenuated the RS-induced BDW loss and hypoglycemia. Interestingly, MG administered mice showed increased BDG levels in the normal and glucose feeding condition. Chronic RS-subjected mice showed immobilized and depressed behaviors. The effect of MG on the depressed behaviors was evaluated using the tail-suspension test (TST) and the forced swimming test (FST). In both tests, RS-induced immobilized behaviors were significantly reversed in MG and fluoxetine administered groups. Taken together, MG significantly attenuated the RS-induced BDW loss, hypoglycemia, and depressed behaviors. Considering that decreased BDG levels (hypoglycemia) can cause depression, MG may exert its anti-depressant like effect by preventing hypoglycemia. Our results suggest that MG isolated from A. barbinerve can exert anti-depressant like effect, and could be used as a new and natural anti-depressant therapy.

Shallow Population Genetic Structures of Thread-sail Filefish (Stephanolepis cirrhifer) Populations from Korean Coastal Waters

  • Genetic diversities, population genetic structures and demographic histories of the thread-sail filefish Stephanolepis cirrhifer were investigated by nucleotide sequencing of 336 base pairs of the mitochondrial DNA (mtDNA) control region in 111 individuals collected from six populations in Korean coastal waters. A total of 70 haplotypes were defined by 58 variable nucleotide sites. The neighbor-joining tree of the 70 haplotypes was shallow and did not provide evidence of geographical associations. Expansion of S. cirrhifer populations began approximate 51,000 to 102,000 years before present, correlating with the period of sea level rise since the late Pleistocene glacial maximum. High levels of haplotype diversities ($0.974{\pm}0.029$ to $1.000{\pm}0.076$) and nucleotide diversities (0.014 to 0.019), and low levels of genetic differentiation among populations inferred from pairwise population FST values (-0.007 to 0.107), support an expansion of the S. cirrhifer population. Hierarchical analysis of molecular variance (AMOVA) revealed weak but significant genetic structures among three groups ($F_{CT}$ = 0.028, p<0.05), and no genetic variation within groups (0.53%; $F_{SC}$ = 0.005, p = 0.23). These results may help establish appropriate fishery management strategies for stocks of S. cirrhifer and related species.

Genetic Variability and Population Structure of Pacific Abalone Haliotis discus hannai Sampled from Stocked Areas Using Microsatellite DNA Markers (종묘방류 해역에서 채집 된 참전복의 microsatellte marker에 의한 유전 다양성 및 집단 구조)

  • Microsatellite DNA markers were used to investigate the genetic diversity and population structure of Pacific abalone Haliotis discus hannai collected from six locations (Uljin, Ulsan, Daechon, Taean, Wando, and Yosu) where hatchery-produced abalone have been released intensively. There was no distinguishable difference in the observed and expected heterozygosities between the six populations and a cultured population. However, there was a difference in the number of alleles per locus: 12.8 for the cultured population and 13.8 to 15.8 for the six populations. The proportion of stocked abalone ranged from 41.1 to 92.7% for wild-caught populations with a decreasing tendency of alleles per locus for an increasing proportion of stocked abalone. A departure from Hardy-Weinberg equilibrium (HWE) assessed using the Markov chain procedure (P<0.05) was observed in the six populations and cultured population at loci Hdh145 and Hdh5l2. The pairwise Fst test (P<0.05) showed a significant difference between the Uljin and Ulsan populations and four remaining populations (Wando, Daechon, Yosu, and the cultured population), among which the Wando population differed less than the other three populations (Daechon, Yosu, and the cultured population).

  • 본 논문에서는 한국어 자연어 정보처리 기술 표준화를 위한 형태소 분석기 및 품사 태거 평가 대회(MATEC99)에 참여한 고려대학교의 형태소 분석기, 품사 태거, 그리고 명사 추출기를 설명하고 평가 결과를 기술한다. 형태소 분석기는 입력된 어절을 우에서 좌로 분석하며 각 상태에 대한 예측 정보를 활용하여 불필요한 분석 후보에 대한 탐색을 수행하지 않도록 한다. 품사 태거로는 띄어쓰기를 고려한 형태소 품사 2-그램 확률과 띄어쓰기를 고려한 형태소 어휘-품사 3-그램 어휘 확률을 이용하는 결합 독립 모형을 사용한다. 고속 명사 추출기는 고속의 FST 사전과 한국어 특성을 반영한 휴리스틱을 이용한다.

Effects of Moutan Cortex on Corticotropin-Releasing Factor, c-Fos and Tyrosine Hydroxylase in Forced Swimming Test (목단피(牧丹皮)가 강제수영부하시험에서 Corticotropin-Releasing Factor, c-Fos 와 Tyrosine Hydroxylase에 미치는 영향)

  • Objectives : The goal of this study was to investigate the effect of Moutan Cortex as antidepressant in forced swimming test(FST) model. Methods : The expressions of corticotropin-releasing Factor(CRF), c-Fos and tyrosine hydroxylase(TH) were measured with immunohistochemical method at paraventricular nucleus (PVN), locus coeruleus(LC) and ventral tegmental area(VTA). Results : The duration of immobility in the forced swimming test was significantly decreased in the Moutan Cortex 100 mg/kg treated group in comparison with the control group(p<0.01). CRF and c-Fos expressions at PVN were decreased in the Moutan Cortex 100mg/kg treated group in comparison with the control group. But only the expression of c-Fos was shown the significance(p<0.05). TH expressions at LC and VTA were significantly decreased in the Moutan Cortex 100mg/kg and 400mg/kg treated group in comparison with the control group(p<0.001). Conclusion : According to the results, Moutan Cortex has the antidepressant effect by showing the reduced immobility through the reduction of c-Fos expression at PVN and the reduction of TH expression at LC and VTA.

Genetic Diversity of Goats from Korea and China Using Microsatellite Analysis

  • Nine microsatellite loci were analyzed in 84 random individuals to characterize the genetic variability of three domestic goat breeds found in Korea and China: Korean goat, Chinese goat and Saanen. Allele diversity, heterozygosity, polymorphism information content, F-statistics, indirect estimates of gene flow (Nm) and Nei's standard distances were calculated. Based on the expected mean heterozygosity, the lowest genetic diversity was exhibited in Korean goat ($H_E$=0.381), and the highest in Chinese goat ($H_E$=0.669). After corrections for multiple significance tests, deviations from Hardy-Weinberg equilibrium were statistically significant over all populations and loci, reflecting the deficiencies of heterozygotes (global $F_{IS}$=0.053). Based on pairwise FST and Nm between different breeds, there was a great genetic differentiation between Korean goat and the other two breeds, indicating that these breeds have been genetically subdivided. Similarly, individual clustering based on the proportion of shared alleles showed that Korean goat individuals formed a single cluster separated from the other two goat breeds.

The Effects of Koamsimsinwhan(without Cinnabarite) on Depression in Chronic Mild Stress(CMS)-treated Rats (고마심신환거주사방(古魔心賢丸去朱砂方)이 우울증(憂鬱症) 모형(模型) 흰쥐의 우울(憂鬱) 성향 및 PVN 의 c-Fos 발현(發顯)에 미치는 효과(效果))

  • This study was performed to evaluate the effects of Koamsimsinwhan (without Cinnabarite) on depression in CMS-treated rats. Rats of trial groups were exposured to chronic mild unpredictable stress for 4 weeks. These CMS-treated rats were stratified into Koamsimsinwhan(without Cinnabarite) group and vehicle(non-drug) group. And there were control rats excluding both CMS and Koamsimsinwhan(without Cinnabarite). Forced swimming test, general activity test and immunohistochemical test(measuring the level of manifestation of c-Fos in PVN) were executed to evaluate the effects of Koamsimsinwhan(without Cinnabarite) on depression after 4 weeks of CMS treatment. The results were as follows 1. In FST, depression was induced from CMS treatment and was supressed with Koamsimsinwhan(without Cinnabarite). 2. The increased activity of nerve cell in PVN, which refers to stress, was induced from CMS treatment and normalized with Koamsimsinwhan (without Cinnabarite).

Genetic Diversity and Population Structure in Native Chicken Populations from Myanmar, Thailand and Laos by Using 102 Indels Markers

  • The genetic diversity of native chicken populations from Myanmar, Thailand, and Laos was examined by using 102 insertion and/or deletion (indels) markers. Most of the indels loci were polymorphic (71% to 96%), and the genetic variability was similar in all populations. The average observed heterozygosities ($H_O$) and expected heterozygosities ($H_E$) ranged from 0.205 to 0.263 and 0.239 to 0.381, respectively. The coefficients of genetic differentiation (Gst) for all cumulated populations was 0.125, and the Thai native chickens showed higher Gst (0.088) than Myanmar (0.041) and Laotian (0.024) populations. The pairwise Fst distances ranged from 0.144 to 0.308 among populations. A neighbor-joining (NJ) tree, using Nei's genetic distance, revealed that Thai and Laotian native chicken populations were genetically close, while Myanmar native chickens were distant from the others. The native chickens from these three countries were thought to be descended from three different origins (K = 3) from STRUCTURE analysis. Genetic admixture was observed in Thai and Laotian native chickens, while admixture was absent in Myanmar native chickens.
